사상성 조류매트 산화지의 수질정화효율 (Pollutant Removal Efficiency in Oxidation Pond with Filamentous Algae Mat)

  • This study was carried out to evaluate of water purification in oxidation pond with filamentous algae mat. It is the water treatment process in the small rural streams to remove the organic materials and nutrients. We used the filamentous algae mat(FAM) which selectively predominate the filamentous algae to prevent the additional contamination by algae outflow. The removal efficiencies of COD, SS, T-N and T-P in Oxidation Pond with Filamentous Algae Mat were -2.5%, 84.7%, 63.9% and 89.2%, respectively. The removal efficiencies of T-N and T-P which are nutrients index were high. Results of this study would help us to determine the possibility of using the water treatment on the contaminated small rural streams.

섬모상 매트에 의한 탁수이동차단에 관한 실험적 연구 (Control of Turbid Water Transport with Filamentous Mat)

  • A lab-scale apparatus for turbid water transport control was tested and examined. The channel had a dimension of $100cm{\times}30cm{\times}15cm$ (length${\times}$hight${\times}$width). And the turbidity water was prepared using two types of particles, bentonite and loess. The channel equipped with filamentous mat was operated under various shock load conditions. In the control channel, instantly, turbid water mixed with the clean water inside the channel and turbidity prevails the entire channel. While in the mat-equipped channel, it increases only at the bottom. Overall, the filamentous mat gave capture efficiency of 70~90% compared with the control group. The capture efficiency of turbid particles decreased with increased input turbidity flux. The result of experimental run on how turbid particles are separated in the mat channel shows that settling, filtration and attachment are the main processes. Meanwhile, turbidity was diffused from the channel bottom due to turbidity gradient before and after mat zone. The particle size before mat zone was lightly coarser than that after mat zone.

  • This study presented a lab-scale apparatus for turbidity control in the lakes or reservoirs. Overall, the filamentous mat had a capture efficiency of 70~90% compared with the control group. Generally, the capture efficiency decreased with improved input turbidity flux. However, the attachment and sedimentation were thought to be the main processes for turbidity water retention and removal. Thus, the increase of hydraulic detention time in the mat zone is very important to improve the capture efficiency of the turbidity water.

저수지 유입하천 현장적용실험을 통한 수질정화효율 평가 (Evaluation of Pollutant Removal Efficiency through Field Test-Bed Experiment in the Rural Small Stream)

  • 본 연구에서는 사상성 조류로 구성된 수질정화시설의 수질정화효율을 평가하고자 충남 서산시 음암면에 소재한 성암저수지 유입하천 홍수부지에 test-bed를 설치하고 유입하천수를 대상으로 현장적용실험을 실시하였다. 수질항목별로 정화효율을 평가한 결과, SS 80.9%, COD 74.6%, T-N 76.8%, T-P 84.4%, DTN 93.8%, DTP 98.3%의 제거효율을 보였다. 수온에 따른 정화효율은 수온이 약 $20^{\circ}C$로 유지되어 수질정화효율에 미치는 영향은 없는 것으로 나타났다. 수리학적 체류시간(HRT)은 SS 및 TP 처리효율과 유의한 상관성이 있는 것으로 나타났고, 체류시간을 5일로 했을 때 SS, TP 처리효율이 좋은 것으로 나타났다. 유입수의 COD/TN에 따른 처리효율은 TN, TP 모두 C/N비가 낮을수록 증가하였고, 유입수의 COD/TP에 따른 처리효율도 C/P비가 낮을수록 TN, TP 모두 처리효율이 증가하는 것으로 나타났다.

New report on cyanophyte in Korea, Microseira wollei (Farlow ex Gomont) G.B.McGregor and Sendall ex Kennis (Oscillatoriaceae)

  • Microseira wollei (Farlow ex Gomont) G.B.McGregor and Sendall ex Kennis, a mat-forming filamentous harmful cyanobacterium, has historically been found in the United States. Microseira wollei produces neurotoxins and hepatotoxins which affect declining water quality. In the present research, we report of unrecorded M. wollei with morphology, TEM anatomy, molecular phylogeny on the Korean population. Based on 16S rRNA gene sequences, Korean population were different by 0.02% (2 bp) to the Japanese population, 1.2-1.3% to the Australian population, and 2.5-3.7% to the United States populations. nifH gene sequences were 8.4-8.7% different to Australian ones and 3.5-3.8% to other population, however molecular phylogenetic analysis of M. wollei living in Korea revealed monophyly with the geographical populations of U.S.A., Australia, and other geographical populations. Since the mat of M. wollei has been reported to be maintained for several years in other countries, it is necessary further investigate the seasonal and regional distribution of this species in Korea.

