후박나무 잎 Flavonoid 성분의 분석 (Analysis of Flavonoid Components from Machilus thunbergii Leaves)

  • Six flavonoid compounds from the ethyl acetate fraction of Machilus thunbergii leaves were identified by HPLC. Separation by reversed phase chromatography on u-Bondapak C18 column was achieved by isocratic elution. The contents of the major flavonoids, guaijaverin and quercitrin were about 0.48% (w/w) and 0.98% (w/w) for the methanol extract, respectively.

Changes in Flavonoid Contents of Safflower Leaf during Growth and Processing

  • Eight flavonoids, apigenin-6-C-β-D-glucopyranosy l-8-C-β-D-glucopyranoside (AGG), quercetin 7-O-β-D­glucopyranoside (QG), luteolin 7-O-β-D-glucopyranoside (LG), quercetin 7-O-(6'-O-acetyl)-β-D-glucopyranoside (QAG), luteolin 7-O-(6'-O-acetyl)-β-D-glucopyranoside(LAG), quercetin (Q), luteolin (L) and acacetin 7-O-β­D-glucuronide (AG) were determined by HPLC in the safflower (Carthamus tinctorius L.) leaf during growth and processing. During growth, levels of five flavonoid glycosides (AGG, QG, LG, QAG, & LAG) in the leaf increased progressively at over time according to growth stages, reached a maximum before June 11, and then decreased sharply, while those of three flavonoid aglycones (Q, L, & AG) increased greatly at the early stage of growth, reached a peak before May 28, and then decreased rapidly. During the steaming process, contents of five flavonoid glycosides increased rapidly with increased steaming time, reached a maximum after 5 min of steaming, and then decreased, whereas those of flavonoid aglycones except for AG decreased sharply with increased steaming time. During the roasting process, contents of three flavonoid glycosides decreased rapidly with increased roasting time, whereas those of two acetylflavonoid glycosides (QAG & LAG) and three flavonoid aglycones increased progressively with increased roasting time, reached a maximum after 3 min of roasting, and then decreased. These results suggest that appropriate steamed and roasted safflower leaves are a rich source of flavonoids, and may be a good source of bioactive components as a functional leaf tea.

데치는 방법에 따른 시금치의 phytochemical 성분 및 위해성 요인 변화 (Changes in Phytochemical Compounds and Hazardous Factors of Spinach by Blanching Methods)

  • 끓는 물에 의한 재래적인 방법과 전자렌지를 이용하여 데치는 방법이 시금치의 phytochemical 성분 및 위해성 요인의 변화에 미치는 영향을 살펴보았다. ${\beta}$-Carotene과 chlorophylls 함량은 데치기에 의해 대조군보다 유의적으로 증가하였으며, 특히 재래적인 방법이 전자렌지에 의한 방법보다 높은 함량을 보였는데 chlorophyll a 함량은 유의적으로 증가한 반면 ${\beta}$-carotene과 chlorophyll b 함량은 단지 함량만 증가하였을 뿐 유의적인 차이는 없었다. Ascorbic acid와 총 flavonoid 및 총 polyphenol 함량은 데치기에 의해 모두 감소하였으며, 전자렌지에 의한 방법이 재래적인 방법보다 높은 함량을 보였다. 이때 전자렌지에 의해 데쳤을 경우 ascorbic acid는 대조군과 유의적인 차이 없이 감소하였으나, 총 flavonoid와 총 polyphenol은 대조군보다 유의적으로 낮은 함량을 보여 다소 영향을 받는 것으로 나타났다. 총균과 대장균군 수는 데치기에 의해 모두 유의적으로 감소하였으며 특히 끓는 물에 의한 재래적인 방법이 전자렌지에 의한 데치기보다 약 20-25% 정도 유의적으로 낮았다. Nitrate는 데치기에 의해 감소하는 경향을 나타냈으며 특히 전자렌지에 의한 방법이 재래적인 방법보다 낮은 함량을 보였다.

  • 헛개나무를 부위별 추출하여 total polyphenol과 flavonoid, Vitamin C equivalent antioxidant capacity를 연구한 결과 헛개나무의 수피와 열매에서 가장 많이 조사되었고 DPPH free radical scavenging activity에서도 수피와 열매에서 활성이 뛰어났다. 식물 추출물인 Polyphenol성 물질이 많이 존재 할 수록 항산화 활성은 뛰어났다.

