홍화유의 triglyceride 분자종에 관한 연구 (Molecular Species of Triglycerides in Safflower Oil)

  • Triglyceride 조성(組成)과 췌장리파제에 의한 PN별(別) 획분(劃分)의 ${\beta}$위치(位置)의 지방산조성(脂肪酸組成)의 결과로부터, 산정가능(算定可能)한 홍화유의 triglyceride 분자종은 23종류였다. 10% 이상을 차지하는 triglyceride분자종은 $C_{18:1}\;C_{18:2}\;C_{18:2}\;OLL;\;13.5%,\;C_{16:0}\;C_{18:2}\;C_{18:2}\;PLL;\;15.0%,\;C_{18:2}\;C_{18:2}\;C_{18:2}\;LLL;\;50.6%$로서 전체의 79.1%이었다.

Bacteriocin을 생산하는 Lactococcus sp. HY 449의 분리와 항균 특성 (Isolation of Bacteriocin-Producing Lactococcus sp.HY 449 and Its Antimicrobial Characteristics)

  • A bacteriocin-producing lactic acid bacteria was isolated from contaminated milk products, which was identified by using the API50 CH kit as Lactococcus lactis subsp. lactis with reliability of 98%. Fatty and analysia of the cell membrane showed that this strain contained same fatty acids profiles as type strain, Lactococcus lactis subsp. lactis ATCC 19435. The bacteriocin of Lactococcus sp. HY 449 showed relatively wide range of inhibition spectrum against gram positive and some gram negative bacteria such as Escherichia coli and maintained the inhibitory activity between pH2.0 and pH9.0 The thermostability of this bacteriocin was higher in acidic solution than in distilled water and was stable at 60$\circ $C for 1 hour.

인슐린 비의존성 당뇨환자의 탄수화물과 지질섭취비율이 혈당과 지질성상에 미치는 영향 (Serum glucose and lipid profiles according to dietary carbohydrate and lipid intake ratio in NIDDM patients)

  • This study aimed at the effect on serum glucose and lipid profiles according to carbohydrate / lipid intake ratio in Korean 191 NIDDM patients. Most of NIDDM patients belong to under 50-64 years. As carbohydrate intake level increased, lipid intake level decreased. The group of higher carbohydrate / lipid intake ratio shows significantly high in fasting glucose level and postprandial glucose in NIDDM patients. The higher carbohydrate / lipid intake ratio the higher dietary linoleic and $\omega$-3 fatty acid level but not in dietary $\omega$3/$\omega$6 ratio. Serum HDL decrease inhigher carbohydrate lipid intake ratio group. Serum total lipid and PUFA level decrease according to ditary total lipid intake decrease in men but not significantly different.

Milk Yield, Composition, and Fatty Acid Profile in Dairy Cows Fed a High-concentrate Diet Blended with Oil Mixtures Rich in Polyunsaturated Fatty Acids

  • To evaluate the effects of feeding linseed oil or/and sunflower oil mixed with fish oil on milk yield, milk composition and fatty acid (FA) profiles of dairy cows fed a high-concentrate diet, 24 crossbred primiparous lactating dairy cows in early lactation were assigned to a completely randomized design experiment. All cows were fed a high-concentrate basal diet and 0.38 kg dry matter (DM) molasses per day. Treatments were composed of a basal diet without oil supplement (Control), or diets of (DM basis) 3% linseed and fish oils (1:1, w/w, LSO-FO), or 3% sunflower and fish oils (1:1, w/w, SFO-FO), or 3% mixture (1:1:1, w/w) of linseed, sunflower, and fish oils (MIX-O). The animals fed SFO-FO had a 13.12% decrease in total dry matter intake compared with the control diet (p<0.05). No significant change was detected for milk yield; however, the animals fed the diet supplemented with SFO-FO showed a depressed milk fat yield and concentration by 35.42% and 27.20%, respectively, compared to those fed the control diet (p<0.05). Milk c9, t11-conjugated linoleic acid (CLA) proportion increased by 198.11% in the LSO-FO group relative to the control group (p<0.01). Milk C18:3n-3 (ALA) proportion was enhanced by 227.27% supplementing with LSO-FO relative to the control group (p<0.01). The proportions of milk docosahexaenoic acid (DHA) were significantly increased (p<0.01) in the cows fed LSO-FO (0.38%) and MIX-O (0.23%) compared to the control group (0.01%). Dietary inclusion of LSO-FO mainly increased milk c9, t11-CLA, ALA, DHA, and n-3 polyunsaturated fatty acids (PUFA), whereas feeding MIX-O improved preformed FA and unsaturated fatty acids (UFA). While the lowest n-6/n-3 ratio was found in the LSO-FO, the decreased atherogenecity index (AI) and thrombogenicity index (TI) seemed to be more extent in the MIX-O. Therefore, to maximize milk c9, t11-CLA, ALA, DHA, and n-3 PUFA and to minimize milk n-6/n-3 ratio, AI and TI, an ideal supplement would appear to be either LSO-FO or MIX-O.

