Changes in Differentially Expressed Genes in the Liver of Oryzias latipes by Binary Exposure to Carcinogenic Polycyclic Aromatic Hydrocarbons

  • The biological effects of carcinogenic polycyclic aromatic hydrocarbons (cPAHs) including benzo[a]pyrene (BaP), dibenzo[a,h]anthracene (DBA), benzo[a]anthracene (BaA), benzo[b] fluoranthene (BbF), benzo[k]fluoranthene (BkF), and indeno[1,2,3-c, d]pyrene (InP) on transcriptomic changes were determined in the liver of Oryzias latipes. Differentially expressed genes (DEGs) by binary exposure to cPAHs (BaP+BaA, BaP+BbF, BaP+BkF, BaP+DbA, BaP+InP) were screened by annealing control primers-based polymerase chain reaction followed by sequence analysis and BLAST searching. The results showed that four DEGs were commonly expressed by cPAHs and they were identified as ribosomal protein S4, coagulation factor II, elongation factor 1 beta, and a predicted protein similar to human immunodeficiency virus type I enhancer binding protein 3. This finding suggests that binary exposure to cPAHs interferes protein synthesis required for fundamental liver functions in fish.

원유와 석유제품 중의 다환방향족탄화수소류[PAHs] 분포특성과 발생원 (Compositional Characteristics and Origin of Polycyclic Aromatic Hydrocarbons[PAHs] of Crude Oils and Petroleum Products)

  • Compositional characteristics and origin of polycyclic aromatic hydrocarbons(PAHs), which should be strongly regulated for environmental protection in the crude oils and petroleum products, have been investigated by gas chromatography-mass spectrometry(GC/MS). In the crude oils analyzed, two-rings compound(naphthalene) of PAHs was detected around $72.3\sim93.5%$, but five- or six-rings compound of PAHs was not detected. In the crude oils analyzed, the molecular ratio indices of Phe/Ant(phenanthrene/anthracene)>15, Fla/Pyr(fluoranthene/pyrene)<1, BaA/Chr(benzo (a)anthracene/chrysene)$\leq0.4$ could be effectively applied, and we found that the origin of PAHs was petrogenic sources. Total PAHs concentrations in the crude oils were increased with increasing API gravity and with decreasing sulfur contents of the analyzed crude oils. Five- or six-rings compound of PAHs were not included in ail petroleum products except bunker-C. Furthermore, the molecular ratio indices of Phe/Ant(phenanthrene/anthracene) vs. Fla/Pyr(fluoranthene/pyrene) could be effectively applied as the standard for the PAHs contamination criterion at the other Korean areas.

Cytotoxicity of Dichloromethane Extracts of Asian Dust

  • The appearance of Asian Dust (AD) originating from China and Mongolia during spring each year is a meteorological phenomenon periodically observed in extensive regions of East Asia. According to a previous epidemiological study, AD has adverse effects on both human beings and ecosystems. In this study, we collected total suspension particles (TSP) in the AD period and Non-AD (NAD) period. We extracted organic components from TSP using dichloromethane (DCM), and the polyaromatic hydrocarbons (PAHs) were analyzed. The DCM extracts contained PAHs such as benzo(b)fluoranthene, benzo[g,h,i]perylene, benzo(k)fluoranthene, benzo(a)pyrene, and pyrene. No significant difference was observed in cytotoxicity of the DCM extracts from AD versus NAD when tested on the human bronchial epithelial cells, BEAS-2B. e also examined the toxic mechanisms of AD extracts in cultured BEAS-2B cells and RAW264.7 cells, and in BEAS-2B cells observed increased levels of reactive oxygen species (ROS), decreased glutathione (GSH), and induced caspase-3 activity. Increased expression of oxidative stress-related and inflammation- related genes were also observed in BEAS-2B cells, while nitric oxide (NO) levels were increased in RAW264.7 cells. Taken together, the results suggest that in these cultured cells, AD may induce cytotoxicity through oxidative stress and pro-inflammatory signals.

서울지역 대기 중의 PAHs 분포 특성 및 발생원 (Distribution and emission source of PAHs in ambient air of Seoul)

  • 본 연구는 화석 연료를 소비하는 산업 등과 같이 인위적인 활동 과정에서 불완전 연소로 인하여 주로 발생하는 PAHs류의 지역적 분포 및 발생원 규명을 위하여 수행하였다. 서울지역 대기시료를 채취하여 분석한 결과, 서울 도심과 외곽 지역의 대기 중 입자상과 가스상을 합한 총 PAHs 평균농도는 관악지역이 $16.52\;ng/m^3$, 서대문 지역은 $59.10\;ng/m^3$로 조사되었으며, 주로 phenanthrene, fluoranthene, pyrene, fluorene이 전체 PAH의 55.6% (관악)와 60.8%(서대문)를 차지하고 있는 것으로 나타났다. 또한, 두 지역의 PAHs의 발생원을 파악한 결과 관악과 서대문 지역 모두 대기 중 PAHs의 발생원은 동일하고, 특히 서대문 지점에서는 자동차에 의한 영향이 큰 것으로 나타났다.

