The Effect of Nitrogen Fertilization to the Sward on Guineagrass (Panicum maximum Jacq cv. Gatton) Silage Fermentation

  • To investigate the effect of nitrogen fertilization on the quality of tropical grass silage, guinea grass grown with 3 types of nitrogen fertilizers, namely, urea, ammonium sulfate, and compound fertilizer 804, at 2 fertilization levels, 0.5 and 2.5 kg $Na^{-1}$ (0.5 N and 2.5 N, respectively), was subjected to silage fermentation. Silage fertilized with 0.5 N showed butyrate-dominant fermentation, irrespective of the type of fertilizer used. On the other hand, fermentation of silage fertilized with 2.5 N was significantly affected by the type of fertilizer used; fertilization with ammonium sulfate and compound fertilizer 804 resulted in silage that contained a large amount of butyrate and no lactate; this silage was considered to be of a significantly low quality as compared with silage fertilized with 0.5 N. Among silage fertilized with 2.5 N, the desirable butyrate-free fermentation was found only in urea-fertilized silage, which had the best quality. Grass material fertilized with a high level of urea accumulated a relatively high concentration of nitrate nitrogen (0.22% dry matter). Our results presented here suggest that nitrogen fertilizer management could affect the quality of tropical grass silage and that a relatively high concentration of nitrate in silage may promote butyrate-free fermentation even in tropical grass silage.

퀴노아를 첨가한 청국장의 발효기간에 따른 품질 변화 (Changes in Quality Characteristics of Cheonggukjang added with Quinoa during Fermentation Period)

  • In this study, the quality characteristics of cheonggukjang with addition of different quinoa were investigated. We evaluated the quality and sensory characteristics of cheonggukjang, including the pH, amino nitrogen, slime contents, color value, and total aerobic bacteria. Moreover, The anti-oxidant activities were measured as total polyphenol content, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, and superoxide dismutase (SOD) activity. The pH and amino nitrogen content were noted to significantly increase during the fermentation period in all the samples. The slime content of cheonggukjang added with quinoa increased with increased fermentation time, but cheonggukjang with addition of 20% of quinoa was decreased. The L value and b value decreased significantly with increased fermentation time, but the a value increased significantly. The microbial tests of cheonggukjang with addition of quinoa showed that the aerobic micro-organisms count was 7.45~9.10 Log cfu/g. Total polyphenol contents increased in all groups during the fermentation period, and activity increased with an increased percentage of added quinoa. The DPPH radical scavenging activity and SOD-like activity of cheonggukjang (with addition of quinoa) were also significantly higher than those of the control. The sensory quality of quinoa 10% cheonggukjang was stronger in flavor, and taste, and demonstrated a higher level of overall acceptability, when compared to the other groups.

Changes in microbial population and chemical composition of corn stover during field exposure and effects on silage fermentation and in vitro digestibility

  • Objective: To effectively use corn stover resources as animal feed, the changes in microbial population and chemical composition of corn stover during field exposure, and their silage fermentation and in vitro digestibility were studied. Methods: Corn cultivars (Jintian, Jinnuo, and Xianyu) stovers from 4 random sections of the field were harvested at the preliminary dough stage of maturity on September 2, 2015. The corn stover exposed in the field for 0, 7, 15, 30, 60, 90, and 180 d, and their silages at 60 d of ensiling were used for the analysis of microbial population, chemical composition, fermentation quality, and in vitro digestibility. Data were analyzed with a completely randomized $3{\times}6$ [corn stover cultivar $(C){\times}exposure$ d (D)] factorial treatment design. Analysis of variance was performed using SAS ver. 9.0 software (SAS Institute Inc., Cary, NC, USA). Results: Aerobic bacteria were dominant population in fresh corn stover. After ensiling, the lactic acid bacteria (LAB) became the dominant bacteria, while other microbes decreased or dropped below the detection level. The crude protein (CP) and water-soluble carbohydrate (WSC) for fresh stover were 6.74% to 9.51% and 11.75% to 13.21% on a dry matter basis, respectively. After exposure, the CP and WSC contents decreased greatly. Fresh stover had a relatively low dry matter while high WSC content and LAB counts, producing silage of good quality, but the dry stover did not. Silage fermentation inhibited nutrient loss and improved the fermentation quality and in vitro digestibility. Conclusion: The results confirm that fresh corn stover has good ensiling characteristics and that it can produce silage of good quality.

