The skin, the largest organ in the body, undergoes age-related changes influenced by both intrinsic and extrinsic factors. The primary external factor is photoaging which causes hyperpigmentation, uneven skin surface, deep wrinkles, and markedly enlarged capillaries. In the human dermis, it decreases fibroblast function, resulting in a lack of collagen structure and also decreases keratinocyte function, which compromises the strength of the protective barrier. In this study, we found that treatment with γ-aminobutyric acid (GABA) had no toxicity to skin fibroblasts and GABA enhanced their migration ability, which can accelerate skin wound healing. UVB radiation was found to significantly induce the production of matrix metalloproteinase 1 (MMP-1), but treatment with GABA resulted in the inhibition of MMP-1 production. We also investigated the enhancement of filaggrin and aquaporin 3 in keratinocytes after treatment with GABA, showing that GABA can effectively improve skin moisturization. In vivo experiments showed that oral administration of GABA significantly improved skin wrinkles and epidermal thickness. After the intake of GABA, there was a significant decrease observed in the increase of skin thickness measured by calipers and erythema. Additionally, the decrease in skin moisture and elasticity in hairless mice exposed to UVB radiation was also significantly restored. Overall, this study demonstrates the potential of GABA as functional food material for improving skin aging and moisturizing.

Ozone, a highly reactive oxidant molecule, is widely used as a complementary therapy for various skin diseases, including wound healing, pressure ulcers, diabetic foot, and infections. However, there is limited research on the effectiveness of ozone for atopic dermatitis (AD). Ozonated sunflower oil (OSO) is an active ingredient obtained from partially ozonated sunflower oil (SO). OSO markedly reduced the LPS-induced increase in IL-1β and nitric oxide (NO) levels in RAW 264.7 mouse macrophage cells. Oxazolone (OXZ) was applied to hairless mice to induce AD-like skin symptoms and immune response. OSO significantly alleviated the OXZ-induced increases in the number of infiltrating mast cells, epidermal thickness, AD symptoms, thymic stromal lymphopoietin (TSLP), and filaggrin, as well as the serum levels of NO, IgE, IL-1β, and TNF-α. Furthermore, OSO inhibited the IL-4/STAT3/MAPK pathway and the expression of NF-κB. Our results suggest that OSO treatment could relieve AD-mediated skin damage through its anti-inflammatory and antioxidant activities. Therefore, it can be used as a therapeutic agent against AD-related skin diseases.
본 연구에서는 이너뷰티용 소재로서의 활용가능성을 확인하기 위하여 생물전환 가시오가피-진피 혼합추출물(B-MEAC)의 항산화 활성, 엘라스타제 억제활성 및 피부보습 효과 등을 평가하였다. B-MEAC의 항산화 활성을 측정한 결과 DPPH radical을 농도의존적으로 소거하여 SC50이 156.1±0.82 ㎍/ml로 나타났다. 또한, 엘라스타제 억제활성을 측정한 결과 B-MEAC 처리 농도 의존적으로 엘라스타제 활성을 억제하는 것으로 확인되었다(p<0.001). 경구투여한 B-MEAC가 마우스 피부 보습에 미치는 영향을 확인하기 위하여 피부 수분량과 경피수분손실량을 측정한 결과 건조유도 된 대조군에 비하여 피부 수분량은 증가하고(p<0.001), 경피수분손실량은 감소하는 것으로 나타났다(p<0.01). 또한, 건조유도 된 마우스 피부의 교원섬유 변화에 미치는 영향을 Masson's trichrome 염색을 통해 관찰한 결과 B-MEAC를 투여한 군에서 콜라겐의 양이 대조군에 비해 상대적으로 증가하여 푸른색으로 보다 진하게 염색되었다. 또한, 건조유도 된 마우스 피부의 보습 기능에 미치는 영향을 확인하기 위하여 보습 관련 인자들의 발현을 Western blot 기법으로 확인한 결과 B-MEAC를 투여한 군에서 대조군에 비하여 MMP-1 단백질의 발현량은 감소하였고, COL1A1, HAS2, filaggrin, AQP3 단백질 발현량은 회복되는 것으로 나타났다(p<0.001). 따라서 B-MEAC의 보습효과는 엘라스타제 활성 억제, MMP-1 발현 감소, 콜라겐 생성 촉진, 피부 보습 관련 단백질인 HAS2 및 AQP3 발현 증가, 피부 장벽기능의 중요인자인 filaggrin 발현 증가를 통한 수분량 증가와 TEWL 감소에 의해 나타나는 것으로 이너뷰티 소재로서의 개발 가능성을 제시한다.