13C NMR 분석 및 지방산 분석을 통한 어유의 종류 구분 및 사용 실태에 관한 연구 (A Study on Classification of Fish Oil Types and Its Usage by 13C-NMR Spectra and Fatty Acids Analysis)

  • This study estimates the classification criteria which distinguishes the types of omega-3 health functional foods, fish oils and fish oil usages through $^{13}C$-NMR spectra and fatty acids contents analysis. The major fatty acids of omega-3, eicosapentaenoic acid (EPA, $C_{20:5}$) and docosahexaenoic acid (DHA, $C_{22:6}$) are being analyzed. 10 ethyl ester (EE) forms and 10 triglyceride (TG) forms are the most common types of fish oils for 20 omega-3 products. Gas chromatography (GC) analysis generally shows the matching EPA and DHA contents of the products listed on the notation. But EE form contents of EPA and DHA are higher and are more varied than the TG form. Most of the samples of EPA/DHA ratio show different content ratios of indicated on the products when comparing with standards. The $^{13}C$-NMR analysis of EPA and DHA on sn-1,3 and sn-2 carbonyl peak position with fish oil triglycerides display whether the reconstituted triglycerides (rTG) are being confirmed or not. As a result of the 9 TG form, the 10 TG products showed similar values: EPA sn-1, 3; 13.46~15.66, sn-2; 3.00~4.52, DHA sn-1, 3; 2.43~4.40, sn-2; 3.84~6.36. But one product showed lower contents (EPA: sn-1, 3; 5.88, sn-2; 2.86, DHA sn-1, 3; 2.29, sn-2; 5.95) of EPA, thus it can be considered a different type of oil and only matched six products according to the label. This study is intended to provide basic materials which identify the status for the types and quality of omega-3 fish oil products according to fatty acids profiles and the $^{13}C$-NMR spectrum confirmed the location specificity of EPA and DHA.

과체중 및 비만인 소아 청소년의 인슐린 저항성, 혈액 특성 및 영양소 섭취량과의 관계 (The Relationship among Insulin Resistance, Blood Profiles and Nutrient Intake in Overweight or Obese Children and Adolescents)

  • The purposes of this study were to investigate blood profiles and nutrient intakes of groups that are different in obese levels, and to find the credible predictor of insulin resistance. The subjects were classified as normal weight (%IBW${\leq}$ 110), obese without MS and obese with MS according to IDF definition of the risk group in metabolic syndrome (MS). Subjects of this study were included 137 (59 boys, 78 girls) free living children and adolescents (mean age $12.6{\pm}3.4$ years) in Gangneung area, South Korea. %IBW of normal weight (94.9%), obese without MS (123.8%) and obese with MS (131.5%) were significantly different among groups. HOMA-IR had positive correlations with TG (r = 0.634), waist circumference (r = 0.553), atherogenic index (r = 0.513), %IBW (r = 0.453) and ALT (r = 0.360), but showed negative correlations with HDL cholesterol (r = -0.417, p < 0.001). HOMA-IR showed positive correlation with polyunsaturated fatty acid intake (p < 0.05). The energy intake of obese with MS was 1762 kcal/day which was not significantly different from those of normal weight and obese without MS. Total fatty acid intakes of two obese groups were significantly higher than that of normal weight. The results of this study suggest that waist circumference and ALT as well as TG, atherogenic index and weight can be credible indices to predict the insulin resistance in children and in adolescents. In addition, nutrition education and adequate diet should be provided to prevent MS in children and in adolescents.