Effects of wilting and additives on the ensiling quality and in vitro rumen fermentation characteristics of sudangrass silage

  • Objective: This study was conducted to investigate the effects of molasses and Lactobacillus plantarum on the ensiling quality and in vitro rumen fermentation of sudangrass silage prepared with or without wilting. Methods: The ensiling experiment, measured with 3 replicates, was carried out according to a 2×4 (wilted stages×additives) factorial treatment structure. Dry matter of the fresh (210 g/kg fresh matter) or wilted (305 g/kg fresh matter) sudangrass were ensiled (packed into 5.0-L plastic jars) without additive (control) or with molasses (M), Lactobacillus plantarum (LP), or molasses + Lactobacillus plantarum (M+LP). After 60 days of ensiling, the silages were analyzed for the chemical, fermentation, and in vitro characteristics. Results: After 60 days of ensiling, the fermentation parameters were affected by wilted, the additives and the interactions of wilted with the additives (p<0.05). The M+LP treatment at wilted had higher lactic acid levels and V-score (p<0.05) but lower pH values and butyric acid concentrations than the other treatments. In comparison with sudangrass before ensiling, after ensiling had lower dry matter and higher non-fibrous carbohydrate. The in vitro gas production, in vitro dry matter digestibility, in vitro crude protein digestibility, and in vitro acid fiber detergent digestibility changed under the effects of the additives. Significant interactions were observed between wilted and the additives in terms of in vitro gas production at 48 h, asymptotic gas production, gas production rate, half time, and the average gas production rate. The total volatile fatty acid levels in the additive treatments were higher than those in the control. Conclusion: Wilting and supplementation with molasses and Lactobacillus plantarum had the ability to improve the ensiling quality and in vitro nutrient digestibility of sudangrass silage. The M+LP treatment at wilted exhibited the strongest positive effects on silage quality and in vitro ruminal fermentation characteristics.

The Effect of Vinegar Fermentation on the Nutritional Quality of Lotus Flower Fermented Product

  • All the parts of lotus, including the seed, rhizome, leaf, stalk, petal, anther, pericarp, and fruit receptacle, have been used in traditional medicine system as a health beneficial supplement. However the most usually used from lotus plant is only the root. Therefore in this study, it will be discussed more the utilization of other parts of the lotus, namely the flower of lotus. The petals and stamens of lotus actually are also rich in bioactive components such as flavonoids and alkaloids, are used in the treatment of tissue inflammation, cancer, skin disease, and also for us as antidotes. One of the biotechnological process that can be used to improve the nutritional content, sensory, and also antioxidant activities is fermentation process. The final product desired from the fermentation process in this study is vinegar. The microbial strain powder used is Uinkin fermented powder with three variations of fermentation. The variations given in this study were initial sugar 32%, 24%, and 14% with the same fermentation temperature, $35^{\circ}C$ for 3 months. The results obtained showed that the pH value and sugar content of products during the fermentation process were decreasing during the fermentation process, with total polyphenol content of $283.7{\pm}97.6mg/100g\;QAE$, and total flavonoid content of $3.3{\pm}0.0mg/100g\;QAE$. For the DPPH radical scavenging ability of the fermentation product also increased in a concentration dependent manner, with ORAC activity of the product showed a high activity of $20.7{\pm}0.41{\mu}M$ TE. Therefore, fermentation process can be the one of method for improving the product. The efficiency of lotus flower vinegar fermentation can be reached with an initial sugar condition of 25% (sample B).

