The Comparison of Food Constituent in Different Parts of Pumpkin (늙은 호박의 부위별 성분 비교)

  • This study was conducted to investigate the constituents of pumpkin, to develop of health food using pumpkin. Crude protein contents of skin (19.82%) and fiber (16.43%) were higher than that of flesh. Crude lipids contents of skin (2.84% ) was higher than those of flesh and fiber. Major free sugars in pumpkin were sucrose, fructose and glucose. Total sugar contents in the flesh was higher than that of fiber. This tendency was opposite to reducing sugar contents. Also, free sugar contents in pumpkin were flesh (30.19 mg/100 g), fiber (22.99 mg/100 g) and skin (15.67 mg/100 g), respectively. The total Phenol and carotenoid contents in 100 g of fiber revealed 379.8$\pm$9.76 mg and 43.71$\pm$1.46 mg, corresponding to 47% and 62% of the total phenol and carotenoid contents of pumpkin. Total flavonoid content in 100 g of skin revealed 81.5$\pm$3.56 mg.

Comparison of Antioxidant and Functional Compounds in Korean Conventional and Chinese Seed Ginger (Zingiber officinale Roscoe) Following Steam Treatment (증숙처리에 따른 재래생강 및 중국종자생강의 항산화 및 기능성물질 비교)

  • The aim of this study was to compare the antioxidant activities and functional contents of Korean conventional and Chinese seed gingers from the Jeollabuk-do Wanju and Chungcheongnam-do Seosan regions. Ginger samples were subjected to steaming treatments for different durations (2-8 h) at 121℃ using an autoclave. The antioxidant activity was evaluated by measuring total polyphenol and flavonoid contents and ABTS and DPPH radical scavenging activities, while functional ingredient contents were analyzed for gingerols and shogaols. The results showed that Wanju conventional seed ginger (WO-2) had the highest total polyphenol (85.24 mg GAE/g) and flavonoid (98.14 RE/100 g) contents, surpassing that of the control in all steamed groups at 6 h. ABTS radical scavenging activity showed a strong correlation with total polyphenol and flavonoid contents. The control groups indicated that Korean conventional seed ginger had 1.0-1.3 times higher gingerol contents compared to Chinese seed ginger. Furthermore, the content of shogaols, considered major functional ingredients, increased significantly with longer steaming durations, reaching the highest content (1,793 mg/kg) at 8 h, which was 1.0-1.8 times higher in Korean conventional seed ginger than that in Chinese seed ginger. These experiments provide valuable data supporting the excellence of Korean conventional seed ginger in the future.

  • An investigation was carried out to study the effects of extraction, heat treatment, and storage temperature on functional ingredients of onion. Extracts of onion paste and freeze dried onion were prepared by treatments with several solvents including hot water, methanol, and ethanol, and then contents of functional ingredients such as total phenol, flavonoid, quercetin, and thiosulfinate were determined. The highest levels of functional ingredients were observed in samples extracted using water at 100$^{\circ}C$ or ethanol. The skin extract contained 60-fold more quercetin than onion paste extract, whereas no thiosulfinate was detected in onion skin extract. Heat treatment of onion extracts at 60, 80, 105, and 121$^{\circ}C$ did not effect on the levels of all functional ingredients tested. During four-week storage, the levels of total phenol, flavonoid, and quercetin were not significantly changed regardless of storage temperature. However, the total thiosulfinate was rapidly reduced as the storage temperature increased.

General Characteristics of Korean Propolis

  • The propolis was collected from the whole part of Korea Peninsula, such as central, southern, and Jeju island, to analyze the general composition including total flavonoid and phenolic contents, heavy metals, colors, amino acids, and crude lipid. The total flavonoid and phenolic contents of middle region in Korea Peninsula (4.26% and 13.06 g/100 g gallic acid) were higher than southern region (2.53%, 10.9 g/100 g gallic acid) and Jeju island (0.03%, 7.22g/100 g gallic acid), respectively. The heavy metals contents showed that Zn contents were found in $0.44{\pm}0.2ppm$ and a harmful heavy metal such as Cr, Cd, Pb, Ni, Cu, As, and Hg were not detected at all in our experiment. The colors of extracted propolis showed a various spectrum from yellow to red purple. Various amino acids were also detected as proline, valine, methionine and the average crude lipid contents of propolis were 42.4%. All data collected in South Korea were very first executed in our institution for the purposes of utilizing and developing the industrial insects, honeybees as well as silkworm.

