The Effect of Fertilization-Promoting Peptide on Fertilizing Ability and Glycosidase Activity In Vitro of Frozen-Thawed Boar Spermatozoa

  • This study evaluated the effect of fertilization-promoting peptide (FPP) on fertilizing ability and glycosidase activity in vitro of spermatozoa frozen-thawed in pig, Using chlortetracycline fluorescence analysis, the various glycosidase analyses and the oocyte penetration test, we have obtained evidence that FPP can promote the fertilizing ability and glycosidase activity of frozen-thawed spermatozoa in vitro. When frozen-thawed spermatozoa was washed with different concentrations of FPP, there were significantly (P<0.05) more acrosome-reacted in medium with 100 nM than 0, 50, 200 and 400 nM. The penetration rates were also highest in medium containing with 100 nM FPP (P<0.05). On the other hand, the $\beta$-N-acetylglucosaminidase activity was at least twofold higher than other glycosidase. In same glycosidase, however, there were no difference in medium with different concentrations of FPP In another experiment, spermatozoa preincubated in medium with or without FPP for 0, 1, 2, 3 and 4 h were inseminated with oocytes matured in vitro. The percentages of spermatozoa that reached acrosome reaction were affected by preincubation and were higher in medium with that than without FPP. When oocytes were inseminated with spermatozoa preincubated in medium with and without FPP during the different periods, however, penetration rates were decreased with preincubation periods of spermatozoa. On the other hand, when the sperm-oocyte were cultured for 4, 8, 12, 16, 20 and 24 h, the penetration rates were higher in spermatozoa preincubated with that than without FPP and had a tendency to increase as time of culture periods. However, The activities of $\alpha$-fucosidase, $\alpha$ -mannosidase, $\beta$-galactosidase and N-acetyl- $\beta$-D-glucosaminidase were higher in medium with that than without FPP regardless of periods of sperm preincubation and sperm-oocyte culture. These results suggest that FPP may play a positive role in promoting of sperm function and glycosidase activity in vitro in pig.

Influences of silicate fertilizers containing different rates of iron slag on CH4 emission and rice (Oryza sativa L.) growth

  • Methane (CH4) is an important greenhouse gas, with a short-term greenhouse effect 80-fold that of carbon dioxide. Blast furnace slag used as a base ingredient for silicate fertilizer, and contained Fe3+, which acts as reduction of CH4 emissions in flooded rice paddy. This study was evaluated the effects of the silicate fertilizer with different rates of the iron slag on CH4 emissions and rice growth. In this study, the SF 0.0% was applied with silicate fertilizer containing 0.0% of the iron slag, while the SF 2.5% and SF 5.0% were treated with silicate fertilizer containing 2.5 and 5.0%, respectively. The CH4 emissions during rice cropping period were assessed using a closed-chamber method and then determined by Gas chromatography. The CH4 fluxes were reduced by 17% (SF 0.0%), 17% (SF 2.5%), and 8% (SF 5.0%) compared to the treatment with only-inorganic fertilization (control). Conversely, rice grain yield increased by 15 - 30% compared to the control owing to the improvement of soil quality by silicate fertilization. In particular, soil pH, available phosphorus and available silicic acid content were increased with the increase in the iron slag rates from 0.0 to 5.0%. These contributed to a significant increase in rice growth such as 1,000-grains weight and percentage of filled grains. Consequently, these findings were indicated that the application of silicate fertilizer containing 2.5 - 5.0% of iron slag would be the most effective in both CH4 reduction and rice growth.

Study on the Cultivation of Vitex rotundifolia According to Different Soils and Fertilization Methods (토양과 비배관리에 의한 순비기나무의 재배방법에 관한 연구)

