한국재래산양의 태아 및 신생아 혀유두 발달에 관한 주사전자현미경적 연구 (Scanning electronic microscopical studies on the development of the tongue in fetuses and neonates of Korean native goats)

  • The morphological development of lingual papillae in fetuses between 60, 90, 120 days gestation and neonates of Korean native goat were investigated by scanning electron microscopy. In 60 day old fetuses, the primordia of lingual papillae were observed on the dorsal surface epithelium and those of papillae were primordial fungiform, vallate and conical papillae. The many of microridges and microplicae were observed on the surface of those epithelial cells. In 90 day old fetuses, the rudiment of lentiform and filiform papillae were appeared. There were microplicaes on the surface of the conical papillae epithelium. In the 120 day old fetuses, the lingual papillaes were well developed. The taste bud were opened on the top of vallate papillae that were compactly many of short microvilli. Moreover, secondary papillae partially were observed on top of vallate papillae. In neonate, the fungiform, vallate and lentiform papillae were similar to the adult lingual papillae, but filiform and conical papillae were different from the mature lingual papillae. The outline of filiform papillae were irregularly in indented, but those of conical papillae were regularly. The diameters of filiform, fungiform, conical, vallate and lentiform papillae were about 80~100, 190~250, 230~470, 360~670 and $730{\sim}1,140{\mu}m$, respectively. The height of filiform and conical papillae were about $130{\sim}140{\mu}m$ and $145{\sim}250{\mu}m$.

Comparative Morphological Study on the Embryonic and Neonatal Development of the Filiform Papillae and Teeth in Mice

  • Background: In the early stages of development, teeth and lingual papillae are induced and developed through special and complex epithelial-mesenchymal interactions. Tooth completion indicates the beginning of the weaning phase, and accordingly, many oral tissues and organs are completed, and it is thought that their developmental completion times are related to each other. The purpose of this study was to clarify the embryonic and neonatal development of the filiform papillae and mandibular molar tooth, and discuss the developmental relationship between these organs by comparing the developmental completion times. Methods: Embryos at embryonic day 15 (EM15), 17 (EM17), and 21 (EM21) and mice at neonatal day 1 (NE1), 5 (NE5), 10 (NE10), and 21 (NE21) were used for experimentation. Tissues dissected from embryos and mice were fixed, and processed for histological analysis. Sections from the tissues were stained with hematoxylin and eosin for observation under a light microscope. Results: Based on the histological analysis results, the developmental process of the lingual epithelium covering the dorsal surface of the tongue was classified into three stages: initiation, morphogenesis, and functional. The development of the filiform papillae begins at EM17; undergoes rapid morphological changes in epithelial cells at EM21, PN1 and PN5, and reaches the functional stage at PN10, which is the sucking phase. Tooth development begins at EM13 or 15 and is completed at NE21 through prenatal and postnatal development. Conclusion: The development of the filiform papillae was initiated late and completed quickly through embryonic and neonatal development in comparison with the mandibular molar tooth. The filiform papillae are considered to play an important role in sucking rather than mastication as it is completed in the sucking phase.

뒤쥐(Sorex caecutiens) 혀유두의 형태학적 연구 (Morphological Study on the Dorsal Lingual Papillae of Sorex caecutiens Laxmann)

