전단 유동 하에서 전기유변유체의 과도응답 특성 (Transient Response of an Electrorheological Fluid in Shear Flow)

  • The transient shear stress response of an electrorheological fluid is investigated experimentally. The characteristic time constants of an electrorheological fluid sheared between two concentric cylinders were obtained under various electric field strengths and shear rates. Also, two experimental modes are adopted to investigate the effect of the shear flow on the dynamic behavior of the fluid; one is that the electric field is induced before shearing, and the other is the electric field is induced after shearing. From the difference in the response time between two modes, the cluster formation time were obtained. The response times were decreased with the increase of the shear rate, irrelatively of the electric field strength. The cluster formation time were monotonically increased with increase of shear rate, and thereafter, were converged with a certain value.

맥동유동하에 있는 탄성혈관에서 벽면운동과 임피던스 페이즈앵글이 벽면전단응력에 미치는 영향 (Influence of Wall Motion and Impedance Phase Angle on the Wall Shear Stress in an Elastic Blood Vessel Under Oscillatory Flow Conditions)

  • 벽면운동과 임피던스 페이즈앵글(압력파와 유량파 사이의 시간차)이 벽면전단응력의 크기와 분포에 미치는 영향을 규명하기 위해 맥동유동하에 있는 직선 탄성혈관에서 전산유체해석을 수행하였다. 탄성을 갖는 직선혈관의 경우에는 벽면운동과 임피던스 페이즈앵글을 고려한 섭동해가 존재하는데, 이를 본 연구의 수치해와 비교함으로 수치해의 타당성을 입증하였다. 해석결과, 혈관의 벽면운동으로 인해 축방향 속도분포와 압력구배의 값에 어떤 추가분이 발생하는 것을 관찰하였다. 이러한 추가분에 의해 벽면전단응력(wall shear stress) 및 압력구배(pressure gradient)의 진폭(amplitude: time-varying component)은 감소하고 평균값(mean: time-averaged component)에도 변화를 보였는데 그 변화의 경향은 임피던스 페이즈앵글에 따라 매우 다른 모습을 보였다. 즉, 임피던스 페이즈앵글이 음의 값을 갖게 될 수록 벽면전단응력의 평균은 감소하고 진폭은 증가하는 경향을 보였다. $\pm$4%의 벽면운동이 있는 경우 대동맥에서 임피던스 페이즈앵글의 변화 가능범위인 0$^{\circ}$에서 -90$^{\circ}$로 페이즈 앵글을 감소시켰을 때 벽면전단응력의 평균값은 10.5% 감소하고 진폭은 17.5% 증가하였다. 그러므로 고혈압환자와 같이 음의 큰 페이즈앵글을 갖는 경우 벽면 전단응력의 시간에 따른 변화량(진폭/평균)이 상대적으로 커지므로 low and oscillatory shear stress 이론에 의하면 동맥경화에 더 민감하게 된다.

  • In-vitro flow characteristics downstream of a polyurethane artificial heart valve and a Bjork-Shiley Monostrut mechanical valve have been comparatively investigated in pulsatile flow using particle image velocimetry (PIV). With a triggering system and a time-delayed circuit the velocity distributions on the two perpendicular measurement planes downstream of the valves are evaluated at any given instant in conjunction with the opening behaviors of valve leaflets during a cardiac cycle. The regions of stasis and high shear stress can be found simultaneously by examining the entire view of the instantaneous velocity and Reynolds shear stress fields. It is known that high shear stress regions exist at the interface between strong axial jet flows along the wall and vortical flows in the central area distal to the valves. In addition. there are large stagnation or recirculation regions in the vicinity of the valve leaflet, where thrombus formation can be induced by accumulation of blood elements damaged in the high shear stress zones. A correlation between the unsteady flow patterns downstream of the valve and the corresponding opening postures of the polyurethane valve membrane gives useful data necessary for improved design of the frame structure and leaflet geometry of the polyurethane valve.

