하계 경기만의 수질오염과 생산력에 관한 연구 1

  • The extent of water pollution was investigated at 4 stations in Kyonggi Bay during the summer seasons in 1970 and 1971. The concentrations of dissolved oxygen, total hardness, ammonia, nitrate, nitrite, phosphate, chemical oxygen demand, salinity, biochemical oxygen demand, coliform bacteria and facel coli were examined together with the measurement of pH and transparency. The relationship between the extent of pollution and the distance from the Inchon Bay also was examined. The concentrations of biochemical oxygen demand, chemical oxygen demand, ammonia, nitrate, nitrite, phosphate, coliform bacteria nad fecal coli were all highest at station 1, and lowest at station 4. Values were somewhat higher at low tide level in general. On the contrary, dissolved oxygen concentration was highest at station 4 and lowest at station 1. The highest and lowest values of biochemical oxygen demand were 10.88 ppm at station 1 and 0.27 ppm at station 4. The chemical oxygen demand concentrations at station 1 and 4 were 1.90 ppm and 0.63 ppm. Ammonia concnetration at station 1 was 0.43 pp, and was nearly 5 times as much as that at station 4. The values were $2.45{\times}10^{-4}$ ppm at station 1, and $6{\times}10^{-4}$ ppm at station 4. Nitrite concnetration at station 1 was $3{\times}10^{-4}$ppm and was the highest, while the lowest was $9.45{\times}10^{-5}$ ppm at station 4. Phosphate value at station 1 too was the highest and was about 4 times as much as that at station 4. Coliform bacteria were most abundant at station 1, and were counted to be 1.$1.7{\times}10^{-5}$cells/ml. At station 4, the number greatly reduced to 8 * 10$^{2}$ cells/ml. The number of fecal coli at station 1 was $2{\times}10^{-4}$ cells/ml. But at station 4, no fecal coli was detected at high tide level. At low tide level, 3 cells/ml were counted at station 4. In all of these, the highest data were obtained at low tide level, while most of the lowest value, at high tide level. Generally, values at statio 1 were 3-5 times as much as those at station 4. Concentration of dissolved oxygen at station 1 was 0.366 mg-atoms/1 and was the lowest. The highest value was 0.420 mg-atoms/1 and was recorded at station 4. The highest at station 4, which certainly were believed to be the result of the dilution by the fresh water of the Han river flowing into the Inchon Bay. As we can see from the data above, the extent of pollution was highest at station 1, the nearest from Inchon harbor, and lowest at station 4, where is the farthest, Station 1 and 2 were proved to be much polluted, but station 3 and 4, not.

Evaluation of FSP (Fermented Soy Protein) to Replace Soybean Meal in Weaned Pigs: Growth Performance, Blood Urea Nitrogen and Total Protein Concentrations in Serum and Nutrient Digestibility

  • A total of one hundred and forty four weaned pigs with an average BW of $8.09{\pm}0.05$ kg were used in a 28 day study to investigate the effects of fermented soy protein on growth performance, blood urea nitrogen and total protein concentrations in serum and nutrient digestibility in weaner pigs. Pigs were blocked by initial body weight and randomly allocated to one of four dietary treatments in a randomized complete block design. There were six replications per treatment. Dietary treatments included: SBM (corn-soybean meal basal diet), F 5, 10 and 15 (fermented soy product was used at 5, 10 and 15% to replace soybean meal in basal diet, respectively). ADG (average daily gain) and ADFI (average daily feed intake) were not affected (p>0.05) by dietary treatments during the entire 4-wk study period. There were linear increments in feed efficiency (p<0.01) as the dietary FSP level increased during the entire feeding period. No significant differences were observed for dry matter and nitrogen digestibility during the experimental period (p>0.05). Digestibilities of histidine, lysine and methionine were increased as the FSP level increased (linear effect, p<0.05, p<0.01). Among non-essential amino acids, alanine, glutamic acid, serine, tyrosine and total non essential amino acid digestibilities were increased linearly (p<0.05, p<0.01). There were quadratic effects in protein digestibility (p<0.05). Total amino acid digestibility of the F15 diet was improved compared with the F5 diet (p<0.05). There were no significant differences in fecal consistency score among the treatments (p>0.05). At the end of experiment, BUN (blood urea nitrogen) concentration was increased as the FSP level increased (linear effect, p<0.01) and total protein concentration was lowest (p<0.05) for pigs fed the SBM diet among treatments. In conclusion, the feeding of 10 or 15% FSP to nursery pigs improved feed efficiency, amino acid digestibility and blood urea nitrogen and total protein concentrations in blood.