발효액종이 빵의 품질에 미치는 영향 (Effects of Fermented Liquid Dough on Bread Quality)

  • This study identifies the factors improving bread quality by using fermented liquid dough. Fermented liquid dough, the main part of bread dough, contains yeasts that are prepared in order to enhance the fermentation rate. This study investigates the fermentation rate after mixing dough, the pH of dough, loaf volume, water activity, hardness, and sensory properties of loaf bread samples with different amounts of fermented liquid dough. The fermentation rate was slightly higher in the bread samples, the control dough and 10% fermented liquid dough, than in samples with more than 20% dough. The pH values of dough decreased with an increase in the content of fermented liquid dough. The loaf volume of bread with 10% fermented liquid dough was the highest. The water activity of loaf bread increased with an increase in amount of fermented liquid dough. For the sensory evaluation of loaf bread, adding 10% fermented liquid dough improved the loaf volume and evenness of baking. These results suggest that 10% fermented liquid dough increased the fermentation rate and bread quality. Further research is required to enhance internal quality characteristics of loaf bread, including taste and flavor.

효모 종류에 따른 참다래 와인의 발효 및 숙성 중 품질 변화 (Quality Changes in Kiwifruit Wines during Fermentation and Aging with Different Yeasts)

  • 국내산 참다래 와인의 고품질화 방안으로 제주도에서 육성된 참다래 품종인 제시골드와 제시스위트를 대상으로 참다래 와인 제조에 적합한 2종의 효모를 이용하여 저온 발효 숙성한 참다래 와인의 품질 특성을 비교 분석하였다. 총산도는 발효 초기에 제시스위트와 제시골드가 각각 0.94%와 1.22%였으나 발효가 진행됨에 따라 조금씩 증가하여 발효 6일에는 각각 1.49%와 1.26%로 가장 높은 값을 나타내었으며 균주별로는 차이가 없었다. 에탄올 함량은 발효 4일부터 급격히 증가하였으며 발효 12일에는 제시스위트의 S. bayanus Lalvin 발효액이 10.2%로 가장 높은 함량을 나타내었고, 제시스위트의 S. cerevisiae Fermivin 발효액이 9.2%, 제시골드의 Lalvin과 Fermivin 발효액이 각각 9.4%와 9.4%였다. 가용성 고형분 함량은 참다래 품종과 효모 종류에 관계없이 비슷한 변화를 보였는데, 발효 초기에 약$24.8^{\circ}Brix$로 발효가 진행됨에 따라 서서히 감소하였으며 발효 6일 후에 약 $9.7^{\circ}Brix$로 가장 낮은 값을 나타내었다. 총페놀 함량은 발효 시작 전에 제시스위트(1,127 mg/L)가 제시골드(848 mg/L)보다 1.32배 높았으며, 발효기간 동안에는 발효 6일 후 급격히 감소하였다가 발효 12일까지 다소 증가하는 경향을 나타내었다. 숙성기간 동안에도 숙성 30일 후 증가하였다가 숙성 120일까지 거의 일정한 값을 유지하였다. 본 연구 결과 효모 종류에 따른 참다래 와인의 품질 특성은 다소 유사한 경향을 보였으나 참다래 품종별로는 차이를 보였으며, 에탄올 함량과 총페놀 함량에 있어서 제시스위트 품종으로 발효한 와인의 품질이 우수하였다.

농가 자가발효에 의한 사과식초의 생산 (Fermentation of Apple Vinegar in the Farmhouse)

  • The study was conducted to research the desirable method for fermentation of acidic apple beverage in the farmhouse. Step by step fermentation(SSF), complex fermentation after crushing of the fruit(CCF), complex fermentation after slicing of the fruit(SCF) and natural fermentation added yeast only after slicing of the fruit(SYNF) were compared. The brief fermentor for using stationary complex fermentation in the farmhouse was made in this experiment. The ability of acid production, flavor, taste and color were measured by sensory evaluation and mechanical methods. The quality of vinegar by SYNF and SCF were estimated by color, flavor, content, composition of sugar, and organic acids, and which was a suitable fermentation method for the farmhouse. The strains of acetobacter, SYNF-1 and 2 were isolated from the SYNF vinegar, and if has been shown that the SYNF-1 was a main strain in this study.

