Fecal sterol을 이용한 울산만과 주변해역 퇴적물내 하수기인 유기물 평가 (Evaluation of sewage-derived organic matter using fecal sterols in the sediments from Ulsan Bay and adjacent areas)

  • Surface sediments (0$\~$4 cm) were sampled at 40 stations from Ulsan Bay and its adjacent areas of Korea, to evaluate the contamination by sewage-derived organic matters using fecal sterols. Some sterols were quantified by gas chromatography coupled to mass spectrometry (GC/MS). Total concentrations of eight sterols (co­prostanol, cholesterol, cholestanol, epicholestanol, epicoprostanol, ${\beta}-sitosterol,$ brassicasterol, and stigmasterol) in the sediments from Ulsan Bay and its vicinity varied from 929 to 23,444 ng/g dry weight. The most predom­inant sterols were cholesterol and coprostanol, accounting for $33\~72{\%}$ of total sterols. The concentration of coprostanol known as the indicator of human feces ranged from 141 to 8,257 ng/g dry weight. In particular, the coprostanol concentrations in the sediments from Jansaengpo Harbor and Taehwa River exceeded the value of 1,000 ng/g dry weight, suggesting that these areas could be considered as the hot-spot zones by municipal sewage contamination. Some molecular indices and multivariate data analysis were used to assess the origin of these sterols in the sediments. Results showed that the major routes of sewage contamination in Ulsan Bay and its vicinity were the input through Jansaengpo Harbor and Taehwa River.

마산만의 하수기인 유기물 fecal sterol의 분포 (Distribution of sewage-derived organic matter using fecal sterol in Masan Bay, Korea)

  • Surface sediments and seawater were sampled at Masan and Haengam Bays of Korea, to evaluate contamination by sewage-derived organic matter using fecal sterols, Six stream-water samples into Masan and Haengam Bays were also sampled. Total concentrations of eight sterols (coprostanol, epichloestanol, epicoprostanol, cholesterol, cholestanol, brassicasterol, stigmasterol and $\beta-sitosterol$) were in the range of $1,274\~4,768{\mu}g/g$ dry weight in suspended particulate from the stream-water, $292\~2,244{\mu}g/g$ dry weight in suspended particulate from the seawater and $4.5\~27.2{\mu}g/g$ dry weight in the sediments. Although sterol compositions in sediments, seawater and stream-water were different, cholesterol was the predominant sterol in all samples. The proportion and concentration of coprostanol, a sewage tracer, in stream-water was much higher than those in sediment and seawater. The sterol levels including coprostanol in the sediments and seawater were higher at inner basin than at the outer bay. Some molecular indices and multivariate statistical analysis were used to assess the origin of these sterols and sewage contamination in the study area. The sterol composition patterns in stream-water were mainly associated with contamination by sewage-derived organic matters and those of seawater were associated with the activities of marine-originated organisms. Sterol levels in the sediments were both from the sewage input through stream-water and the marine- originated organisms. This survey suggests that the main source/route of sewage-derived organic matters in Masan Bay is the input of stream-water into the bay.

  • Canine parvovirus(CPV) is a very highly contagious virus causing hemorrhagic enteritis and myocarditis mainly in young dogs. The diseases were first recognized in 1978, and then spread throughout the world by 1980. The main source of the infection seems to be the feces of infected dogs, at the same time feces are suitable materials for detection of virus in the enteric form exactly for the same reasons. Recently, a new technique of in vitro DNA amplification, Known as the polymerase chain reaction (PCR), has been widely applied to clinical viral diagnosis because of its sensitivity, specificity and rapidity. In this research, we attemped to set up the PCR for the detection of CPV in fecal samples and conformed the canine parvpviral enteritis by PCR. To increase the sensitivity and specificity of a PCR, the nested PCR (two-step PCR) was performed. We also surveyed the contamination status of CPV in the research using fecal specimen was highly sensitive and specific. Of the 100 fecal specimens suspected canine parvoviral enteritis, 45 fecal specimens were positive in HA test, 64 fecal specimens were positive in the first PCR, and 87 fecal specimens were positive in the second PCR. CPV contamination status of animal clinics and breeding centers was serious, wo hygienic management of environment in which dogs are reared is required. The nested PCR described here seems to be a rapid, sensitive and specific for the detection of canine parvovirus.

