This study was performed to establish the condition and the methods for the techniques of insertion the isolated blastomere cells into cytoplasm, in order to research the develop-mental ability of bovine embryo blastomere cells in vitro produced. After 24h in vitro ovary maturation with the ovaries from a slaughter house, in vitro fertilization was performed to the vital sperms which their mobility were decided by percoll gradient method, with 2~8 cell stage embryos, the blastomeres were isolated in $Ca^2$+. $Mg^2$+-free PBS, and following that embedded into agar and alginate solution, respectively. The rates of in vitro develop-ment are as follows ; in agar embedded 11 among 120(9.2%) 1 /2~1 /3 blastomers cleaved and 6 among 93(6.5%) 1 /4~1 /8 blastomeres cleaved. In sodium alginate-embedded 14 among 84(16.7%) 1 /2~1 /3 blastomeres cleaved and 6 among 85(7.1%) 1 /4~1 /8 blastomeres cleaved. In case of Na-alginate, the rate of the cells were better than those of agar. The results suggest that the techniques for embeeding the isolated blastomeres into gel may help cloning of bovine early embryo without nuclear transplantation.
High rates of nitrogen fertilization dangerously increase the nitrate content of vegetable crops, and the accumulation of nitrate in edible crops is undesirable because of potential risks to human health. Micronutrient solution containing Cu, Mn, Mo, Zn was tested for the suppression of nitrate accumulation in lettuce grown in pots treated with Mg fertilizer under a greenhouse condition. The micronutrient solution was sprayed on leaves at 3 and 4 weeks after transplanting of 20-day old seedlings. Plants were harvested after 5-week growth, and yield, contents of chlorophyll, sugar, micronutrient and nitrate, and also nitrate reductase activity were measured. Fresh weight of lettuce was significantly increased by the application of Mg and micronutrients, and the effect was the most significant in the Mg+micronutrient treatment. Also contents of chlorophyll and micronutrients were higher in the plants of micronutrient treatments. Contents of nitrate were reduced by about 14-18% in lettuce with Mg and/or micronutrient applications. Compared to the plants of control treatment, nitrate reductase activity was also higher in those plants treated with micronutrients, and in the treatment of Mg+micronutrients the enzyme activity was six times as high as that of control treatment. Although the effect of mineral nutrients on the suppression of nitrate accumulation in lettuce was relatively small in this study, an appropriate supply of mineral nutrients could be one of the solutions for the nitrate accumulation in vegetables.

The freemartinism is the most frequent form of intersexuality found in cattle, and females of heterosexual twins become sterile. With increase of twinning rates due to transfer of multiple embryos derived from in vitro fertilization, it is of great economic value to establish early diagnosis of freemartins to remove infertile individuals from breeding stock. In the present study polymerase chain reaction (PCR) of two different Y-chromosome specific segments (BRY.l and AMX/Y) was performed to identify freemartins from twins and less common single born freemartins in Korean Native Cattle (KNC). Two male-specific sequences were amplified in all heterosexual twins tested (n=5). In addition, Y-specific PCR products were detectable in one of the single born females (n=4) with visible genital abnormalities. These results suggest that the sensitivity of PCR-based assay may be sufficient to detect freemartinism in single born females as well as female partners of heterosexual twins in KNC.
It has been known that spermatozoa should obtain their fertilizing ability through capacitation and acrosome reaction, and that in these processes of fertilization, Ca2+ platys an important role for their conjugation. Therefore the present study has been designed in order to clarify the effect of fluctuation of the media Ca2+ level and the intracellular concentration of the spermatozoa on the acrosomes. During the incubation of spermatozoa, a considerable fluctuation in the media Ca2+ level has been observed after the BSA administration and the media concentration of Ca2+. It is deduced that these fluctuation rates may have an effect on the acrosome reaction. The fluctuation of K+ flux has been observed in accordance with the incubation period over time, and it's concentration seems to be closely related with the acrosomal reaction. The respiratory exchange rate (RERI of the spermatozoa is kept more regular in the BSA and Cacl2 administration groups than the non-administration group. Based on the experimental findings, it is possible to deduce a hypothesis from these findings that physiological activities of the acrosome reaction are not functionally related to the media Ca2+ level and the intracellular influx of Ca2+ concentration, although Ca2+ platys an important role as a stimulating factor in the acrosome reaction.
The morphology, seed germination, and life history of Salix nipponica were investigated in the wetland of the Nam-River, Jinju, Gyeongsangnam-do, Korea from 2000 to 2001 to provide the basic data necessary for riverine ecosystem restoration through understanding the growth characteristics of Salix nipponica. Salix nipponica had stomata on only lower side and stomata type was paracytic. Salix nipponica produced many small and light seeds. The seed number per mature ament was 1599.4, seed mass of 0.04 mg, and fertilization rate of 66.1%. Seed germination was little affected by light. Germination rate was high and mean germination time was short Under flooding condition, seeds were germinated normally and were viable after as much as 14 days of flooding. But there were no differences under various water depths on germination rates. Tree age was closely correlated with more stem diameter than tree height and there was no difference of growth rate between male and female tree. Growth rate was most rapid for 2 to 3 years after germination, and length growth was almost stopped for more than 11 years even though mass growth was done. Besides even a second-year indivisual was flowered.