Acute Toxicity of Heavy Metals, Tributyltin, Ammonia and Polycyclic Aromatic Hydrocarbons to Benthic Amphipod Grandidierella japonica

  • Benthic amphipod, Grandidierella japonica widely inhabits the Korean coastal waters and is developed as a standard test species for sediment toxicity tests. We exposed G. japonica to various pollutants including 4 kinds of inorganic metals (Ag, Cd, Cu and Hg), tributyltin [TBT], ammonia and 7 polycyclic aromatic hydrocarbon (PAH) compounds (acenaphthene, chrysene, fluoranthene, fluorene, naphthalene, phenanthrene and pyrene) to estimate the no observed effect concentration (NOEC) and the median lethal concentration (LC50) of each pollutant during the 96-hour acute exposure. Among all tested pollutants, TBT was most toxic to G. japonica, and Rg was most toxic among inorganic metals. The toxicity of pyrene to G. japonica was greatest among PAH compounds, followed by fluoranthene, phenanathrene, acenaphthene, fluorene and naphthalene. The toxicity of PAH compounds was closely related to their physico-chemical characteristics such as $K_ow$ and water solubility. G. japonica responded adequately to pollutant concentrations and exposure durations, and the sensitivity of G. japonica to various inorganic and organic pollutants was generally comparable to other amphipods used as standard test species in ecotoxicological studies, indicating this species can be applied in the assessment of environments polluted by various harmful substances.

Kinetic Biodegradation of Polycyclic Aromatic Hydrocarbons for Five Different Soils under Aerobic Conditions in Soil Slurry Reactors

  • In this study, soil slurry bioreactors were used to treat soils containing 16 polycyclic aromatic hydrocarbons (PAHs) for 35 days. Five different soil samples were taken from manufactured gas plant (MGP) and coal tar disposal sites. Soil properties, such as carbon content and particle distribution, were measured. These properties were significantly correlated with percent biodegradation and degradation rate. The cumulative amount of PAH degraded (P), degradation rate (Km), and lag phase (𝜆) constants of PAHs in different MGP soils for 16 PAHs were successfully obtained from nonlinear regression analysis using the Gompertz equation, but only those of naphthalene, anthracene, acenaphthene, fluoranthene, chrysene, benzo[k]fluoranthene, benzo(a)pyrene, and benzo(g,h,i)perylene are presented in this study. A comparison between total non-carcinogenic and carcinogenic PAHs indicated higher maximum amounts of PAH degraded in the former than that in the latter owing to lower partition coefficients and higher water solubilities (S). The degradation rates of total non-carcinogenic compounds for all soils were more than four times higher than those of total carcinogenic compounds. Carcinogenic PAHs have the highest partitioning coefficients (Koc), resulting in lower bioavailability as the molecular weight (MW) increases. Good linear relationships of Km, 𝜆, and P with the octanol-water partitioning coefficient (Kow), MW, and S were used to estimate PAH remaining, lag time, and biodegradation rate for other PAHs.

Heavy metal profiles of agricultural soils in Sakarya, Turkey

  • Sakarya is famous for cucurbit productions in Turkey and cucurbits can grow as big as 560 kg of weight per fruit in its agricultural areas. There is no or limited information about contaminant levels and profiles of the agricultural fields in Sakarya. The purpose of this study is to investigate the levels of polycyclic aromatic hydrocarbons (PAHs) (naphthalene, phenanthrene, pyrene, and fluoranthene) and heavy metal (As, Cd, Cu, Cr, Ni, Pb, Zn) concentrations of the selected fields. Total 33 soil samples were collected from 12 counties of Sakarya where both cucurbits have been produced and organochlorine pesticides have been applied to the fields for more than 30 y during the historical plantation periods. Heavy metal and PAH contents in the soil samples were measured by an Inductively Coupled Plasma Emission Spectroscopy and a Gas Chromatography-Mass Spectrometry. The highest phenanthrene, pyrene, and fluoranthene concentrations were measured as 63.50 ng/g, 134.34 ng/g, 140.0 ng/g, respectively in the soil samples from Geyve County. Cu, Ni, and Cr concentrations were measured as 108.2 mg/kg, 219.9 mg/kg, and 173.1 mg/kg, respectively in Geyve's samples which were also the highest and 2-7 times more than the limit values given in the Turkish Soil Pollution Control Regulation. Precautions need to be taken for Sakarya's agricultural fields which are an important milestone of Turkey's cucurbit and fruit productions since the contaminants can be accumulated in the fruits and edible parts of the plants.

