In this study, whether Giemsa staining solution can accurately determine bacterial contamination of liquid spawn for Flammulina velutipes in a short period of time was investigated. Giemsa solution staining cells of blood, bone marrow, lymph node, malaria parasites, rickettsia et al. was prepared by dissolving basic methylene azul and methylene blue, and acidic eosine in methyl alcohol-glycerine. Supernatant samples of Flammulina velutipes liquid spawn cultured under explosive aeration were placed on a slide, mixed with Gimesa solution and examined with optical microscope after staining. In 40 to 60 seconds bacterial cells were distinguishable from soybean meal residual and hyphal cell fragments. Thus we conclude that microscopy using Gimesa staining solution is a quick, simple and accurate method for the mushroom growers to effectively use to detect bacterial contamination of the liquid spawn.
The experiments were conducted to provide information on the chemical concentrations and cultural characteristics in the periods of hyphal incubation, primordial formation, and fruiting bodies yield of winter mushroom, Flammulina velutipes at the mixture ratio of the raw materials. Substrates were analyzed for pH, total carbon (T-C), total nitrogen (T-N), and C/N ratio. In case of Flammulina velutipes, yield of fruiting bodies were 190.5 g/850 ml in the substrates, [Corn-cob + Rice bran + Soybean curd residue (75:20:5)], which was increased 20.4% more than the control, [Douglas fir sawdust + Rice bran (75:25)]. But the periods of hyphal incubation took 28 days, which was 7 days longer than the control. Also, in the substrates [Douglas fir sawdust + Rice bran + Soybean curd residue (75:20:5)], the yields was 172.7 g/850 ml and the periods of hyphal incubation was 21 days.

To provide functional properties in fish meat paste, enoki mushroom (Flammulina velutipes) having anticarcinogenic effect and reduction of blood pressure was added at 1, 5, 10, 15, and 20%. The fried fish meat pastes were investigated for color, rheological properties, and sensory evaluation. Increasing the amount of mushroom in the fish meat paste tended to decrease the lightness (L) in Hunter color value, but to increase the redness (a) and yellowness (b). All test samples with 3mm thickness had good flexibility and did not break even after 4 times folds. The sample containing 15% mushroom showed the highest values in strength, gumminess and brittleness. In sensory evaluation, the higher amounts of mushroom obtained favorite score in color and flavour and 5% mushroom sample had the best score in overall acceptance. Therefore, this results suggest that enoki mushroom can be applied to fish meat paste for the purpose of high quality and functionality.
This study were carried out to analyze nutritional ingredients of pre-inoculation and post-harvest Flammulina velutipes media. pH, moisture content, total carbon(T-C), total nitrogen(T-N), crude protein, crude fiber, and crude fat content on the 8 raw materials using Flammulina velutipes bottle cultivation medium, and after sterilization(pre-inoculation) and post-harvest medium was analyzed. This result is expected to be utilized as the basis for recycling post-harvest media.

