Phytochemical Compounds from the Underground Parts of Gardenia jasminoides var. radicans Makino (꽃치자 지하부의 식물화학적 성분)

  • The phytochemical study of the underground parts of Gardenia jasminoides var. radicans Makino (Rubiaceae) led to the isolation of five coumarins. On the basis of spectroscopic evidences, the structures of these compounds were charaterized as ferulic acid(l), 5,8-di-(3-methyl-2,3-dihydroxybutyloxylpsoralen)(2), skimmin(3), uracil(4), $3-0-{\alpha}-D-glucopyranosyl-(1{\rightarrow}4)-{\beta}-D-glucopyranosyloxypeucedanin$ hydrates(5), respectively.

Analysis of Phenolic Acids in Tobacco Leaf by GC/MS (GC/MS에 의한 잎담배중 Phenolic Acid의 분석)

  • A GC/MS method was developed to analyze phenolic acid extract from tobacco leaf. Extracted acids were converted to their methyl esters by refluxing with 3M hydrogen chloride in methanol, and the esters were reacted with his (trimethylsilyl) trifluoroacetamide plus 10% trimethylchlorosilane to silylate the phenolic groups. Derivatives of standard salicylic, p-hydroxybenzoic, vanillic, gentisic, p-coumaric, syringic, ferulic, and sinapic acids prepared by this procedure were analyzed by GC/MS on $20m{\times}0.2mm$ column of SE-54 glass capillary. GC/MS analysis of the extract from tobacco leaf revealed the presence of salicylic, p-hydtoxybenzoic, vanillic, gentisic, protocatechuic, p-coumaric, syringic, gallic, ferulic, caffeic, sinapic, and quinic acids, respectively. The quantitative analysis of these phenolic acids were achieved by using multiple ion selection technique.

Antioxidative Effectiveness of Terminalia chebula Rets Extracts (가자(Terminalia chebula Retz)추출물의 항산화 효과)

  • In this study, antioxidative effectiveness of BHA, BHT at 0.02%(w/w) was compared with those of separated free phenolic acid, ester form and insoluble bound phenolic acid which were extracted from 50 g of Terminalia chebula Retz by MeOH/aceton solvents. Antioxidative effectiveness was measured by peroxide values and TBA values for 7 days, storaging respective substrates and contrast tube at $45{\pm}1^{\circ}C$ for 35days. Laboratory tubes was added by BHA, BHT, separated free, soluble and insoluble phenolic acid extracts and peroxide value of contrast tube after 21 day storage were 60, 30, 14, 11, 100. On the other hand, at the same conditions, TBA values of each antioxidants were 0.150, 0.108, 0.105, 0.073, 0.078, 0.185. This results remarkably appeared antioxidative effectiveness in meal soybean oil substrates. Phenolic acid separated and identificated were p-coumaric acid, Ferulic acid, Phloroglucinol, Pyrogallol, Vanillic acid and Caffeic acid.

