Immunomodulatory Effects of Fucoidan on NK Cells in Ovariectomized Rats

  • In this study we investigated the effects of supplementation with fucoidan from brown alga on the function of natural-killer (NK) cells to evaluate the possibility as an immunomodulator in ovariectomized (OVX) rats. A total of 18 female Wistar rats (six weeks) were used this study and 12 rats were OVX, and the rest of rats were sham-operated. The sham and one OVX group were fed standard diet, and the remaining OVX group received fucoidan (0.05% supplemented diet). After 12 weeks of supplementation, rats were sacrificed to assess the tumoricidal activity of the NK cells and the NO-iNOS regulation from splenocytes. The mass of body and the immune organs such as spleen and thymus were also studied. In OVX rats, body and thymus weights increased, however fucoidan supplementation did not change the body mass and organs weight compared to OVX group. Fucoidan supplementation increased NK cell activity and reduced NO-iNOS production in OVX rats. Ex vivo treatment of fucoidan increased NK cell activity in splenocytes from shame and OVX rats. Ex vivo, we confirmed that fucoidan partially reduced the NK cell activity in the presence of iNOS inhibitors in OVX-splenocytes. These results indicate fucoidan supplementation has a NK cell tumoricidal activity, which are regulated by the iNOS production in OVX rats. This suggests that fucoidan is useful for potential therapeutic strategies as a nutrient in regulating the NK cells in postmenopausal osteoporosis patients.

Comparative antiplatelet and antithrombotic effects of red ginseng and fermented red ginseng extracts

  • Background: Fermentation may alter the bioavailability of certain compounds, which may affect their efficacy and pharmacological responses. This study investigated the antiplatelet effects of red ginseng extract (RGE) and fermented red ginseng extract (FRG). Methods: A rodent model was used to evaluate the antiplatelet and antithrombotic effects of the extracts. Rats were orally fed with human equivalent doses of the extracts for 1 week and examined for various signaling pathways using standard in vivo and ex vivo techniques. Light transmission aggregometry was performed, and calcium mobilization, dense granule secretion, integrin αIIbβ3-mediated signaling molecules, cyclic nucleotide signaling events, and various protein molecules were evaluated ex vivo in collagen-stimulated washed platelets. Furthermore, antithrombotic properties were evaluated using a standard acute pulmonary thromboembolism model, and the effects on hemostasis were investigated using rat and mice models. Results: Both RGE and FRG significantly inhibited platelet aggregation, calcium mobilization, and dense granule secretion along with integrin-mediated fibrinogen binding and fibrinogen adhesion. cAMP levels were found to be elevated in RGE-treated rat platelets. Ginseng extracts did not exert any effect on prothrombin time and activated partial thromboplastin time. RGE-treated mice showed significantly better survival under thrombosis than FRG-treated mice, with no effects on hemostasis, whereas FRG-treated mice exhibited a slight increment in bleeding time. Conclusion: Both extracts, especially RGE, are remarkable supplements to maintain cardiovascular health and are potential candidates for the treatment and prevention of platelet-related cardiovascular disorders.

평생이모자반 젖산 추출물이 성장기 암컷흰쥐의 골 형성에 미치는 영향 (Effect of Lactic Acid Extracts of Sargassum horneri on Bone Formation in Female Sprage-Dawley Rats)

  • We investigated the effect of a lactic acid extract of Sargassum horneri (ExSL) as a calcium supplement on bone formation in 48 female Sprague-Dawley rats for 4 weeks of their growth phase. The rats were divided into four groups based on diet: two calcium-sufficient and two calcium-deficient diets. The normal control group (NC) was fed AIN-93G; the NCS group was fed the same diet containing 1% extract; the calcium-deficient control (DC) diet was based on AIN-93G; and the DCS group received the same calcium-deficient diet plus 1% extract. Bone formation in the rats was evaluated using the wet weight, length, diameter, and bone mineral density (BMD) of the femur. Serum parameters were also examined. The food intake among the groups did not differ significantly (P<0.05). The NCS group gained the most body weight, while the DC group gained much less weight than the other groups. The feeding efficiencies of the groups that received the extract (NCS and DCS) were slightly higher than those of the control groups (NC and DC). The calcium intakes of all groups depended on the amount of calcium in the feed; the NCS and DCS diets contained 12.15 mg more calcium than the NC and DC diets. The calcium absorption was lower in NCS than in DC and DCS, but significantly higher than in NC (P<0.05). The BMDs in the calcium-sufficient groups were not significantly different (P<0.05), while in the calcium-deficient groups the BMD was significantly higher in DCS than in DC (P<0.05). The serum calcium and phosphorus levels in all groups were not associated with markers of bone growth related to the extract. The osteocalcin content and alkaline phosphatase (ALPase) activity were higher in the calcium-deficient groups than in the normal groups (P<0.05). Ultimately, the osteocalcin content and ALPase activity were lower in DCS compared to DC. These results suggest that the addition of ExSL promotes bone formation and calcium absorption in growing rats.