Computational Analysis on Calcium Dynamics of Vascular Endothelial Cell Modulated by Physiological Shear Stress

  • Flow-induced dilation of blood vessel is the result of a series of bioreaction in vascular endothelial cells(VEC). Shear stress change by blood flow in human artery or vein is sensed by the mechanoreceptor and responsible for such a chain reaction. The inositol(1,4,5)-triphophate($IP_3$) is produced in the first stage to elevate permeability of the intercellular membrane to calcium ions by which the cytosolic calcium concentration is consequently increased. This intracellular calcium transient triggers synthesis of EDRF and prostacyclin. The mathematical model of this VEC calcium dynamics is reproduced from the literature. We then use the Computational Fluid Dynamics(CFD) technique to investigate the blood stream dictating the VEC calcium dynamics. The pulsatile blood flow in a stenosed blood vessel is considered here as a part of study on thrombogenesis. We calculate the pulsating shear stress (thus its temporal change) distributed over the stenosed artery that is implemented to the VEC calcium dynamics model. It has been found that the pulsatile shear stress induces larger intracellular $Ca^{2+}$ transient plus much higher amount of EDRF and prostacyclin release in comparison with the steady shear stress case. It is concluded that pulsatility of the physiological shear stress is important to keep the vasodilation function in the stenosed part of the blood vessel.

Transient rheological probing of PIB/hectorite-nanocomposites

  • Clay suspensions in liquid polymers exhibit a time-dependent behaviour that includes viscoelastic as well as thixotropic features. Because of the presence of interacting clay platelets, particulate networks can develop, which are broken down during flow and rebuild upon cessation of the flow. Here, the use of thixotropic techniques in probing flow-induced structures in nanocomposites is explored with data on a hectorite-poly(isobutylene) model system. By means of fast stress jump measurements the hydrodynamic contributions to the steady state stresses are determined as well as those caused by the stretching of the clay floes. Flow reversal measurements do not provide a clear indication of flow-induced anisotropy in the present case. The recovery of the clay microstructure upon cessation of flow is followed by means of overshoot and dynamic measurements. The development of a particulate network is detected by the appearance and growth of a low frequency plateau of the storage moduli. The modulus-frequency curves after various rest times collapse onto universal master curves, regardless of the pre-shear history or temperature. The scaling factors for this master curve are the crossover parameters. The crossover moduli are nearly a linear function of the crossover frequency, the relation being identical for recovery after shearing at different shear rates. This function depends, however, on temperature.

혈관내피세포에 열 충격 부과시 죽력이 stress proteins의 발현에 미치는 영향 (Effects of Bambusae Caulis in Liquamen on the Stress Proteins Induced by Heating in Endothelial Cells)

  • We have previously observed that Bambusae Caul is in Liquamen (BCL) stimulates the adipose conversion of 3T3-L1 cells and molecular chaperones were involved in the process of the assembly and replacement of laminin subunits in Bovine aortic endothelial cells(BAEC). Endothelial cells are exposed to continuous shear stress due to the blood flow. Heat shock protens(hsp) are a well-known stress response protein, namely, stress proteins. To investigate effects of BCL on the stress proteins induced by heating in endothelial cells, we have analyzed synthetic amounts of stress proteins in sodium dodecyl sulfate gel electrophoresis under reducing conditions. Under the condition of heating stress, BCL inhibited the synthesis of stress proteins in endothelial cells. These results suggest that BCL may have an important role for expression of stress proteins induced by heating in endothelial cells.

Effects of resveratrol on laminar shear stress-induced mitochondrial biogenesis in human vascular endothelial cells

  • [Purpose] The purpose of the study was to determine the combined effects of resveratrol supplementation with high-flow LSS on mitochondrial biogenesis in human vascular endothelial cells. [Methods] Cultured human umbilical vein endothelial cells were treated with 20 μM of RSV. For the shear experiments, cells grown to a >90% confluence were exposed to physiological levels of LSS (5 to 20 dyne/cm2) for 12 to 36 hours using a cone and plate shear apparatus. Gene expressions were analyzed by western blotting. [Results] Depletion of mitochondrial integrity was directly associated with increase in endothelial activation/dysfunction. The expressions of mitochondrial biogenesis regulator genes, such as SIRT1, PGC-1α, and TFAM, and the mitochondrial contents were significantly increased after treatment with both resveratrol and high-flow LSS for 12 hours. However, supplementation of resveratrol to high-flow LSS for a prolonged duration had no synergistic effect on the levels of mitochondrial biogenesis regulator gene expressions and mitochondrial content compared to the LSS treatment alone. [Conclusion] The present study demonstrated that the supplementation of resveratrol to high-flow LSS has no synergistic effects on enhancing mitochondrial integrity in human vascular endothelial cells.