서울지역 교육시설 및 공원 내 토양 중 분원성대장균군 오염실태 (Survey of fecal coliform contamination in sands of educational facilities and parks in Seoul)

  • Fecal coliform bacteria are indicators of fecal contamination and of the potential presence of pathogens associated with animal excreta, wastewater, sludge. This study examined the distribution of fecal coliform in sand of the playground at educational facilities and parks in Seoul. Fecal coliform were detected from 61 (22.1%) places out of 276 playgrounds. Escherichia (E) coli (75.4%) was found most frequently, followed by Cronobacter (Cro) sakazakii (8.7%), Klebsiella (K) pneumoniae (7.2%) and Actinobacter baumannii (4.3%). We also tested the bacteria for their susceptibility to 16 antibiotics using disk diffusion method. All the isolates were highly susceptible to cefepime, gentamicin, tetracycline, ciprofloxacin, enrofloxacin, nalidixic acid and sulfamethoxazole/ trimethoprim, but resistant to apramycin and cephalothin. E. coli and Cro. sakazakii were highly resistant to apramycin (100% and 83.3%), followed by cephalothin (51.9% and 50%) and ampicillin (19.2% and 16.7%). K. pneumoniae was highly resistant to ampicillin and apramycin (100%, respectively) but the remaining antibiotics were susceptible.

  • The direct detection of intestinal pathogens and viruses often requires costly, tedious, and time-consuming procedures. These requirements developed a test to show that the water was contaminated with sewage-borne pathogens by assessing the hygienic quality of water based on indicator microorganisms whose presence indicates that pathogenic microorganisms may also be present. Various groups of microorganisms have been suggested and used as indicator microorganisms. Proposed and commonly used microbial indicators are total coliforms, fecal coliforms, fecal streptococci, Clostridium perfringens, heterotrophic plate count, bacteriophage, and so on. Unfortunately, most, if not all, of these indicators are not ideal because of the sensitivity and resistance to environment stresses and disinfection. However, the development of gene probes and PCR technology may give hope for the discovery of rapid and simple methods toy detecting small number of fecal pathogens in various environments.

연안 하구역 내의 비점오염부하량 산정을 위한 수학모델의 적용 (A Mathematical Framework for Estimating Non-point Waste Load at Enclosed Beaches)

  • Beaches in estuaries, bays, and harbors are frequently contaminated with indicators of human pathogens such as fecal indicator bacteria. Tracking down the sources of contamination at these enclosed beaches is complicated by the many point and non-point sources that could potentially degrade water quality along the shore. A mathematical framework was developed to test quantitative relationships between fecal indicator bacteria concentration in ankle depth water at enclosed beaches, the loading rate of fecal indicator bacteria from non-point sources located along the shore, physical characteristics of the beach that affect the transport of fecal indicator bacteria across the beach boundary layer, and a background concentration of fecal indicator bacteria attributable to point sources of fecal pollution that impact water quality over a large region of the embayment. Field measurements of fecal indicator bacteria concentrations and water turbulence at an enclosed beach were generally consistent with predictions and assumptions of the mathematical model, and demonstrated its utility for assessing waste load of non-point sources, such as runoff, bather shedding, bird droppings, and tidal washing of contaminated sediments.

도심지역 인공호의 수질관리를 위한 지표세균에 관한 연구 (A Study on Indicator Bacteria for Water Quality Management of Urban Artificial Lakes)

  • Distribution of fecal pollution indicator bacteria and environmental parameter were investigated of urban artificial lakes. An average concentration of temperature, pH, SS, DO, $COD_{Mn}$, T-P, T-N, Turbidity, Chl-a were $21.5^{\circ}C$, 8.07, 116.70 mg/l, 8.66 mg/l, 2.24 mg/1, 0.52 mg/l, 1.71mg/l, 80.54 NTU, and 52.12 mg/l respectively. From the results of bivariate correlation analysis, fecal contamination indicator bacteria were found to be mutually correlated. And turbidity and suspended solid were correlated. From the results of principal component analysis, four factors were extracted. And four factors of variance explained up to 81.5 percentage. Factor 1 was pollution pattern by fecal contamination, factor 2 was physical pollution pattern by pollution source, factor 3 was natural pollution by precipitation, and factor 4 was artificial pollution pattern by organism.

