연안해수와 지하해수를 사용하는 제주 넙치 양식장의 수질과 미생물 변동 (Investigation of water qualities and microbials on the flow-through olive flounder, Paralichthys olivaceus farms using coastal seawater and underground seawater in Jeju)

  • This study assessed the levels of water qualities and microbials contamination of inland olive flounder farms in Jeju in the summers from 2015 to 2017. Three farms (A-C) located in a concentrated area using mixing coastal seawater and underground seawater and one farm (D) located in an independent area using only coastal seawater were selected. Total ammonia nitrogen (TAN) reached a maximum of 0.898 ± 1.024 mg/L as N in the coastal seawater of A-C, which was close to the limit of the water quality management goal of the fish farm. TAN in the influent from A-C was up to three times higher than that of D, so that the discharged water did not spread to a wide range area along the coast and continued to affect the influent. TAN of the effluent in A-C increased by 2.7-4.6 times compared to the influent, resulting in serious self-pollution in the flounder farm. Heterotrophic marine bacteria in the influent of A-C was about 600 times higher than D, and the discharge of A-C was increased by about 30 times compared to the influent.

Rapid Detection of Virulence Factors of Aeromonas Isolated from a Trout Farm by Hexaplex-PCR

  • The detection of virulence factors of Aeromonas is a key component in determining potential pathogenicity because these factors act multifunctionally and multifactorially. In this study water samples were collected from a trout farm on a seasonal basis, and diseased fish and Aeromonas species were isolated and identified. For rapid detection of six virulence factors of isolated Aeromonas, a hexaplex-polymerase chain reaction (hexaplex-PCR) assay was used. The detected virulence factors include aerolysin (aer), GCAT (gcat), serine protease (ser), nuclease (nuc) lipase (lip) and lateral flagella (laf). The dominant strain found in our isolates was Aeromonas sobria, and the dominant virulence factors were aer and nuc for all seasons. We confirmed that A. sobria and two of the virulence genes (aer and nuc) are related. We proposed a method by which one can identify the major strains of Aeromonas: A. hydrophila, A. sobria, A. caviae, and A. veronii, using hexaplex-PCR.

제주도 어류양식장에 대한 미생물학적 오염도 조사 (Survey of the level of Microbial Contamination in Fish Farms on the Jeju-Island)

  • 본 연구는 2006년 3월부터 8월까지 제주도에서 성업 중인 육상 수조식 양식장 21개소를 대상으로 하여 미생물학적 위해요소를 양어 용수, 양어 사료, 양식 넙치에서 조사하였다. 식품 병원성 미생물 중 일반세균과 총 대장균군, 그리고 Salmonella spp., Escherichia coli O157:H7, Staphylococcus aureus, Bacillus cereus, Vibrio parahaemolyticus를 조사하였다. 이번 연구를 진행하는 동안 특이할만한 점은 여름에 들어서면서 집중적이면서도 빈번하게 비가 내렸다. 양어용수를 검사한 결과 S. aureus, B. cereus and Salmonella spp.는 각각 $0-3.3{\times}10^2 CFU/ml,\;0-2.2{\times}10^2CFU/ml\;and\;0-5.1{\times}10^1 CFU/ml$ 검출이 되었다. 또한 사료 내 미생물을 조사하여 본 결과 조사한 MP사료 85%에서 S. aureus, B. cereus and total coliforms이 검출이 되었다 반면 양식 넙치를 조사한 결과 Escherichia coli O157:H7, total coliforms은 검출이 되지 않았다. 추후 이와 관련하여 좀더 체계적인 연구 시스템을 양식현장에 적용한다면 근래 들어 문제가 되고 있는 식품위생상 수산물이 야기하는 문제점들이 해결이 되어 안전한 식품으로서의 가치를 인정받을 수 있을 것 같다.

양식장 배출수에 첨가된 항생제 제거 위한 수중 비열 유전체장벽 방전 플라즈마 처리 효과 (Effect of plasma treatment using underwater non-thermal dielectric barrier discharge to remove antibiotics added to fish farm effluent)

