Effects of Citronellol and Thymol on Cell Membrane Composition of Candida albicans (Citronellol 및 Thymol이 Candida albicans 세포막 조성에 미치는 영향)

  • Phospholipids are crucially important in a cell membrane function and could thereby influence antibiotic susceptibility. In order to investigate the antifungal mechanism the total lipid was extracted from C. albicans treated with citronellol or thymol in concentration of their minimum inhibiting concentration and the changes in phospholipids composition were analyzed using ketoconazole as control. The cell growth and total lipid synthesis in cell walls of C. albicans were inhibited by treatment with citronellol. The levels of total lipids were decreased by 35.85% compared to the control. They also showed a significant decrease in the contents of phospholipid, phosphatidylcholine(PC), phosphatidyl ethanolamine(PE) and phosphatidylinositol(PI). As the result of GC assay for total fatty acid methyl esters of PC, PE and PI in C. albicans treated with citronellol, it was found that the major fatty acid composed of three phospholipid were palmitic acid, stearic acid and oleic acid. Moreover, the pattern of the fatty acid compositions of PC, PE and PI were changed by the oil. Based on the results, the anti-Candida mechanism of citronellol or thymol might be closely associated with disrupting the permeability barriers of the fungal cell wall composition or construction.

Evaluation of Microbial Community Composition in Cultivated and Uncultivated Upland Soils by Fatty Acids (지방산에 의한 경지 및 미경지 토양의 미생물군집평가)

  • We examined the relationships among community composition, microbial population, and microbial biomass to determine whether different land use leads to differences in microbial community composition. And also the relationships between soil characteristics and microbial community composition were investigated. There was no difference in pH between uncultivated and cultivated soils, but electrical conductivity, and contents of organic matter, available P and exchangeable cations were greater in the cultivated soil compared to the uncultivated soil. A linear correlation ($r^2=0.557$, n=18, p<0.01) was found between biomass-C estimated with fumigation extraction technique and total amount of fatty acids. An increase of fatty acid methyl esters (FAMEs) for bacteria, actinomycetes, fungi and protozoa was observed in cultivated soil.

  • 재생 가능한 자원인 동 식물성 기름으로부터 제조되는 수소용 연료 바이오디젤은 낮은 대기오염 배출과 $CO_2$ Neutral 특성으로 환경 친화적인 연료로 인정을 받으며 전 세계적으로 그 생산량이 급격히 증가하고 있다. 바이오디젤 생산 기술에는 직접이용법, 마이크로 에멀젼법, 열분해법, 에스테르화법, 초임계 메탄올 이용 생산법 등이 있으며 현재의 대부분의 상용 공정은 전이에스테르화법에 근거하고 있다. 이 공정은 크게 나누어 원료 유지 물질의 전처리 단계, 촉매를 사용하여 알콜과 에스테르화시키는 단계, 그리고 생성된 바이오디젤/글리세린의 분리와 정제 단계로 이루어지며 각 단계의 세부기술은 바이오디젤 생산비에 직접적인 영향을 미친다. 본 연구에서는 대두 원유의 전처리 반응, 전처리된 대두원유의 전이에스테르화 반응, 그리고 분리 및 정제 공정의 운전 변수들의 영향에 대한 연구결과와 본 연구를 통해 확립된 생산 공정으로 생산한 연료 grade의 바이오디젤 연료 물성 평가하였다.

Optimization of Biodiesel Production from Rapeseed Oil Using Response Surface Methodology (반응표면분석법을 이용한 유채유로부터 바이오디젤 생산의 최적화)

  • Biodiesel (fatty acid methyl esters) have used to as substitutes for petro-diesel by mixed-form with petro-diesel. In several processes of biodiesel production, alkali-catalyst transesterification produced to biodiesel of high contents with short reaction time. In this study, we investigate the optimal condition of alkali-catalyst transesterification of rapeseed oil produced at Jeju island in Korea using response surface methodology. The optimal condition of biodiesel production is reaction temperature 59.7$^{\circ}C$, catalyst amount 1.18%, oil to methanol molar ratio 1:8.75, and reaction time 5.18 min. At that reaction condition, the fatty acid methyl ester contents of product are above 97%. Our results may provide useful information with regard to the development of more economic and efficient biodiesel production system.

