The gDNA isolated from Korean cuttle fish (Sepia esculenta Hoyle) from Sockcho (SOCKCHO), Seocheon (SEOCHEON), Incheon (INCHEON) and Vietnamese cuttle fish (VIETNAM), were amplified by PCR. Here, the seven selected primers (BION-A07, BION-A09, BION-A11, BION-A20, BION-B04, BION-B06, and BION-B14) were used to generate the unique shared loci to each population and shared loci by the four cuttle fish populations. In this study, the primer BION-A11 detected 112 shared loci by the four populations, major and/or minor fragments of sizes 300 bp, 400 bp, 700 bp and 1,000 bp, respectively, which were identical in all samples. The dendrogram obtained by the seven primers indicates five genetic clusters: cluster 1 (SOCKCHO 01-SOCKCHO 07), cluster 2 (SEOCHEON 08-SEOCHEON 10), cluster 3 (SEOCHEON 11-SEOCHEON 14), cluster 4 (INCHEON 15-INCHEON 21), and cluster 5 (VIETNAM 22-VIETNAM 28). The shortest genetic distance that displayed significant molecular differences was between individuals 25 and 26 from the Vietnamese cuttle fish (0.025), while the longest genetic distance among the twenty-eight cuttle fishes that displayed significant molecular differences was between individuals SOCKCHO no. 02 and SEOCHEON no. 12 (0.640). Individual of Seocheon and Incheon cuttle populations was somewhat closely related to that of Vietnamese cuttle fish population. Even though it could not be affirmed by a single case, such a result seems to be closely connected that the Korean peninsula is subject to climate changes by global warming. In conclusion, our PCR analyses revealed a significant genetic distance among the four cuttle fish populations.

We determined the morphologic characteristics (body weight and degree of abdomen inflation) of the red spotted grouper, Epinephelus akaara, mother fish producing healthy eggs. Experimental fish were chosen from fish reared in a sea cage. The fish were divided into four size groups by body weight: 400~600, 600~800, 800~1,000, and 1,000~1,200 g and four stages (I~IV) of the degree of abdomen inflation. After hormone treatment, we observed the amount of ovulation-induced eggs, and rates of buoyancy, fertilization, embryonic survival, and hatching. As a result, mother fish with a body weight of 600 g or more spawned, and the fertilization rate, embryonic survival rate, and hatching rate were high in the 800~1,000 g range, thus showing effective ovulation induction. As a result of dividing the degree of abdomen inflation based on the anal fin of the mother fish into I-IV stages and determining hormone treatment time, the GSI was $0.9{\pm}0.2%$ at stage I, $2.3{\pm}0.2%$ at stage II, $5.6{\pm0.2%$ at stage III, and $7.9{\pm}0.9%$ at stage IV. The flotation rate and hatching rate were highest at stage III, and the fertilization rate and embryonic survival rate were highest at stage IV. Therefore, in terms of egg quality, the amount of eggs collected per mother fish, maturation, and histology were different depending on the degree of abdomen inflation. At stage III, where the abdomen inflation degree of the mother fish was based on the basal part of the dorsal fin relative to the height of the anal fin was 1, the egg quality was highest.