Distribution of Fecal Sterols, Nonylphenol, and Polycyclic Aromatic Hydrocarbons in Surface Water from Masan Bay, Korea

  • Fecal sterols, nonylphenolic compounds (NPs), and polycyclic aromatic hydrocarbons (PAHs) were determined in surface water from Masan Bay and its adjacent rivers in February 2005. Concentrations of coporstanol (Cop), an indicator of fecal pollution, in surface water ranged from <10 to 13,853 ng/L, and concentrations of nonylphenol, the most toxic of the NPs, ranged from 10.2 to 481 ng/L, and concentrations of PAHs ranged from 8.61 to 223 ng/L. The concentrations of the compounds measured in this study were lower than or comparable to those at other locations in Korea and other countries. The contamination of Cop and PAHs in surface water was associated with the discharge from rivers passing through cities and/or industrial complexes. The NP contamination was associated with wastewater treatment plant (WWTP) effluents through outfalls as well as riverine discharge. Compared to ecotoxicological values, the concentrations of NPs from rivers, the mouths of rivers, and WWTP outfall areas exceeded guidelines, suggesting that hot spot areas may pose a potential risk to sensitive species.

A Preliminary Study: Antibiotic Resistance of Escherichia coli and Staphylococcus aureus from the Meat and Feces of Various South African Wildlife Species

  • This study determined the antibiotic resistance patterns of Escherichia coli and Staphylococcus aureus from the raw meat and feces of three game species from three different farms across South Africa. The Kirby-Bauer disk diffusion method was used according to the Clinical and Laboratory Standards Institute 2018 guidelines. E. coli was tested against ampicillin, ceftazidime, chloramphenicol, streptomycin, sulphafurazole and tetracycline. S. aureus was tested against tetracycline, erthromycin, vancomycin, penicillin, oxacillin and cefoxitin. There were no significant differences in the E. coli antibiotic resistance profiles between the meat and fecal samples (except towards ceftazidime where 5% of the meat isolates were resistant and 0% of the fecal isolates). The S. aureus meat isolates showed high (75%) resistance towards penicillin and on average, 13% were resistant to oxacillin/ cefoxitin, indicating methicillin resistance. The results from this study indicate that there is incidence of antibiotic resistant bacteria from the feces and meat of wildlife species across South Africa, suggesting that cross contamination of the meat occurred during slaughter by antibiotic resistant bacteria from the abattoir personnel or equipment and or from carcass fecal matter. In addition, the results highlight the importance of food safety and hygiene procedures during slaughter to prevent cross-contamination of antibiotic resistant bacteria, as well as pathogens, onto raw meat.

  • Bovine mastitis is an important disease causing serious economic loss in dairy production and food poison in public health. Escherichia coli and Staphylococcus aureus are the major causative agents of bovine mastitis. These bacteria were found in milk and environmental condition such as feces, water, soil and so on. Bovine mastitis causative micro-organisms can survive in 1-2 weeks in feces and bed complexes. Low level of percentage of water content (PWC) of feces and bed complexes can reduce the spreading of bovine mastitis incidence from environmental contamination. In this study, we developed the fecal dehydrating system and determined the elimination rates of bovine mastitis causative agent from feces and bed complexes. To develop the fecal dehydrating system, the screw pressurized dehydrating system was used and the maximum rate of dehydrating was reached to 52% PWC using 90% PWC (wet base) of fecal and bed complexes. The elimination rates of the dehydrating system for E. coli and S. aureus were reached at 41.19 $\pm$ 7.84% to 62.55 $\pm$ 8.71% in various percentages of PWC of feces and bed complexes (80, 85 and 90%). These results suggested that the application of fecal dehydrating system would reduce the exposure of dairy cattle to bovine mastitis causing agents contaminated feces and bed complexes, and can be used for environmental bovine mastitis control avoiding misuse or abuse of chemical disinfectants and antibiotics in dairy farm.