  • Biodiesel is a fuel produced in the form of a fatty acid methyl ester by using raw materials such as animal fat, vegetable oil and its by-products, and is being seen as a biofuel that can replace petroleum energy. However unsaturated fatty acid methyl esters in biodiesel causes to oxidize during storage and distribution, resulting in poor fuel quality and corrosion of vehicle engine components. In this study, the influence of quality and oxidation characteristics of biodiesel on the oxidation stability is investigated and the evaluation method related it is described. We also propose a method to improve the drawback of oxidation stability in biodiesel.

  • Fatty acid methyl esters, also referred to as biodiesel, have been determined to have a great deal of potential as substitutes for petro-diesel. In order to enhance productivity in the biodiesel production process, feedstocks were previously recommended to be anhydrous, with a free fatty acid content of less than 0.5%. In this study, the effects of several catalysts, methanol molar ratio, catalyst amount, and reaction time on the reduction of free fatty acid level were studied with a simulated feedstock consisting of 20% oleic acid in rapeseed oil. Ferric sulfate was selected as the best catalyst. Increasing the catalyst amount and methanol molar ratio is very effective in decreasing the acid value of the simulated mixture. Our results may provide useful information with regard to the development of more economic and efficient free fatty acid removal system.

  • A Comparison of the analysis of the green and roast coffee of Arabica and Robusta compositions and regular instant coffee has been investigated by chromatography. The coffee oil were obtained by extracting the green and roast coffee with ethyl ether by soxhlet methood. Instant coffee samples were accurately weighted into 100ml beaker (ca. 0.5g regular coffee and 1.5g decaffeinated coffee) and add ca. 50ml water, heat and boil, remove from heat, and mechanically stried ca. 15min. and filtered of one sample and another sample were without filtrated and proceed with liquid chromatographic separation. The fatty acid compositions of green and roast coffee were compared by gas liquid chromatography and general chemical compositions of sample were analysed. Some similarities between green and roast coffee fatty acids were found in the case of green and roast coffee of both kinds acid methyl esters. They contained stearic, oleic, linoleic, and unknown fatty acid, and palmitic ana linoleic acid were rich.

  • The production of biodiesel by transesterification of waste frying oil was conducted on various zeolite catalysts with different acidity and pore structure. $H^+$ ion exchanged MOR, MFI, FAU, and BEA zeolites were employed in the reaction with silicalite which has no strong acid sites. $H^+$ ion exchanged MOR(10) zeolite, which has more acid sites and stronger acid strength than other zeolites, exhibited the highest methyl esters yield as 95%. Dealumination to the HMOR zeolite induced decreasing of acid amount and acid strength. It brought about the decrease of fatty acid methyl esters (FAME) yield. The yield increased linearly with enhancing of acid strength and increasing of amount of strong acid sites. The yields were independent on pore structure of the zeolites.

  • Emulsified transesterification of soybean oil into biodiesel was investigated using potassium hydroxide and sodium methoxide catalysts with methyl glucoside oleic polyester as a methanol-in-oil emulsifier. The transesterification reaction conditions were optimized to obtain high yields of fatty acid methyl esters of the quality defined by biodiesel standards. The developed process resulted in $95{\sim}96%$ of overall yield from soybean oil by alkali-catalyzed methanolysis at $45^{\circ}C$ of reaction temperature with 6:1 of methanol-to-oil molar ratio and 1(v/v)% methyl glucoside oleic polyester in the presense of 0.8wt% KOH and 1.2wt% $NaOCH_{3}$.

  • The free fatty acids were prepared from the ethereal fraction of Panax ginseng. The prepared acids were methylated with diazomethzne. The methyl esters of saturated and unsaturated fatty acids were separated by the means of mercuric acetate method and column chromatography. The separated methyl esters were gaschromatographed and analyzed. The obtained conclusions were as follows. 1. The root of six-year old Korean Panax ginseng contains 0.28% of free fatty acids. 2. It was found that 24 kinds of free fatty acids existed in Panax ginseng. Among them, 22 kinds of free fatty acids were indentified by the gas chromatogram and the graphical method but the rest, 2 kinds of them were not identified by the only gas chromatographical data. The amount of each free fatty acid which was not identified was predominant and they were supposed to be unusual free fatty acids which would not commonly exist in nature. These results were shown in Table III. 3. $L_{EE}$ and $L_{EE}$ reported that n18:3 existed in Panax ginseng. However, in this experiment, n18:3 did not exist in Panax ginseng, and instead, peak XVI appeared between n18:2 and n18:3 as shown in Fig. 9.