  • 뒤쥐(Sorex caecutiens) 혀유두의 형태적 특징을 알아보기 위해 주사전자현미경을 이용하여 관찰하였다. 혀유두는 실유두, 버섯유두 그리고 성곽유두의 세 가지 유형으로 관찰되었다. 실유두는 혀측면을 제외하고는 혀의 전역에 걸쳐 분포하고 있었으며, 형태와 구조적 특징을 기초로 하여 세 가지의 유형으로 구분되어졌다. 다수의 버섯유두는 혀 전체에 걸쳐 분포하였으며, 특히 혀의 양 측면에 걸쳐 존재하고 있었으며, 혀의 앞쪽보다는 뒤쪽으로 갈수록 크기가 더 컸었다. 버섯유두는 쌍을 이루며 분포하지는 않았지만, 대부분 일정하게 분포하였다. 큰 2개의 성곽유두는 혀의 후방부의 끝 가까이에 존재하고 있었다. 성곽유두는 바깥쪽(측면)이 두꺼운 두겹의 pad가 둥글게 둘러싸고 있었으며, 중앙에는 2개 혹은 3개 정도의 돌출된 둥글게 둘러싼 pad가 존재하고 있었다. 본 연구 종은 일반적인 뒤쥐류에서 볼 수 있는 혀의 형태와 거의 흡사한 구조를 가지고 있었음을 알 수가 있었다. 본 연구에서 성곽유두는 Sorex 종과는 차이가 있었다. 즉, 박쥐류와 Sorex 종에서와 같이 바깥쪽의 pad가 환상의 형태를 취하고 있다는 점에서는 동일하나 박쥐류와 Sorex 종의 경우는 바깥쪽의 pad가 한 겹인데 반해 본 연구 종은 두 겹의 환상 구조를 취하고 있다는 점에서 차이가 있었다.

  • 주사전자현미경을 이용하여 한국산 긴가락박쥐(Miniopterus schreibersi fuliginosus)와 검은집박쥐(Pipistrellus savii)의 혀유두의 형태를 관찰하였다. 본 연구에서 두 종 모두 세 가지의 혀유두, 즉, 실유두, 버섯유두 그리고 성곽유두가 관찰되었다. 실유두는 다시 두 종류로 세분화되어졌으며, 침상의 돌기물을 갖는 유형1은 혀의 전반부의 1/2 지점까지 분포하며, 유형2는 밋밋하고 두꺼운 돌기물을 갖고 있으며 혀의 후반부의 1/2에 분포한다. 두 종 모두 35~45개의 버섯유두가 혀의 표면에서 관찰된다. 이들 버섯유두는 혀의 측면과 혀의 전반과 후반부의 끝 쪽에서 나타난다. 두 종의 버섯유두는 각각 두 개 혹은 세 개의 매우 큰 유두로 혀의 후반부 중앙에 일직선으로 배열되어 있다. 두 개의 성곽유두는 혀의 후반부 끝쪽 가까이에 분포하고 있다. 이들 성곽유두는 크고 둥글며 각각은 두 층의 pad를 갖고 있다. 긴가락박쥐와 검은집박쥐의 혀유두의 전체적인 형태는 다른 익수류와 유사하였다. 그럼에도 불구하고, 두 종 사이에 드물지만 주목할 만한 차이점이 실유두와 버섯유두에서 나타났다. 검은집박쥐와는 달리 긴가락박쥐의 유형2의 실유두는 두 개 혹은 세 개의 돌기를 가지고 있었다. 혀의 후반부 중앙에 위치하고 있는 큰 버섯유두의 경우, 긴가락박쥐에서는 3개, 검은집박쥐에서는 단지 2개만 관찰되었다.

한국재래산양 혀에 관한 형태학적 연구 (Morphological study on the tongue of Korean native goat)