Effects of Feed Processing and Feeding Methods on Growth and Carcass Traits for Growing-Finishing Pigs

  • The present experiment was conducted to evaluate the effects of feed processing and feeding methods on growth performance and carcass characteristics of growing-finishing pigs. A total of 72 pigs (LYD, 22.24 kg BW) were employed for a 90-d feeding trial. Treatments were 1) mash dry feeding (MD), 2) mash wet feeding (MW), 3) pellet dry feeding (PD), and 4) extruded pellet dry feeding (EPD). Corn, soybean meal and wheat bran in the basal diets were extruded before mixing and pelleting for EPD diet production. Ileal or fecal digestibility and carcass traits including lean meat percentage and weights of stomach ulcer were also examined. During the growing period, pigs fed PD showed improved (p < 0.05) average daily gain (ADG) and feed conversion (F/G) over those fed MD, whereas no significant differences in average daily feed intake (ADFI) were found among dietary treatments. Finisher pigs fed MD showed lover, but not significant, ADG and F/G than those fed MW, PD or EPD. For the overall period, pigs fed PD grew faster (p < 0.05) than those fed MD or EPD. Feed intake was different between the two feeding methods (MD vs MW), and between the two processed feeds (PD vs EPD). The digestibility of crude fat was higher (p < 0.05) in pigs fed EPD than in pigs fed mash feeds. NFE digestibility of EPD treatment was also higher (p < 0.05) than that of PD. Back fat (10th rib area) was thicker (p < 0.05) tn pigs fed MD than in pigs fed EPD. other carcass traits including incidence of esophagogastric ulcers were not different among treatments. In conclusion, pelleting appeared to bo the desirable processing methods and wet feeding could also be recommended for growing-finishing pigs.

Growth, Nutrient Utilization and Amino Acid Digestibility of Dairy Calves Fed Milk Replacers Containing Different Amounts of Protein in the Preruminant Period

  • This study was designed to examine the effects of different dietary protein levels on growth performance, nutrient utilization, amino acid (AA) digestibility and serum biochemical parameters of preruminant dairy calves. Fifteen healthy new-born calves were randomly allotted to three experimental groups and fed with different milk replacer that contained 18% (LP), 22% (MP) or 26% (HP) of protein. The results showed that final BW, net gain and ADG were significantly higher in the MP group than in LP and HP groups (p<0.05). In addition, the apparent digestibility of CP in the MP group was significantly higher than in the other two groups (p<0.05). The values of N intake and fecal N excretion were significantly increased following the increase of dietary protein content (p<0.05). However, in all three groups of animals, dietary protein content had no significant effect on urinary N concentration (p>0.05). BUN concentration, on the other hand, decreased as calves grew but increased following the increase of dietary CP content. Furthermore, no significant differences in digestibility of amino acids were observed among these three groups of animals (p>0.05). We concluded that calves fed with milk replacer containing 22% of protein had better growth performance and nutrient utilization as compared to animals treated with milk replacer containing either 18% or 26% of protein.

Biotransformation of Pueraria lobata Extract with Lactobacillus rhamnosus vitaP1 Enhances Anti-Melanogenic Activity

  • Isoflavone itself is less available in the body without the aid of intestinal bacteria. In this study, we searched for isoflavone-transforming bacteria from human fecal specimens (n = 14) using differential selection media. Isoflavone-transforming activity as the production of dihydrogenistein and dihydrodaidzein was assessed by high-performance liquid chromatography and we found Lactobacillus rhamnosus, named L. rhamnosus vitaP1, through 16S rDNA sequence analysis. Extract from Pueraria lobata (EPL) and soy hypocotyl extract were fermented with L. rhamnosus vitaP1 for 24 and 48 h at $37^{\circ}C$. Fermented EPL (FEPL) showed enhanced anti-tyrosinase activity and antioxidant capacities, important suppressors of the pigmentation process, compared with that of EPL (p < 0.05). At up to $500{\mu}g/ml$ of FEPL, there were no significant cell cytotoxicity and proliferation on B16-F10 melanoma cells. FEPL ($100{\mu}g/ml$) could highly suppress the content of melanin and melanosome formation in B16-F10 cells. In summary, Lactobacillus rhamnosus vitaP1 was found to be able to biotransform isoflavones in EPL. FEPL showed augmented anti-melanogenic potential.