Antioxidant activity and anti-inflammatory activity of ethanol extract and fractions of Doenjang in LPS-stimulated RAW 264.7 macrophages

  • BACKGROUND/OBJECTIVES: Fermentation can increase functional compounds in fermented soybean products, thereby improving antioxidant and/or anti-inflammatory activities. We investigated the changes in the contents of phenolics and isoflavones, antioxidant activity and anti-inflammatory activity of Doenjang during fermentation and aging. MATERIALS/METHODS: Doenjang was made by inoculating Aspergillus oryzae and Bacillus licheniformis in soybeans, fermenting and aging for 1, 3, 6, 8, and 12 months (D1, D3, D6, D8, and D12). Doenjang was extracted using ethanol, and sequentially fractioned by hexane, dichloromethane (DM), ethylacetate (EA), n-butanol, and water. The contents of total phenolics, flavonoids and isoflavones, 2,2-diphenyl-1 picryl hydrazyl (DPPH) radical scavenging activity, and ferric reducing antioxidant power (FRAP) were measured. Anti-inflammatory effects in terms of nitric oxide (NO), prostaglandin (PG) E2 and pro-inflammatory cytokine production and inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 expressions were also measured using LPS-treated RAW 264.7 macrophages. RESULTS: Total phenolic and flavonoid contents showed a gradual increase during fermentation and 6 months of aging and were sustained thereafter. DPPH radical scavenging activity and FRAP were increased by fermentation. FRAP was further increased by aging, but DPPH radical scavenging activity was not. Total isoflavone and glycoside contents decreased during fermentation and the aging process, while aglycone content and its proportion increased up to 3 or 6 months of aging and then showed a slow decrease. DM and EA fractions of Doenjang showed much higher total phenolic and flavonoid contents, and DPPH radical scavenging activity than the others. At $100{\mu}g/mL$, DM and EA fractions of D12 showed strongly suppressed NO production to 55.6% and 52.5% of control, respectively, and PGE2 production to 25.0% and 28.3% of control with inhibition of iNOS or COX-2 protein expression in macrophages. CONCLUSIONS: Twelve month-aged Doenjang has potent antioxidant and anti-inflammatory activities with high levels of phenolics and isoflavone aglycones, and can be used as a beneficial food for human health.

Antioxidant and QR Inductive Activities of Novel Functional Soybean 'Agakong3'

  • In order to evaluate the bioactivity of 'Agakong3', which was newly bred, quinone reductase (QR) inductive activity and antioxidant activity were both assessed. The methanol extract of 'Agakong3' showed a significantly stronger QR inductive activity than other soybeans. The methanol extract of 'Agakong3' also showed a significantly stronger cytotoxicity on hepa1c1c7 than other soybeans. 'Agakong3' exhibited the most potent antioxidant activity in the Trolox equivalent antioxidant capacity (TEAC) assay whereas it showed significantly weak antioxidant in the DPPH assay. In total phenol and flavonoid contents, 'Agakong3' showed the highest contents regarding phenol and flavonoid compounds. Major isoflavones such as daidzein and genistein were quantitified by high performance liquid chromatography. 'Agakong3' also showed the highest total isoflavone contents. Results of correlation analysis showed that there were high correlation coefficients between the contents of isoflavone and TEAC and the contents of isoflavone and QR inductive activity, respectively. These results suggest that 'Agakong3' will be a promising and functional food material.

Studies on the Antioxidative Activities and Active Components of the Extracts from Pleurotus ostreatus (느타리버섯 추출물의 항산화 활성과 그 성분에 대한 연구)

  • Antioxidative components and activities of the extracts from Pleurotus ostreatus extracted at different ethanol concentration were analyzed and their correlation were investigated. Ergothioneine, total phenolic compounds, and flavonoid contents of the extracts from P. ostreatus extracted with hot water (0% ethanol) were the highest ($2.98{\pm}0.05$, $9.51{\pm}0.45$, and $2.83{\pm}0.03mg/g$, respectively) and the contents were decreased according to increase of ethanol concentration for extraction. DPPH and ABTS radical scavenging activities of the extracts from P. ostreatus extracted with hot water were the highest ($80.41{\pm}0.56$ and $91.47{\pm}0.11%$, respectively). FRAP value also showed the highest reducing power by $8.86{\pm}0.33\;FeSO_4\;eq$. mM in hot water extracts. Positive correlations were found between ergothioneine contents and antioxidative active components and antioxidant activity of the extracts from P. ostreatus. Results indicate that hot water extraction was most efficient for the extracts with high antioxidative activities from P. ostreatus.

