Feeding a Diet with Precise Lysine Level improved Laying Performance and Feed Efficiency of Broiler Breeder Hens at the Early Laying Stage

  • A dose-response experiment was conducted to determine the lysine requirement for broiler breeder hens during pre-peak production. Totally, one hundred and twenty-six flock 23-week-old Ross 308 broiler breeder hens with similar body weight were selected ($2,188{\pm}32g$) for a 6-week experiment. Hens were fed with a basal diet of corn-wheat-soybean meal formulated to achieve the Ross 308 breeder nutrient specifications (2016), except for lysine. The 7 graded, daily lysine intake levels used in this experiment were 732, 785, 838, 891, 944, 997, and 1,050 mg, and hens were restricted to 133 g of feed throughout this experiment. Pen based egg production were recorded once a day and all eggs were weighed daily. Age at sexual maturity was determined when the hens attained age at 25% production. Body weight at 23~29 weeks of age was not affected (P>0.05) by lysine levels. By fitting a linear-plateau model, the daily lysine requirements for feed conversion ratio, total produced egg weight, and age at sexual maturity at 23~29 weeks of age were estimated as 865, 907, and 891 mg, respectively. Using a quadratic-plateau model, the daily lysine requirement at 23~29 weeks of age were estimated as 974, 964, and 950 mg for feed conversion ratio, total produced egg weight, and age at sexual maturity, respectively. These results suggested that the daily lysine requirement for modern broiler breeder hens according to the National Research Council (1994) are insufficient for higher total produced egg weight, sexual maturity, and feed efficiency, and 120% of the NRC recommendation level would improve hen productivity when data are fitted under linear- and quadratic-plateau models.

Effect of time and depth of insemination on fertility of Bharat Merino sheep inseminated trans-cervical with frozen-thawed semen

  • Background: Artificial insemination (AI) can serve as a powerful tool to the sheep owners for making rapid genetic progress of their flock. The AI in sheep is mostly performed using fresh semen with two reasons i) lambing rate following trans-cervical AI with frozen semen is limited by the inability of frozen-thawed sperm to transit the cervix and ii) the need of circumventing the cervical barrier through laparoscope aided intrauterine AI. Therefore, AI with frozen-thawed semen is not as widespread in sheep as it is in other domestic species. However, to get maximum benefits through the use of AI, frozen-thawed semen is a prerequisite because instead of high fertility, the short shelf life of fresh semen coupled with a limitation on the number of insemination doses achievable per unit time restricts the widespread use of individual sires. Therefore, in order to enhance lambing rate, a total of 240 trans-cervical artificial inseminations with frozen-thawed semen were performed in Bharat Merino ewes during autumn season either once in the evening (G-I, 10 h after onset of estrus, n = 100) or twice (G-II, 14 h and 22 h after onset of estrus, n = 140) i.e. once in the morning and again in the evening. Results: The pregnancy rate (proportion of pregnant ewes confirmed by ultrasonography at day 40) and lambing rate (proportion of ewes lambed) were higher in G-II as compared to G-I (26.4 vs 20% and 19.3 vs 10%, respectively). The difference in lambing rates was statistically (P < 0.05) significant. The depth of insemination within cervico-uterine tract had no significant effect on pregnancy and lambing rates. Conclusions: The results indicate that lambing rate in sheep following TCAI with frozen-thawed semen was significantly influenced by time of inseminations. Two inseminations after 14 and 22 h of onset of estrus enhanced the lambing rates of Bharat Merino sheep as compare to single insemination after 10 h of onset of estrus. The TCAI technique with frozen-thawed ram semen is promising and may serve as a valuable tool for genetic improvement of sheep breeds. Research efforts are going on worldwide to overcome the poor fertility following TCAI with frozen-thawed semen.

Studies on the Ecological Characteristics of Marine Bacteria Isolated from Deep Sea (심해 해양미생물의 분리 및 분리균의 생태적 특징에 관한 연구)

