Synthesis and Characterization of Aluminosilicate Prepared Using Structure Directing Agent Containing Piperidine Moiety (피페리딘형 구조유도분자를 이용한 알루미노실리케이트 제올라이트의 합성 및 특성 연구)

  • Zeolites with different structures were synthesized from the hydrothermal synthesis condition employing simple structure directing agent (SDA) containing piperidine moiety. The gel containing $1.0SiO_2$:0.9SDA:$0.062NaAlO_2$:0.217NaOH:$20H_2O$ was subject to hydrothermal synthesis at 413~453 K for 7 days. FER type zeolite was obtained at 433 K when piperidine was employed as SDA, whereas TON and MFI type zeolites were also obtained at 433 K when 2,6-dimethylpiperidine and 2,2,6,6-tetramethylpiperidine were used, respectively. Further increase of hydrothermal synthesis temperature to 453 K resulted in the formation of TON type zeolite when 2-mtheylpiperidine was used. The structural analysis of powder X ray diffraction pattern over FER type zeolite suggested that the SDA, piperidine interacted intimately with the zeolite where it located close to the framework.

Optimization of the Expression of the Ferritin Protein Gene in Pleurotus eryngii and Its Biological Activity (큰느타리버섯에서 석충 페리틴 단백질 유전자의 발현 최적화 및 생물학적 활성)

  • To optimize the expression and secretion of ferritin protein associated with ion storage in the mushroom, Pleurotus eryngii, a recombinant secretion vector, harboring the ferritin gene, was constructed using a pPEVPR1b vector under the control of the CaMV 35S promoter and signal sequence of pathogen related protein (PR1b). The ferritin gene was isolated from the T-Fer vector following digestion with EcoRI and HindIII. The gene was then introduced into the pPEVPR1b secretion vector, and it was then named pPEVPR1b-Fer. The recombinant vector was transferred into P. eryngii via Agrobacterium tumefaciens-mediated transformation. The transformants were selected on MCM medium supplemented with kanamycin and its expression was confirmed by SDS-PAGE and western blotting. Expression of ferritin protein was optimized by modifying the culture conditions such as incubation time and temperature in batch and 20 L airlift type fermenter. The optimal conditions for ferritin production were achieved at 25℃ and after incubating for 8 days on MCM medium. The amount of ferritin protein was 2.4 mg/g mycelia, as measured by a quantitative protein assay. However, the signal sequence of PR1b (32 amino acids) seems to be correctly processed by peptidase and ferritin protein may be targeted in the apoplast region of mycelia, and it might not be secreted in the culture medium. The iron binding activity was confirmed by Perls' staining in a 7.5% non-denaturing gel, indicating that the multimeric ferritin (composed of 24 subunits) was formed in P. eryngii mycelia. Mycelium powder containing ferritin was tested as a feed additive in broilers. The addition of ferritin powder stimulated the growth of young broilers and improved their feed efficiency and production index.

Study on Development of Horticultural Media Using Macroporous Calcium-Silicate Mineral (다공성규산칼슘계 화합물을 이용한 원예용 상토개발에 관한 연구)

  • The experiment was conducted to investigate the agricultural utility of macro-porous calcium-silicate rnineral(CellCaSi) as topsoil mixture, as well as to estimate as a soil conditioner. The bulk density of CellCaSi which is consisted of various particle sizes ranging from 1mm to 3.35mm was about $0.42g/cm^3$, and its maximum porosity was approximately 81.4%. We also investigate gerrnination rates for Cabbage and Lettuce to obtain the suitable mixing ratios of CellCaSi with topsoil. Among 4 different mixing ratios, the germination rates of ropsoil mixed with 10% of CellCaSi were 94.1% and 64.6% for Cabbage and Lettuce, respectively resulted in the mosr suitable for germination. The growth rates for Cabbage and Lettuce showed thar 10% and 20% of CellCaSi treatments signification influenced the fresh weight. To observe the adsorption capacity of CellCaSi, CellCaSi was treated with a chemical fertlizer(N:P:K=18:18:18). lncreasing the contents of N, P and K, the amounts of adsorption by CellCaSi for these element also increased. The most suitable types nutrient resources for growth condition of Cabbage, and Lettuce were Fer-1 and Fer-0.5.