본 연구에서는 천연물 6 종 앵두, 으름, 살구, 야콘, 금귤 및 핑거라임을 활용하여 스트레스 완화에 의한 수면 개선 및 피부장벽 개선 효과를 검증하였다. 실험 결과, 6 종의 천연물 중 앵두, 핑거라임, 으름 및 금귤이 유의적으로 스트레스 호르몬인 cortisol의 생성을 억제하였다. 또한, 수면 조절에 관여하는 GABA 생성 효소인 GAD67의 발현은 앵두 정제수 추출물 및 핑거라임 50% 에탄올 추출물에서 유의한 증가 효과를 보였으며 앵두 용매별 추출물은 총 폴리페놀의 함량이 가장 높게 확인되었다. 스크리닝 결과 활성이 가장 높은 앵두 추출물을 활용하여 후속 실험을 진행하였다. 앵두 용매별 추출물 중, 30% 에탄올 추출물이 가장 높은 DPPH radical 소거 활성을 보였으며, 정제수 추출물은 GABA 생성 및 피부 장벽 인자인 filaggrin과 claudin-1의 발현을 가장 높게 증가시켰다. HPLC 분석을 통해 앵두 추출물의 주성분으로 quercitrin이 확인되었고 추출 용매별 quercitrin 함량은 30% 에탄올 > 정제수 > 70% 에탄올 > 50% 에탄올의 추출 용매 순으로 높은 함량을 보였다. Quercitrin은 cortisol의 생성을 농도의존적으로 억제하였으며, cortisol에 의해 감소되었던 GAD67 발현 및 GABA 생성을 유의적으로 증가시켰다. 연구를 통해 앵두 추출물이 스트레스를 완화시켜 수면 개선 및 피부 장벽 강화에 도움을 주는 화장품 소재로서 활용이 가능함을 입증하였다.
본 연구에서는 lactic acid (LA) 및 gluconolactone (GL)의 피부 장벽 개선 효능을 평가하기 위하여 피부세포에서 필라그린, 로리크린, hyaluronic acid (HA), hyaluronan synthase-2 (HAS2), aquaporine-3 (AQP3)의 발현량 및 임상시험을 통한 수분 함유량과 경표피수분손실량(transepidermal water loss, TEWL)을 평가하였다. 장벽 기능(필라그린, 로리크린)과 보습 기능(HA, HAS2, AQP3)에 관련된 인자들의 발현량을 qRT-PCR과 Western blot으로 측정한 결과, H2O2 처리에 의해 감소된 인자들의 발현량이 LA, GL 및 혼합물 처리로 mRNA전사량과 단백질 발현량을 유의적으로 증가하는 것으로 나타났다(p<0.05). LA와 GL 혼합물을 함유하는 나노에멀젼을 에멀젼 반전법으로 제조한 결과 평균 입자 크기는 299.9±0.287 nm로 확인되었다. 나노에멀젼의 TEWL을 Vapometer로 측정한 결과, 제품 사용 2주 후 및 제품 사용 4주 후에 제품 사용 전에 비하여 각각 15.53%, 26.73% 정도 TEWL이 감소하는 것으로 나타났다(p<0.001). 나노에멀젼의 피부 수분함유량을 Corneometer로 측정한 결과, 제품 사용 2주 후 및 제품 사용 4주 후에 제품 사용 전에 비하여 각각 15.40%, 26.59% 정도 수분 함유량이 유의적으로 증가하는 것으로 나타났다(p<0.001). 따라서 LA 및 GL의 피부 장벽 기능 및 보습 효과는 피부 보습 관련 단백질인 HA, HAS2 및 AQP3 발현 증가와 피부 장벽 기능의 주요인자인 필라그린과 로리크린의 발현 증가를 통한 수분 함유량 증가 및 TEWL 감소에 의해 나타나는 것으로 기능성 화장품 소재로서의 개발 가능성을 제시한다.