  • This study is focused on the cultivation of seedlings and cuttings of roundleaf chastetree(Vitex rotundifolia L. f.) according to the sorts of soil and fertilization methods. The results are summarized as follows: When roundleaf chastetree seedlings(aged 1-0) and cuttings(aged 1-0) were cultivated in three different kinds of soil- sand, granite soil, and yellow silt- without any fertilization, 70% of the seedlings survived in sand and granite soil, and 35% did in yellow silt; 90% of the cuttings survived in granite soil and 80% did in sand and yellow silt. The general growth of both seedlings and cuttings was good in granite soil, and sand but relatively inferior in yellow silt. Seedlings showed good stem growth while cuttings had fine root growth. In case of cultivation of seedlings with different kinds and quantities of fertilizer adjusted and with granite soil as basic soil, seedlings and cuttings survived in the experimental section where only the organic fertilizer was used as basic fertilization; in contrast, seedlings grown in the experimental section were all dead where both inorganic and liquefied fertilizer were used. Cuttings showed considerably higher survival rates than seedlings at the experimental fertilization section; particularly, the survival rate of cuttings was twice as much as that of seedlings at the experimental section with low fertilization. This study results revealed that cuttings are desirable than seedlings in general rootage, growth, adaptability to soil, and cuttings have lower demand for soil fertility in time of growing roundleaf chastetree saplings.

Responses of Young 'Fuyu' Persimmon Trees to Summer Fertilization Rate and Leaf-fruit Ratio

  • Small-sized persimmons produced by high crop load are better accepted in the export markets. However, maintaining high crop load frequently results in weakness of tree vigor, deterioration of fruit quality, and increase of the risks for alternate bearing. This experiment was conducted to determine the combined effects of fertilization rate and leaf-fruit (L/F) ratio on container-grown 3-year-old 'Fuyu' persimmon trees. Application of 3.6-g N, 2.1-g $P_2O_5$, 2.7-g $K_2O$, 2.7-g CaO, and 0.6-g MgO was for the control fertilization rate (CF) and that of a 3-fold CF was for the high fertilization rate (HF). Commercial fertilizers were surface-applied to a container on July 6, July 17, and August 10 in three equal aliquots. Single tree for each fertilization rate was assigned for 12 L/F ratios (5, 6.3, 7.7, 9, 10.4, 13, 15.5, 18, 21, 24, 27, and 33) mostly by fruit thinning or rarely by defoliation on July 1. HF did not affect the yield, weight and soluble solids of the fruits but decreased skin color. As L/F ratio increased, yield decreased but average weight, skin color, and soluble solids of fruits increased. With HF, N and K concentrations in leaves, fruits, and shoots increased to some extent but soluble sugars in dormant shoots decreased. Many shoots were cold-injured with low L/F ratio especially at the HF. HF did not increase number of flower buds the next spring either on a shoot or on a tree basis but increased shoot length, compared with the CF. Increasing L/F ratio markedly increased number of flower buds and shoot growth the following year at both fertilization rates. Therefore, an appropriate combination of fertilization rate and L/F ratio should be necessary to maintain stable fruit production and tree vigor at high crop load.

Studies on the Viability of In Vitro-Matured Bovine Oocytes Vitrified by Microdrop and Straw Method (Microdrop과 Straw 방법으로 초자화 동결한 소 난자의 생존율에 관한 연구)

  • Studies on the Viability of In Vitro-Matured Bovine Oocytes Vitrified by Microdrop and Straw Method To establish vitrification method for bovine oocytes, mature bovine oocytes were vitrified by microdrop (MD) or straw (Straw) method and the viability of vitrified oocytes with or without cumulus cells (CC) were examined by several methods; a) parthenogenetic activation; b) pronuclear formation after in vitro fertilization (IVF); and c) embryonic development after IVF. The survival rate of vitrified oocytes by MD was significantly higher than by Straw (92.50 vs. 74.19%, p<0.05). Most of the oocytes survived from vitrification using the MD methods. Cleavage and blastocyst development of parthenogenetically activated oocytes were higher in MD (45.05% and 10.81%, respectively; p<0.05)) than those in Straw method (27.17% and 6.52%, respectively; p<0.05). Male and female pronuclear formation of vitrified-thawed oocytes with or without cumulus cells (CC) after IVF were examined, respectively. The survival rate of vitrified oocytes by MD without CC was no difference between MD and Straw (80.368.14% vs. 67.31%). Normal fertilization (2PN) rates were not different among groups (Fresh; 54.55% vs. MD; 42.22% vs. Straw; 37.14%, p>0.05). While no fertilization (<1PN) rates were significantly different between fresh and vitrified-thawed groups (Fresh; 32.47% vs. MD; 57.78% and Straw 62.86%, p<0.05). The polyspermy (3PN) was appeared in the fresh (12.99%), but no appeared in the vitrified-thawed groups. In the without CC, normal fertilization (2PN) rates were significantly different between fresh and vitrified-thawed oocytes (Fresh; 59.38% vs. MD; 17.31% and Straw; 30.43%, p<0.05). Moreover, no fertilization (<1PN) rates were significantly different between fresh and vitrified-thawed groups (Fresh; 23.44% vs. MD; 73.08% and Straw 58.70%, p<0.05). The polyspermy (3PN, >4PN) was appeared not only fresh but vitrified-thawed groups. After IVF, two-cell developmental rates of vitrified oocytes with CC by MD and Straw were significantly low compared to fresh oocytes (Fresh; 81.76% vs. MD; 22.22% and Straw; 11.36%, p<0.05). Blastocyst developmental rates of vitrified oocytes also were significantly low compared to fresh oocytes (Fresh; 28.38 vs. MD; 1.71% and Straw 0%, p<0.05). In the without CC, two-cell developmental rates were no difference between Fresh and MD (27.59% vs. 19.25%, p<0.05), while blastocyst rates were difference between Fresh and MD or Straw (4.31% vs. 0.62% and 0%, respectively; p<0.05). In conclusion, the results indicate that the vitrified bovine oocytes have the ability to develop to the blastocyst stage after IVF.