도정도를 달리한 백미 및 미강에 함유된 phenolic acids와 총 폴리페놀 함량

  • 최근 쌀 소비량이 감소되고 쌀중심 식생활의 영양적 우수성을 인식하지 못하여 쌀은 주식으로서 위치가 흔들리고 있다. 그동안 쌀의 영양특성에 관한 연구로서 주로 탄수화물 급원으로서의 당질에 관한 연구와 미강을 중심으로한 식이섬유, 미강유등에 관한 연구결과들이 발표되었다. 특히 미곡 부산물인 미강으로부터 다양한 유효성분들이 확인되어 그 효능이 보고되고 있으며 일부가 기능성 소재로서 제품화되기도 하였다. 그러나 실제 우리의 식생활에서 섭취되고 있는 백미 또는 현미상태에서의 유효성분의 분포 및 함량에 대한 연구는 미진한 상태이다. 본 연구에서는 쌀의 영양적우수성을 규명하고자 유효성분 중 phenolic acid와 폴리페놀 함량을 품종 및 도정도에 따라 조사하였다. 조생종인 오대와 만생종인 남평 품종으로부터 현미와 7분도, 10분도 및 12분도로 도정도를 달리한 백미, 도정도별 미강을 각각 제조하였다. Ferulic acid 등 쌀과 미강에 존재하는 phenolic acid 는 알칼리로 추출한 후 pH를 조정하고 에틸아세테이트로 반복 추출하여 HPLC로 분석하였다. 현미에 함유된 phenolic acid는 ferulic acid가 가장 많아 50% 내외를 차지하였으며 p-coumaric acid, benzoic acid, sinapinic acid 순으로 검출되었다. 오대 현미의 총 phenolic acid 함량은 65.9 mg%로서 남평 현미의 57.2 mg%보다 높았으며 도정도가 증가할수록 백미 중의 총 phenolic acid 함량은 감소되었다. 미강 중에는 백미의 10배 량에 해당하는 phenolic acid가 검출되었고 benzoic acid와 m-hydroxy benzoic acid는 미강 시료에서만 검출되었다. 도정한 백미 중의 phenolic acid는 28.8∼51.7 mg%, 미강에서 321.4∼438.4 mg% 범위로 나타났다. 현미, 백미 및 미강에 함유된 총 폴리페놀의 함량을 표준 페놀화합물로 카테친을 사용하고 비색법에 의하여 측정하였을 때 오대 현미의 폴리페놀 함량은 78.4 mg%, 남평 현미 88.8 mg% 였다. 도정한 백미 중의 총 폴리페놀 함량은 30.3∼56.9 mg%, 미강이 541.5∼472.6 mg%의 범위였다. 이상과 같이 쌀에는 phenolic acid 및 총 폴리페놀이 상당량 함유되어 있으며 특히 배유보다는 강층에 많이 존재하므로 이들 성분의 효율적인 이용을 위한 쌀의 섭취방안이 필요한 것으로 나타났다.

Phenolic Compounds and Antioxidant Activity in Cell Wall Materials from Deodeok (Codonopsis lanceolata) (더덕 부위별 세포벽 물질의 페놀성화합물과 항산화 활성)

  • This study measured the amounts of uronic acid, total sugars, non-cellulosic neutral sugars, phenolic compounds as well as antioxidants activity in cell wall materials (CWM) derived from different parts of deodeok (Codonopsis lanceolata). The values of the uronic acid (UA): neutral sugars (NS) ratio in polymers extracted from the CWM of the flesh and skin were 4 and 6, respectively. The total sugar contents of the flesh and skin were 788.6 and 824.9 ${mu}g/mg$ of CWM, respectively. Galactose and arabinose were the main noncellulosic neutral sugars. The chemical structure of five phenolic compounds from the CWM were analyzed and identified as vanillic acid, p-OH-benzaldehyde, vanillin, ferulic acid, and 8-O-4' diferulic acid by HPLC spectral data. Among them, p-OH-benzaldehyde, vanillin, and 8-O-4' diferulic acid were the first compounds identified from the deodeok. The content of 8-O-4' diferulic acid in the skin CWM was 56.1 ${mu}g/g$ AIR (alcohol insoluble residue). The ethanol-NaOH fractions from CWM had the highest oxygen radical absorbance capacity (ORAC) activities, followed by the AIR fractions and ethanol fractions.

Influence of ferulic acid and clinoptilolite supplementation on growth performance, carcass, meat quality, and fatty acid profile of finished lambs

  • This study was conducted to evaluate the effect of ferulic acid (FA) and clinoptilolite (CTL) supplementation on the growth performance, carcass characteristics, and meat quality of hair-breed lambs. Twenty-eight Kathadin male lambs (33.72 ± 3.4 kg) were randomly allocated to one of the four diets (n=7) under a 2 × 2 factorial arrangement to evaluate the effect of FA (0 or 300 ppm) and CTL (0% or 1%) during the last 40 days of the finishing phase. No interaction between additives was shown for growth performance, carcass characteristics and meat quality, with exception of the fatty acid profile (p < 0.05). FA reduced feed intake and carcass conformation (p < 0.05). Wholesale cuts were not affected by FA or CTL (p > 0.05). The L*, a*, and C* color parameters and some intramuscular fatty acids of the longissimus thoracis muscle were positively modified by CTL supplementation (p < 0.05). While there was no FA × CTL interaction, each additive could be used individually in animal nutrition to improve the feedlot performance and meat quality of the lambs.

