Conjugated Linoleic Acid를 급여한 돼지의 호중구 탐식능 증강 (Supplement of Conjugated Linoleic Acid Increases Neutrophil Phagocytosis in Pigs)

  • CLA를 급여시킨 돼지에 있어 말초 혈액 세포의 변화와 탐식세포의 탐식반응을 조사하였다. 100일령, 평균체 중 80kg인 거세 수퇘지에게 10t-l2c CLA와 CLA mixture를 각각 사료 중에 5% 첨가하여 총 4주간에 걸쳐 자유급여 시켰다. 급여 전 (0주), 급여 2주후, 4주후 그리고 급여 중지 후인 5주차에 각각 혈액세포 수치(PCV 백혈구수 및 백혈구 감별계산치)의 변화와 탐식세포의 탐식활성을 측정하였다. CLA의 급여 따른 PCV의 수치에 있어서는 아무런 변화가 없었다. 그러나 10t-l2c CLA 또는 CLA mixture가 함유된 사료를 급여한 돼지에 있어서 CLA를 급여하지 않은 돼지에 비해 백혈구수 특히 호중구 수가 현저히 증가하였다. 말초혈액 단핵구세포(mononuclear cells; MNC)와 다형핵백혈구(polymorphonuclear cells; PMN)의 탐식능를 유세포분석기로 분석한 결과, 10t-l2c CLA 또는 CLA mixture 급여와 상관없이 MNC와 단구세포들(monocyte-rich cells)의 탐식능에는 변화가 없었다. 그러나 PMN의 탐식 활성은 CLA 무급여 대조돼지들에 비해 CLA 급여군은 2주에서 5주차에 걸쳐 증가된 탐식능를 나타내었다. 이상의 결과로부터 돼지에 CLA 급여는 호중구수 증가와 호중구 탐식능 증가를 유도할 수 있음을 제시하였으며 향후 생체방어 증강을 위해 CLA를 임상적으로 투여할 수 있을 것으로 보인다.

Anti-fibrotic effects of Orostachys japonicus A. Berger (Crassulaceae) on hepatic stellate cells and thioacetamide-induced fibrosis in rats

  • BACKGROUND/OBJECTIVE: Orostachys japonicus A. Berger (Crassulaceae) has been used in traditional herbal medicines in Korea and other Asian countries to treat various diseases, including liver disorders. In the present study, the anti-fibrotic effects of O. japonicus extract (OJE) in cellular and experimental hepatofibrotic rat models were investigated. MATERIALS/METHODS: An in vitro hepatic stellate cells (HSCs) system was used to estimate cell viability, cell cycle and apoptosis by MTT assay, flow cytometry, and Annexin V-FITC/PI staining techniques, respectively. In addition, thioacetamide (TAA)-induced liver fibrosis was established in Sprague Dawley rats. Briefly, animals were divided into five groups (n = 8): Control, TAA, OJE 10 (TAA with OJE 10 mg/kg), OJE 100 (TAA with OJE 100 mg/kg) and silymarin (TAA with Silymarin 50 mg/kg). Fibrosis was induced by treatment with TAA (200 mg/kg, i.p.) twice per week for 13 weeks, while OJE and silymarin were administered orally two times per week from week 7 to 13. The fibrotic related gene expression serum biomarkers glutathione and hydroxyproline were estimated by RT-PCR and spectrophotometry, respectively, using commercial kits. RESULTS: OJE (0.5 and 0.1 mg/ mL) and silymarin (0.05 mg/mL) treatment significantly (P < 0.01 and P < 0.001) induced apoptosis (16.95% and 27.48% for OJE and 25.87% for silymarin, respectively) in HSC-T6 cells when compared with the control group (9.09%). Further, rat primary HSCs showed changes in morphology in response to OJE 0.1 mg/mL treatment. In in vivo studies, OJE (10 and 100 mg/kg) treatment significantly ameliorated TAA-induced alterations in levels of serum biomarkers, fibrotic related gene expression, glutathione, and hydroxyproline (P < 0.05-P < 0.001) and rescued the histopathological changes. CONCLUSIONS: OJE can be developed as a potential agent for the treatment of hepatofibrosis.