Reproduction of the goby fish Periophthalmus magnuspinnatus was examined using 298 specimens collected from April to October 2005 in a mud flat of Suncheon Bay, Korea. Specimens ranged in body length(BL) from 1.3 to 9.1 cm. The gonadosomatic index(GSI) of females was highest in June and decreased until August. The hepatosomatic index(HSI) and fatness index of females were high in April and then decreased to nadirs in July. Spawning season lated from April to August and the ratio of females to males did not significantly differ($x^2$-test, p>0.05). The first spawning length was 4.5 cm BL, and the size of 50% maturity was estimated at 4.98 cm BL. Fecundity(F) ranged from 1,316 to 4,768 eggs, and the relationship between F and BL was estimated as $F=0.1562BL^{1.4068}(R^2=0.59)$.
Habitat, reproduction and feeding habit of endangered fish Koreocobitis naktongensis from Jaho stream, Gyeongsangbuk-do, Korea were investigated from March 2010 to June 2011. This species inhabits at the upper and middle parts of stream, which were composed of cobbles and pebbles bottom. The current velocities in these microhabitat were 5~10 cm/sec and depth were 50~110cm. The sex ratio of female to male was 1 : 0.92. It is presumed that the spawning season is from May to June when water temperatures were 17 ~ 20$^{\circ}C$. The average number of mature eggs in ovary were about 22,643 (15,909~30,323), mean egg diameter was 0.87${\pm}$0.05mm. K. naktongensis fed mainly on Chironomidae (IRI, 72.0%) and Ephydridae (26.9%) of Diptera.
Improvement and possible commercialization of a home-made electroporation apparatus(home-made) were further tried to establish a simple and effective introduction of foreign gene into sperm followed by in vitro fertilization. Expressions of introduced pJJ9 and pNT plasmids were shown in all fertilized eggs with electroporated spermatozoa. In particular, with this gene transfer system all the fry showed a consistently transient expression in the syncytium of the yolk sac. This fact is important since some required, minute quantity of human proteins can be produced from the established transient expression on the yolk sac of all fry derived from in vitro fertilization with electroporated spermatozoa. To explore tissue-specific expression in fish, which we will use a similar system later, we targeted the nerve tissue to see whether tissue-specific promoter is working in fish properly. pNT plasmid containing a nerve cell-specific tubulin promoter gene demonstrated consistently exact targeted expressions among the developing nerve cells in later stages of embryos and hatched fry. Finally, liver-specific genes are now being cloned by using already selected primers for useful human protein gene fusion.
Fish ovarian germline stem cells (OGSCs) that have the abilities to self-renew and differentiate into functional gametes can be used in various researches and applications. A main issue to be solved for effective utilization of fish OGSCs is the development of their stable in vitro culture condition, but only few researches about fish OGSC culture have been reported so far. In this study, in order to find the clues to develop the culture condition for OGSCs from Japanese medaka (Oryzias latipes), we tried to establish somatic cell lines as a candidate for the feeder cells and evaluated its supporting effects on the culture of ovarian cell populations from O. latipes. As the results, the somatic cell lines could be established only from the embryonic tissues among three tissues derived from embryos, fins and ovaries. Three embryonic cell lines were tested as a feeder cell for the culture of ovarian cell population and all three cell lines induced cell aggregation formation of the cultured ovarian cells whereas the feeder-free condition did not. Furthermore, a significant cellular proliferation was observed in the ovarian cells cultured on two of three cell lines. As a trial to increase the capacity of the cell lines as a feeder cell that supports the proliferation of the cultured ovarian cells, we subsequently established a stable line that expresses the foreign O. latipes fibroblast growth factor 2 (FGF2) from an embryonic cell line and evaluated its effectiveness as a feeder cell. The ovarian cells cultured on FGF2 expressing feeder cells still formed cell aggregates but did not show a significant increase in cellular proliferation compared to those cultured on non-transformed feeder cells. The results from this study will provide the fundamental information for in vitro culture of medaka OGSCs.
Present study was aimed to summary the recent reports of chromosomal technology such like a polyploidv, sex differentiation, gynogenesis, transgenic fish and gene manipulation. Triploid cells for rainbow trout and channel catfish were induced through thermal shocks of varying temperature levels and produced as a industrial use. A monosex fish with homogametic females of 15 species of high valued fish were produced by exposing to irradiation. It seemed that different irradiation was suitable to inactivate the sperm and block the formation in producing the gynogenetic diploids. Since 1985, transgenic fish have been successfully produced by microinjecting or electroporating desired foreign DNA into unfertilized or newly fertilized eggs using about 40 fish species. More recently, transgenic fish have also been produced by infecting newly fertilized eggs with pantropic, defective retroviral vectors carrying desired foreign DNA. These transgenic fish can serve as excellent experimental models for basic scientific investigations as well as in marine biotechnological applications.

In fish, light (photoperiod, intensity and spectra) is main regulator in many physiological actions including growth. We investigate the effect of light spectra on the somatic growth and growth-related gene expression in the rearing tiger puffer. Fish was reared under different light spectra (blue, green and red) for 8 weeks. Fish body weight and total length were promoted when reared under green light condition than red light condition. Expression of somatostatins (ss1 and ss2) in brain were showed higher expression under red light condition than green light condition. The ss3 mRNA was observed only higher expression in blue light condition. Expression of growth hormone (gh) in pituitary was detected no different levels between experimental groups. However, the fish of green light condition group was showed more high weight gain and feed efficiency than other light condition groups. Our present results suggest that somatic growth of tiger puffer is induced under green light condition because of inhibiting ss mRNA expression in brain by effect of green wavelength.
Differential gene expression profiling was carried out in the hepatic tissue of medaka fish, Oryzias latipes, after exposure to an organophosphorus pesticide (OPP), Iprobenfos (IBP), a widely used pesticide in agri- and fish-culture, using a medaka cDNA micro array. Twenty six kinds of differentially expressed candidate genes, with 15 and 11 induced and repressed in their gene expressions, respectively, were associated with cytoskeleton (3.8%), development (7.7%), immune (7.7%), metabolism (30.8%), nucleic acid/protein binding (42.3%) and reproduction (7.7%). Of these genes, changes at the transcription level of five were re-evaluated by real-time quantitative PCR (qRT-PCR). Considering the known function of authentic genes, the effects of IBP on the biological activity and pathological aspects in medaka fish were discussed. The identified genes could be used as molecular biomarkers for biological responses to OPPs contamination in an aquatic environment.

This study was examined the ovogenesis of Ussurian bullhead, Leiocassis ussuriensis and the morphological development of its larvae and juveniles and to use the results as basic information for the preservation of species and resource enhancement. For artificial egg collection, human chorionic gonadotropin (HCG) was injected at a rate of 10 IU per gram of fish weight. During breeding period, water temperature maintained at $24.5{\sim}26.5^{\circ}C$ (mean $25.0{\pm}0.05^{\circ}C$). The process of ovogenesis reached the two-cell stage in 50 minutes after fertilization. In 73 hours of fertilization the movement of the embryoid body became active state and the larvae began to hatch from the tail through the oolemma. Length of prelarvae were 6.33~6.50 mm long (mean $6.40{\pm}0.06mm$) just after hatching having yolk with their mouth not opened. After thirty eight days of hatching, juveniles were 30.6~32.5 mm long (mean $31.5{\pm}0.65mm$). The color was dark yellowish brown throughout the entire body, and the number of caudal fin rays developed to thirty six perfectly.