Evaluation of human embryo development in in vitro fertilization- and intracytoplasmic sperm injection-fertilized oocytes: A time-lapse study

  • Objective: We investigated whether the insemination method (in vitro fertilization [IVF] or intracytoplasmic sperm injection [ICSI]) affected morphokinetic events and abnormal cleavage events in embryonic development. Methods: A total of 1,830 normal fertilized embryos were obtained from 272 IVF and ICSI cycles that underwent ovum retrieval culture using a time-lapse system (Embryoscope) from June 2013 to March 2015. All embryos were investigated by a detailed time-lapse analysis that measured the developmental events in the hours after IVF or ICSI insemination. Results: No significant differences were observed between the two groups regarding clinical outcomes (p>0.05). ICSI-derived embryos showed significantly faster morphokinetics than those derived from conventional IVF, from the time to pronuclear fading to the time to 6 cells (p<0.05). However, no significant differences were found from the time to 7 cells to the time to expanded blastocyst (p>0.05). There were no differences in abnormal cleavage events between the two groups (p>0.05); they showed the same rates of direct cleavage from 1 to 3 cells, 2 multinucleated cells, 2 uneven cells, and reverse cleavage. Conclusion: The morphokinetics of embryo development was found to vary between IVF- and ICSI-fertilized oocytes, at least until the 6-cell stage. However, these differences did not affect the clinical outcomes of the embryo. Additionally, no significant differences in abnormal cleavage events were found according to the fertilization method.

Tendency and Problems in Porcine in-vitro Fertilization (돼지체외수정의 연구동향과 문제점)

  • In vitro culture has provided new information on the mechanisms involved in fertilization how sperm and oocyte fuse together. At the same time, results obtained in vitro have led to new questions. Techniques for In vitro maturation of porcine oocytes have progressed such that the problem of the low rate of pronucleus formation with in vitro matured oocytes after in vitro fertilization has been nearly improved. On the other hand, porcine spermatozoa have been shown to be capacitated if the fertilization medium contains caffeine and Ca$^2+$, but the incidence of polyspermy in IVM-IVF oocytes is still high. To prevent polyspermy, co-culture with oviductal cells, sperm preincubation with porcine follicular fluid or control of sperm concentration, have been examined with significant effects but still remarkably high rates of polyspermy. The under standing of these influences is a prerequisite to enhancing in vitro production of porcine em bryos.

Fertilization Efficiency of Livestock Manure Composts as Compared to Chemical Fertilizers for Paddy Rice Cultivation

