  • The extract of white rose petals has an antioxidant effect and can be used to treat allergic disease. The purpose of this study was to identify optimal conditions for extracting antioxidative compounds from white rose petals with 2,2-diphenyl-1-picrylhydrazyl scavenging activities. A response surface methodology based on a central composite design was used to investigate the effects of three independent variables: ethanol concentration ($X_1$), extraction temperature ($X_2$), and extraction time ($X_3$). The estimated optimal conditions for obtaining phenolic compounds with antioxidant activities were as follows: ethanol concentration of 42% ($X_1$), extraction time of 80 min ($X_3$), and extraction temperature of $75^{\circ}C$ ($X_2$). The estimated optimal conditions for obtaining flavonoid compounds with antioxidant effects were an ethanol concentration of 41% ($X_1$), extraction time of 119 min ($X_3$), and an extraction temperature of $75^{\circ}C$ ($X_2$). Under these conditions, predicted response values for the phenolic and flavonoid contents were 243.5 mg gallic acid equivalent/g dry mass and 19.93 mg catechin equivalent (CE)/g dry mass, respectively.

  • Objectives : The objective of this research was to investigate the antioxidant activities of stem bark of Maackia amurensis extract. Methods : Stem bark of Maackia amurensis extract were prepared using 70% methanol. Methanol extracts were fractionated to hexane, chloroform, ethyl acetate, butyl alcohol, water fractions and investigated. The antioxidant activities of fractions was evaluated by four different assays as total polyphenol contents, total flavonoid contents, DPPH(1,1-diphenyl-2-picrylhydrazyl) free-radical scavenging activity and ABTS(2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) scavenging ability. Results : The yield of methanolic extracts from stem bark of Maackia amurensis was 10.16%, whereas those of its solvent fractions (hexane, chloroform, ethyl acetate, butyl alcohol, and water) were 5.45, 11.39, 13.88, 26.07, and 40.80%, respectively. The total polyphenol contents and electron donating ability of 70% methanol extracts from stem bark of Maackia amurensis were 15.44 mg/g and 194.15 μg/mL of its IC50, respectively. The 70% methanol extracts showed the highest antioxidant activity. The total polyphenol content and total flavonoid content of chloroform fractions were higher in each of 201.98 mg/g and 13.55 mg/g. The chloroform fraction showed the lowest levels of DPPH(IC50, 183.95 μg/mL) and ABTS scavenging activity(IC50, 10.0 μg/mL). The antioxidant activity was detected in methanol extract, chloroform fractions. Conclusions : These results indicate that 70% methanol extract and its fractions of stem bark of Maackia amurensis, especially chloroform fraction, have the properties of anti-oxidant suggesting stem bark of Maackia amurensis may be a candidate for natural and functional materials.

  • Chungsimyeonja-tang (CSYJT) is an herbal decoction that consists with 12 herbal medicines. CSYJT has been shown to have anti-stress, anti-allergic and anti-oxidant effects. The aim of this study was to determine flavonoid contents in CSYJT and evaluate its protective effect against cisplatin induced nephrotoxicity using both in vitro (porcine renal epithelial cell; PK15 cell) and in vivo (Sprague Dawley rat) experiments. In the present study, thee mean contents of baicalin, wogonoside and baicalein in CSYJT were 14.65, 5.27 and 0.02 mg/g, respectively. The CSYJT extract treatment attenuated the following alteration in porcine renal epithelial (PK15) cell: the increase in reactive oxygen species (ROS), the glutathione depletion and the increase in p53 expression induced by cisplatin treatment. In the in vivo study, rats were orally treated with CSYJT extract once a day for 28 days. Five days before the last treatment, cisplatin (5 mg/kg) was intraperitoneally injected to induce acute renal failure. Increased blood urea nitrogen (BUN) and creatinine (CRE) levels after cisplatin treatment were ameliorated by pretreatment of CSYJT extract. In addition, lipid peroxidation was decreased and antioxidant enzyme (glutathione) was recovered in CSYJT pretreated kidney tissue. In histopathological examination, CSYJT pretreated group showed ameliorated pathological alteration after cisplatin injection with decreased apoptosis. Taken together, pretreatment of CSYJT could ameliorate cisplatin-induced nephrotoxicity.

  • All the parts of the Cedrela sinensis A. Juss., including the seeds, roots, and leaves, have been known to exert medicinal effects. The C. sinensis and its major compound, quercetin, were previously reported to exhibit the anti-inflammatory and anti-oxidative activities. However, the hepatoprotective effects of the C. sinensis leaves against the alcohol-induced oxidative stress in the HepG2 cells have not been studied. In this study, we investigated the antioxidant activities and analyzed the flavonoid contents of the C. sinensis-leaf extract (CE). The total flavonoid contents of the CE is 1,874.5 mg/100 g dry weight (DW), while the total quercetin 3-O-rhamnoside (quercitrin) contents, which was identified as the major flavonol in the CE, is 1,456.0 mg/100 g DW. In the ethanol-stimulated HepG2 cells, the CE effectively prevented the cytotoxic effect and increased the gene expression of the antioxidant enzymes, such as the heme oxygenase-1 (HO-1) and the glutathion peroxide (GPx). The level of the reactive oxygen species (ROS) production was significantly decreased in the CE-treated HepG2 cells. In conclusion, the C. sinensis extract suppressed the alcohol-induced oxidative stress in the HepG2 cells via the induced GPx and HO-1 gene expressions. It is expected the CE positive effects will likely be attributed to the flavonoids, like the quercetin, within the CE.