우수관거 흐름 제어를 위한 관망 설계에 관한 연구 (A Study of Sewer Layout to Control a Outflow in Sewer Pipes)

  • 최적의 우수관망 설계를 위한 기존의 연구 모형들은 관경, 관 경사 등을 포함한 관거의 설계에 있어서 설계 강우에 대하여 최소의 비용을 목적으로 하여왔다. 그러나 이러한 모형들은 우수관망 관거 내 흐름의 중첩효과에 대한 고려는 하지 않고 있다. 관거 내 흐름의 특성은 관경, 관 경사 등에 따라서 달라지며 특히 관망의 구성이 달라짐에 따라서 크게 영향을 받는다. 본 연구에서는 우수관망을 구성함에 있어서 관거 내 흐름을 분산시키고 제어함을 목적으로 하고 있다. 이것은 관망 구성에 따른 관거 내 흐름의 중첩효과를 제어함으로써 가능하며, 이러한 흐름의 제어를 통하여 우수관망에서의 내수침수 위험도가 저감될 수 있다. 본 연구에서는 맨홀간 관거의 연결 방향을 조정함으로써 관내 유입량을 분산시키고 그 결과로서 유출구에서의 첨두 유출량은 저감되도록 우수관망을 구성하였다. 이때 관망 내 흐름에 대한 수리학적 분석은 SWMM(Storm Water Management Model)을 본 모형과 연계 구성함으로써 가능하였다. 관망 구성에 따른 내수침수 위험도의 저감을 검증하기 위하여 본 연구에서는 서울시 가락 배수분구의 현재 구성된 관망에 대하여 유출구에서의 첨두 유출량을 최소화하기 위하여 맨홀간 관거의 연결 방향을 변화시켰으며, 그 결과 30분 지속시간의 설계빈도 강우에 대하여 유출구 첨두 유출량은 약 20% 감소하였다. 또한 수정된 관망 구성은 설계빈도를 초과하는 강우에 대해서도 월류량 발생량이 저감되는 결과를 나타내었다.

플라스틱 단동온실의 천창 종류에 따른 자연환기 효과 (Efficiency of Different Roof Vent Designs on Natural Ventilation of Single-Span Plastic Greenhouse)

  • 여름철에 자연환기는 온실의 온도를 낮추는데 중요한 역할을 한다. 온실의 형태, 환기창 종류, 환기창의 위치 등은 자연환기 성능에 큰 영향을 미친다. 본 연구에서는 전산유체역학(CFD)을 이용하여 다양한 천창구조에 대하여 측창에 따른 부력환기 효과를 비교분석 하였다. Boussinnesq 가정을 사용하여 전체 계산영역에 대한 부력효과를 시뮬레이션 하였다. 또한 RNG $K-{\varepsilon}$ 난류모델을 사용하였다. 일사량 효과를 시뮬레이션 하기 위해 Solar ray tracing과 함께 Discrete originates (DO) radiation 모델을 사용하였다. 실험온실 내부의 온도를 측정하여 CFD모델을 검증하였으며, 실험값과 계산값이 잘 일치하는 것으로 나타났다. 7가지의 천창구조에 대하여 온실의 내외부 온도차이와 환기횟수를 비교하였다. 내외부온도의 차이는 $3.2{\sim}9.6^{\circ}C$ 범위로 나타났고, 환기횟수는 $0.33{\sim}0.49min^{-1}$ 범위로 나타났다. 고깔형 천창구조 온실의 경우 내외부 온도차이가 $3.2^{\circ}C$로 가장 낮았고 환기횟수도 $0.49min^{-1}$로 가장 높게 나타나 환기효과가 가장 우수한 것으로 나타났다.

개 정액의 냉동보존 시 κ-Carrageenan이 정자 성상에 미치는 영향 (Effect of κ-Carrageenan on Sperm Quality in Cryopreservation of Canine Semen)