  • This studies were carried out to identify the characteristics of the tongue of Korean native goat(Capra hircus) by macroscopy, microscopy and scanning microscopy. Korean native goat had torus linguae, median lingual sulcus, lingual fossa and ventral median fissure but did not have glossoepiglottic fold and terminal sulcus in the tongue. The whole length of tongue was $11.51{\pm}0.76cm$. The length of tongue apex, tongue body, tongue root and the torus linguae were $2.62{\pm}0.28$, $7.39{\pm}0.27$, $1.56{\pm}0.26$ and $6.37{\pm}0.29cm$, respectively. The width of tongue apex, torus linguae and tongue root were $3.41{\pm}0.24$, $3.74{\pm}0.29$ and $3.68{\pm}0.11$, respectively. The thickness of tongue apex was $1.60{\pm}0.10$, and the height of torus linguae was $1.52{\pm}0.15cm$. Filiform papillae were present at the tongue apex and the tongue body rostral to torus linguae. Fungiform papillae were scattered from tongue apex to rostral portion of torus linguae, being in abundance at the tongue apex. Vallate papillae were showed at the lateral portion of torus linguae, while lentiform papillae were present at its central portion. Conical papillae were located between vallate and lentiform papillae. The numbers of filiform, fungiform, conical, vallate and lentiform papillae were $46,980{\pm}1070.98$, $446.8{\pm}36.97$, $818.4{\pm}43.99$, $34.8{\pm}2.77$, and $255.6{\pm}39.30$, respectively. The average numbers of taste bud were $8.3{\pm}2.04$ in a fungiform papilla and $247.3{\pm}37.44$ in a vallate papilla. The filiform papilla had secondary and tertiary papillae. The height of filiform papilla was about $150{\mu}m$ and the diameter was $100{\mu}m$. The diameters of fungiform papillae were 350 to $550{\mu}m$. The long and short diameters of maximum-sized lentiform papilla were 4000 and $3000{\mu}m$, respectively, while those of minimum-sized papilla were 700 and $600{\mu}m$, respectively. The height of conical papillae was 450 to $600{\mu}m$ and diameter was 250 to $450{\mu}m$. The vallate papilla was round or oval in shape and its diameter was 500 to $850{\mu}m$. It had well-developed papillary groove around itself. The modified conical papillae were not observed in the tongue of Korean native goat.

Scanning electron microscopy of filiform papillae development in Korean native goats (Capra hircus)

  • The aim of this study was to investigate morphological development of filiform papillae (FP) in Korean native goats by using scanning electron microscopy. Tongues were removed from goat fetuses (days 60, 90, and 120), neonates, and juveniles (days 30, 60, 90, 120, 150, and 180 after birth). During the prenatal period, primordia of FP appeared at fetal day 60 and were observed to be developed at day 90. At fetal day 120, the FP were observed like flower leaves of a double flower bud. In neonates, FP were shaped like an obliquely sectioned cylinder with secondary papillae irregularly arranged in a saw blade-like manner. In 60-day-old juvenile goats, the FP were densely distributed at the inner base of 1/3-1/2 degrees. In 90-, 120-, and 150-day-old goats, FP were compacted at the inner base of 1/2-2/3, 3/4, and 4/5 degrees, respectively. In 180-day-old goats, FP were found to be completely compacted on the inner surface with complete morphogenesis. Microridges, microplicae, and micropits were well-developed on the epithelial surface of lingual papillae from embryonic day 120 to juvenile day 180. These results indicate that FP of goats have different shapes and sizes during development both before and after birth.

Differential Expression of Taste Receptors in Tongue Papillae of DBA Mouse

  • The tongue has 4 kinds of papillae, which are filiform, fungiform (FU), foliate (FO) and circumvallate papilla (CV). Tongue papillae except filiform papilla include taste buds. The papillae differ in taste sensitivities, likely due to differential expression of taste receptors. In this study, we evaluated differences in the expression levels of taste receptors in FU, FO and CV. Male DBA2 mice, 42-60 days old, were used in the study. Messenger RNAs were extracted from the murine epithelial tissues including FU, FO and CV. Cloned DNAs were synthesized by reverse transcription. Quantitative PCRs (qPCRs) were performed to determine mRNA expression levels of taste receptors. Results of qPCR revealed that the relative expression levels and patterns were different among FU, FO and CV. All three type 1 taste receptors were expressed FU, FO and CV at varying relative expression levels. All 35 kinds of type 2 taste receptors showed higher expression in FO and CV than in FU. Tas2r108 and Tas2r137 showed the two highest expression levels in all tested papillae. The differential expression levels and patterns of taste receptors among the three papillae could contribute to the different physiological sensitivities by tongue areas. Additional studies such as in situ hybridization or taste receptor cell activity recording is necessary to elucidate the functional relationship between expression levels of taste receptors and taste sensitivity.