Ophiostomatoid Fungi in Pine Wilt Disease and Oak Wilt Disease in Korea

  • Pinewood nematode (PWN, Bursaphelenchus xylophilus) is a serious pathogenic worm that quickly dry pine trees to death. Recently, PWN has been devastating huge amounts of conifer trees in Korea. As a first step to explore the association and ecological roles of fungi in PWN life cycle in Korea, in this study we first isolated and indentified fungi from PWN-infested Korean pine and Japanese black pine wood sampled in Jinju, Sacheon, Pocheon, Chuncheon, Gwangju, and Hoengseong in Korea. A total of 144 fungal isolates were obtained from Japanese black pine wood and 264 fungal isolates from Korean pine wood. Their morphology and nucleotide sequences of the ITS rDNA and ♌-tubulin gene were examined for species identification. Ophiostoma ips, Botrytis anthophila, Penicillium sp., Hypocrea lixii, Trichoderma atroviride, O. galeiforme, Fusarium proliferatum were identified from Japanese black pine wood. Leptographium koreanum, L. pini-densiflorae, Ophiostoma ips, Penicillium raistrick, Trichoderma sp. were isolated from Korean pine wood. O. ips and L. koreanum were the major species on the two different PWN-infected pine tree. The cultivation of PWN on fungal mat of the identified species did some enhance PWN reproduction. The ambrosia beetle, Platypus koryoensis, is a serious pest of oak trees in Korea. In this study we investigated filamentous fungi present in the body of the beetle. Fourteen genera of filamentous fungi belonging to Ascomycota and Basidiomycota were isolated. All the obtained genera were isolated in the mitosporic state. The identified fungi were classified in 11 distinct orders including the Ascomycota (Eurotiales, Hypocreales, Microascales, Ophiostomatales, Pleosporales, and Sordiales) and Basidiomycota (Agaricales, Corticiales, Polyporales, and Russulales Xylariales). Within Ascomycota, 13 species were found. Meanwhile five species were found within Basidiomycota. The results showed the presence of diverse fungi in P. koryoensis. Among the isolated fungi, some were able to produce wood degrading enzymes. Further fungal isolation was performed with P. koryoensis infested Quercus mongolica trees sampled at Kumdan mountain in Hanam-Si, Gyeonggi province from June of 2009 to June of 2010. Penicillin spp. and Trichoderma spp. were the major species of mold fungi group. Pichia guilliermondii was the major species of mold yeast group. Raffaelea quercus-mongolicae was also isolated, but its isolation frequency was not high. Other species identified were Ambrosiella xylebori, Fusarium solani, Cryphonectria nitschke, Chaetomium globosum, and Gliocladium viride, Candida kashinagacola, C. maritima, C. vanderkliftii, Saccharomycopsis crataegensis.

Colletotrichum fructicola, a Member of Colletotrichum gloeosporioides sensu lato, is the Causal Agent of Anthracnose and Soft Rot in Avocado Fruits cv. "Hass"

  • The filamentous Ascomycota Colletotrichum gloeosporioides sensu lato is a fungus that has been reported worldwide as a causal agent of anthracnose disease in avocado and other crops. In Mexico, this species affects fruits from an early stage of development in the orchard until the post-harvest stage. Although fungicides are continuously applied to control Colletotrichum species, pericarp cankers and soft rot mesocarp in fruits are still frequently observed. Considering the lack of a precise description of the causative agent, the aim of the current study was to determine the pathogens involved in this symptomatology. Twenty-four isolates were consistently obtained from the pericarp of avocado fruits cv. "Hass" collected in the central avocado-producing area of Mexico. Morphological features such as colony growth, conidia size, and mycelial appressorium were assessed. Bayesian multilocus phylogenetic analyses were performed using amplified sequences of the internal transcribed spacer region of the nuclear ribosomal DNA; actin, chitin synthase, glyceraldehyde-3-phosphate dehydrogenase partial genes; and APn2-Mat1-2 intergenic spacer and mating type Mat1-2 partial gene from the nine selected isolates. In addition, fruits were inoculated with a conidial suspension and reproducible symptoms confirmed the presence of Colletotrichum fructicola in this area. This pathogenic species can now be added to those previously reported in the country, such as C. acutatum, C. boninense, C. godetiae, C. gloeosporioides, and C. karstii. Disease management programs to reduce the incidence of anthracnose should include C. fructicola to determine its response to fungicides that are routinely applied, considering that the appearance of new species is affecting the commercial quality of the fruits and shifting the original population structure.