Optimization of Procedure for Efficient Gene Transfer into Porcine Somatic Cells with Lipofection

  • The objective of this study was to establish conditions for transfection of a foreign gene into somatic cells using cationic lipid reagents and to evaluate the effects of transfection on in vitro development of somatic cell nuclear transfer (SCNT) embryos. Green fluorescent protein (GFP) gene was used as a foreign gene and a non-transfected somatic cell was utilized as a control karyoplast. Monolayers of porcine cells were established and subsequently transfected with a GFP-expressing gene (pEGFP-N1) using three types of transfection reagents (LipofectAMINE PLUS, FuGENE 6 or ExGen500). Donor cells used for SCNT included transfected fetal or adult fibroblasts and oviduct epithelial cells, either serum-fed or serum-starved. Oocytes matured in vitro for 42 h were reconstructed with either transfected or non-transfected porcine somatic cells by electric fusion and activation using a single DC pulse of 1.8 kV/cm for $30{\mu}s$ in $Ca^{2+}$ and $Mg^{2+}-containing$ 0.26 M mannitol solution. Reconstructed oocytes were subsequently cultured in NCSU-23 medium for 168 h and the developmental competence and cell number in blastocyst were compared. There were no significant differences (P>0.05) in fusion, cleavage rates or development to the blastocyst stage between non-transfected, transfected, serum-fed and serum-starved cells. However, the rates of GFP-expressing blastocysts were higher in the FuGENE 6 group (71.4%) among transfection reagents and in the fetal fibroblasts group (70.4%) for donor cells. These results indicate that fetal fibroblasts transfected with FuGENE 6 can be used as donor cells for porcine SCNT and that GFP gene can be safely used as a marker of foreign genes in porcine transgenesis.

감국약침이 과산화지질을 급여한 흰쥐의 지질대사, 항산화 및 면역계에 미치는 영향 (Effects of gamgook(Chrysanthemum indicum L.) herbal-acupuncture on lipid lowering effect, anti-oxidative capacity and anti-inflammatory effect in rat fed high oxidized fat)

  • Objectives: To investigate the effects of Chrysanthemum indicum L. pharmacopuncture on lipids, antioxidant capacity and anti-inflammation in rats fed high-fat diet. Methods: Hyperlipidemic rats induced by high-fat diet were divided into 5 groups: no treatment control (normal, n=8), high-fat diet only control (control, n=8), high-fat diet and Chrysanthemum indicum L. pharmacopuncture at CV4 group (TI, n=8), high-fat diet and Chrysanthemum indicum L. pharmacopuncture at CV17 group (TII, n=8), and high-fat diet and Chrysanthemum indicum L. pharmacopuncture at EX-HN3 group (TIII, n=8). They were given pharmacopuncture accordingly every other day for two weeks followed by analyses of lowering lipids effects, oxidative capacity and anti-inflammatory effects. Results: Compared with the control, pharmacopuncture groups showed significantly decreased plasma total cholesterol (TC), liver thiobarbituric acid reactive substance (TBARS), catalase, glutathione peroxidase, neutrophils, monocytes, plasma and liver IL-$1{\beta}$, and plasma and liver IL-6. In other parameters including plasma and liver triglyceride, liver TC, LDL-cholesterol, HDL-cholesterol, liver TBARS, supraoxide dismutase, total protein, albumin, blood cell analysis, plasma and liver TNF-$\alpha$, and IL-10, there was no significant difference between control and pharmacopuncture groups. No clear acupoint-specificity was observed. Conclusions: Chrysanthemum indicum L. pharmacopuncture may improve control of hyperlipidemia.

