Strength of E. coli Floc as indicated by Shear Index (Shear Index로 표시된 E. coli Floc의 강도)

  • 침강제에 의해 형성된 E. coli floc들의 강도를 측정하기 위해 floc 의 shear index를 측정하였다. 형성된 E. coli floc은 10/sec 같이 낮은 shear rate에서도 분쇄되거나 변형되었 다. 측정된 shear index의 감소에서 보듯이 E. coli floc의 강도는 염의 농도가 증가함에 EK 라 감소하였다. E. coli floc의 shear index는 NaCl의 농도가 0에서 100 mM로 증가함에 따 라 0.47에서 0.09로 줄었다. 발효배지의 조성에서 형성된 E. coli floc들은(shear index=0.18-0.24 with BPA-1000. 0.13-0.22 with BPA-1050 and 0.37-0.42 with BPA-5020) 염이 없을 때 형성된 floc에(shear index=0.47 with BPA-1000 and 0.46 with BPA-1050) 비 해 약하였다. 따라서 발효배지에서 형성된 floc은 생물공정 중 쉽게 shear에 의해 분쇄되거 나 변형될것이다.

The Production of Biopolymer by Zoogloea ramigera (Zoogloea ramigera에 의한 생물고분자 생산에 관한 연구)

  • Zoogloea ramigera 115 was cultured for biopolymer production and its bioflocculant usages. Cultural conditions of the organism were examined with regard to high production of the microbial polysaccharide. The most suitable medium was found to contain glucose as a carbon source, $NaNO_3$ as a nitrogen source, and yeast extract as an organic nutrient. The initial pH of 6.0 proved to optimal. The biopolymer was extracted effectively using ultrasonication and high speed centrifugation, followed by propanol addition. Jar test results indicate that the polysaccharide produced by the organism is an effective flocculant.

  • A gram(+) bacteria that produced microbial flocculant was isolated from soil and classified as a Bacillus species and named as Bacillus sp. A56. The culture conditions of the strain for fluocculant production were studied in a shake flask. Optimum temperature and initial pH for flcculant production were 30C and 6.5, respectively. The optimized medium has gollowing composition: glucose 20g/l, NH4NO3 0.5g/l, K2HPO4 1.0g/l, KH2PO4 0.8g/l, MgSO4.7H2O 0.2g/l, MnSO4.4-6H2O 0.3g/l, CaCO3 0.5g/l, yeast extract 0.3g/l, tryptone 0.3g/l in tap water.

  • The consequence of water system closure and reduced water consumption in Paper Mills is increased white-water conductivity associated with increased total dissolved solids. This leads to difficulties man-aging the wet end chemistry of paper machines, mainly due to stearic hindrance effects on wet end chemical additives. This in turn causes poor productivity and Inefficient chemicals usage. The success of a number of projects is reported. The application and development of new multi-component micro-particle systems which can further assist in achieving a significant degree of system closure or Zero Effluent is described.

  • In this study, dewaterability, one of the important properties of wastewater sludge, was investigated using a simple capillary suction time (CST) measurement method. It turned out that one could use the results of CST to find optimum flocculants ratio to improve drainage in wastewater treatment for the printing paper, tissue paper, and newsprint paper mill. Since the optimum ratio of flocculants could be determined with the value of CST, COD removal efficiency could be improved and optimum floc strength could be achieved with precise ratio of flocculants. It was thus that using CST for determining the optimum ratio of flocculants could be economical by reducing the amount of flocculants. Dewaterability could be measured within several seconds using the values of CST in a precise way. The dewaterability could also be useful in investigating the optimum ratio of flocculants.

Production of High Fructose Syrup by Flocculated Actinoplanes missouriensis KCTC 1780 (Actinoplanes missouriensis KCTC 1780의 응집 균체에 의한 과당생산)

  • The whole cells of Actinoplanes missouriensis KCTC 1780 which produce glucose isomerase was immobillized by flocculation method for the effective production of high fructose syrup using packed-bed bioreactor system. Among the flocculation methods used In this study, the glucose Isomerase activity of flocculated cells using 5% polyethylenimmine and 0.2% glutaraldehyde was the highest as 46.3 unit, and the flocculant was 10.3g(wet weight) per 100m1 of broth, and the residual activity was 92.5%. In the batch operation of glucose isomerization using the flocculated cells, the optimum pH, temperature and isomerization ratio were 7.0, 75$^{\circ}C$ and 31%, respectively. The optimum concentration of Mg2+ which was activator on the glucose isomerization of flocculated cells was 0.1M, and glucose isomerase activity was increased by about 40% compared to none of Mg2+. In the packed-bed bioreactor system with 1.2 hour of residence time at 7$0^{\circ}C$, the reaction stability maintained until 96 hour without toss of activity, and the equilibrium was kept up to 120 hours of the operation.