Antioxidant Activities and Monacolin K Production of Fermented Chinese Yam by Monascus sp. MK805 (Monascus sp. MK805 균주를 이용한 발효마의 Monacolin K 생산과 항산화 활성)

  • This study was carried out to investigate the characteristics of fermented chinese yam and rice (Dioscorea japonica, Dioscorea batatas, Dioscorea opposita, Rice, FCYR) using Monascus sp. MK805. The extracts from FCYR were measured to examine pigments, antioxidant activities were investigated through DPPH radical scavenging activity, total polyphenol and flavonoid contents, reducing power. Also it was investigated monacolin K productivity by FCYR. Pigments productivity (yellow, orange and red) were 26.2, 13.9, 17.3 at Dioscorea japonica, 41.9, 22.6, 53.2 at Dioscorea batatas, 12.5, 7.5, 9.7 at Dioscorea opposita and 10.1, 7.7, 10.2 at rice, respectively. DPPH radical scavenging activity of FCYR was about 69.7, 79.6, 57.8, and 42.3%, total polyphenol contents of FCYR was about 480.6, 658.7, 379.3, and 212.9 mg/kg, total flavonoid contents of FCYR was about 342.5, 448.4, 235.2, and 168.7 mg/kg, reducing power of FCYR was about 1.57, 2.14, 1.14, and 0.35 (OD at 700 nm), respectively. And then monacolin K productivity of FCYR was about 453.8, 509.5, 332.2, and 263.2 mg/kg, respectively.

Effects of Hot Air Drying on the Antioxidant Capacity of Actinidia arguta × A. deliciosa cv. Mansoo, a Hardy Kiwifruit (열풍 건조 가공 공정이 다래 과실(품종명 만수)의 산화방지능에 미치는 영향)

  • A hardy kiwifruit is a good source of phenolics and antioxidants. This study was aimed at investigating the effects of hot air drying at three different temperatures (35, 50, and $70^{\circ}C$) for 24 h on the total phenolic and flavonoid contents and the antioxidant capacity of hardy kiwifruits of Actinidia arguta ${\times}$ A. deliciosa cv. Mansoo. Dried kiwifruit extracts were produced using 80% (v/v) aqueous ethanol under homogenization. Hot air drying lowered the total phenolic and flavonoid contents and decreased the antioxidant capacity in the dried kiwifruits compared with their fresh counterparts. The results also revealed high positive linear correlations of antioxidant capacity with total phenolic and flavonoid contents, indicating that phenolics are the major contributors to antioxidant capacity. Our results suggested that an optimal drying process must be determined for industrial application in order to obtain dried kiwifruits with increased antioxidant capacity for consumers.

Evaluation of Individual Glucosinolates, Phytochemical Contents, and Antioxidant Activities under Various Red to Far-Red Light Ratios in Three Brassica Sprouts (적색/원적색광 조사 비율에 따른 3종 배추과 채소 새싹의 Glucosinolate 함량 및 항산화 기능성 평가)

  • The aim of this study was to evaluate the individual glucosinolate (GSL), total phenol, total flavonoid, and vitamin C content, and antioxidant activity under various light quality condition, mainly focusing on red (R) to far-red (FR) light ratios in three Brassica sprouts (radish, Chinese cabbage, and broccoli). Three R/FR ratio of 0.6, 1.3, and 2.0 were exposed to 5-day old sprouts for 48 h in a controlled environment, and the targeted phytochemical contents and antioxidant activities were compared with three separate control plot of dark, fluorescent, and red:blue 8:2 conditions. Total GSL content was highest in broccoli among the cultivars throughout the respective treatments, and increased with the increasing of R/FR ratio in the broccoli sprouts, while the content showed non-significant results in the Chinese cabbage sprouts. The progoitrin, a major GSL in Chinese Cabbage and broccoli, content decreased by upto 38% and 69%, respectively, with decreasing the R/FR ratio compared to the control plots (fluorescent, red:blue 8:2, and dark condition). The contents of phenol, flavonoid, and vitamin C were lowest in dark condition in all the three Brassica sprouts. The total phenol content and antioxidant activities increased with decreasing the R/FR ratio in all the Brassica sprouts, while total flavonoid and vitamin C content showed different patterns depending upon the Brassica sprouts. These results suggest that additional use of FR is expected to improve the functional quality of Brassica sprouts in different ways.
