Metamorphic Changes of the Neurosecretory Cells in the Brains of Wax Moth Gafferfa mellonella during Metamorphosis (꿀벌부채명나방 뇌신경분비세포의 변태에 따른 변화)

  • The paraidehyde-fuchsin(PAE)-positive neurosecretory cells in the brains of wax moth Galleria mellonella have been morphologically examined at the six different metamorphic stages. During the metamorphosis, neurosecretory cells in the brain can be found in the five unclei of pars intercerebralis, lateral region of protocerebrum, optic lobe, deutocrebrum and tritocerebrum. The five nuelel include one to seven neurosecretory cells. On the bases of cell sizes and histochemical specificities of neurosercretion within cells, all the PAF-positive neurosercretory cells included in the six different metamorphic brains can be recognized as four species of neurosecretory cells as follows; (1) large (about 25 $\mu$m), neurosecretion-parcked cell (type I neurosecretory cell), (2) large, granule-dispersed cell (type II neurosecretury cell), (3) small (about 15 $\mu$m), neurosecretion-packed cell (type III neurosecretory cell), and (4) small, granule-dispersed cell (type IV neurose-cretory cell). The three tb seven medial neurosecretory cells are included in the pars intercerebralis of the six different metamorphic brains. With the increase of days from the late larva to the adult the type I cells of medial neurosecretory cells gradually decrease in number, but the respective three type II neurosecretory cells appear in the five different metamorphic brains except in pupa 2 day before the emeregnce of the adult which has only one type II. The one to five lateral neurosecretory cells are observed in the lateral region of protocerebrum from thepupa just after pupation to the adult. The type IV neurosecretory cells are the most in number of lateral neurosecretory cells. The one type Ineurosecretory cells are included near the optic lobe of only the 4-day-old pupa. the one deutocerebral neurosecretory cell, type II, appears only in the adult. The tritocerebrum includes both three neurosecretory cells in the late larva and one neuresecretory cell in the adult. In the late larva the two tritocerebral neurosecretory cells are type Ill neurosecretory cell and the one is type IV. The remaining one tritocerebral neurosecretory cell is type IV.

Cytologic Analysis of Adenocarcinoma of the Cervicovaginal Smears (자궁경부질 도말상 선암종의 세포학적 분석)

  • We have investigated the cervicovaginal smears in order to define the cytologic features of uterine adenocarcinomas. Total 22 cases were reviewed(12 cases from the Sanggye Paik hospital, Inje university and 10 cases from the Korea Cancer Center Hospital) from January 1992 to December 1997. Five cases were endometrial adenocarcinomas and the remaining 17 cases were cervical adenocarcinomas. Seventeen cases of cervical adenocarcinomas were divided into endometrioid(6 cases), endocervical(7 cases), mixed endometrioid and endocervical(1 case), papillary (2 cases), and adenosquamous(1 case) carcinomas. The background of endocervical adenocarcinoma was hemorrhagic or inflammatory. The tumor diathesis was less prominent than that of the squamous cell carcinoma. The prominent features of the endocervical type adenocarcinomas were large and loose clusters, large intracytoplasmic vacuoles, and prominent overlapping and peripheral palisading of nuclei. In contrast, the endometrioid adenocarcinomas showed small and compact clusters, and small intracytoplasmic vacuoles. The detection rate of endometrial adenocarcinoma was lower than that of the endocervical adenocarcinoma.

Fine Needle Aspiration Cytology of the Sclerosing Hemangioma of the Lung - A Report of Five Cases - (폐의 경화성 혈관종의 세침흡인 세포학적 소견 - 5예 보고 -)

  • Sclerosing hemangioma is a rare, benign neoplasm of the lung, usually presented as a solitary pulmonary nodule in an otherwise asymptomatic middle-aged woman. Cytologically it shows papillary, sheet, and cyst-like arrangements representing three main histologic patterns of papillary, solid, and angiomatous ones, respectively. Herein, we report the fine needle aspiration cytology of 5 cases of sclerosing hemangioma of the lung. The most characteristic finding is cyst-like spaces Intimately related to the papillary or solid cell nests. The tumor cells are relatively monotonous round to oval, small to medium in size. They have small amount of eosinophilic cytoplasm. The nuclei are uniform, round and have small but conspicuous nucleoli. The tumor cells in one of the presenting cases are large with abundant cytoplasm and show moderate nuclear pleomorphism. The nuclear chromatin, however, is fine and even without exception, even in the case showing nuclear pleomorphism. Major differential diagnoses based on the cytologic findings are well-differentiated papillary adenocarcinoma, bronchioloalveolar carcinoma, and carcinoid tumor.