  • 본 연구의 목적은 양식장 배출수 내에 포함된 5종의 항생제(tetracycline, doxycycline, oxytetracycline, clindamycin 및 erythromycin)를 제거하기 위해 사용한 수중 비열 유전체장벽 방전 플라즈마(Dielectric Barrier Discharge plasma, DBD plasma) 장치 안으로 공기와 산소를 각각 주입했을 때 항생제의 제거효율을 비교하는 것이다. DBD plasma를 발생시키기 위해서 주어진 전압은 27.8 kV이었고, 처리간격은 0, 0.5, 1, 2, 4, 8, 16 및 32분이었다. 3종의 tetracycline계 항생제는 공기를 주입했을 때는 4분만에 유의하게 감소하였고, 산소를 주입했을 때는 30초만에 유의성을 나타내었다. 32분째 공기와 산소를 각각 주입한 결과, tetracycline은 78.1%와 95.8%, doxycycline은 77.1%와 96.3% 그리고 oxytetracycline은 77.1%와 95.5% 감소하였다. Clindamycin은 공기를 주입했을 때 59.6%가 감소되었고, 산소는 83.0% 감소되었다. 또한, erythromycin은 공기주입 시 53.3%가 감소되었고 산소 주입 시 74.3%가 감소하여 두 항생제 모두 tetracycline계 항생제보다 낮은 제거 효율을 보였다. 결론적으로 수중 DBD plasma는 양식장 배출수 내에 포함된 5종의 항생제를 감소시킬 수 있고, 제거 효율은 공기보다 산소를 주입하는 것이 더 효과적이다.

제주도 연안 해역의 오염지표세균의 분포 (Distribution of Indicator Bacteria in Seawater off the Coast of Jeju Island)

  • We examined correlations of the density of fish farms with the distributions of indicator bacteria (Escherichia coli, fecal streptococci) and a bacterial fish pathogen (Streptococcus parauberis) off the coastline of Jeju Island. Seawater samples were collected at four coastal sites on the Island [Aewol (control), Gujwa, Pyoseon and Daejeong] in June, August and October 2016. The indicator bacteria were generally more frequently isolated from samples taken in August when water temperatures and human activities on nearby beaches were highest. Although fish farms were least common at Daejeong, the numbers of isolated fecal indicator bacteria were highest in the seawater and effluent water collected from this site. Hence, fish farms were not likely major contributors of indicator bacteria at Daejeong. We found discrepancies between the isolated bacterial counts and the predicted bacterial copy numbers deduced from our qPCR results, indicating that this pathogen may exist in a viable but non-culturable (VBNC) state in seawater. Thus, livestock wastewater and chemical fertilizer loading off Jeju Island may negatively impact seawater quality more than the effluent released from fish farms does.

미꾸라지 (Misgurnus mizolepis)의 뒤집힘병에 관한 연구 (An Overturn disease of Cultured Mud Loach (Misgurnus mizolepis))

  • 양식 미꾸라지 (Misgurnus mizolepis)가 수중으로 잠입하지 못하고 양어지의 물표면에 배를 위로 한 채 힘없이 회전 유영을 하는 질병이 발생하였다. 병어는 복부가 팽만되어 있었으며, 해부하여 보면 소화관에 커다란 공기방울의 축적, 장염이 현저하였다. 병어를 혈액학적으로 조사한 결과 적혈구수, Ht치 및 Hb 농도는 건강어와 차이가 없었다. 병리조직학적으로는 아가미의 호흡상피의 증생과 새변의 유착, 소화관 상피세포의 출혈, 괴사 및 박리가 관찰되었다. 뒷지느러미와 항문사이를 실로 결박하여 항문으로부터 공기의 배출을 억제한 재현 실험에서 발병어와 같은 증상이 재현되었다. 따라서 이 병은 빠른 성장을 목적으로 고단백질 사료를 투여함으로서 장호흡을 위하여 마신 공기가 소화관내에 축적되고, 축적된 공기의 부력 때문에 물속으로 하강하지 못하게 되고, 또 소화관을 압박하여 소화관에 순환장애를 일으킨 결과 폐사한 것으로 추정된다.

강도다리(Platichthys stellatus) 배합사료 내 곤충분 첨가에 따른 성장 및 사료가치 비교 (Effect of Dietary Inclusion of Black Soldier Fly Larvae Hermetia illucens Meal on Growth Performance of Starry Flounder Platichthys stellatus and Feed Value )

  • The dietary inclusion of black soldier fly larvae meal Hermetia illucens (BSL) for starry flounder Platichthys stellatus was examined in a four-month trial at a Pohang fish farm. Two diets were prepared: a fish meal-based commercial diet (CD) and an experimental diet containing 7% BSL in the CD (BCD). Fish (average weight, 125.2 g) were reared in a square concrete tank (7×7 m) in triplicates. Feed efficiency was significantly higher in fish fed BCD compared to that of the fish fed CD. The DHA/EPA ratio in dorsal muscle was not significantly affected by the fatty acid composition of the experimental diets. Based on the growth performance, the feed cost for producing 1 kg of starry flounder was 1,797 won for CD and 1,814 won for BCD. With government subsidies the feed cost for producing 1 kg of starry flounder was 1,481 won for CD and 1,309 won for BCD. The results indicate that BSL can be included at 7% without adverse effects on growth performance, fillet composition, or feed cost. However, further research is needed to determine the optimum percentage of BSL as a replacement of FM in starry flounder diets.