국내 PAHs 오염 우려지역의 환경 시료 중 PAHs 잔류량 모니터링 (Monitoring of Polycyclic Aromatic Hydrocarbon Residues in Environmental Samples in Korea)

  • 본 연구는 국내 지역별 농작물과 농업환경 중 16종의 PAHs 잔류 양상을 파악하고자 수행하였다. 국내 PAHs 오염 우려지역인 공업단지 및 화력발전소 주변에서 주요 농작물(26점), 농작물 재배 토양(46점) 및 농작물 재배 주변 물(15점) 시료를 채취하여 GC/MSD로 PAHs 잔류량을 분석하였다. 농작물의 경우 최대 5종의 PAHs가 $4.5{\sim}52.2{\mu}g\;kg^{-1}$ 수준으로 검출되었으며, 토양에서는 최대 13종의 PAHs가 $4.3{\sim}662.9{\mu}g\;kg^{-1}$ 범위로 검출되었다. 이 중 발암성이 강한 물질로 알려진 benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene, dibenzo(a,h)-anthracene 등 총 6가지 성분이 $14.2{\sim}167.8{\mu}g\;kg^{-1}$ 범위로 검출되었다. 특히 토양 중 PAHs 잔류함량과 검출빈도는 제철, 중공업단지가 밀집되어있고 교통량이 많은 지역이 다른 지역보다 최대 3배 이상 높게 나타났으며, 산불이 발생했던 지역에서는 최대 6종의 PAHs가 $73{\sim}36.0{\mu}g\;kg^{-1}$ 수준으로 검출되었다. 자동차 배기가스 및 주거난방 시설에서 주로 발생하는 naphthalene, phenanthrene, fluoranthene, pyrene 등의 검출빈도가 다른 성분에 비해 높게 나타났다. 채취한 모든 물 시료에서는 PAHs가 검출되지 않았다. 이는 PAHs의 수용해도가 비교적 낮고, 친유성의 특성을 가지고 있어 물보다는 유기화합물이 풍부한 저니토에 잔류할 가능성이 크기 때문인 것으로 판단되었다. 본 연구 결과 국내농작물과 농업환경 중 일부 PAHs가 분포되어 있는 것을 확인하였으며, 향후 환경 중 PAHs의 농작물 오염경로 및 잔류 특성 등을 규명하는 연구가 지속적으로 이루어져야 할 것으로 사료된다.

PAH가 CYP1B1 유전자 발현에 미치는 영향 (Effect of PAH on CYP1B1 Gene Expression)

  • Cytochrome P4501B1(CYP1B1) is known to be inducible by xenobiotic compounds such as policyclic aromatic hydrocarbon(PAH) and dioxins such as 2,3,7,8-tetrachloro-dibenzo-p-dioxin(TCDD). And these induction of CYP1B1 is also regulated by many categories of chemicals. In order to investigate the effects of several chemicals on CYP1B1 gene expression in Hepa-I and MCF-7 cells, 5' flanking DNA of human CYP1B1 was cloned into pGL3 basic vector containing luciferase gene, and then transfected into these cells. After treatment of chemicals, the luciferase activity was measured. CYP1B1 enzyme metabolize PAHs and estradiol. CYP1B1 metabolize estradiol to 4-hydrozyestradiol that is considered as carcinogenic metabolite. Luciferase activity was induced about 20 folds over that control by 1 nM TCDD (2,3,7,8-tetrachloto-p-dioxin). Recent industrialized society, human has been widely been exposed to widespread environmental contaminants such as PAHs(polycyclic aromatic hydrocarbon) that are originated from the imcomplete combustion of hydrocarbons. PAHs are known to be ligands of the AhR(aryl hydrocarbon receptor). Induction of cytochrome P4501B1(CYP1B1) in cell culture is widely used as a biomarker for PAHs. Therefore we have studied the effect of PAHs in the human breast cancer cells MCF-7 to evaluate bioactivity of PAHs. We have used the United State of America EPA selected 13 different PAHs, PAHs mixtures and extracts from environmental samples to evaluate the bioassay system. We examined effects of PAHs on the CYP1B1-luciferase reporter gene and CYP1B1 mRNA level. Benzo(k)fluoranthene and dibenzo(a, h)anthracene showed strong response to CYP1B1 promoter activity stimulation, and also CYP1B1 mRNAs increase in MCF-7 cells in a concentration-dependent manner. Acenaphthene, anthracene, benzo(b)fluoranthene, fluorene, fluoranthene, anphthanlene, pyrene, phenanthrene and carbazole were weak responders in MCF-7 cells. RT-PCR analysis indicated that PAHs significantly up-regulate the level of CYP1B1 mRNA.