The effects of addition of dried apple pomace to sawdust substrate on the growth of Flammulina velutipes were investigated during the period of 2 years $(1994{\sim}1995)$. Dried apple pomace used in this study consisted of 44.6% C, 0.54% N, 0.16% CaO, 0.16% $P_2O_5$, 0.11% MgO and 1.29% $K_2O$ (pH 4.4). The addition of 15% apple pomace (v/v) increased the yield of the mushroom fruitbody by 9%, and period of primordia formation was similar regardless of the treatments. In economical analysis, the addition of 15% apple pomace (v/v) increased 29% compared to the control treatment in relative income.
Agrobacterium-mediated transformation was conducted in order to generate DNA insertional mutants of Flammulina velutipes. Agrobacterium tumefaciens AGL-1 harboring pBGgHg was transformed into gill tissues of Flammulina velutipes strain KACC42777. The transformants resistant on hygromycine ($30\;{\mu}g/ml$) were confirmed by PCR. The targeted insertional sites were amplified by inverse PCR and sequenced. To find the phenotype variation of all generated transformants, bottle cultivation which followed by the standard cultivation protocol were conducted. Color variation was observed on the cultivated fruiting bodies. Furthermore, the transformant pool will be used as a good genetic resources for studying gene function.
Nutritional requirements for the growth of Flammulina velutipes were studied. Mannitol, glutamic acid and ammonium nitrate were chosen for the maximum mycelial growth when various carbon and nitrogen sources tested. Optimum C : N ratio for the mycelial growth was 20 : 1. Potassium dihydrogen phosphate was selected among the phosphate sources. Magnesium sulphate and thiamine HCl stimulate mycelial growth. Final compositions of optimized chemically defined medium were 1.5% mannitol, 0.082% $NH_4NO_3$, 0.312% glutamic acid, 0.25% $KH_2PO_4$, 0.06% $MgSO_4{\cdot}7H_2O\;and\;0.3\;{\mu}g/l$ thiamin HCl. This medium not only support mycelial growth but also induce fruit body formation.

Various studies on the effect of mushroom, a member of phytonutrients, on inflammatory diseases have been reporter over the years, Among various species mushrooms, King oyster mushroom (Pleurotus eryngii) and winter mushroom (Flammulina velutipes) are popular dietary ingredients for Asian cuisine. The extracts from all the three mushrooms were used to determine the antioxidative effect and the cytotoxicity. Result analysis were repeated more than three times to get an average${\pm}$standard deviation, and statistical significance were confirmed by SPSS. As a result, total phenol content of the king oyster mushroom and the winter mushroom were $19.66{\pm}0.10mg/g$ and $22.08{\pm}1.10mg/g$, respectively. Also, the total flavonoid content was $15.21{\pm}1.31mg/g$ for the king oyster mushroom and $20.50{\pm}4.52mg/g$ for winter mushroom. The results in winter mushroom showed higher values in total phenol and flavonoid content than in the king oyster mushroom. All samples of extracts showed free radical DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging activity with their 10 mg/mL concentration. These results indicate that the extract of the king oyster mushroom (Pleurotus eryngii) and the winter mushroom (Flammulina velutipes) may be possible phytonutrients with the anti-inflammatory and the anti-cancer effect.
This study was conducted to determine the proximate compositions, nutritional components, and antioxidant effects of white and brown enoki mushrooms (Flammulina velutipes). The crude protein and carbohydrate contents were higher in the brown than white mushrooms, whereas the moisture, crude ash, crude lipid, and dietary fiber levels were lower. The mineral contents of the white mushroom was higher than levels obtained in the brown mushroom for the detected components (Ca, Cu, K, Mn, Na, and P). The amount of vitamin B3 in the brown mushroom was 1.51 mg/100 g, which was 4.5 times higher than that in the white mushroom. The major fatty acids detected were palmitic acid, linoleic acid, and α-linolenic acid. The total polyphenol and flavonoid contents were highest in 70% ethanol extracts of the white and brown mushrooms, respectively. For the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, a 70% methanol extract of the white enoki mushrooms showed an activity of 76.4% (p<0.05). For the ferric-reducing antioxidant power (FRAP) activity, a 70% methanol extract of the brown enoki mushrooms showed the highest value. Further, the total flavonoid contents were significantly correlated with the DPPH and FRAP activities.
Agricultural and forest wastes were tested as substrate for the production of Flammulina velutipes. Among the meranti, pine tree, mixed sawdust, coffee waste and peanut hull tested, coffee waste was the best basal substrate for the production of basidiocarps. When various supplements, such as ricebran, brewer's grain, defatted corn flour, defatted soybean flour and defatted rapeseed flour were tested, defatted corn flour was chosen for the production of basidiocarps. Maximum yield of basidiocarps(129.38 g/bottle) was obtained from the mixed medium at 4 : 1 ratio of coffee waste and defatted corn flour.
