Rapid Detection of Virulence Factors of Aeromonas Isolated from a Trout Farm by Hexaplex-PCR

  • The detection of virulence factors of Aeromonas is a key component in determining potential pathogenicity because these factors act multifunctionally and multifactorially. In this study water samples were collected from a trout farm on a seasonal basis, and diseased fish and Aeromonas species were isolated and identified. For rapid detection of six virulence factors of isolated Aeromonas, a hexaplex-polymerase chain reaction (hexaplex-PCR) assay was used. The detected virulence factors include aerolysin (aer), GCAT (gcat), serine protease (ser), nuclease (nuc) lipase (lip) and lateral flagella (laf). The dominant strain found in our isolates was Aeromonas sobria, and the dominant virulence factors were aer and nuc for all seasons. We confirmed that A. sobria and two of the virulence genes (aer and nuc) are related. We proposed a method by which one can identify the major strains of Aeromonas: A. hydrophila, A. sobria, A. caviae, and A. veronii, using hexaplex-PCR.

Survey of the level of Microbial Contamination in Fish Farms on the Jeju-Island (제주도 어류양식장에 대한 미생물학적 오염도 조사)

  • The purpose of this study was to monitor and compare the contamination levels of heterotrophic bacteria and pathogenic bacteria (total coliforms, Escherichia coli O157:H7, Salmonella spp., Vibrio parachaemolyticus, Staphylococcus aureus and Bacillus cereus). The samples collected at fish farms located in the Jeju-island were investigated from March to August in 2006. The contamination levels of S. aureus, B. cereus and Salmonella spp. ranged $0-3.3{\times}10^2 CFU/ml,\;0-2.2{\times}10^2 CFU/ml\;and\;0-5.1{\times}10^1CFU/ml$, respectively, and 85% of moist pellet feeds were contaminated with S. aureus, B. cereus and total coliforms. But total coliforms and E. coli O157:H7 were not detected in oliver flounder (Paralichthys olivaceus). For the production of safe oliver flounder (Paralichthys olivaceus), education of sanitation for employees, control of fish feed, and continuous monitoring for microorganism will be required.

Effect of plasma treatment using underwater non-thermal dielectric barrier discharge to remove antibiotics added to fish farm effluent (양식장 배출수에 첨가된 항생제 제거 위한 수중 비열 유전체장벽 방전 플라즈마 처리 효과)

  • The purpose of this study was to compare the efficiency of air and oxygen injected into the underwater non-thermal dielectric barrier discharge plasma (DBD plasma) device used to remove five types of antibiotics (tetracycline, doxycycline, oxytetracycline, clindamycin, and erythromycin) artificially contained in the fish farm discharge water. The voltage given to generate DBD plasma was 27.8 kV, and the measurement intervals were 0, 0.5, 1, 2, 4, 8, 16 and 32 minutes. Tetracycline antibiotics significantly decreased in 4 minutes when air was injected and were reduced in 30 seconds when oxygen was injected. After the introduction of air and oxygen at 32 minutes, 78.1% and 95.8% of tetracycline were removed, 77.1% and 96.3% of doxycycline were removed, and 77.1% and 95.5% of oxytetracycline were removed, respectively. In air and oxygen, 59.6% and 83.0% of clindamycin and 53.3% and 74.3% of erythromycin were removed, respectively. The two antibiotics showed lower removal efficiency than tetracyclines. In conclusion, fish farm discharge water contains five different types of antibiotics that can be reduced using underwater DBD plasma, and oxygen gas injection outperformed air in terms of removal efficiency.