유체에 의해 유발된 전단력이 치은 섬유아세포 유전자 발현 변화에 미치는 영향에 관한 연구 (GENE EXPRESSION AFTER THE APPLICATION OF THE FLUID-INDUCED SHEAR STRESS ON THE GINGIVAL FIBROBLAST)

  • The oral cavity is humid environment mainly due to the continuous salivary flow. The reaction of oral mucosa to fluid flow is important for homeostasis and pathogenesis. The objective of this study is the screening the change of gene expression after the application of fluid induced shear stress (FISS) on the gingival fibroblast using cDNA microarray assay. The immortalized human gingival fibroblasts were grown and FISS was applied using a cone viscometer at a rotational velocity of 40 rpm, respectively for periods of 2 and 4 hours. The synthesis of cDNA was done from the extracted total RNA and cDNA microarray assay was done subsequently. The genes that showed over 1.6 in the Cy3/Cy5 or the Cy5/Cy3 value were regarded as genes influenced significantly by the FISS application ion (/M/>0.7). The " RUNX-1" was increased its expression in 2 hours group and " RUN and SH3 domain containing 1" was increased its expression in 4 hours group. The "CC020415", "cyclin L1", "interferon regulatory factor1", "early growth response 1", "immediate early response 2", and "immediate early response 3" genes were increased their expression in 2 and 4 hours after FISS application. In conclusion, we could find many genes that were probably related to the FISS application. Interestingly, most of them were placed in similar molecular pathways and these findings improve the reliability of chip data and usefulness in overall screening. From this experiment, we could find many items for further study and it will make improvement in the understanding of intracellular events in response to FISS.

  • This paper presents the time responses of calcium ($Ca^{2+}$) ion concentration of MG-63 cells induced by a constant shear stress in micro channel were observed in the real time. Most of cells have similar rising time. There were some time delays because of the initial position of the cell in the micro channel along the pressure-driven fluid flow. The concentration of $Ca^{2+}$ exponentially decreased while time constant of each profile did not have any relation to the peak value of concentration.

Change of Fractured Rock Permeability due to Thermo-Mechanical Loading of a Deep Geological Repository for Nuclear Waste - a Study on a Candidate Site in Forsmark, Sweden

  • Opening of fractures induced by shear dilation or normal deformation can be a significant source of fracture permeability change in fractured rock, which is important for the performance assessment of geological repositories for spent nuclear fuel. As the repository generates heat and later cools the fluid-carrying ability of the rocks becomes a dynamic variable during the lifespan of the repository. Heating causes expansion of the rock close to the repository and, at the same time, contraction close to the surface. During the cooling phase of the repository, the opposite takes place. Heating and cooling together with the, virgin stress can induce shear dilation of fractures and deformation zones and change the flow field around the repository. The objectives of this work are to examine the contribution of thermal stress to the shear slip of fracture in mid- and far-field around a KBS-3 type of repository and to investigate the effect of evolution of stress on the rock mass permeability. In the first part of this study, zones of fracture shear slip were examined by conducting a three-dimensional, thermo-mechanical analysis of a spent fuel repository model in the size of 2 km $\times$ 2 km $\times$ 800 m. Stress evolutions of importance for fracture shear slip are: (1) comparatively high horizontal compressive thermal stress at the repository level, (2) generation of vertical tensile thermal stress right above the repository, (3) horizontal tensile stress near the surface, which can induce tensile failure, and generation of shear stresses at the comers of the repository. In the second part of the study, fracture data from Forsmark, Sweden is used to establish fracture network models (DFN). Stress paths obtained from the thermo-mechanical analysis were used as boundary conditions in DFN-DEM (Discrete Element Method) analysis of six DFN models at the repository level. Increases of permeability up to a factor of four were observed during thermal loading history and shear dilation of fractures was not recovered after cooling of the repository. An understanding of the stress path and potential areas of slip induced shear dilation and related permeability changes during the lifetime of a repository for spent nuclear fuel is of utmost importance for analysing long-term safety. The result of this study will assist in identifying critical areas around a repository where fracture shear slip is likely to develop. The presentation also includes a brief introduction to the ongoing site investigation on two candidate sites for geological repository in Sweden.