Effect of Dietary Structural to Nonstructural Carbohydrate Ratio on Rumen Degradability and Digestibility of Fiber Fractions of Wheat Straw in Sheep

  • The effect of different dietary structural carbohydrate (SC) to nonstructural carbohydrate (NSC) ratios on fiber degradation, digestion, flow, apparent digestibility and rumen fluid characteristics was studied with a design using 18 wethers fitted with permanent rumen and duodenum cannulae. All sheep were divided into six groups randomly, receiving six diets with varying SC to NSC ratios. All diets contained the same proportion of wheat straw and concentrate. The dietary SC to NSC ratios were adjusted by adding cornstarch to the concentrate supplements. The duodenal and fecal flows of dry matter (DM), neutral detergent fiber (NDF), acid detergent fiber (ADF), hemicellulose (HC) and cellulose (CEL) were estimated using chromium-mordanted wheat straw as a flow marker. The degradation parameters of wheat straw DM, NDF, ADF, HC and CEL were determined by incubating the ground wheat straw in nylon bags in the rumen for different periods of time. There was no effect (p>0.05) of the different dietary SC to NSC ratios on rumen pH or $NH_3$-N, but acetate, propionate and butyrate concentrations were significantly affected (p<0.05 or p<0.01) by dietary SC to NSC ratios in the rumen fluid. When the dietary SC to NSC ratio was 2.86, the highest rumen degradability of wheat straw DM, NDF, ADF and CEL was found, but the highest apparent rumen digestibilities of DM, NDF, ADF, HC and CEL occurred at a 2.64 SC to NSC ratio. However, because of compensatory digestion in the hindgut, the apparent digestibilities of DM, NDF, ADF, HC and CEL were highest when the dietary SC to NSC ratio was 2.40. In conclusion, there is a optimal range of dietary SC to NSC ratios (between 2.86 and 2.40) that is beneficial to maximize wheat straw fiber degradation and apparent digestibility.

Long-term Bias of Internal Markers in Sheep and Goat Digestion Trials

  • Two digestion trials, one with sheep and another with goats, were conducted to evaluate the long-term bias (LTB) of the indigestible dry matter (iDM), indigestible neutral detergent fiber (iNDF) and indigestible acid detergent fiber (iADF) internal markers. The study used eight Santa In$\hat{e}$s castrated male sheep (average body weight of 16.6 kg) distributed in two $4{\times}4$ Latin squares and eight Saanen castrated male goats (average body weight of 22.6 kg) distributed in two $4{\times}4$ Latin squares. The experiments were conducted simultaneously, and the animals were housed in 1.2 $m^2$ individual pens with wood-battened floors equipped with individual feeders and drinkers. The animals received isonitrogenous diets that were offered ad libitum and contained 14% crude protein and 70% sugar cane (with 0, 0.75, 1.5 or 2.25% CaO, in natural matter percentage), corrected with 1% urea and 30% concentrate. The experiment consisted of four experimental periods of 14 d each, with the feed, leftovers and feces sampled on the last four days of each period. The marker concentrations in the feed, leftovers and fecal samples were estimated by an in situ ruminal incubation procedure with a duration 240 h. The relationship between the intake and excretion of the markers was obtained by adjusting a simple linear regression model, independently from the treatment (diets) fixed effects and Latin squares. For both the sheep and goats, a complete recovery of the iDM and iNDF markers was observed (p>0.05), indicating the absence of LTB for these markers. However, the iADF was not completely recovered, exhibiting an LTB of -9.12% (p<0.05) in the sheep evaluation and -3.02% (p<0.05) in the goat evaluation.

Seasonal Variations of Water Quality in the Lower Part of the Nagdong River (낙동강 하류수질의 계절적 변화)