  • Aluminium based inorganic flocculants are extensively used in this country in the removal of fine particles present in the raw water for the production of drinking water. These inorganic flocculants, however, have potential hazard of high residual aluminium ions in the treated waters, which is known to be one of the reasons of alzheimer's disease. Inorganic flocculants alone are sometimes incapable of treating water when there are excessive turbidity in the raw water sources due to flooding. A polyamine type polymeric flocculant has long been used to treat raw water in the drinking water production in the European countries and United State of America. The optimum reaction conditions such as mole ratio of epichlorohydrin(EPI) to dimethylamine(DMA), reaction temperature and time for each stage for the pilot scale preparation of polyamine from EPI-DMA was studied in this work. The variation of intrinsic viscosity and flocculating efficiency in the water treatment of the synthesized polyamines were evaluated. The residual materials after polymerization reaction were analyzed by gas chromatography to study the effect of variation of reaction conditions.

Optimization of Bioflocculant Production Conditions for Organic Wastewater Treatment with Aeromonas hydrophila KH-54 (Aeromonas hydrophila KH-54가 분비하는 유기폐수처리용 생물응집제 생산조건의 최적화)

  • For the need of bio-degradable flocculant in stage of wastewater treatment, some cultural conditions of bioflocculant production were optimized with Aeromonas hydrophila KH-54. About 260 strains of type culture and bacteria isolated from marsh, pond, activated sludge, etc were examined for their ability to flocculate kaolin particles and swine wastewater. Among them, KH-54 showed the highest flocculating activity and was identified as Aeromonas hydrophila according to the cultural, morphological and physiological properties. The maximum production of the flocculant secreted by Aeromonas hydrophila KH-54 was observed in culture medium containing 2.0% mannitol, 0.05% ammonium chloride, 0.02% potassium phosphate dibasic, 0.01% $MgSO_4{\cdot}7H_2O$ and 0.05% yeast extract at initial pH 7.0 when cultured on rotary shaker controlled at $25^{\circ}C$ and 150 rpm. Under the optimized condition, the flocculating ability reached to 770 units/ml of kaolin flocculating activity and 81% of NTU removal efficiency against swine wastewater after 4 days cultivation. The bioflocculant was also effective on various organic wastewaters other than swine wastewater, showing NTU removal rate ranging from 92% to 34%.

  • For this study, we polymerized polyacrylamide base flocculants (PAA) and tested their properties and settling efficiency as a treatment for potable water. The most common chemicals for potable water treatment in Korea are alum or PAC. However, due to various reasons (such as rainy season or algae), inorganic flocculants cannot be solely depended on to solve all the problems caused by the poor quality of inflow water. When PAA coupled with coagulants in a potable water purification process is used, the turbidity removal efficiency increases by a factor of three on a single chemical system using PAC (Raw water: 5.21 NTU; Treated PAA+PAC: 0.34 NTU; and, Treated PAC: 1.04 NTU). It is possible to offset the toxic effect of residual monomers in treated water using PAA, because the concentrations of residual acrylamide are less than 400 mg/L in the polymer itself and less than $0.04\;{\mu}g/L$ in the treated water base at a dosage of 0.1 mg/L. Therefore, PAAs may be a workable, and dependable, potable water treatment process for the high pollutant level of resource water.

Dewatering Characteristics of Sewage Sludge Produced by the Biological Treatment Process (도시하수 슬러지의 탈수특성 연구)

  • The dewatering characteristics of the sewage sludge was investigated through the experimental observations and model simulations. The activated sludge and the anaerobically digested sludge were examined for the dewaterability evaluation within the pressure range of $0{\sim}10^6N/m^2$. Modified Buchner funnel test and compression test by the consolidometer were conducted to evaluate average specific resistance, porosity, and moisture percentage of filter cake. Shirato's technique of compression-permeability test was followed for the pressure range lower than about $10^2N/m^2$. The flocculation effects on sludge dewatering was also examined for ferric chloride and polymeric flocculant. The application of hydrated lime which can be used for flue-gas desulfurization showed improved moisture percentage, and was thought to have positive feasibility in combined system of sludge dewatering and incineration. Determined characteristic constants were applied to Tiller's cake filtration model to simulate liquid pressure distribution and porosity distribution in cake. Model simulations showed a sharp drop of the porosity close to the cake-medium interface for the highly compressible material such as the activated sludge and the anaerobically digested sludge.