Distribution of Indicator Bacteria in Seawater off the Coast of Jeju Island (제주도 연안 해역의 오염지표세균의 분포)

  • We examined correlations of the density of fish farms with the distributions of indicator bacteria (Escherichia coli, fecal streptococci) and a bacterial fish pathogen (Streptococcus parauberis) off the coastline of Jeju Island. Seawater samples were collected at four coastal sites on the Island [Aewol (control), Gujwa, Pyoseon and Daejeong] in June, August and October 2016. The indicator bacteria were generally more frequently isolated from samples taken in August when water temperatures and human activities on nearby beaches were highest. Although fish farms were least common at Daejeong, the numbers of isolated fecal indicator bacteria were highest in the seawater and effluent water collected from this site. Hence, fish farms were not likely major contributors of indicator bacteria at Daejeong. We found discrepancies between the isolated bacterial counts and the predicted bacterial copy numbers deduced from our qPCR results, indicating that this pathogen may exist in a viable but non-culturable (VBNC) state in seawater. Thus, livestock wastewater and chemical fertilizer loading off Jeju Island may negatively impact seawater quality more than the effluent released from fish farms does.

An Overturn disease of Cultured Mud Loach (Misgurnus mizolepis) (미꾸라지 (Misgurnus mizolepis)의 뒤집힘병에 관한 연구)

  • In late summer of 2001, a new disease "overturn disease" occurred among mud loach (Misgurnus mizolepis) cultured in a farm Chonbuk Province. The fish exhibited a lethargic, erratic and spiral swimming at the water surface. Affected fish showed swollen abdomen with accumulation of air bubbles in the intestinal canal. The bubbles were much larger in the anterior part than in the posterior part. Hematological and histopathological examinations were done. No significant differences were found in RBC count, Ht value and Hb concentration between the diseased and the healthy fish. Gill filaments showed proximal hyperplasia of the respiratory epithelia and fusing of the gill lamella. Hemorrhage and necrosis occurred in the intestinal epithelia and within the lamina propria, and the mucosal epithelia were separated from the muscularis. Hepatocytes underwent atrophy. In the fish experimentally tied between the annal fin and the anus by a thread to confirm accumulation of air bubbles within intestine, accumulation of air bubbles was confirmed. This disease in mud loach appeared to be induced by circulatory disturbances in the intestinal wall owing to accumulation of air bubbles in the alimentary canal after fed high protein diet throughout all culturing period.

Effect of Dietary Inclusion of Black Soldier Fly Larvae Hermetia illucens Meal on Growth Performance of Starry Flounder Platichthys stellatus and Feed Value (강도다리(Platichthys stellatus) 배합사료 내 곤충분 첨가에 따른 성장 및 사료가치 비교)

  • The dietary inclusion of black soldier fly larvae meal Hermetia illucens (BSL) for starry flounder Platichthys stellatus was examined in a four-month trial at a Pohang fish farm. Two diets were prepared: a fish meal-based commercial diet (CD) and an experimental diet containing 7% BSL in the CD (BCD). Fish (average weight, 125.2 g) were reared in a square concrete tank (7×7 m) in triplicates. Feed efficiency was significantly higher in fish fed BCD compared to that of the fish fed CD. The DHA/EPA ratio in dorsal muscle was not significantly affected by the fatty acid composition of the experimental diets. Based on the growth performance, the feed cost for producing 1 kg of starry flounder was 1,797 won for CD and 1,814 won for BCD. With government subsidies the feed cost for producing 1 kg of starry flounder was 1,481 won for CD and 1,309 won for BCD. The results indicate that BSL can be included at 7% without adverse effects on growth performance, fillet composition, or feed cost. However, further research is needed to determine the optimum percentage of BSL as a replacement of FM in starry flounder diets.

  • The Serranidae is high-quality fish species with good meat quality and is traded at high price, and is attracting attention in South Korea as a cultured species that creates high added value. However, the high-density fish farming for mass production increases the risk of mass mortality due to infectious diseases, leading to enormous economic losses. Therefore, in order to safely prevent and protect farmed fish from serious infectious diseases, it is necessary to conduct disease monitoring on a regular basis. In this study, Hyporthodus septemfasciatus, Epinephelus moara, and the hybrid longtooth grouper (E. moara ♀×E. lanceolatus ♂) were collected once a month from fish farm of Garorim and Aquabiotech Co., Ltd for a total of six months, from July to December 2023. We investigated infections of five species of bacterial diseases, including Flavobacterium columnare, six species of viral diseases, including LCDV (lymphocystis disease virus), and parasitic pathogens in grouper farms. As the result, Vibrio vulnificus and V. harveyi were detected in H. septemfasciatus in August, in the case of viral diseases, NNV was detected in H. septemfasciatus from July to August using RT-PCR or PCR. Finally, In the case of parasitic diseases, Tricodina sp. was detected in E. moara and the hybrid longtooth grouper from August to December.