자외선(Ultraviolet radiation)은 피부 주름 형성과 피부 건조를 특징으로 하는 광노화의 주요 원인이다. 모자반과(Sargassaceae)에 속하는 모자반(Sargassum fulvellum)은 항염증, 항산화, 항아토피 활성을 갖고 있다. 특히, 모자반은 인간 각질세포(keratinocyte)와 실험동물의 피부에서 UVB에 의한 염증 반응을 완화시키는 것으로 보고되었다. 그러나, 현재까지 모자반 추출물의 광노화 억제 효과에 대해서는 보고된 바가 없다. 본 연구에서는 광노화를 유도하기 위하여 UVB를 SKH-1 무모 마우스의 등 부분에 8주간 처리하였으며, 동시에 모자반 추출물을 300 mg/kg의 농도로 매일 경구 투여하였다. 현상학적으로, 모자반 추출물은 주름 형성, 홍반 및 피부 두께를 감소시켰으며, 피부 탄력을 증가시켰다. 모자반 추출물은 콜라겐 합성 유전의 발현을 증가시키고, matrix metalloproteinase의 발현을 감소시킴으로써 콜라겐의 하이드록시프롤린(hydroxyproline) 함량을 증가시켰다. 추가적으로, 모자반 추출물의 경구 투여는 무모 마우스의 피부 수분 함량을 증가시켰으며, 경피수분손실(transepidermal water loss)을 감소시켰다. 모자반 추출물은 각질세포막 형성에 관여하는 involucrin, loricrin, transglutaminase의 발현과 자연보습인자 관련 생체지표인 filaggrin 및 caspase-14의 발현을 증가시켰다. 종합적으로, 모자반 추출물은 피부의 콜라겐 손실과 건조를 완화함으로써 광노화 억제 활성을 제공하였다. 모자반 추출물은 피부 광노화를 억제하기 위한 기능성 식품 소재로서 활용될 수 있을 것으로 판단된다.
천연보습인자(NMF, natural moisturizing factor)는 정상 피부 내의 각질층에 존재하는 수용성, 친수성 성분을 총칭하는 말로 외부로부터 수분을 흡수하고 각질층의 수분보유능을 증가시키며 피부가 건조해지는 것을 방지하여 피부의 유연성과 가수성을 유지하는 기능을 한다. NMF는 주로 아미노산과 아미노산의 대사산물로 구성되며 이들은 필라그린(filaggrin)이 분해되면서 생성된다. 본 연구에서는 피부 각질층의 22종의 개별 아미노산의 함량분석을 통하여 피부보습능과 개개의 아미노산과의 상관관계를 연구하였다. 15명의 건강한 지원자들의 전박과 이마로부터 tape stripping한 각질 시료 중 22종의 아미노산 함량을 UPLC-PDA를 이용하여 동시 분석하였다. 그 결과 각질 표면보다는 안쪽에서의 아미노산들의 함량이 높음을 확인하였다. 또한 신체 부위별 비교에서 전박이 이마에 비해 아미노산 함량이 1.5배 정도 높음을 확인하였다. 전박부위의 아미노산 총 함량과 보습능(TEWL)과의 상관 관계분석을 통해서는 보습능이 클수록 총아미노산 함량이 높은 것으로 확인되었으며 특히 Ser, Glu, Gly, Ala 및 Thr은 각질 내 존재하는 주요 아미노산 성분으로 보습에 긍정적인 영향을 주는 성분으로 확인되었다. 이상의 결과로써 NMF의 주성분인 아미노산의 함량은 각질 생리기능과 밀접한 관련을 가지고 있음을 확인할 수 있었으며 본 연구를 통해 얻은 각질층의 아미노산의 통합적인 분석법은 보습, 노화, 미백, 피부 염증질환 등 다양한 피부 상태와 아미노산류들의 변화와의 관계를 깊이 이해하고 피부 기능과 관련된 새로운 분자적 타겟을 발굴하고 증명하는데 기초적인 방법으로써 사용될 수 있을 것으로 사료된다.