The Effect of Three Different Grain Diets on the Growth of Albino Rats (곡식 혼식이 흰쥐의 성장에 미치는 영향)

  • The effect of three different grains supplemented diet as a dietary carbohydrate source on the growing rat, growth, food intake, FER and PER value, excretion of urinary arid fecal nitrogen and glucose and the level of glucose and cholesterol in serum blood was studied. Forty males and same number of females of Albino rats weighing male $58{\pm}2g$, female$62{\pm}2g$ were divided into eight experimental group, five male and female in each and fed rice diet, rice supplemented with wheat and rice supplemented with barley diet for 14 weeks. Under this condition wheat supplemented diet group shows the best FER, PER and body weight gain value. Especially in female group shows better FER, PER and body weight gain value compare to control diet group. Under the male groups wheat supplemented diet group shows lower FER, PER value and body weight compare to the control group but higher than those of barley diet group or rice alone. In female group the nitrogen retention shows also highest value by wheat supplemented diet group. By male group the wheat supplemented diet group shows not better value compare to the control group, but higer than barley supplemented group or rice alone. The nitrogen excretion through feces was significantly higher by grain supplemented diet group compare to grain the 20% sugar casein diet. The blood cholesterol content was slightly decreased by the control group which was fed disaccaride, compare to groups which were fed polysaccaride. But the free cholesterol to esterified cholesterol ratio shows lower value by control group than grain groups except the 70% rice supplemented with 30% barley group.

The Effect of Pore Structure of Zeolites on their Product Distribution and Deactivation in the Catalytic Cracking of n-Octane (n-옥탄의 촉매 분해반응에서 제올라이트의 세공구조가 생성물 분포와 활성저하에 미치는 영향)

  • The catalytic cracking of n-octane over FER, MFI, MOR and BEA zeolites was studied by the protolytic cracking mechanism in order to understand the effect of pore structure of zeolites on their product composition and deactivation. The selectivities for $C_3$ and $C_3{^=}$ were high over the zeolites with medium pores due to additional cracking, while those for $C_4$ and $C_4{^=}$, the initial products, were high over the zeolites with large pores. MFI zeolite showed slow deactivation due to small carbon deposit, while FER zeolite with small pores deactivated rapidly with severe carbon deposit. The deactivation of BEA zeolite was slow even with a large amount of carbon deposit, but MOR zeolite showed a rapid deactivation even with a small amount of carbon deposit. The conversion measured along with the time on stream on these zeolite catalysts was simulated by a mechanism based on the simplified reaction path of n-octane cracking and the deactivation related to the pore blockage by carbon deposit.

Once correspondences determined, the depth information of those points are easily computed form the pairs of points in both image.   • Until a recent day, degradation of PEMFC MEA (membrane and electrode assembly) has been studied, separated with membrane degradation and electrode degradation, respectively. But membrane and electrode were degraded coincidentally at real PEMFC operation condition. Therefore in this work, AST (Accelerated Stress Test) of MEA degradation was done at the condition that membrane and electrode were degraded simultaneously. There was interaction between membrane degradation and electrode degradation. Membrane degradation reduced the decrease range of catalyst active area by electrode degradation. Electrode degradation reduces increase range of the hydrogen crossover current and FER (Fluoride Emission Rate) by membrane degradation.

Characteristics of the surface ozone concentration on the occurrence of air mass thunderstorm (기단성 뇌우 발생시 지표오존농도의 변화 특성)

  • This study was performed to research ozone concentration related to airmass thunderstorm using 12 years meteorological data(1990~2001) at Busan. The occurrence frequency of thunderstorm during 12 years was 156 days(annual mean 13days). The airmass thunderstorm frequency was 14 days, most of those occurrence at summertime(59%). In case August 4, 1996, increase of ozone concentration was simultaneous with the decrease of temperature and increase of relative humidity, In case July 23, 1997, ozone concentration of western site at Busan increased, while its of eastern site decreased as airmass thunderstorm occurred(about 1500LST). It is supposed that these ozone increases are the effect of ozone rich air that is brought down by cumulus downdrafts from height levels where the ozone mixing ratio is larger. Thunderstorms can cause downward transport of ozone from the reservoir layer in the upper troposphere into planeta교 boundary layer(PBL). This complex interaction of source and sink processes can result in large variability fer vertical and horizontal ozone distributions. Thus a variety of meteorological precesses can act to enhance vertical mixing between the earth's surface and the atmospheric in the manner described fer thunderstorm.