Effects of the Extruded Pellets and Raw Fish-Based Moist Pellet on Growth and Body Composition of Flounder, Paralichthys olivaceus L. for 10 Months (상업용 부상사료와 어분기초생사료가 넙치의 성장과 체조성에 미치는 영향)

  • Effects of the commercially available or formulated extruded pellets (EP) and raw fish-based moist pellet (MP) on growth and body composition of flounder Paralichthys olivaceus L. were evaluated on commercial scale for 10 months. Survivals of flounder fed the MP, EP 1 and EP3 were not significantly different from those of fish fed the EP2 or EP5, but significantly (P<0.05) higher than that of fish fed the EP4. Weight gain of fish fed the MP was not significantly different from that of fish fed the EP3 and EP5, but significantly (P<0.05) higher than that of fish fed the EP1, EP2 or EP4. Improvement in weight gain of flounder fed the high lipid diets (EP1, EP2, EP3) compared to low lipid diet (EP4) in the $55\%$ protein level, and weight gain of fish fed the high lipid diet (MP) compared to low lipid diet (EP5) in the $61\%$ protein level indicated protein-sparing effect of lipid. FER for flounder fed the EP3. which was not significantly different from that for fish fed the EP1 or EP5 was significantly (P<0.05) higher thar. that for fish fed the EP2, EP4 and MP. FER significantly (P<0.05) changed over time. CF of flounder fed the MP was significantly (P<0.05) higher than that of fish fed EP1 or EP4. Moisture content of dorsal muscle in flounder fed the EP2 was significantly (P<0.05) higher than that in fish fed the EP1 or EP4. Crude protein content of dorsal. muscle in flounder fed the EP1, EP4 and EP5 was significantly (P<0.05) higher than that in fish fed the EP2. How- ever, crude lipid content of dorsal muscle, and moisture, crude protein and crude lipid content of liver in flounder was not significantly different among the diets. In considering performance of flounder, it can be concluded that the use of EP3 and EP5 is more recommendable than the MP, which commonly used in flounder farm.

Occurrence of Atypical Aeromonas Salmonicida in Mandarin Fish (SSOGARI: Korean Name) Siniperca Scherzeri (양식 쏘가리에서 Atypical Aeromonas Salmonicida의 발병)

  • Mandarin fish (SSOGARI: Korean name) has emerged as new aquaculture and aquarium fish among Korean native fish species. Unfortunately, this fish could not exempt from infectious agent invasion. As an epidemiological survey, high mortality of mandarin fish (Siniperca scherzeri) observed from a fish farm in Gyeongnam province of South Korea. The major macroscopic sign of the diseased fish was severe multifocal dermal ulceration. Histological observation revealed inflammation, necrosis and colonization of bacteria in various tissues. Seven bacterial isolates from the diseased fish were identified as Aeromonas salmonicida subsp. salmonicida (typical A. salmonicida) using API 20E and 20NE system. However, the polymerase chain reaction (PCR) assay using two different primers (AP and MIY) revealed that the bacterial strains were not typical A. salmonicida, but atypical A. salmonicida. This study demonstrates that PCR assay is a reliable method for the confirmation of atypical A. salmonicida in mandarin fish as compared to biochemical tests. Furthermore, this is the first account of extensive dermatitis in mandarin fish due to atypical A. salmonicida infection, which has high potential in aquaculture and aquarium fish among native fish species.

  • Form, increasing the income of fishermen and internationally competitive high value-added seafood merchandising consumers for the purpose of improving the reliability of the food as do Tongyeong Gyeongsangnam-piping of the high quality marine products traceability system for farm businesses. Real-time traceability of the seafood supply chain to manage RFID technologies and related IT technology was introduced in progress. In this study, real-time traceability of fish introduced to the features of RFID technology and to recognize its effects. And efficient distribution system of fish suggests ways to improve and execute simulation about supply chain.