  • Flavobacterium spp., Pseudomonas spp., and Vibrio spp. were isolated from samples(sediments) of Sagami Bay and Suruga Bay(in Japan) at 810-4,000m in depth. Among isolated strains, Vibvio sp.-86 and sp.-87 strains were identified as barophilic and psychrophilic ones. They grew in 400 atm and showed best growth at 100 atm. Marine bacteria grown at 400 atm were long rod shape and 30 to 50times longer than those grown at 1 atm. which were short rod shape and formed flocks (aggregates). Vibrio sp,-86 strain grew at $5-37^{\circ}C\;and\;0,5-9.0\%\;NaCl\;(3.0\%\;of\;optimum\;concentration),$ while Vibrio sp.-87 strain grew at $1-7\%\;NaCl\;(2,0\%\;of\;optimum\;concentration).$ The fatty acid compositions of Vibrio sp.-86 strain grown at 1 atm were $C_{20}-C_{22:0},\;C_{l6:1},\;and\;C_{16:0}$ in the order of their abundance and at 400 atm the order were $C_{18:1},\;C_{18:0},\;and\;C_{20}-C_{22}$, whereas those of Vibrio sp.-87 strain at 1 atm were $C_{6:1},\;C_{14:1},\;and\;C_{20}-C_{22}$ and at 400 atm the order were $C_{14:1},\;C_{12:0},\;and\;C_{16:1}$ The amino acids compositon of Vibrio sp.-86 strain grown at 1 atm were abundant in the order of aspartic acid, methionine, and glutamic acid and those at 400 atm were abundant in the order of methionine, glutamic acid, and aspartic acid. The amino acids composition of Vibrio sp.-87 strain grwon at 1 atm were abundant in the order of methionine, glutamic acid, and aspartic acid and those at 400 atm were abundant in the order of methionine, glutamic acid, and isoleucine.

Time Trends in Estimates of Genetic Parameters in a Population of Layer Breeders (난용종계 집단에서의 선발에 의한 유전모수 변화 양상)

  • This study was carried out to investigate the time-trends of genetic parameters of the dosed flock population which selected for improving egg production. Data for two layer pure lines, Line-W (Single Comb White Leghorn) and Line-B (brown layer) which have been maintained at the Mani Breeding Farm were collected from 1980 to 1985 during 5 generations. The effective number of parents per generation ranged from 148 to 366 in Line-W and 85 to 355 in Line-B, and the cumulative expected inbreeding coefficients during 5 generations of selection were 15% and 1.6%. So inbreeding could not be considered a critical factor on estimating the genetic parameters, heritabilities and genetic correlations Heritabilities of EN 300 and EN 400, primary two selected traits were significantly decreased during 5 generations but the estimates of the other 03its not showed the consistent decreasing pattern significantly. No time trends of probable consequence were evident in the genetic correlation coefficients of the traits studied. The reason for that situation was attributed to the fact that selection was conducted for multiple objectives and the relative importance of selection for the studied traits were not consistent by generations.

A Survey on the Immune Status and Productivity of Vaccinated Poultry Flocks against Newcastle Disease in the Epizootic Area (뉴캣슬병백신 접종계군에 있어서 면역상태와 ND 발생시 닭의 생산성에 미치는 영향)

  • During the 1978-79 Newcastle disease (ND) epizootic period, a detailed survey was conducted on the five representative farms which had been following one of the recommended vaccination programs. When the disease broke out during laying period, clinical symptoms were mild to moderate respiratory distress and greenish diarrhea. Affected flocks experienced weekly mortality from less than 1% to 17%. Egg production returned to normal 18 to 36 days after the initial signs appeared although some flocks never returned to normal. On postmortem examination,, most affected chickens showed severe hemorrhagic lesions in the duodenum, hematoma on ova, and heavy fat accmulation on various visceral organs. Most of the NO affected flocks had geometric mean hemagglutination inhibition antibody(HIA) titers of 7 log$_2$ or higher two to three weeks after the appearance of clinical signs. These HIA titers were at least 16-fold higher than those before infection. Flock mean HIA titers before infection were usually lower than 3 log$_2$. Severity of clinical signs and anamnestic antibody response were maximum in the flocks whose vaccination immunity was insufficient or waned considerably. Observations showed that even young birds, if properly vaccinated, could get effective protection from field ND exposure.

Protocols for Estrus Synchronization in Awassi Ewes under Arid Environmental Conditions