Effects of Dietary Supplementation with Ferulic Acid or Vitamin E Individually or in Combination on Meat Quality and Antioxidant Capacity of Finishing Pigs

  • This study aimed to evaluate the effects of vitamin E (VE), ferulic acid (FA) and their combination supplementation on meat quality and antioxidant capacities of finishing pigs. Sixty barrows were randomly allocated to four experimental diets using a $2{\times}2$ factorial arrangement with 2 VE supplemental levels (0 or 400 mg/kg) and 2 FA supplemental levels (0 or 100 mg/kg) in basal diets. After 28 days, six pigs per treatment were slaughtered. The results showed that VE supplementation increased loin eye area of pigs (p<0.05) and FA supplementation increased $pH_{45min}$ value (p<0.05). The interaction of $FA{\times}VE$ was observed in shear force of longissimus dorsi muscle (p<0.05). Moreover, supplementation with VE decreased hepatic and sarcous malondialdehyde (MDA) content, increased hepatic glutathione (GSH) content and sarcous glutathione peroxidase (GSH-Px) activity (p<0.05). Additionally, supplementation with FA increased hepatic GSH-Px activity and decreased sarcous MDA content (p<0.05). However, dietary treatment did not affect the expression of genes related to nuclear factor, erythroid 2-like 2 (NFE2L2) pathway. These results suggest that dietary FA and VE could partially improve meat quality and antioxidant capacity of finishing pigs, but not by activating NFE2L2 pathway under the normal conditions of farming.

Synergistic Antibacterial Effect and Antibacterial Action Mode of Chitosan-Ferulic Acid Conjugate against Methicillin-Resistant Staphylococcus aureus

  • We evaluated the synergistic antibacterial effect in combination with the chitosan-ferulic acid conjugate (CFA) and β-lactam antibiotics, such as ampicillin, penicillin, and oxacillin, against methicillin-resistant Staphylococcus aureus (MRSA) using fractional inhibitory concentration (FIC) indices. CFA clearly reversed the antibacterial activity of ampicillin, penicillin, and oxacillin against MRSA in the combination mode. Among these antibiotics, the combination of oxacillin-CFA resulted in a ΣFICmin range of 0.250 and ΣFICmax of 0.563, suggesting that the oxacillin-CFA combination resulted in an antibacterial synergy effect against MRSA. In addition, we determined that CFA inhibited the mRNA expression of gene mecA and the production of PBP2a, which is a key determinant for β-lactam antibiotic resistance, in a dose-dependent manner. Thus, the results obtained in this study supported the idea on the antibacterial action mechanism that oxacillin will restore the antibacterial activity against MRSA through the suppression of PBP2a production by CFA.

Extractives from the Allium victorials (산마늘(Allium victorialis)의 추출성분)

  • Allium victorials var. platyphyllum were ground, extracted with 95% EtOH, concentrated, and fractionated with a series of n-hexane, methylene chloride, ethyl acetate, butanol and water on a separatory funnel. A portion of ethyl acetate soluble fraction was chromatographed on a Sephadex LH-20 and silica gel column using various solvent system as eluents. The isolated compounds were identified by TLC, $^1H-$, $^{13}C$-NMR, HMBC and EI-MS. Astragalin (compound 1), kaempferol (compound 2), quercetin (compound 3), ferulic acid (compound 4) were isolated from the ethyl acetate soluble fraction of A. victorials.

Antioxidants Isolated from Kalopanax pictus (엄나무 유래 신규 항산화 활성물질)

  • Screening of new antioxidants form oriental medicines resulted in the isolation of a new antioxidative compound and eight known compounds from the stem bark of Kalopanax pictus. On the basis of various spectrosopic studies, the structure of the new compound was determined to be 4-rhamnose-3,5-dimethoxybenzoic acid methly ester. Other known compounds were identified as ferulic acid, 4,5,6,-trihydroxyflavanone, 2', 4',4' -trihydroxychalcone, caffeic acid, coniferyl alcohol, syringin, 1,3-di-O-caffeoylquinic acid. These compounds showed lipid peroxidation inhibitory acitivity in rat liver microsomes and free radical scavenging acitivity.