To develop policies regarding fertility and pregnancy that will be effective in preventing further declines in fertility rates in the context present-day Korea, current policies in Japan were analyzed and compared with those now being implemented in Korea. This study was structured to involve (a) comparison of maternal health projects in Korea and Japan, and (b) comparison of infertility support policies based in regional cities in Korea and Japan. Korea's Health Plan 2030 emphasizes strengthening healthcare for high-risk pregnant women, expanding investments to benefit vulnerable groups, and establishing a support system for infertile couples. In Japan, government programs involving treatment targeting infertility specifically were implemented nationwide in 2006. Wide dissemination of accurate knowledge related to pregnancy is emphasized. Also, counseling centers specializing in infertility were established by 67 local governments. We have confirmed that Korean policies include decentralization, while Japan is implementing the central government's infertility policy uniformly in all regions. Japan also adjusted its policy out of concern that problems related to infertility and childbirth will worsen due to the social disaster of COVID-19. The results indicate that providing additional support for psychological counseling may be preferable to increasing the number of in-vitro fertilization procedures. The physical burden on women may be minimized by benchmarking policies in Japan. Step-by-step application of these procedures should be systematically supported to achieve the best results.

Solitary bees, such as Osmia cornifrons, O. pedicornis, O. satoi, and O. taurus (Hymenoptera: Megachilidae), have the potential for cost-effective and sustainable pollination, necessitating a comprehensive understanding of their ecological traits to implement effective fertilization strategies for various crops. This study investigated the nesting rate of Osmia spp. in different regions and altitudes, using various trap types, and found that the highest nesting rate occurred at altitudes of 300-399 m a.s.l. and showing a preference for bamboo-type traps, with the Andong region having the highest nesting rate overall, indicating the influence of altitude, habitat area, and trap type on the density of Osmia spp. nests. The distribution and diversity of the four Osmia spp. in different regions and altitudes revealed variations in their occurrence, with O. pedicornis having the broadest distribution rate, particularly at altitudes above 300 m a.s.l.. The present study found significant differences between species in the cocoon masses of O. cornifrons, O. pedicornis, and O. taurus, with region and altitude influencing the masses of each species too.
Yams (Dioscorea spp), which are edible or medicinal tuber crops, are a important crop in South Korea. Yams require a high level of soil fertility. The various cultural practices such as fertilizing and plowing were tested for marketable tuber production in Chinese yam (Dioscorea opposita). Tuber yield was also affected by organic matters in soil. Application higher level of organic matters result in increased each tuber weight and tuber yield per unit area. The nutrient absorption quantity of the plant such as nitrogen, phosphoric acid, calcium and potassium was increased from 100~120 days after planting, which time to begin tuber enlargement. The tuber yield was increased when the fertilization increased in quantity. Total yield and marketable ratio were the highest in 31~32 kg/10a of nitrogen fertilizer. From above result, income become larger with increase of marketable yield and quality improvement at 63% (27 kg/10a) level of conventional N fertilization (43 kg/10a). The tuber yield was not significantly different between with in various application level of potash fertilizer. Tuber size and weight decreased accordingly to decreased fertilizing level, so the rates of small tubers increased greatly at cultivation without chemical fertilizer. In considering the accumulation rates of allantoin in Chinese yam tubers, the apt harvest season was after October. The allantoin quantity of it was not influenced with nitrogen fertilizing. Moreover it was advantageous with decrease of chemical fertilizer and appropriate fertilizing in soil environment protection. Commercial tuber's number and yield were increased in trenching before planting with trencher compared with rotavating with tractor.

With a purpose of finding out the effects of magnesium and boron enriched organic-compound fertilizer application on the dry matter yield, yield components and changes in the botanical composition on the hilly pasture, a field experiment was arranged with five different treatments as a randomized block desigh and lasted from September, 1984 to the end of growing season in 1986. The results obtained are summarized as follows: 1. As a early plant growth and development, winter hardiness, growth vigour and coverate of grasses at the plots with organic-compound fertilizer application were better comparing to single dressing. 2. Average dry matter yields for two years were shown significantly high due to increasing amount of fertilizer. This trend was same both single and organic-compound fertilizer. And dry matter yields with organic-compound fertilizer application of low (8,693.1 kg/ha) and conventional level (12,758.7 kg/ha) were appeared to increase by 10 and 15% than those of single dressing of Low (7,930.6 kg/ha) and conventional level (11,122.6 kg/ha), respectively. But it was not significant difference. 3. Dry matter yield of grasses was significantly gained by increasing amount of fertilizer. The yield of legumes at the plots with organic-compound fertilizer application the yields of grasses were a litle more increased by 8-14%, and legumes were much more gained by 26-29% than those of the same species groups with single dressing, but it was not significant between the differnet kinds of fertilizer in the same fertilizing level. 4. At the grassland management, the rate of legumes tended to dominate at the plot without fertilization gradually. On the other hand, the botanical compositions and the rates of grasses were much better maintained at the plots with fertilization. But the rates of legumes with organic-compound fertilizer application tended to increase a little more than those of single dressing gradually. 5. All of the soil chemical properties after the experiment were much more improved comparing to before the experiment. But the average contents of soil organic matter and available $P_2O_5$ were slightly higher at the plot with organic-compound fertilization, on the other hand, pH and exchangeable cations tended to a little less than those of single dressing.