  • The Nagdong is one of the biggest rivers in Korea, which is very important water source not only for tap water of Pusan city but also for the industrial water. Therefore, authors tried to check the water quality year by year. In this experiment one hundred and twenty water samples collected from August 1983 to July 1984 were analyzed bacteriologically and physiologically. Fifteen sampling stations were established between near Samrangjin and estuary of the river. To evaluate the water quality, temperature, pH, chloride ion, salinity, chemical oxygen demand (COD), electrical conductivity, nutrients, total coliform, fecal coliform, fecal streptococcus, viable cell count and bacterial flora were observed. The variation of water temperature was ranged $-1.5{\sim}29.0^{\circ}C$ (Mean value $13.9{\sim}16.5^{\circ}C$), it in spring was higher as $10{\sim}15^{\circ}C$ about $10^{\circ}C$ than in winter and it in autumm was very stabilized as about $20^{\circ}C$ at each station. The pH variation of the samples was ranged $6.68{\sim}9.15$. The range of concentration of chloride ion and salinity varied $7.4{\sim}l,020.5$ mg/l and $1.05{\sim}33.0\%0$, respectively. Especially, salinity of the 3rd water war was the higher than others as $25.76{\sim}31.58\%0$. COD was ranged $1.45{\sim}14.94$ mg/l and the lower part of the Nagdong River was heavily contaminated by domesitc sewage and waste water from the adjacent factor area. The range of electrical conductivity was $1.360{\times}10^2{\sim}5.650{\times}10^4{\mu}{\mho}/cm$ and that was by far higher the estuary than the upper. Concentration of nutrients were $0.008{\sim}0.040$ mg/l (Mean value $0.019{\sim}0.068$ mg/l) for $NO_2-N,\;0.038{\sim}5.253$ mg/l ($0.351{\sim}2.347$ mg/l) for $NO_3-N,\;0.100{\sim}2.685$ mg/l($0.117{\sim}1.380$ mg/l) for $NH_4-N,\;0.003{\sim}0.084$ mg/l($0.014{\sim}0.065$ mg/l) for $PO_4-P$ and $0.154{\sim}6.123$ mg/l ($1.165{\sim}3.972$ mg/l) for $SiO_2-Si$, respectively. Usually nutrients contents of the water in the upper part(included station 1 to 5) were higher than those of the estuarine area. The bacterial density of the samples ranged 7.3 to 460,000/100 ml for total coliforms, 3.6 to 460,000/100 ml for fecal coliform, $0{\sim}46,000/100ml$ for fecal streptococcus and $<30{\sim}1.2{\times}10^5/ml$ for viable cell count. Composition of coliform was $28\%$ Escherichia coli group, $18\%$ Citrobacter freundii group, $31\%$ Enterobacter aerogenes group and $22\%$ others. Predominant species among the 659 strains isolated from the samples were Pseudomonas spp. ($42\%$), Flavobacterium spp. ($20\%$) and Moraxella spp. ($12\%$).

Preparation of Fermented Milk Added with Powder of Opuntia ficus-indica var. saboten and Its Sensory Characteristics (백년초 분말을 첨가한 발효유 제조 및 관능적 특성)

  • The effects of Opuntia ficus-indica var. saboten powder on the growth of lactic acid bacteria (LAB) were investigated in order to explore the possibility of manufacturing fermented milk containing the powder. Differences in pH, acidity, LAB counts, viscosity, and sensory evaluation were measured. Also the effects of dietary supplementation on the growth of piglets were evaluated by feeding fermented milks containing 0.1%, 0.2%, 0.5%, and 1.0% Opuntia ficus-indica var. saboten powder. Levels of pH, titratable acidity, viable LAB counts and viscosity were significantly different by the addition of the powder. When fermented milks containg 0.1%, 0.2%, 0.5% and 1.0% Opuntia ficus-indica var. saboten powder were kept at $4^{\circ}C$ for 30 days, viable LAB counts remained high after 30 days of storage. The effects of dietary supplementation of 0.2% Opuntia ficus-indica var. saboten fermented milk were investigated by examining piglet growth rate and fecal ammonia gas release. The piglets were fed 100g/herd of the fermented milk for 14 days. Average daily body weight gain was significantly (p<0.05) improved (110%) with dietary supplementation of the fermented milk, compared to a control group. More over, fecal ammonia gas emissions were reduced by dietary supplementation of the Opuntia ficus-indica var. saboten fermented milk. Sensory evaluation results showed that the samples containing 0.1% and 0.2% Opuntia ficus-indica var. saboten powder had the most parameters similar to those of the control.

Use of Pyrosequencing for Characterizing Microbial Community at Phylum Level in Yeongsan River Watershed during Early Summer (Pyrosequencing을 이용한 하절기 영산강 유역의 Phylum 계층의 세균 군집 조사)

  • We have conducted pyrosequencing for freshwater microbial community analyses. Fourteen sites along the Yeongsan river were selected for this study, and samples were collected monthly from May to July, 2012. Total 987,380 reads were obtained from 42 samples and used for taxonomic classification and OTU distribution analysis. Our results showed that high geographical and temporal variation in the phylum level bacterial composition, suggesting that microbial community is a very sensitive parameter affected by the surrounding environments including tributaries and land use nearby. In addition, we conducted an OTU-based Microbial Source Tracking to identify sources of fecal pollution in the same region. From this study Firmicutes was found to be the most influential taxa in this region. Here, we report that the use of pyrosequencing based microbial community analysis may give an additional information on freshwater quality monitoring, in addition to the currently used water quality parameters, such as BOD and pH.