산초 (Zanthoxylum schnifolium)열매 추출물의 항산화 및 $\alpha$-Glucosidase 저해 활성 (Antioxidant and $\alpha$-glucosidase Inhibition Activity from Different Extracts of Zanthoxylum schnifolium Fruits)

  • This study was performed to the activity of antioxidant and anti-diabetic from Zanthoxylum schnifolium fruit. The Zanthoxylum schnifolium fruits were extracted with water, 100% methanol, 100% ethanol, 70% methanol and 70% ethanol. The activities of each extracts were measured by antioxidant tests, such as total phenolic contents, total flavonoid contents, hydroxyl radical activity, DPPH radical scavenging activity, reducing power and $\alpha$-glucosidase inhibition activity assay. The water extract represented the highest activity in the antioxidants properties in vitro, including hydroxyl radical activity, DPPH radical scavenging activity, reducing power. Furthermore, water extract also showed significantly high total phenolic contents but the 100% methanol extract showed high total flavonoid contents estimated as $196.48{\mu}g/mg$. The $IC_{50}$ values for $\alpha$-glucosidase inhibition activity of the 100% ethanol and 100% methanol extracts were $275.66{\mu}g/mg$ and $261.44{\mu}g/mg$. Our results indicated that the Zanthoxylum schnifolium fruit expected to be useful as antioxidants.

Free radical scavenging activity and kinetic behavior of the Galgeuntang water extract

  • Galgeuntang water extract exhibited a strong free radical scavenging activity and reducing power determination. However, a gradual increase in the free radical scavenging activity and reducing power determination was obtained with increasing concentrations. The highest radical scavenging activity was shown by the water extract from Galgeuntang (116.93 ${\mu}g$/mL) and the water extract from Cinnamonum cassia Presl. (95.01 ${\mu}g$/mL). These results of phenolic and flavonoid contents of the extracts indicated that the strong radical scavenging activity of the Cinnamonum cassia Presl. extract might be in part due to the phenolic compounds. The correlation coefficient between TPC and DPPH ($r^2$ = 0.9312), TFC and DPPH ($r^2$ = 0.9677), showed positive correlation among total phenolic/flavonoid contents and antioxidant activity. These results suggest that Galgeuntang has a potential antioxidant activity.

Changes in the Antioxidant Activity of Onion (Allium cepa) Extracts with Heat Treatment

  • We evaluated the effects of heat treatment on various properties of onion extracts (Allium cepa). Onion was heated at various temperatures ($110-150^{\circ}C$) for various times (1-5 hr), and the total polyphenol, flavonoid, and free sugar contents, and antioxidant activity were investigated. With increased heating temperatures and exposure times, the total contents of polyphenols and flavonoids, as well as antioxidant activity increased. The highest total polyphenol content (189.80 mg/100g) occurred after heating for 2 hr at $140^{\circ}C$. The highest total flavonoid content (252.51 mg/100g) occurred after heating for 3 hr at $150^{\circ}C$. The antioxidant activity assessed using the DPPH method was highest, at 2.1 mg/mL ($IC_{50}$ value), after heating for 2 hr at $150^{\circ}C$. Correlations between antioxidant activity and the total polyphenol, total flavonoid, and fructose contents were highly significant (all p<0.01). The optimal heating time and temperature were 2 hr and $130^{\circ}C$.

층층갈고리둥굴레 (Polygonatum sibiricum Redoute) 뿌리줄기와 잎의 증숙 시간과 증숙 온도에 따른 생리활성 변이 연구 (Changes of Biological Activities of Rhizome and Leaves of Polygonatum sibiricum Redoute according to Steaming Time and Temperature)

