Application of Engineered Zinc Finger Proteins Immobilized on Paramagnetic Beads for Multiplexed Detection of Pathogenic DNA

  • Micro-scale magnetic beads are widely used for isolation of proteins, DNA, and cells, leading to the development of in vitro diagnostics. Efficient isolation of target biomolecules is one of the keys to developing a simple and rapid point-of-care diagnostic. A zinc finger protein (ZFP) is a double-stranded (ds) DNA-binding domain, providing a useful scaffold for direct reading of the sequence information. Here, we utilized two engineered ZFPs (Stx2-268 and SEB-435) to detect the Shiga toxin (stx2) gene and the staphylococcal enterotoxin B (seb) gene present in foodborne pathogens, Escherichia coli O157 and Staphylococcus aureus, respectively. Engineered ZFPs are immobilized on a paramagnetic bead as a detection platform to efficiently isolate the target dsDNA-ZFP bound complex. The small paramagnetic beads provide a high surface area to volume ratio, allowing more ZFPs to be immobilized on the beads, which leads to increased target DNA detection. The fluorescence signal was measured upon ZFP binding to fluorophore-labeled target dsDNA. In this study, our system provided a detection limit of ≤ 60 fmol and demonstrated high specificity with multiplexing capability, suggesting a potential for development into a simple and reliable diagnostic for detecting multiple pathogens without target amplification.

TIG 용접에서 열유속이 용융효율과 용입형상에 미치는 영향 (Effect of Heat Flux on the Melting Efficiency and Penetration Shape in TIG Welding)

  • The characteristics of arc pressure, current density and heat flux distribution are important factors in understanding physical arc phenomena, which will have a marked effect on the penetration, size and shape of a weld in TIG welding. The purpose of this study is to find out the effect of the heat flux on the melting efficiency and penetration shape in TIG welding using the results of the previous investigators. The conclusions obtained permit to draw a proper method which derived the heat flux distributions by arc pressure distribution measurements, but previous researchers calculated heat flux and current distribution with the heat intensity measurements by the calorimetry. Heat flux of Ar gas arc was concentrated at the central part and distributed low from the arc axis to the radial direction, that of He mixing arc was lower than that of Ar gas, and it was wide distributed to radial direction. That showed a similar characteristic with the Nestor's by calorimetry calculated values. Throughout heat flux drawn in this study was discussed melting efficiency and penetration shape on Ar gas and He mixing gas arc.

메탄의 완전산화 반응을 위한 중공사형 페롭스카이트 촉매 제조 (Preparation of the Hollow Fiber Type Perovskite Catalyst for Methane Complete Oxidation)

  • 선행 연구를 통해서 우수한 메탄 완전 산화 특성을 보인 $La_{0.1}Sr_{0.9}Co_{0.2}Fe_{0.8}O_{3-{\delta}}$ (LSCF1928) 페롭스카이트 분말 촉매로 비드형과 중공사형 촉매를 제조하였다. 중공사형 촉매는 내부가 비어있는 원기둥 형태이며, 상전이법을 통해 기공을 형성시킬 수 있어 비표면적을 획기적으로 향상시킬 수 있다. Methyl Cellulose (MC)를 넣어 제조한 비드형 촉매의 경우 MC에서 배출되는 $CO_2$와 촉매 성분인 Sr이 반응하여 원래 촉매 조성이었던 LSCF1928 조성 외에 $SrCO_3$가 생성되었다. 중공사형 촉매의 경우 불순물 없이 단일상의 페롭스카이트 구조를 얻었다. $700{\sim}900^{\circ}C$에서 하소한 중공사형 촉매는 손가락구조-스펀지구조-손가락구조의 기공 형태를 보였으며, 모두 $475^{\circ}C$에서 메탄의 완전 산화 반응을 통해 99.9% 산소 전화율을 달성하였다. 중공사형 촉매의 기공을 제어하기 위하여 에어 갭과 방사압력 조건을 변경하였다. 2 cm 에어 갭, 7 bar의 방사 압력으로 제조한 중공사형 촉매가 가장 우수한 촉매 성능을 보였으며, $425^{\circ}C$, $450^{\circ}C$$475^{\circ}C$에서 각각 70.65%, 93.01%, 99.99% 이상의 산소 전화율을 달성하였다.