Extract of Curcuma zedoaria R. prevents atherosclerosis in apolipoprotein E-deficient mice

  • BACKGROUND/OBJECTIVES: Curcuma zedoaria R. (Zingiberaceae) has been used to treat headache, fever, and hypertension-related symptoms in Asian countries, including Korea, China, and Japan. We investigated whether dietary intake of a C. zedoaria extract (CzE) affected atherosclerosis in vivo. MATERIALS/METHODS: Apolipoprotein E-deficient (ApoE-/-) mice (n = 32) were fed a normal diet (ND), a high-cholesterol diet (HCD), an HCD containing CzE (100 mg/kg/day), or an HCD containing simvastatin (10 mg/kg/day) for 12 weeks. The anti-atherosclerotic effects were evaluated by observing changes in fatty streak lesions, immunohistochemical analysis, ex vivo fluorescence imaging, lipid profiles, and western blot analysis. RESULTS: The CzE-fed group showed a 41.6% reduction of atherosclerosis. Furthermore, CzE significantly reduced the levels of serum triglyceride, high-density lipoprotein, the chemokine (C-X3-C-motif ) ligand 1, the adhesion molecules vascular cell adhesion molecule-1, intracellular adhesion molecule-1, and E-selectin; down-regulation of tumor necrosis factor-α, interleukin-6, high mobility group box-1, and cathepsin levels in the aortic sinuses and aortas of ApoE-/- mice were also observed. CONCLUSIONS: The results suggest that the inclusion of a water extract of C. zedoaria in a HCD is closely correlated with reducing the risk of vascular inflammatory diseases in an ApoE mouse model.

블루베리나무에서 작은상제집나방 피해 보고 (The Report of the Damage for Saridoscelis sphenias (Lepidoptera: Yponomeutidae) on Blueberry Trees)

  • 집나방과(Yponomeutidae)에 속한 작은상제집나방(Saridoscelis sphenias Meyrick)은 2016~2017년 전남 진도와 완도에서 채집된 개체를 동정하여 2020 국내 미기록 종으로 보고되었으며, 마취목(Pieris japonica)과 진달래과에 속한 모새나무(Vaccinium bracteatum Thunb.)와 Leucothoe grayana Maxim. var oblongifolia (Miq.)가 기주식물로 알려져 있었다. 이 종은 2014년 8월 제주특별자치도 블루베리 재배지에서 처음 발견된 후 미정착된 것으로 취급되었으나, 2018년에 이어 2019년에도 제주시와 서귀포시 여러 블루베리 농장에서 발생이 확인되었다. 그 후 2020년부터는 노지 과원에서도 발견되고 있다. 부화유충이 처음 신초 줄기 속으로 침입하여 가해하다가 중간크기로 자란 후 탈출하여 몇 개의 신초를 그물로 철하고 집단적으로 가해하는 등 특이한 피해증상이 나타난다. 향후 피해가 우려되는 해충으로 피해증상과 생활사를 간단히 보고한다.

박하의 in vivo 생리활성 (In vivo Physiological Activity of Mentha viridis L. and Mentha piperita L.)

