  • 본 실험은 alcohol 사료 처리에 따른 in vitro 발효 pattern의 변화를 검토하여 alcohol 사료 처리가 반추위내 발효 pattern에 미치는 영향을 규명코자 실시하였다. 시험구 처리는 배합사료를 첨가하는 대조구(control), 배합사료와 1, 3 및 5%의 alcohol 사료(Alcoholic feed; AF)를 첨가하는 처리구를 각각 AF-1구, AF-3구 및 AF-5구로 나누어 실시하였다. 배양 12시간 동안 ammonia 농도는 AF-1, AF-5구에서 대조구에 비해 낮았다(p<0.05). 배양액의 alcohol 농도는 alcohol 첨가 수준이 증가할수록 높았다(p<0.05). 배양 12시간에 TVFA 농도는 AF-1, AF-3 및 AF-5에서 대조구에 비해 현저히 높았다(p<0.05). 배양 8~12시간 사이에 acetate 비율은 대조구에서 빠른 속도로 감소하였으며, AF-1, AF-3 및 AF-5구에서는 일정한 수준을 유지하는 것으로 나타났다. Propionate 비율은 배양 8~12시간 사이에 AF-1, AF-3 및 AF-5에 비해 대조구에서 빠른 속도로 증가하였다(p<0.05). Butyrate 및 valerate 비율은 AF-1, AF-3 및 AF-5에서 대조구에 비해 높았다(p<0.05). Caproate 비율은 배양 8시간에 AF-1구, AF-3구 및 AF-5구에서 각각 0.05, 0.58 및 0.47M%로 나타났으나, 대조구에서는 검출되지 않았다. 본 실험의 결과에서 alcohol 사료의 처리는 in vitro 반추위액의 ammonia, alcohol 및 휘발성지방산 농도와 pH에 긍정적인 영향을 미치는 것으로 판단된다. 또한 본 실험의 결과에서는 5%의 alcohol 첨가 수준이 여타 alcohol 첨가 수준에 비해 반추위 발효에 보다 효과적인 것으로 사료된다.

  • 포도당 20%와 효모엑기스가 함유된 배지에서 $40^{\circ}C$에서 48시간에 걸친 에탄올발효 과정 중에 일어나는 고온성 알코올 발효 효모균주 Saccharomyces cerevisiae KNU5377의 세포 내에서 일어나는 생리적 변화를 살펴보고자 했다. 그 결과, 이 발효 효모균주는 $40^{\circ}C$ 48시간의 발효로써 11.4% alcohol을 생성하여, 고온발효인데도 불구하고 우수한 발효능을 가진 것을 확인할 수 있었다. 또한 12시간 발효과정동안 배지의 pH가 시작단계의 pH 6.0이 4.1로 내려갔으며, 이후 에탄올발효가 완성될 때까지 거의 변화하지 않고 이 값으로 유지되었다. 발효 12시간이 지나면서 세포막의 지방산의 조성은 불포화지방산인 $C_{16:1}$ (palmitoleic acid)가 포화지방산인 $C_{16:0}$ (palmitic acid)보다 1.5배나 증가하였으며, $C_{18:1}$보다는 2배 가까이 증가하여 구성되어 있었다. 48시간 배양한 세포의 2차 전기영동(2-D)을 통한 세포내 단백질의 발현정도를 proteomics분석법으로 살펴본 바, phosphoglycerate kinase가 가장 크게 발현했음을 Mass Spectrometry를 통해 알 수 있었다. 그 뒤를 이어 adenylate kinase, Cys3p, Tdh3p, translational elongation factor 등이 크게 발현된 것을 알게 되었다. 이들은 직간접적으로 해당과정에 관여하는 인자들 이어서, 고온 장시간에 걸친 에탄올발효를 하는 이 발효 효모균주의 세포에게는 생존과 에탄올발효를 위하여 해당과정 관여 인자가 중요한 역할을 하고 있다는 것을 강력히 시사하였다.

  • 천연계 비이온 계면활성제 APG(Alkyl Polyglucoslde)는 옥수수 성분의 글루코스와 팜, 야자유에서 얻어지는 지방 알코올을 이용하여 합성된다. 친수성인 글루코스의 중합도(D.P.=1.2~1.8)와 소수성인 alkyl chain 길이 (C8-C14)에 따른 계면특성(표면 계면장력, cmc, 기포력, effectiveness 등)을 조사한 바 중합도에는 영향이 적으나, alkyl chain 길이에는 큰 영향을 받았다. 지방산 오염에 대한 APG alkyl chain 길이별 세정력 평가를 한 결과 계면 장력이 낮을수록, 단위 면적당 흡착량이 많을수록 우수한 세정력을 나타내었으며, 그 순서는 APG 0814> APG 1214> APG 10> APG 0810> APG 08과 같다.

