The trans $\rightarrow$ cis Photoisomerization and Fluorescence of trans 1,2-Bispyrazylethylene: pH, Salt and Heavy Atom Effects

  • The $trans{\rightarrow}cis$ photoisomerization and fluorescence of trans-1,2-bispyrazylethylene were investigated in various conditions. The quantum yields of the $trans{\rightarrow}cis$ photoisomerization and the fluorescence intensity of trans-1,2-bispyrazylethylene decrease on going from neutral to acidic or basic solutions. The quantum yields of photoisomerization, however, are little affected by changing the concentration of salt while the fluorescence intensity increases as the concentration of salt increases. pH and salt effects on the energy levels of $^1(n,\;{\pi}^*)\;and\;^1({\pi},\; {\pi}^*)$ states lead to opposing changes in photoisomerization and fluorescence quantum yields. The heavy atom effect on the fluorescence of 1,2-bispyrazylethylene was also investigated.

Maillard Reaction of Pidan White as Inhibited by Chinese Black Tea Extract (Camellia sinensis) in the Pickling Solution

  • Changes in Maillard reaction of pidan white were monitored with $A_{294}$, fluorescence intensity, and browning intensity during pickling in the absence and presence of Chinese black tea extract (Camellia sinensis) at levels of 2% and 5% together with 0.2% $ZnCl_2$ or 0.2% $CaCl_2$ up to 3 wk, followed by ageing for another 3 wk. Browning intensity and $A_{294}$ of pidan white increased with increasing pickling/ageing, while fluorescence intensity decreased during ageing (p<0.05), irrespective of treatments. At wk 6, pidan white treated with 0.2% $ZnCl_2$ and 0.2% $CaCl_2$ showed slightly higher browning intensity, fluorescence intensity and $A_{294}$ than those treated with divalents together with Chinese black tea (p<0.05). Free amino group and sugar contents showed continuous decrease during pickling and ageing irrespective of tea and cations used. However, pidan treated without Chinese black tea extract showed significantly lower free amino group and sugar during the ageing of 6 wk (p<0.05). Thus, Chinese black tea extract had an inhibitory effect on the Maillard reaction during ageing of pidan white.

Proposal and design of reflecting optical system to improve detection intensity in fluorescence confocal scanning microscopy (형광 공초점 주사 현미경의 측정 강도 향상을 위한 반사 광학계의 제안 및 설계)

  • Confocal microscopy is very popular technology in bio-medical inspection due to its ability to reject background signals and to measure very thin slide of thick specimens, which is called optical sectioning. But intensity of detected signal in fluorescence type confocal microscopy is so small that only 0.2% of emitted fluorescence light can be detected in the best case. In this paper, we proposed the reflecting optical system to improve the detection intensity and designed the optical system by optimal design method. At the end of the paper, we analyzed the characteristics of the proposed reflecting optical system.

PROTEIN CONFORMATIONS OF OCTOPUS RHODOPSIN AND ITS DEPROTONATED PHOTOCYCLE INTERMEDIATE MONITORED BY ABSORPTION AND PROTEIN FLUORESCENCE

  • Picosecond time-resolved and static protein fluorescence spectra and absorption spectra of octopus rhodopsin, a photorecepting protein, are measured and compared with those of bacteriorhodopsin, a photon-induced proton pumping protein, to understand the protein conformations and functions of octopus rhodopsin and its deprotonated photocycle intermediate. The bluer and weaker absorption of retinal indicates that octopus rhodopsin is better in thermal noise suppression but less efficient in light harvesting than bacteriorhodopsin. The protein fluorescence of octopus rhodopsin shows the characteristic of Trp only and the uantum efficiency and lifetime variations may result primarily from variations in the coupling strength with the retinal. The stronger intensity by four times and larger red shift by 12 nm of fluorescence suggest that octopus rhodopsin has more open and looser structure compared with bacteriorhodopsin. Fluorescence decay profiles reveal two decay components of 300 ps (60%) and 2 ns (40%). The deprotonation of protonated Schiff's base increases the shorter decay time to 500 ps and enhances the fluorescence intensity by 20%. The fluorescence and its decay time from Trp residues near retinal are influenced more by the deprotonation. The increase of fluorescence intimates that protein structure becomes loosened and relaxed further by the deprotonation of protonated Schiff's base. The driving force of sequential changes initiated by absorption of a photon is too exhausted after the deprotonation to return the intermediate to the ground state of the begun rhodopsin form.

  • In present study, a temperature field of specimens which was coated with fluorescence dye such as Rhodamine-B(Rh-B) has been measured, based on the fluorescence intensity. Silica(SiO2) nano porous structure with 1um thickness was constructed on a cover glass, and fluorescence dye was digested into these porous thin films. To optimize manufacturing coating process, various solvents, Rh-B concentration, and other chemical materials were applied to fabricate the specimen and all specimens were measured on the various temperature conditions. For the measurement, a 14 bit cooled CCD camera with 1600 by 1200 spatial resolution is equipped with epifluorescence microscope to obtain only fluorescence intensity from 1.2 mm by 0.9 mm field of view of the illuminated coated specimen.