  • M. viridis와 M. piperita의 흰쥐에서의 생리활성을 확인하기 위해 수행한 실험 결과는 다음과 같다. 1. ADH 활성은 M. viridis ext. 1g/kg body weight of rat/day 투여군 (이하 M. viridis ex. 1 g 투여군)과 M. piperita ext. 0.2g/kg body weight of rat/day 투여군 (이하 M. piperita ext. 0.2g 투여군)이 음성대조군에 비해 각각 147% 및 144% 증가하여 양성대조물질인 Silymarin 투여군 (95%)보다 매우 효과적으로 알콜의 빠른 분해에 기여하는 것으로 확인되었다. 2. ALDH활성은 M. viridis ext. 1g 투여군과 M. piperita ext. 0.2g 투여군이 음성대조군에 비해 각각 154%, 158%의 활성증가를 나타냈으며 이는 Silymarin 투여군 (153%)보다 높은 활성으로 2종의 박하가 모두 효과적으로 알콜을 분해할 것으로 생각되었다. 3. CAT 활성은 M. viridis ext. 투여군이 M. piperita ext. 투여군보다 유의적으로 높았으며 특히, M. viridis ext. 투여군은 정상군이나 음성대조군 및 Silymarin 투여군보다도 활성이 높아 M. viridis ext. 투여군이 CAT에 의한 알콜분해에 있어서 M. piperita ext. 투여군보다 우수한 것으로 확인되었다. 4. Mn-SOD 활성은 M. viridis ext. 0.2g 및 1g 투여군은 음성대조군에 비해 각각 69% 및 75%를, M. piperita ext. 0.2g 및 1g 투여군은 각각 94% 및 86%를 나타내어 M. viridis ext. 투여군이 M. piperita ext. 투여군보다도 알콜투여에 의한 산화적 스트레스를 효과적으로 방어하는 것을 알 수 있었으며 특히, M. viridis ext. 투여군은 SiIymarin 투여군(84%)보다도 MN-SOD의 활성 회복에 효과적이었다. 5. GSH-px의 활성은 음성대조군에 비해 M. viridis ext. 0.2g 및 1g 투여군이 각각 81% 및 88%를, M. piperia ext. 0.2g 및 1g 투여군이 88% 및 67%를 나타내 알콜에 의해 유도된 GSH-px의 활성증가를 2종의 박하 모두 효과적으로 회복시켜주었다. 6. 혈청 GPT의 활성은 음성대조군에서 정상군에 비해 유의적으로 증가 (126%)하였으나 M. viridis와 M. piperita 투여군은 음성대조군이 나타낸 활성에 대해 $69%{\sim}72%$의 활성을 나타내 우수한 GPT활성 회복력을 가졌으며 이는 Silymarin(93%) 보다도 우수하였다. 2종의 박하 추출물 투여군들 사이에는 혈청 GPT 활성에 대한 유의적인 차이가 나타나지 않았다. 7.과산화물질 (TBARS)의 함량은 정상군에 비해 음성대조군에서 약간의 함량 증가가 관찰되었으나 유의적인 수준은 아니었으며 대부분의 박하 추출물 투여군과 Silymarin 투여군에서 과산화지질이 비슷한 수준으로 증가하여 박하는 알콜에 의한 과간화지질생성에 대한 저해효과가 낮은 것으로 확인되었다. 8. 상대간장중량은 각 실험군별로 상대간장중량에서의 차이는 다소 있었으나 유의성은 없었다.

Novel Antioxidants and Atherosclerosis

  • Coronary heart disease (CHD) has been the number one killer in western society for a long time, and CHD in most instances is due to atherosclerosis. One of the earliest events in atherogenesis is the intracellular accumulation of lipids, particularly cholesterol esters, in the aortic intima. The lipids presumably came from the uptake of plasma lipoproteins, particularly from LDL. These foam cells were identified as being predominantly as macrophages. Currently, it is believed that oxidation of low density lipoprotein (LDL) might contribute to the generation of foam cells. An outcome of the oxidation hypothesis is that the consumption of antioxidants would be beneficial. In this study, Boldine, an alkaloid of Peumus boldus was tested for their antioxidant potency both in, in vitro oxidation system and in mouse models. Boldine decreased the ex-vivo oxidation of Low-density lipoprotein (LDL). In vivo studies were performed to study the effect of these compounds on the atherosclerotic lesion formation in LDL r-/- mice. Three groups of LDL r-/- mice (N=12 each) were fed an atherogenic diet. Group 1 was given vehicle and group 2 and 3 were given 1 and 5 mg of Boldine/day in addition to the atherogenic diet. The results indicated that there was a decrease in lesion formation reaching a 40% reduction due to Boldine compared to controls. The in vivo tolerance of Boldine in humans (has been used as an herbal medicine in other diseases) should make it an attractive alternative to vitamin E.

항만 옥외저장소 위험물의 안전관리 개선 방안 (A Study on the Improvement of Dangerous Goods Safety Management in Maritime Terminal)

  • According to the research and investigations performed on Port Facilities for cargo-works of hazardous materials around the Port of Busan, a conclusion was drawn that the manually operating foam tower monitors are ineffective as they are hard to access due to the radiant heat under contingency circumstances such as a fire. Moreover, in some cases, the effective range of Foam Tower Monitors are insufficient to reach docked ships and manifolds where hazardous materials are being unloaded. In addition, Dangerous Goods freights are inadequately and inefficiently stored regardless properties of hazardous substances due to complicated local regulations, and some are equipped with inapplicable facilities for docked ships. Therefore, in order to effectively counteract the contingencies and obtain safety, it would be recommended to install adequate facilities.