  • To promote the practical use of livestock manure compost (LC) for paddy rice cultivation, the fertilization efficiency of nutrients in LCs was investigated compared to that of chemical fertilizer. This experiment was conducted at rice field in Hwaseong, Korea, with 6 treatments by each of 3 kinds of tested LCs, cattle manure compost (CaC), swine manure compost (SwC) and chicken manure compost (ChC). The treatments consisted of 3 application levels of LCs and 3 chemical fertilizer treatments having the same application levels with LCs. $NH_4$-N content in soil became higher according to the increase in the urea application rate, while it became lower in LC plots than in urea plots, and statistically had no significant difference among LC plots. There was a close relationship between phosphate fertilization rate and the increment of soil available phosphate content after experiment resulting y = 0.1788x - 6.169 ($R^2=0.9425$) when applied fused superphosphate fertilizer, and y = 0.0662x - 2.689 ($R^2=0.9315$) when applied LC at the equivalent rates to phosphate input (x: phosphate application rate, kg $ha^{-1}$, y: increment in soil available phosphate content, mg $kg^{-1}$). And from these two equations, the correlation on the phosphate application rate between fused superphosphate fertilizer and LC could be obtained as y = 2.7056x - 52.492 (x: $P_2O_5$ application rate of fused superphosphate, kg $ha^{-1}$, y: $P_2O_5$ application rate of LC, kg $ha^{-1}$). Plant height, number of tillers, nutrients uptake by rice, and rice yield showed higher levels in N 100% and N 150% application plots of chemical fertilizers, while every LC plots exhibited lower values and no significant difference among them. Relative nitrogen fertilization efficiencies of LCs compared to urea was 12.3% for CaC, 8.8 for SwC and 24.6 for ChC, respectively.

Expression of Yolk Processing Enzyme Genes in Fertilized Eggs from Artificially Matured Female Eel, Anguilla japonica

  • Large quantity of eggs fail to be fertilized and many of fertilized eggs are unable to hatch in the eel, Anguilla japonica. Larvae of eel absorb egg yolk up to 8 days after hatching but the majority of hatched larvae die before they reach the stage of first feeding in this species. Genes of key enzymes for yolk processing (cathepsin B, D, L and lipoprotein lipase - abbreviated as ctsb, ctsd, ctsl and lpl, respectively) could be associated with egg quality. In this study, we investigated differences in the expression of these genes between floating eggs and sinking eggs, and also the relationship between the gene expressions of the enzymes and fertilization rates in the fertilized eggs obtained from artificially matured female eels. Expressions of yolk processing enzyme genes did not show significant difference between floating and sinking egg groups. Expression of ctsb decreased when fertilization rate was high. Expression of ctsd, ctsl and lpl, however, did not show any significant differences. These results suggest that ctsb expression could be an indicator of egg quality, and that some proteins prone to be digested by ctsb could be very important in the process of fertilization and normal cleavage in this species. Further study should identify these critical proteins to improve our understanding on the quality of fish eggs.

Effects of Rice Straw Compost Application on Exchangeable Potassium in Long-term Fertilization Experiments of Paddy Soils

  • In an experiment conducted at the research field of the National Institute of Agricultural Science, we investigated the effects of mineral fertilizer and rice straw compost on exchangeable potassium and K balances, and rice grain yield under a rice single system. The treatments were no fertilization (No fert.), inorganic fertilization (N), inorganic fertilizer (N, P, K) plus rice straw compost at rates of 7.5, 15.0, 22.5, and $30.0ton\;ha^{-1}$ (NPKC7.5, NPKC15.0, NPKC22.5, and NPKC30.0, respectively). The inorganic fertilizers(N, P, K) were added with standard fertilizer application rate in which nitrogen (N), phosphate ($P_2O_5$), and potassium ($K_2O$) were applied with $75{\sim}150kg\;ha^{-1}$, $70{\sim}86kg\;ha^{-1}$, $75{\sim}86kg\;ha^{-1}$, respectively. Exchangeable potassium for NPKC15.0 NPKC22.5, and NPK30.0 treatments was higher by $0.05{\sim}0.19cmol_c\;kg^{-1}$ than that of NPKC7.5 treatment. Increasing levels of rice straw compost resulted in an increase in the K balance from - $19.9kg\;ha^{-1}yr^{-1}$ (No fert.) to $41.9kg\;ha^{-1}yr^{-1}$ at NPKC22.5 treatment and $62.9kg\;ha^{-1}$ at NPKC30.0 treatment. Continuous application of rice straw compost with NPK fertilizers affected significantly the rice grain yields. The result of the study imply that the application of more than $22.5ton\;ha^{-1}$ of rice straw compost with NPK fertilizers are recommended as the best fertilization practice for enhancement of crop production and K supplying power of soil in the continuous rice cropping system.