Photometric Reverberation Mapping of Active Galactic Nuclei with Medium-band Filters and a Large FOV Telescope.

  • We present a noble method to determine BH mass of many AGNs directly through reverberation mapping using a small telescope with wide-field of view. In 2017 August we installed five medium-band filters to a 0.25m diameter $5deg^2$ FOV telescope at the McDonald observatory. The width of these filters (FWHM ~ 50nm) are matched to the broad line width of type-1 AGNs at various redshifts. From recently obtained data, about r ~ 19 magnitude AGNs can be detected in line component with 150s exposure. With this magnitude limit, about 20~30 AGNs can be studied in one field. We plan to carry out at one day cadence observation over 20~30 fields, enabling us to monitor up to ~1000 AGNs over a wide range of variability. This poster presents out plan and early results from test observation.

Pathological findings and virus detection by in situ hybridization in the Korean native goats experimentally infected with Aujeszky's disease virus (오제스키병바이러스 인공감염 한국재래산양의 병리학적 소견 및 절편내 in situ hybridization 바이러스 동정)

  • Aujeszky's disease virus(ADV) was inoculated intranasally into the Korean native goats to investigate pathological findings and pathogenesis of ADV infection by using of histological and immunohistochemical methods and in situ hybridization(ISH). Clinical signs of salvation, pyrexia, pruritus and staggering were followed by death with five days after inoculation, Pathoanatomical findings were edema of the lung and the urinary bladder with hemorrhage and congestion, petechial hemorrhages on the endo-and epicardium, renal congestion, moderate splenomegaly and cystic edema. Main microsocpic lesions observed in all infected goats were confined to the CNS and charcterized by perivascular cuffing with lymphocytes and macrophages, focal gliosis, neuronal degeneration and necrosis, and intranuclear inclusion bodies in the neurons and glial cells. Positive reactions to ADV were detected more frequently in the nuclei than in the cytoplasms of infected nerve cells in the CNS by immunohistochemistry and ISH. Frequenctly localized sites of ADV in the CNS were olfactory bulb, prietal cortex, callosal sulcus and corpus callosum. Positive reactions were also detected in the tonsillar epithelium, and alveolar macrophage and sloughed epithelium of the lung.

  • Structures of the adventitious root meristem induced from callus culture of tobaco (Nicotiana tabacum cv. NC 82) leaves were investigated by light and transmission electron microscopy. Structural differences between in vitro root and callus cells were also examined by the microscopy. The submicroscopic features of the in vitro root cells were as follows. Intercellular spaces were not developed and nuclei with two nucleoli were observed occasionally. Plasmodesmate were found in groups or sing1y on transverse and longitudinal walls. Amyloplast solely filled with starch grains, with one to five electron - dense bands, was surrounded by single membrane. in the callus cells, vacuolization of central part in the cytoplasm having mitochondria with swollen cristae and starch grains like those of in vitro root cells was a distinct feature. Vesicles which were found between cell wall and plasma membrane may be arisen by a process of protoplasmic invagination. By comparing of ultrastructures between the cells of callus and in vitro roots we found that the distinct differences lied on thickened cell walls and hypertrophed vacuoles in the former, and less thickened cell walls and several small vacuoles in the later.

Reverberation Mapping of Nearby AGN with Medium-band and LSGT

  • Reverberation mapping is one of the best ways to investigate the physical mechanism of broad-line regions around central supermassive black holes of active galactic nuclei (AGNs). It is usually used to estimate the masses of supermassive black holes. Although spectroscopic reverberation mapping has used to study dozens of AGN, spectroscopic monitoring campaign of large sample is expansive. Here, we present results of photometric reverberation mapping with medium-band photometry. We monitored five nearby AGN which were already studied with H-alpha emission line. Observation has been performed for ~3 months with ~3 days cadence using three medium-band filters installed in LSGT (Lee Sang Gak Telescope; 0.43m). We found 0.01-0.08 magnitude variations from differential photometry. Also time-lags between continuum light-curves and H-alpha emission line light-curves are found using JAVELIN software. The result shows that our study and previous studies are consistent within uncertainty range. In the near future, medium-band photometric reverberation mapping seems useful to study large AGN samples. We will present preliminary result of following study that report new time lag measurement of six AGNs in the similar way.