각종 가축 설근부의 조직학적 구조에 대한 비교 연구 (Comparative Study of Histological Structure in the Tongue Root of Domestic Animals)

  • In order to know the comparative histological structure of the tongue of some domestic and laboratory animals, the root of the tongue has been investigated by light microscopy. The filiform papillae were mainly distributed in the root of the tongue of most animals and many foliate papillae were also found in the dog and rabbit. On the other hand, only filiform papillae were found in the fowl. The very well-developed mucosa was found in the cattle and in the dog, fowl, rat and mouse was fair. The lamina propria was well developed in the cattle, dog and pig, The muscle layer was well developed in the cattle. The adipose tissue which distributed between the muscle fibers was mainly found in the dog. The lingual glands were well developed in the pig, fowl and rabbit. The properties of these glands were shown strung acid mucopolysaccharide in the dog and pig, weak acid mucopolysaccharide in the fowl, rabbit and rat and neutral or acid mucopolysaccharide in the mouse. The species differences on the histological structure of the tongue were also ascertained by light microscopical observations.

A RADIOAUTOGRAPHIC STUDY OF POSTNATAL DEVELOPMENT OF THE TONGUE FOLLOWING 5-FLUOROURACIL ADMINSTRATION IN MICE

  • 신생의 Balb/C strain 백서20두를 사용하였고, 실험군과 대조군으로 구분하여 실험군에는 5-fluorouracil의 체중 25 mg/kg씩 2회를 복강내주사하였다. 실험군가 대조군에 모두 희생 2시간전에 체중그람당 5μ Ci의 thymidine-H³ (Specific activity는 9.0 Ci/mM 이상)를 복강내 주사하였다. 각군은 5-fluorouracil 최종 주사후 1, 3, 7, 14, 21일 간격으로 희생시키고, 두부를 4% formalin에 고정하였다. 조직을 0.5M EDTA에 탈회하고 parlodion과 paraplast에 이중 매몰을 하여 Parasagittal serial section 을 10μ의 절편을 만든 후 자기방사용표본을 제작하였다. 그 결과는 다음과 같다. 1. 5-fluorouracil이 백서설의 기저세포층의 핵산합성 및 단백합성에 미치는 영향은 실험초일인 제 1일부터 억제하기 시작하여 (80.9%) 제3일 76.9%이고, 제 7일이 가장 극심하고 (66.2%) 그후부터는 다소회복되기 시작하여 제14일이 92/3%이고, 제21일인 98.9%로서 서의 대조군수치에 접근하였다. 2. 5-fluorouracil은 설유두중 nallate papillae 성장에 가장 억제적 현상을 보였고, 그 다음이 fungiform papillae, filiform papillae의 순위였다. 3. 5-fluorouracil 이 백서설의 기저세포층의 핵산합성을 억제함을 알 수 있고, 아울러 백서설의 조기정상 발육에 영향을 줌울 알 수 있다.

  • This study was carried out to investigate the morphological development of the tongue in the 60-, 90- and 120-day-old fetuses and neonates of Korean native goats by light microscopy. In 60-day-old fetuses, the tongue tissues were differentiated into epithelium, lamina propria, and muscle layer. The primordia of filiform, conical, lentiform, fungiform, and vallate papillae appeared by this time, and rudiments of taste buds were observed in the epithelia of the primordia gustatory papillae. The dorsal lingual epithelia were PAS positive. Collagenous fibers and blood vessels were present in the lamina propria. In 90-day-old fetuses, the gustatory glands were moderately positive for PAS and muscle fibers, and connective tissues were developed. In 120-day-old fetuses, many taste buds were observed in the gustatory papillae. The muscle bundles, collagenous fibers, blood vessels and gustatory glands were well developed. In neonates, over 40 taste buds were found in a transverse section of the vallate papillae. Muscle layers, blood vessels, collagenous fibers and gustatory glands were very well developed.