순환여과시스템에서 양식되는 바리과 어류 3종[능성어, (Hyporthodus septemfasciatus), 자바리(Epinephelus moara), 교잡종 대왕자바리(E. moara ♀× E. lanceolatus ♂)]의 감염성 질병 모니터링 (Monitoring of infectious diseases in three grouper species (Hyporthodus septemfasciatus, Epinephelus moara and E. moara ♀× E. lanceolatus ♂) cultured in recirculating aquaculture systems)

  • The Serranidae is high-quality fish species with good meat quality and is traded at high price, and is attracting attention in South Korea as a cultured species that creates high added value. However, the high-density fish farming for mass production increases the risk of mass mortality due to infectious diseases, leading to enormous economic losses. Therefore, in order to safely prevent and protect farmed fish from serious infectious diseases, it is necessary to conduct disease monitoring on a regular basis. In this study, Hyporthodus septemfasciatus, Epinephelus moara, and the hybrid longtooth grouper (E. moara ♀×E. lanceolatus ♂) were collected once a month from fish farm of Garorim and Aquabiotech Co., Ltd for a total of six months, from July to December 2023. We investigated infections of five species of bacterial diseases, including Flavobacterium columnare, six species of viral diseases, including LCDV (lymphocystis disease virus), and parasitic pathogens in grouper farms. As the result, Vibrio vulnificus and V. harveyi were detected in H. septemfasciatus in August, in the case of viral diseases, NNV was detected in H. septemfasciatus from July to August using RT-PCR or PCR. Finally, In the case of parasitic diseases, Tricodina sp. was detected in E. moara and the hybrid longtooth grouper from August to December.

상업용 부상사료와 어분기초생사료가 넙치의 성장과 체조성에 미치는 영향 (Effects of the Extruded Pellets and Raw Fish-Based Moist Pellet on Growth and Body Composition of Flounder, Paralichthys olivaceus L. for 10 Months)

  • Effects of the commercially available or formulated extruded pellets (EP) and raw fish-based moist pellet (MP) on growth and body composition of flounder Paralichthys olivaceus L. were evaluated on commercial scale for 10 months. Survivals of flounder fed the MP, EP 1 and EP3 were not significantly different from those of fish fed the EP2 or EP5, but significantly (P<0.05) higher than that of fish fed the EP4. Weight gain of fish fed the MP was not significantly different from that of fish fed the EP3 and EP5, but significantly (P<0.05) higher than that of fish fed the EP1, EP2 or EP4. Improvement in weight gain of flounder fed the high lipid diets (EP1, EP2, EP3) compared to low lipid diet (EP4) in the $55\%$ protein level, and weight gain of fish fed the high lipid diet (MP) compared to low lipid diet (EP5) in the $61\%$ protein level indicated protein-sparing effect of lipid. FER for flounder fed the EP3. which was not significantly different from that for fish fed the EP1 or EP5 was significantly (P<0.05) higher thar. that for fish fed the EP2, EP4 and MP. FER significantly (P<0.05) changed over time. CF of flounder fed the MP was significantly (P<0.05) higher than that of fish fed EP1 or EP4. Moisture content of dorsal muscle in flounder fed the EP2 was significantly (P<0.05) higher than that in fish fed the EP1 or EP4. Crude protein content of dorsal. muscle in flounder fed the EP1, EP4 and EP5 was significantly (P<0.05) higher than that in fish fed the EP2. How- ever, crude lipid content of dorsal muscle, and moisture, crude protein and crude lipid content of liver in flounder was not significantly different among the diets. In considering performance of flounder, it can be concluded that the use of EP3 and EP5 is more recommendable than the MP, which commonly used in flounder farm.

양식 쏘가리에서 Atypical Aeromonas Salmonicida의 발병 (Occurrence of Atypical Aeromonas Salmonicida in Mandarin Fish (SSOGARI: Korean Name) Siniperca Scherzeri)

  • 경남 산청지방에서 쏘가리를 양식하던 어장에서 대량폐사가 발생하였다. 주요 증상으로 전신 피부에서 궤양이 진행되었고 조직학적으로는 주요 실질 장기에서 세균의 집락과 주변부위로 염증 및 괴사가 관찰되었다. 분리된 세균을 API 20E 및 API 20NE를 이용하여 생화학적 검사를 실시한 결과 Aeromonas salmonicida subsp. salmonicida (typical A. salmonicida)로 진단할 수 있었다. 그러나 atypical A. salmonicida와 typical A. salmonicida로 구분할 수 있는 PCR 기법을 실시한 결과, 본 세균은 atypical A. salmonicida임을 확인할 수 있었다. 본 연구는 우리나라 쏘가리에서 A. salmonicida로 인한 발병의 최초 보고임과 동시에, A. salmonicida의 정확한 진단을 위해선 생화학적 검사가 아닌 PCR 검사를 이용한 진단을 실시해야 함을 제시하였다.