UV irradiation is the main factor contributing to skin damages that are associated with an excessive production of matrix-degrading metalloproteinase (MMP)-1 and a deficient expression of collagens. To date, red ginseng has been revealed to possess many biomedical effects, such as anti-aging, anti-oxidation, and anti-inflammatory. In this study, we prepared the Korean Red Ginseng extracts treated with enzyme (KRGE) and investigated the effects of dietary KRGE on the formation of wrinkles generated by UVB irradiation in hairless mice. It was found that KRGE inhibited the UVB-induced formation of wrinkles, epidermal thickness, and skin dryness in hairless mice. Further results also showed that KRGE attenuated UVB-induced MMP-${\beta}$1 level, while accelerated procollagen type I, transforming growth factor-${\beta}$1 secretion. Interestingly, the expression of profilaggrin and filaggrin in both the epidermis and dermis were decreased due to UVB exposure and reversed by KRGE. The KRGE 0.06% was prior to KRGE 0.24%. In view of these results, which indicated that KRGE protected skin from UVB-induced photodamages, which may not only mediated by regulating of MMP-1 and procollagen type I, but also by increasing the production of profilaggrin and filaggrin. In conclusion, our results suggest that KRGE may be a promising agent for the treatment of skin photodamages. The challenge of KRGE will be expected as cosmeceuticals and nutraceuticals in order to intervene in aging-related degenerative skin changes.
Objectives : This study was designed to examine the effects of Daecheonglyong-tang(DCL) on atopic dermatitis induced by DNCB in mice Methods : The Nc/Nga mice were divided into 5 groups, and four groups excluding the normal group were applied by 2,4-dinitrochlorobenzene(DNCB), to cause AD and were orally administered with distilled water(negative control), dexamethasone(positive control), and DCL 200 or 400mg/kg once a day for 4 weeks respectively. The visual changes on skin, changes in skin tissue thickness and eosinophil infiltration were observed. IgE, Histamine, Cytokines, immune cells and the amount of gene expression of filaggrin, VEGF, $TGF-{\beta}1$, EGF were measured. Results : Dermatitis score showed a gross improvement on all DCL groups, similar to or better than positive control. All DCL groups showed no significant change in the basophils, while neutrophils and eosinophils decreased. In only DCL 400 mg/kg groups, white blood cells and mononuclear cells were decreased and lymphocytes were increased. In particular, neutrophils had similar or better effects than the positive control. In all DCL groups, IgE, Histamine, $IL-1{\beta}$, IL-4, IL-5, IL-6 and $TNF-{\alpha}$ were decreased and IL-2 was increased. In only DCL 400 mg/kg groups, IL-10 decreased and $IFN-{\gamma}$ increased. In particular, $IL-1{\beta}$ and $IFN-{\gamma}$ showed a similar rate of increase and decrease comparing positive control in DCL 400 mg/kg. $TGF-{\beta}$1 was increased in all DCL groups, filaggrin and VEGF were increased in only DCL 400 mg/kg groups. EGF did not make any changes. Epidermis, dermis thickness and eosinophil infiltration were also decreased in all DCL groups. Conclusions : By increasing Th1 cytokine and decreasing Th2 cytokine, DCL extracts appear to be effective in controlling immune response imbalances, anti-inflammatory and skin regeneration and are likely to be available as a treatment for AD.
레몬그라스와 자소엽 추출물은 다양한 생리 효과를 나타내는 것으로 알려져 있지만, 피부보습과 피부장벽에 미치는 영향은 아직까지 연구되지 않았다. 본 연구에서는 레몬그라스와 자소엽 추출물의 피부보습과 피부장벽에 미치는 영향과 페놀성 화합물을 분석하였다. 피부각질형성세포에서 각 추출물이 피부보습에 미치는 영향을 조사한 결과, 두 추출물 모두 물보다 에탄올 추출물에서 히알루론산 생성이 많았다. HPLC를 이용한 19종 페놀성 화합물 분석 결과는 레몬그라스 에탄올 추출물(CCE)에서 chlorogenic acid와 p-coumaric acid가 검출되었으며 자소엽 에탄올 추출물(PFE)에서 rosmarinic acid와 caffeic acid가 검출되었다. 피부보습에 관련된 HAS1, HAS2, HAS3 및 AQP3와 피부장벽에 관련된 filaggrin, loricrin 발현은 PFE보다 CCE에서 높았다. 또한, CCE, PFE 모두 피부보습과 표피분화 조절에 관여하는 $PPAR-{\alpha}$ 단백질의 발현이 농도 의존적으로 증가하였으며 CCE의 주요성분인 chlorogenic acid와 p-coumaric acid가 $PPAR-{\alpha}$ 발현을 증가시켰다. 결론적으로 피부보습과 피부장벽보호 효과에 있어서 CCE가 PFE보다 우수한 효과를 나타내었고, 두 추출물은 피부보습과 피부장벽개선에 대한 기능성 소재로써 활용될 수 있을 것이라 생각된다.

회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