  • This study was conducted to find out the effect that ${\kappa}-Carrageenan$ has on the properties of dog sperm when it was added to the cryoprotectant. Extender basically was contained 1.21 g Trizma base, 0.67 g citric acid, 0.4 g glucose, 0.03 g penicillin G, 0.05 g streptomycin sulfate. Extender1 was added with 0.1%, 0.2%, 0.3%, and 0.5% carrageenan, while extender2 was supplemented with glycerol. After freezing-thawing, the motility, viability, acrosome integrity, apoptosis, and ROS (reactive oxygen specifications) of sperm were measured to analyze the effects of the supplementation of carrageenan. Total Motile (TM), Rapid Progressive Motile (RPM), Medium Progressive Motile (MPM), and Immotile were measured through the CASA system after thawing in 37 degree water. Extender with 0.2% ${\kappa}-carrageenan$ ($64.26{\pm}0.49$) was significantly higher than control ($40.24{\pm}8.27$) (p < 0.05). RPMs of extender with 0.1%, 0.2% ${\kappa}-carrageenan$ ($57.64{\pm}6.34$, $56.47{\pm}1.35$) were significantly higher than the other groups (p < 0.05). Acrosome integrity was measured by dyeing to PSA-FITC with an epifluorescence microscope. Normal acrosome ratio of extender with 0.5% ${\kappa}-carrageenan$ ($61{\pm}8.03$) was higher than the other groups (p < 0.05). Apoptosis was measured with a FACSCalibur flow cytometer using FITC (FITC Annexin V Apoptosis Detection Kit). Treated groups of ${\kappa}-carrageenan$ of 0.1% ($0.81{\pm}0.05$), 0.2% ($0.85{\pm}0.05$) were significantly higer (p < 0.05) than control. Modified SYBR/PI staining was used for determination of viability and DCF staining was used for evaluation of ROS. Viability and ROS were not significantly different from other groups. In conclusion, adding a certain concentration of carrageenan to the extender of cryopreservation, carrageenan contributes to the improvement of the sperm motility, acrosome integrity and prevention of apoptosis.

마이크로버블을 이용한 수경재배 물냉이의 글루코시놀레이트 함량 증대 (Microbubbles Increase Glucosinolate Contents of Watercress (Nasturtium officinale R. Br.) Grown in Hydroponic Cultivation)

  • 본 실험은 물냉이 수경재배 시 양액 내 발생 시킨 마이크로버블이 물냉이의 생육과 glucosinolate 축적에 미치는 영향을 알아보기 위해 수행되었다. 본엽 4매의 물냉이 유묘(파종 2주 후)를 마이크로 버블과 비-마이크로버블을 발생시킨 오오츠카 배양액을 이용하여 환경조절룸에서 3주간 재배하였다. 물냉이 초장은 대조구처리가 마이크로버블처리보다 41% 증가하였으며, 유의적으로 높게 나타났다. 그러나, 지상부 생체중과 건물중, 근장, 엽장, 엽폭, SPAD, 량자수율값은 두처리간 유의적 차이는 나타나지 않았다. Glucosinolate 함량을 분석 결과 4-methoxygluco-brassicin을 제외한 glucoiberin, glucobrassicin, gluconapin, gluconasturtiin의 경우 마이크로버블 처리구가 대조구보다 유의적으로 높게 나타났으며, 물냉이 한주 당 총 glucosinolate 함량은 마이크로버블 처리구가 대조구 보다 $85%({\mu}mol/g\;DW)$$65%({\mu}mol/plant)$ 더 높게 나타났다. 본 연구 결과는 담액재배시 양액 내 마이크로버블이 물냉이의 glucosinolate 함량을 증가시킬 수 있을 것으로 나타났다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. Our department has developed an information system for routine hematology using down-sized computer system. The operating system and database management system were SCO UNIX and SCO foxbase, respectively. For program development, we used Xbase language provided by SCO foxbase. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer. The frame and leaflet of the polymer valve were made from polyurethane, To evaluate the hemodynamic performance of the polymer valve a comparative study of flow dynamics past a polymer valve and a St. Jude Medical prosthetic valve under physiological pulsatile flow conditions in vitro was made. Comparisons between the valves were made on the transvalvular pressure drop, regurgitation volume and maximum valve opening area. The polymer valve showed smaller regurgitation volume and transvalvular pressure drop compared to the mechanical valve at higher heart rate.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Also, we propose the real time trajectory planning method which generates continuous accelleration paths using 3 stage linear filters to minimize the impact to human body induced by arm motions and to reduce the muscle fatigue. We use combination of MLP and fuzzy filter for pattern recognition to estimate the direction of a muscle and Hogan's method for the force estimation. EMG signals are acquired by using a amputation simulator and 2 dimensional joystick motion. The bedside monitor consists of a main unit and module cases with various parameter modules. The main unit includes a 12.1" TFT color LCD, a main CPU board, and peripherals such as a module controller, Ethernet LAN card, video card, rotate/push button controller, etc. The main unit can connect at maximum three module cases each of which can accommodate up to 7 parameter modules. They include the modules for electrocardiograph, respiration, invasive blood pressure, noninvasive blood pressure, temperature, and SpO2 with Plethysmograph.