  • In July, three trials were conducted to evaluate the best sponge type and optimum PMSG dose to be administered to sheep under the Jordanian Badia (arid) conditions. In trial 1, three flocks (n=77, n=18 and n=47 for flocks 1, 2 and 3, respectively) were administered with 40 mg fluorogestone acetate (FGA) intravaginal sponges for 12 days before receiving 600 IU of PMSG at the time of sponge removal. In trial 2, 95 ewes were assigned to 4 groups to receive 300 (n=25), 450 (n=27), 600 (n=22) or 750 (n=21) IU of PMSG following a 12 d FGA 40 mg sponge insertion period. In trial 3, 60 ewes were assigned to 3 groups (n=20) to receive either FGA 30 mg, FGA 40 mg or medroxyprogesterone acetate (MAP) 60 mg intravaginal sponges all followed by an administration of 600 IU of PMSG at sponge removal. In all trials, rams were isolated 1 day before sponge insertion and were allowed back with the ewes at sponge removal. Estrual responses and lambing data were collected. The effects of treatment, milking status and face color on estrual responses and lambing data were examined. In trial 1, greater first cycle conception rate (p<0.05), twinning rate (p<0.01) and the number of lambs born/served ewe (p<0.01) were observed in flock 2 compared with flocks 1 and 3. Neither face color nor milking status had any influence on the measured parameters (p>0.05). Despite low lambing rate in trial 2, ewes receiving 600 IU of PMSG had greater (p<0.05) number of lambs born/served ewe compared with ewes receiving 450 IU of PMSG. Regardless of PMSG dose, intervals to detected estrus occurred 10 h earlier (p<0.01) in dry than lactating ewes. Similar to trial 2, lambing rate was depressed in trial 3. The expression of estrus was advanced (p<0.05) in ewes receiving MAP 60 mg sponges compared with those receiving FGA 30 and FGA 40 mg sponges (42$\pm$3.1, 49$\pm$3.1 and 49$\pm$3.1 h post sponge removal in ewes receiving MAP 60 mg, FGA 30 mg and FGA 40 mg sponges, respectively). Other parameters were not influenced (p>0.05) by sponge type, milking status and face color. Data show that a 600 IU dose of PMSG tends to give the best lambing results. In addition, results indicate that the use 60 mg MAP sponges for estrus synchronization may be more appropriate under the Jordanian Badia conditions during late seasonal anestrus.

The change of core habitats of the cranes due to release of the civilian control zone; CCZ and construction disturbance (서식지 교란 및 민간인통제지역 해제에 의한 두루미와 재두루미의 핵심서식지 변화)

  • The endangered species, The Red-crowned Crane and the White-naped Crane are vulnerable species to the disturbance for human beings. We examined the importance of CCZ for the cranes by comparing the crane's density in some CCZ-released areas, which are cranes' wintering site in Cheorwon. We also assessed influence of disturbance such as construction and greenhouses on core habitats of cranes. Our study results suggested that the construction and reclamation in the CCZ shrunk core habitat area while increasing core area of far from the construction and reclamation area. The CCZ has been set since March, 2011 and the number of greenhouse has rapidly increased after 2012. As the number and size of greenhouse in the area where designation of CCZ was cancelled increased, foraging area of cranes diminished. Although the area where designation of CCZ was cancelled seemed to have more human disturbance than CCZ, the foraging density of cranes did not decline and even that of white-naped cranes increased. This could be the influence of artificial food supply at their roosting site. In conclusion, if the area of CCZ decline continuously in the future, density of cranes would decrease.

Prevalent Characteristics of Fowl Typhoid and Antibiotic Susceptibility of Salmonella gallinarum (닭 티푸스의 발생상황과 Salmonella gallinarum의 항균제 감수성)

  • The present study was conducted to investigate the prevalent characteristics of Fowl Typhoid and antibiotic susceptibility of Salmonella gallinarum isolated from 56 infective or dead chickens of 20 egg laying farms in Kyung Buk province during the period from August to December 1994. 1. Among 416, 000 chickens of 92 flocks in 20 egg laying farms, 17, 360 chickens of 31 flocks were died of Fowl Typhoid. 2. Salmonella gallinarum was isolated from 56 chickens in liver and spleen, and then blood of infective chickens was positive to Pullorum antigen. 3, In the survey of gross lesion of 56 chickens, 43 chickens(76.8%) were swelled at liver, 39(69.6%) were swelled at spleen, 12(21.4%) were changed with bronze, 3(5.4% ) were hemorrhagic in peritoneal cavity. 4. In transmission pattern, 4 farms were outbreaked the entrance of chicken house at first, but the others were outbreaked at various place. They were transmitted at right and left directions in flock. 5. 2 farms confirmed at the early stage of infection were eradicated by removing infective chickens and administrating antibiotics, but 18 farms at chronic stage were not. 6. The biochemical properties of 112 Salmonella gallinarum from chickens were generally identical to those of the referance, but H$_2$S was not productive, cellobiose was fermentive. 7 Minimum inhibitory concentration(MIC) of 20 isolates was performed by using 21 antibiotics, MICs of Amikacin(Ak), Gentamicin(Gm), Kanamycin(Km), and Tetracycline (Tc) were below 1.6 ug/ml, Ampicillin(Am), Furazolidone(Fu) and Neomycin(Nm) were below 3.1 ug/ml, Cephalothin(Ce), Cefazoline(Cf) and Chloramphenicol(Cm) were below 6.3 ug/ml, Nalidixic acid(Na), Polymyxin(Po) and Rifampicin(Rf) were below 12.5 ug/ml, Penicillin (Pm) was below 25 ug/ml, Colistin(Co) and Streptomycin(Sm) were below 50 ug/ml, Sulfamerazine(Sr) and Sulfamethazine (St) were below 200 ug/ml, Lincomycin(Lm) and Spiramycin(Sp) were below 400 ug/ml, Bacitracin(Ba) was below 800 ug/ml. 8. Among the 20 isolates, all(100%) of those were sensitive to Ak, Am, Ce, Cf, Cm, Fu, Gm, Km, Na, Nm, Po, Rf, Sr, St and Tc, but 6 isolates(30%) were resistent to Co, 20(100% ) to Ba, Lm, Pm, Sm, and Sp. The drug resistance patterns were simple which 6 strains were BaCoLmPmSmSp type, and 14 were BaLmPmSmSp type.