  • Five subcellular fractions were obtained by successive centrifugation from the liver of rats within 6 hours of life and characterized by comparing marker compound or marker enzyms. After incubating $3{\beta}$-hydroxy-$5{\alpha}$-pregnan-20-one with the each fraction, the steroids were analyzed by TLC, GLC and GC-MS. A $6{\alpha}$-hydroxylase which hydroxylizes the tetra-hydrogenated compound of progesterone, $3{\beta}$-hydroxy-$5{\alpha}$-pregnan-20-one, was localized in the crude plasma membrane fraction, but not in the microsome fraction. The maximum 6α-hydroxylation was observed at pH 7.0. While this 6α-steroid hydroxylase was not able to hydroxlyze the progesterone, the $3{\alpha}$-isomer was hydroxylized at the $6{\alpha}$-position.

Effects of Herbal Bath on Acetic Acid-induced Somato-visceral Pain in Mice (현호색, 창출, 천수근 약욕이 체성내장통에 미치는 영향)

  • As an effective non-pharmacological method of pain relief, hydrotherapy was widely used. And bath additive has been used for enhancing the efficacy of hydrotherapy, In the present study, as a bath additive, the analgesic activity of HAC, which composed of Corydalis turtschaninovii, Atractylodes japonica, and Harpagophytum procumbens(HAC), was investigated in the ventrolateral periaqueductal gray (VIPAG), lateral PAG (IPAG), central nuclei of amygdala (CeA), and the paraventricular nucleus of the hypothalamus (PVN) in mice, using writhing test and immunohistochemistry for c-Fos. Male C57BU6 mice weighing $25{\;}{\pm}{\;}2g$ (8 weeks of age) were used for this experiment. The animals were divided into five groups: the control group, the acetic acid treatment group, the acetic acid treatment and 0.01 g/L HAC-immersed group, the acetic acid treatment and 0.1 g/L HAC-immersed group, and the acetic acid treatment and 1.0 g/L HAC-immersed group. To induce somato-visceral pain in the experimental animals, a single intraperitoneal (i.p.) injection of acetic acid was administrated to each animal, and the animals of the control group received injections of equivalent doses of normal saline. The animals of the HAC-immersed groups were immersed the water with HAC powder at the respective doses deep enough to cover the mice body, and those of the control group and the acetic acid treatment group immersed the water without HAC powder at 10 min immediately after the acetic acid injection. Our present study has shown that the HAC reduced the acetic acid-induced abdominal constrictions and the acetic acid-Induced increase of numbers of c-Fos-positive cells in the VIPAG, IPAG, PVN, and CeA. The most potent analgesic effect appeared with the treatment of 1.0 g/L KB-immersed group. Based on our present results, it is very possible that HAC can be a potent therapeutic bath additive for alleviating pain without the fear of addiction to the drugs and side-effects associated with the prescription of multiple analgesic drugs.

Intergeneric Transfer of Isolated Nuclei from Lentinus edodes into Protoplasts of Pleurotus florida (사철느타리버섯 원형질체내에 표고 핵의 전이)

  • Transfer of the isolated nuclei from Lentinus edodes into protoplasts of Pleurotus florida was induced with polyethlene glycol (PEG) and $CaCl_2$. The intergeneric transfer products were classified into nuclear hybrid, heterokaryon or synkaryon, and reconstituted cell. These progenies except nuclear hybrids formed mature fruiting bodies on sawdust rice bran medium. Formation of fruit bodies was influenced by several factors such as light, temperature, nutrition and physic state of the culture media. Most of fruiting body characters were similar to those of P. florida in synkaryon and L. edodes in reconstituted cell, respectively. All these basidiocarps had clamp connections though initial heterokaryon colonies were lacking. Isozyme patterns of intergeneric progenies were quite different from those of parents. DNA polymorphisms of transfer products were also compared by random amplified polymorphic DNAs (RAPD) analysis based on polymerase chain reaction. The RAPD patterns were different from those of donor and recipient. DNA fingerprints ranged in size from 0.25 to 4.0 Kb. On the basis of RAPD, the transfer products were classified into five groups. Two synkaryon were analysed with distribution of progenies and segregation of genetic markers by random spore analyses. The genetic markers were segregated into wild type and riboflavine requiring auxotrophs.