도립$(-6^{\circ})$이 혈장 Catecholamine 및 심장혈관계에 미치는 영향 (Effects of Head-Down Tilt$(-6^{\circ})$ on Hemodynamics and Plasma Catecholamine Levels)

  • Head-down tilt (HDT) at $-6^{\circ}$ has been commonly used as the experimental model in both man and animals to induce the blood shift toward the head or central protion of the body, demonstrating similar physiological effect encountered in the weightlessness in the orbital flight. There are few reports about the physiological response upon the cardiovascular regulatory system or the tolerance to the $(-6^{\circ})$ HDT within a relatively short period less than 1 hour. Therefore, the purpose of this study way to observe the effects of $-6^{\circ}$ HDT on cardiovascular system within 30 minutes and to evaluate early regulatory mechanism for simulated hypogravity. Ten mongrel dogs weighing 8-12 kg were anesthetized with the infusion of 1% ${\alpha}-chloralose$ (100 mg/kg) intravenously, and the postural changes were performed from the supine to the $-6^{\circ}$ head-down Position, then from the head-down to the supine (SUP), and each posture was maintained for 30 minutes. Blood flow $({\dot{Q}})$ through common carotid and femoral arteries were determined by the electromagnetic flowmeter. Mean arterial pressure (MAP), heart rate (HR), respiratory rate , and pH, $P_{O_2}$, $P_{CO_2}$ and hematocrit (Hct) of arterial and venous blood were also measured. The peripheral vascular resistance was calculated by dividing respective MAP values by ${\dot{Q}}$ through both sides of common carotid or femoral arteries. The concentration of plasma epinephrine and norepinephrine was determined by Peuler & Johnson's radioenzymatic method. The results are summarized as follows: In the initial 5 minutes in $-6^{\circ}$ HDT, HR was significantly (p<0.05) increased and MAP slightly decreased. Although ${\dot{Q}}$ and carotid peripheral artery resistance were not significantly changed, ${\dot{Q}}$ through femoral artery was diminished and femoral peripheral artery resistance was elevated. In the SUP, the initial changes of MAP and HR were increased (p<0.05), but those of ${\dot{Q}}$ and peripheral vascular resistance through both common carotid and femoral arteries were not significant. After 10 minutes of each postural change in both HDT and SUP, MAP was maintained almost equal to that of the level of pretilting control. During 60 minutes of both postural changes of HDT and SUP, $P_{O_2}$ and Hct were not changed significantly. However pH tended to increase slowly and $P_{CO_2}$ was gradually decreased. The pH and $P_{CO_2}$ seemed to be related to the increased respiratory rate. Plasma epinephrine concentration was not changed significantly and plasma norepinephrine concentration was slightly decreased in the course of HDT and also at 10 minutes of SUP. However these concentration changes were statistically insignificant. From these results, it may be concluded that the effect of $-6^{\circ}$ HDT for 30 minutes on the cardiovascular system and plasma catecholamine levels of the dog is minimum and it is suggestive that the cardiovascular regulatory mechanism, possibly mediated by so called gravity receptors including baroreceptor and volume receptor, has been properly and adequately operated.

액체헬륨을 이용한 위성시험용 극저온패널 냉각시스템 개발 및 검증 (Development and Validation of Cryopanel Cooling System Using Liquid Helium for a Satellite Test)

  • 인공위성 적외선 탑재체의 열싱크 역할을 위해, 액체헬륨을 이용하여 극저온패널(가로 약 800 mm, 세로 약 700 mm)을 4.2 K까지 냉각시키는 시스템을 설계, 개발, 검증하였다. 유효직경 8 m, 유효 깊이 10 m의 대형열진공챔버에서 검증된 본 냉각시스템은 500리터 용량의 액체헬륨용기 두 개(극저온 패널로의 액체헬륨 또는 저온헬륨가스 주 공급용기 및 주공급용기로의 재충진용기)를 사용하였는데, 목표인 극저온패널의 냉각 및 온도제어는 주 공급용기 내부의 미세압력조절을 통해 액체헬륨 공급유량을 제어함으로써 이루었다. 극저온패널에 공급된 후 배기되는 저온 헬륨가스는 특별히 설계, 제작된 사중진공배관의 제3층을 흐르며 열차단막의 역할을 수행함으로써, 액체헬륨 공급 라인인 제1층(중심배관)으로의 열유입을 최소화하였다. 극저온패널을 상온에서 40 K(합성표준불확도 194 mK)까지 냉각시키는데 약 3시간이 소요되었으며, 20 W의 열을 발산하는 극저온패널을 40 K 주변 온도에서 1 K 이내의 온도균일도를 가지며 유지할 수 있었다.