Genetic and Non-genetic Factors Affecting Mortality in Lori-Bakhtiari Lambs

  • Data and pedigree information for Lori-Bakhtiari sheep used in this study were 6,239 records of lamb mortality from 246 sires and 1,721 dams, collected from 1989 through 2007 from a Lori-Bakhtiari flock at Shooli station in Shahrekord. The traits investigated were cumulative lamb mortality from birth up to 7 days, up to 14 days, up to 21 days, and up to 1, 2, 3, 4, 5, 6, 7, 8, 9, 10, 11 and 12 months of age. The models included fixed factors that had significant effects and random direct genetic, maternal genetic and maternal permanent environmental effects. Variance components were estimated using the restricted maximum likelihood procedure applying three animal models with and without maternal and common environmental effects. The overall mean of cumulative lamb mortality rate was 22.95% from birth to 1 year of age, while the overall mortality rate up to 3 and from 3 to 6 months of age was 6.14% and 12.76%, respectively. The mortality rate after 6 months of age declined as the lambs grew older. The age of dam had no important effect on lamb mortality. The type of birth was more important during the preweaning period than at later ages, and lamb mortality rate was higher in twins. The year of birth, month of birth and sex of lamb significantly (p${\leq}$0.01) affected the cumulative lamb mortality rate at all ages. The least square mean of mortality during the final one-third of the lambing period was higher than the first and middle onethird of the lambing period. Male lambs were found to be at a higher risk of mortality than females. Birth weight of the lamb had a highly significant (p${\leq}$0.01) effect on lamb mortality at all ages as a quadratic regression. Direct and maternal heritability estimates of lamb mortality ranged from 0.01 to 0.13 and 0.01 to 0.05, respectively. Direct heritability increased with age of lamb, while maternal effects (genetic and common environmental) were important in the preweaning period. These results indicate that lamb mortality can be reduced first through farm management practices and secondly by genetic selection. Both animal and maternal effects should be considered in breeding programmes for reducing lamb mortality at preweaning.

Effect of Low Ambient Temperature on the Concentration of Free Radicals Related to Ascites in Broiler Chickens

  • A flock of Arbor Acres chickens were reared in cages and provided with high energy pelleted feed. At 14 d of age, a total of 350 birds were separated into 3 groups randomly as follows: 100 birds were exposed to ambient temperature of 20$^{\circ}C$ as a control group, 150 birds were exposed to lower ambient temperature of 11$^{\circ}C$ to induce ascites (group I), and another group of 100 birds were exposed to lower ambient temperature of 11$^{\circ}C$ and fed diet containing 1% L-arginine for ascitic prophylactic treatment (group II). Blood and tissue samples (lung and liver) were collected from chickens at 3, 4, 5, 6 and 7 wk of age subsequently, to analyze the concentration and activities of free radicals, mononaldehyde (MDA), superoxide dismutase (SOD), Nitric Oxide (NO) and Nitric oxide synthase (NOS). The results showed that the prevalence of ascites in the control, group I and group II was 3%, 9.33% and 3% respectively (p<0.01). The concentration of free radicals in the lungs of 3 wks old preascitic broilers in group I was significantly higher than in the corresponding control group (p<0.05). The concentrations of free radicals in lung and liver in the 7 wk period, and that of NO and SOD in the plasma were significantly lower in group I than in the control group (p<0.01). However, the accumulated MDA contents in group I were higher than in the control group and group II (p<0.05), respectively. In the same way, the activity of NOS in group II was higher than both group I and control group (p<0.01) during the 7 wk period. There was no significant difference between SOD activities of group II and the control group (p>0.05), and also insignificant difference between NOS in group I and the control group (p>0.05). The results of this study indicate that there was a significant decrease in the concentration of MDA in group II. On the other hand, the concentration of free radicals decreased and MDA concentration increased in group I during the 7 wk period. The reduction in concentration of MDA in group II, following arginine supplementation may be associated with the scavenging activity of NO.