An Apparatus for Monitoring Real-time Uranium Concentration Using Fluorescence Intensity at Time Zero

  • An apparatus for detecting remote real-time uranium concentration using an optrode was developed. An optrode to detect uranium fluorescence as remote real-time control was designed. Fluorescence intensity at time 2ero was derived by the fluorescence signal processing and the algorithm to exclude the quenching effect of various quenchers and temperature fluctuations. This apparatus employing the above deriving method and the optrode has an error range within 6% in spite of serious fluorescence lifetime changes due to the quenching effect and temperature fluctuations. The detection limit is 0.06 ppm and the linearity is excellent between 0.06 ppm and 2 ppm on the aqueous uranium solution.

Effect of membrane lipid peroxidation on rat liver microsomal enzyme activity (막지질 과산화와 간세포내 마이크로솜 및 리덕타제 기능과의 상관성에 관한 연구)

  • The effects of membrane lipid peroxidation and retinyl palmitate on rat liver microsomal functions were investigated in vitro. Rat liver homogenates exposed to oxygen tension for 0, 3, 6, 9 or12 hours and lipid peroxidation levels were evaluated by the measurements of fluorescence intensity, malondialdehyde (MDA) and retinyl palmitate. The fluorescence intensity of homogenates and microsomes were elevated and retinyl palmitate concentrations were decreased. But the concentration of MDA was not affected to exposure time. Therefore, fluorescence intensity and retinyl palmitate concentration were used to analyze the correlation between lipid peroxidation and microsomal functions. To investigate the liver microsomal functions, the microsome was isolated from rat liver homogenates exposed to oxygen. The concentration of cytochrome P450 and the activity of NADPH-cytochrome P450 reductase in liver microsomes were gradually decreased with increasing the exposure time. The correlation between fluorescence intensity of microsomes showed a very high inverse correlation of -0.97 and -0.93, respectively. The decrease of cytochrome P450 concentration was due to the regeneration of cytochrome P450 to cytochrome P420. Also, the activities of cytochrome P450-dependent aminopyrine demethylase and benzpyrene hydroxylase of liver microsomes were gradually decreased with increasing the exposure time. The correlation with fluorescence intensity of microsome showed a high inverse correlation of -0.97 and -0.91, respectively. The retinyl palmitate concentrations of rat liver homogenates were decreased with increasing the exposure time. The decrease of retinyl palmitate concentration was followed by a low concentration of cytochrome P450 and activity of NADPH-cytochrome P450 reductase. The correlation indicated high direct correlation of 0.92 and 0.93, respectively. The decrease of retinyl palmitate concentration was also accompanied by the reduction of aminopyrine demethylase and benzpyrene hydroxylase activities. The correlation was analyzed a high direct correlation of 0.90 and 0.85, respectively. In conclusion, these studies have shown that the membrane lipid peroxidation of rat liver microsome proportionally decreased microsomal enzyme activities in vitro experiments.

Differences in in vivo Fluorescence Yield for Netplankton and Nanoplankton Size Classes (Netplankton과 Nanoplankton 크기별 in vivo Fluorescence의 차이)

  • In the South Sea of Korea, in vivo fluorescence intensity (IVF) and extractable chlorophyll a concentration were measured to determine whether there was significant defference in in vivo fluorescence per unit chlorophyll a (R) between netplankton and nanoplankton size classes (less than $22{\mu}m$). IVF and chlorophyll a were linearly related for both size classes, but R's were significantly different between two size classes. The R of nanoplankton was about 7 times higher than that of netplankton. Therefore, the size dependency of R must be taken into consideration when size fraction of phytoplankton biomass is determined from the measurements of in vivo fluorescence intensity.

Change in Fluorescence Characteristics of Dissolved Organic Matter at Inflow Stream per Catchment of Different Land Use (토지이용도가 다른 소유역별 유입하천에서 용존유기물 형광특성 변화)

  • The Lake Sihwa watershed includes industrial, urban, and rural areas simultaneously. In this study, we analyzed the characteristics of dissolved organic matter (DOM) in spatial-temporal runoff patterns at representative sites having different land use in the watershed of Lake Sihwa. The result of synchronous and 3D-EEMs(3-Dimensional Excitation Emission Matrix Spectroscopy) analysis in 4TG (industrial area), fluorescence distribution and variation clearly appeared in the Fulvic-like fluorescence (FLF) and Humic-like fluorescence (HLF) regions along with the Protein-like fluorescence (PLF) region. A characteristic that Peak A (HLF) region fluorescence intensity did not decrease and the HLF region of fluorescence intensity and spatial-temporal changes clearly appeared during rainfall in AS (urban area). The results of fluorescence analysis in MS did not show great changes in PLF and FLF while showing that fluorescence intensity changes over time in the Terrestrial-like fluorescence (THLF) region increased greatly. In conclusion, our results showed significant differences in the runoff characteristics of DOM particularly in industrial, urban and rural area, and these differences should be considered for the efficient controlling of DOM in the watershed.

  • Effects of light intensity on the steady-state fluorescence quenching kinetics are examined for general cases where the bimolecular quenching can occur via long-range energy transfer processes and the potential of mean force between the energy donor and acceptor molecules is not negligible. Approximate analytic expressions are derived for the steady-state quenching rate constant and for the ratio of the steady-state intensity of unquenched to quenched fluorescence. The analytic results are compared with the exact results obtained from numerical analysis and the results of conventional theories.