Effects of calcium propionate on the fermentation quality and aerobic stability of alfalfa silage

  • Objective: To assess the potency of calcium propionate (CAP) used as silage additive, an experiment was carried out to evaluate the effect of CAP on the nitrogen transformation, fermentation quality and aerobic stability of alfalfa silages. Methods: Alfalfa was ensiled with four levels of CAP (5, 10, 15, and 20 g/kg of fresh weight [FW]) in laboratory silos for 30 days. After opening, the silages were analyzed for the chemical and microbiological characteristics, and subjected to an aerobic stability test. Results: The increasing proportion of CAP did not affect pH, lactic acid (LA) concentrations and yeast counts, while linearly decreased counts of enterobacteria (p = 0.029), molds (p<0.001) and clostridia (p<0.001), and concentrations of acetic acid (p<0.001), propionic acid (p<0.001), butyric acid (p<0.001), and ethanol (p = 0.007), and quadratically (p = 0.001) increased lactic acid bacteria counts. With increasing the proportion of CAP, the dry matter (DM) loss (p<0.001), free amino acid N (p<0.001), ammonia N (p = 0.004), and non-protein N (p<0.001) contents were linearly reduced, whereas DM (p = 0.048), water soluble carbohydrate (p<0.001) and peptide N (p<0.001) contents were linearly increased. The highest Flieg's point was found in CAP10 (75.9), represented the best fermentation quality. All silages treated with CAP improved aerobic stability as indicated by increased stable hours compared with control. Conclusion: The addition of CAP can suppress the undesirable microorganisms during ensiling and exposure to air, thereby improving the fermentation quality and aerobic stability as well as retarding the proteolysis of alfalfa silage. It is suggested that CAP used as an additive is recommended at a level of 10 g/kg FW.

Effect of Additives, Storage Temperature and Regional Difference of Ensiling on the Fermentation Quality of Napier Grass (Pennisetum purpureum Schum.) Silage

  • The effects of addition of celulases (A cremonium cellulolyticus and Trichoderma viride, CE), a commercial inoculum containing lactic acid bacteria (Lactobacillus casei, LAB), fermented green juice (macerated napier grass with water was incubated anaerobically with 2% glucose for 1 day, FGJ) and glucose (G), and regional difference of ensiling on napier grass (Pennisetum purpureum Schum.) silage were studied by using 900 ml laboratory glass bottle silos under 30 and $40^{\circ}C$ storage conditions in 1995 and 1996. Experiment 1 was carried out to compare the addition of CE, LAB, FGJ and the combinations. Silages were stored for 45 days after ensiling. Experiment 2 studied the effects of applications of CE, LAB, FGJ and G. Experiment 3 was carried out using the similar additives as experiment 2 except for LAB. Silages were stored for 60 days in the experiments 2 and 3. Experiments 1 and 2 were done in Nagoya, and experiment 3 in Okinawa. Sugar addition through CE or G improved the fermentation quality in all the experiments, which resulted in a greater decrease in the pH value and an increased level of lactic acid, while butyric acid contents increased under $30^{\circ}C$ storage condition in CE addition. LAB and FGJ additions hardly affected the silage fermentation quality without additional fermentable carbohydrate. But the combination of LAB, FGJ and glucidic addition (CE and G) improved the fermentation quality. The effect of the regional difference of ensiling between temperate (Nagoya; $35^{\circ}$ N) and subtropical (Okinawa; $26.5^{\circ}$ N) zones on silage fermentation quality was not shown in the present study.