Identification and Antibacterial Activity of Volatile Flavor Components of Cordyceps Militaris

  • Flavor characteristics of raw Cordyceps militaris significatntly different from those of dried one. In the case of raw Cordyceps militaris , major flavor components were composed of 5 alcohols, 3 ketones, 4 phenols, 9 alkanes , and 3 alkenes. The major alcohol was 1-octen-3-ol(22.56%, 1147.3% ng/ml), which contributed to the characteristic green flavor. Ketones (3-ocatone, inparticular )were present in the highest concentration in raw Cordyceps militaris . In contrast, major flavor components of dried Cordyceps militaris were composed of 4 alcohols, 4 ketones, 3 furans, 4 pyrizines, 2 dithiazines, 5 phenols , 8alkenes , 17 alkanes, and 8 fatty acids. Dried Cordyceps militaris had unique sweet aroma of sesame as wella s a milky flavor. Green or fruit flavor were rarely detected . In alkanes , 10 cosanes, component fo wax were present. Typical flavor components of alkanes such as $\beta$-caryophyllen and Δ-cadinene were also detected. Fatty acids of dried Cordyceps militaris ranged from myristic acid (14 :0) to linoleic acid (18 ; 2). The sweet aroma of dried Cordyceps militaris was mostly due to pryazines, dithaiazines, and furans. Two dithaizines were identified and characteristics of these flavor components was a roasted bacon flavor. Strong antibacterial acitivity was observed toward Vibrio spp. such as V. vulnificus, V.cholerae, V. parahaemlyticus. Relatively high antibacterial acitivity was shown toward Bacillus subtilis , B,cereus, Staphyllococcus aureus, and Corynebacterium xerosis.

한국 성인에서 혈청 Alanine Aminotransferase 농도의 상승과 대사증후군과의 관계 (Relationship between Elevated Serum Alanine Aminotransferase Concentration and Metabolic Syndrome in Korean Adults)

  • Metabolic syndrome has been strongly associated with elevated alanine aminotransferase (ALT), a surrogate of nonalcoholic fatty liver disease. We investigated the relationship between metabolic syndrome and elevated ALT in the general Korean population. The study sample was comprised of 4,781 Korean adults who had participated in the 2005 Korean National Health and Nutrition Examination Survey. Metabolic syndrome was defined by National Cholesterol Education Program for Adult Treatment Panel III. Elevated ALT was defined as an enzyme activity > 40 IU/L for men, and > 31 IU/L for women. ALT was measured by enzymatic methods. Among participants, 425 (8.9%) subjects displayed elevated ALT. The odds ratios (ORs) for elevated ALT increased in subjects with obesity or one of components of metabolic syndrome such as abdominal obesity, high blood pressure, high fasting glucose, high triglyceride, and low HDL cholesterol after adjusting for age and sex. The unadjusted OR for elevated ALT increased according to the number of components of metabolic syndrome (OR = 1.5, 95% CI: 0.96-2.32 for 1 component; OR = 3.0, 95% CI: 1.98-4.61 for 2 components; OR = 6.3, 95% CI: 4.29-9.35 for ${\geq}3$ components; p for trend < 0.0001). This trend did not differ after adjustments for putative risk factors including age, sex, BMI, smoking status, and alcohol intake. Metabolic syndrome is implicated as a strong risk factor of elevated ALT in Korean adults.

Analysis of the Chemical Constituents of Agaricus brasiliensis

  • This study examined the chemical composition of A. blasiliensis and the chemical structural properties of an immuno-stimulating polysaccharide. The amino acids, free sugars, and organic acids by HPLC and fatty acids by GC were analyzed. The immuno-stimulating substance from A. blasiliensis was extracted with hot water and purified by ethanol precipitation. It underwent ion exchange chromatography on DEAE-cellulose and gel filtration on Toyopearl HW 65F. Through GP-HPLC, the substance was found to be homogeneous. Its chemical structure was determined by $^{13}C-NMR$. Fatty acids, organic acids, and sugar alcohol composition consisted exclusively of linoleic acid, fumaric acid and mannitol, respectively. The amino acids were mainly glutamic acid, glycine, and arginine. By $^{13}C-NMR$ analysis, the immuno-stimulating substance was identified as ${\beta}-(1{\rightarrow}3)\;(1{\rightarrow}6)$-glucan, composed of a backbone with $(1{\rightarrow}3)$-linked D-glucopyranosyl residues branching a $(1{\rightarrow}6)$-linked D-glucopyranosyl residue. The ${\beta}$-glucan from A. blasiliensis showed pronounced immuno-stimulating activity on the antibody-production ability of B-lymphocytes by the hemolytic suspension assay. In these results, A. blasiliensis was estimated to have potent pharmacological properties and potential nutritional values.