  • Background: The aim of this study was to analyze the total phenol and total flavonoid contents and antioxidant activity of steam-treated leaves and rhizomes of Polygonatum sibiricum Redoute. In addition, we aimed to confirm their potential use as cosmetic materials by investigating their anti-aging and skin-whitening activity. Methods and Results: The leaves and rhizomes of P. sibiricum were treated with steam at different temperatures for different durations, and the antioxidant activity (DPPH and ABTS radical scavenging activity) and total phenol and total flavonoid contents of each sample were tested. The steam temperature and treatment duration siginificantly affected the antioxidant activity and, total phenol and total flavonoid content of the leaves and rhizome of P. sibiricum. Treating the P. sibiricum samples with steam at 120℃ for 12 h, yielded higher total phenol and total flavonoid contents. Comparatively, the samples treated with steam at 120℃ for 12 to 24 h showed significantly higher antioxidant activity. Further, the steamed samples of P. sibiricum demonstrated collagenase and tyrosinase inhibition activity, which indicated their anti-aging and skin-whitening properties. The samples steamed at 120℃ for 12 h, exhibited higher collagenase and tyrosinase inhibition activity. Conclusions: Leaves and rhizomes of P. sibiricum steamed at 120℃ for 12 h, showed highest antioxidant activity and, total phenol and total flavonoid contents than all other samples. Our results indicate the potential of using P. sibiricum as a cosmetic material by confirming its excellent anti-aging and whitening activity.

메밀순의 증숙 및 건조에 따른 침출차 특성 모니터링 (Monitoring on the Tea with Steaming and Drying Process of Germinated Buckwheat)

  • 메밀순을 이용한 침출차를 제조하고자, 증숙시간과 건조온도를 달리 처리하여 가용성 고형분 함량, 총 플라보노이드 함량 및 관능적 특성을 살펴보았다. 메밀순차 침출액의 가용성 고형분 함량은 증숙시간 6.93 min 및 건조온도 73.59$^{\circ}C$에서 최대값으로 나타났으며, 총 플라보노이드 함량은 증숙시간 5.22 min 및 건조온도 79.05$^{\circ}C$에서 높은 함량을 나타내는 것으로 나타났다. 메밀순 침출차의 전반적 기호도에 대한 높은 관능평점은 증숙시간 6.00 min 및 건조온도 77.33$^{\circ}C$인 것으로 나타났다. 가용성 고형분 및 총 플라보노이드 함량이 높고 전반적인 기호도도 우수한 조건은 증숙시간 5.4∼7.0 min 및 건조온도 75∼8$0^{\circ}C$로 예측되었으며 최적 조건에서의 예측된 관능평점은 최적 조건에서 실제 실험한 결과와 유사하였다.

[Retracted] Optimization of Jirisan Mountain Cudrania tricuspidata leaf substance extraction across solvents and temperatures

  • Objective: The aim of this study is to optimize the extraction of beneficial substance from Cudrania tricuspidata leaves grown at Jirisan Mountain in South Korea by three different solvents depending on extraction time and at different temperature. Methods: The total phenolic contents were determined by the method reported by $S{\acute{a}}nchez$-Moreno et al. The total flavonoid contents were analyzed by Slinkard and Singleton. The DPPH radical scavenging activity was determined according to the method reported by Blois Results: The extraction yield for each solvent is 9.05-14.1%, 2.17-5.67%, and 2.3-3.9% for D.W., ethanol, and hexane, respectively. The overall results were maximized for the extract obtained with D.W. for 5 min at $100^{\circ}C$. The average phenol contents were 77.11, 45.64, and 0.343 mg/g at $100^{\circ}C$ in water, $78^{\circ}C$ in ethanol, and $68^{\circ}C$ in hexane, respectively. The flavonoid contents were the highest in the materials extracted with D.W., and were increased with increasing temperature, regardless of the extraction solvents, whether water (green), polar organic ethanol, or nonpolar organic hexane. In the ethanol extract, the flavonoid contents are increased gradually from 5.66 mg/g to 7.73 mg/g. The total flavonoid contents were proportional to the concentrations of the water extracts, ranging from 4.14 mg/g to 48.89 mg/g. The antioxidative activities of the water-extracted compounds are generally increased with increasing temperature from 42.5% to 85.5%. Those of the hexane extracts are increased slowly from 3.79% to 8.8%, while those of ethanol extracts are increased from 29.8% to 47.4%. Conclusion: The extraction yields were dependent upon solvents for extraction as well as extraction time and the temperature. The optimal extraction time was 5 min and the extraction yields were increased with increasing temperature excepted hexane. Of the three tested extraction solvents, the greenest solvent of water shows excellent results, suggesting that water is among the most effective solvents for natural sample extractions for general medicinal, pharmaceutical, and food applications.