설문지에 의한 식이종류 및 식이행동의 비만과의 상관성에 관한 연구 -비만클리닉에 내원한 사무직 여성을 중심으로- (Diet, Eating Behavior and Their Associations with Obesityin Korean Office Ladies by Questionnaire)

  • Objectives : This study is to examine associations between diet and eating behavior with body mass index(BMI) in Korean office ladies. Methods: From OCT 2004 to OCT 2005, we collected obese office ladies who visited to the oriental obesity clinic and control group who were matched by age, occupation, working hours. Weight and height were self-reported. Subjects were divided to obese and non obese group by body mass index(BMI). Diet, eating behavior were determined from 21-item self-administered questionnaire. Independent paired t test was used for analyzing associations between diet, eating behavior and body mass index(BMI) Results: There were 141 participants in obese group and 50 participants in control group. Eating fast, skipping breakfast, night eating habit(p<.001), eating fatty food, drinking alcohol were highly positively associated with body mass index(BMI) values. There was no conclusive evidence that diet except fatty food were related to obesity. Conclusions: Eating behaviors are strongly related in obesity but associations between diet and obesity are not clear.

Antioxidant and Antibacterial Activity of Caprylic Acid Vanillyl Ester Produced by Lipase-Mediated Transesterification

  • Vanillyl alcohol (VA), which is abundant in Vanilla bean, has strong antioxidant activity. However, the use of VA in the food and cosmetics industries is limited, due to its low solubility in emulsion or organic solvents. Meanwhile, medium chain fatty acids and medium chain monoglycerides have antibacterial activity. We synthesized butyric acid vanillyl ester (BAVE) or caprylic acid vanillyl ester (CAVE) from VA with tributyrin or tricaprylin through transesterification reaction using immobilized lipases. BAVE and CAVE scavenged 2,2-diphenyl-1-picrylhydrazyl radicals in organic solvents. In addition, BAVE and CAVE decreased the production rate of conjugated diene and triene in the menhaden oil-in-water emulsion system. While BAVE showed no antibacterial activity, CAVE showed antibacterial activity against food spoilage bacteria, including Bacillus coagulans. In this study, the antibacterial activity of vanillyl ester with medium chain fatty acid was first revealed. Zeta potential measurements confirmed that BAVE and CAVE were inserted into B. coagulans membrane. In addition, the propidium iodide uptake assay and fluorescent microscopy showed that CAVE increased B. coagulans membrane permeability. Therefore, CAVE is expected to play an important role in the food and cosmetics industries as a bi-functional material with both antioxidant and antibacterial activities.

The hepatoprotective effects of silkworm: Insights into molecular mechanisms and implications

  • The liver, a multifunctional organ, plays a vital role in maintaining overall health and well-being by regulating metabolism, detoxification, nutrient storage, hormone balance, and immune function. Liver diseases, such as hepatitis, cirrhosis, fatty liver disease, and liver cancer, have significant clinical implications and remain a global health concern. This article reviews the therapeutic potential of silkworm larvae (Bombyx mori) and explores their underlying molecular mechanisms in protecting against liver diseases. Silkworm larvae are rich in proteins, vitamins, minerals, and n-3 fatty acids, making them a promising candidate for therapeutic applications. The anti-inflammatory mechanisms of silkworm larvae involve modulating the production of cytokine such as TNF-α and interleukins, inflammatory enzymes including cyclooxygenase-2 and macrophage polarization, thereby attenuating liver inflammation. Silkworm larvae also exhibit anti-oxidative effects by scavenging free radicals, reducing intracellular reactive oxygen species and enhancing the liver's antioxidant defense system. Moreover, silkworms have been reported to decrease the serum alcohol concentration and lipid accumulation. Understanding the therapeutic properties of silkworm larvae contributes to the development of innovative strategies for liver injury prevention and treatment. Further research is warranted to elucidate the precise signaling pathways involved in the anti-inflammatory and anti-oxidative effects of silkworm larvae, paving the way for potential therapeutic interventions in liver diseases.