The Effects of a High-fat or High-sucrose Diet on Serum Lipid Profiles, Hepatic Acyl-CoA Synthetase, Carnitine Palmitoyltransferase-I, and the Acetyl-CoA Carboxylase mRNA Levels in Rats

  • The purpose of this study was to investigate the effects of altering relative intakes of fat and carbohydrates on serum lipid profiles, hepatic acyl-CoA synthetase (ACS), carnitine palmitoyltransferase-I (CPT-I), and the acetyl-CoA carboxlyase (ACC) mRNA level in Sprague-Dawley rats. For four weeks the rats were fed either an AIN-76 diet or one of its modified diets that were supplemented with 20% beef tallow (high-fat diet, HF) and 66.3% sucrose (highsucrose diet, HS). The HS group had significantly higher serum triglyceride and total cholesterol concentrations when compared with the other groups. Serum LDL-cholesterol concentrations in the HS and HF groups were significantly higher when compared to the normal diet (ND) group. Serum HDL-cholesterol levels of the ND and HS groups were significantly higher than those of the HF group. The hepatic total lipid level of the HF group was significantly higher than those of other groups; triglyceride levels of the HS and HF groups were significantly higher than those of the ND group. Hepatic ACS mRNA levels of the HF group were significantly higher than those of the ND group. Hepatic CPT-I mRNA levels were higher in the HF group than other groups. Also, ACC mRNA levels in the liver increased in the HF group. In conclusion, changes in the composition of dietary fat and carbohydrates could affect the hepatic ACS, CPT-I, and ACC mRNA levels. These results facilitate our understanding of the coordinated regulation of the ACS, CPT-I, and ACC mRNA levels and will serve to enhance our understanding of the molecular mechanisms that underlie the regulation of fatty acid metabolism.

Nuclear magnetic resonance-based metabolomics analysis and characteristics of beef in different fattening periods

  • Beef quality is influenced by the fattening period. Therefore, meat metabolomics profiles from the different fattening periods (e.g., short-term vs. long-term) were analyzed for identify potential indicators using nuclear magnetic resonance. Additionally, blood, free fatty acid, sensory, and mineral compositions in Korean steers were determined. Blood, free fatty acid, and mineral concentrations showed significant differences between short-term and long-term groups that were fed different diets. However, there were no sensory differences in the two fattening groups. Additionally, the metabolic profiles of meats were clearly separated based on multivariate orthogonal partial least square discriminant analysis. Six metabolites of variable importance in the projection plot were identified and showed high sensitivity as candidate markers for meat characteristics. In particular, lactate, carnosine, and creatine could be directly linked to scientific indicators of the fattening stage (31 vs. 28 mo) of meat. Our findings suggest that the metabolomics approach could be a powerful method for the detection of novel signatures underlying the managing period of beef.

녹비 시용이 초기 논 토양 미생물군집에 미치는 영향 (Response of Soil Microbial Communities to Applications of Green Manures in Paddy at an Early Rice-Growing Stage)

  • 논 토양에서 청보리, 자운영과 트리티케일을 시용하고 벼생육초기에 토양 미생물 생태계의 변화에 미치는 영향을 검토하였다. 녹비를 시용한 처리구는 관행에 비해 토양 유기물 함량은 유의적으로 증가하였으나 토양의 나트륨 함량은 유의적인 감소를 나타냈다. 내생균근균의 함량과 구성비율은 녹비 시용으로 관행 보다 유의적으로 높아졌으며 특히 청보리 처리구가 가장 높았다. 또한 토양의 그람음성 세균과 그람양성 세균의 비율과 불포화지방산과 포화지방산 함량의 비율은 청보리 처리구가 다른 처리구에 비해 유의적인 증가를 보였다. 그리고 주성분 분석에서 PC 1에 가장 큰 영향을 미친 미생물 군집은 그람음성 세균인 것으로 나타났으며 청보리 처리구는 관행과 가장 큰 차이를 나타냈다. 이러한 결과를 종합한 결과 청보리는 논 토양의 초기 미생물 다양성을 증대시킬 수 있는 최적의 녹비작물로 판단된다.

Fungal Production of Single Cell Oil Using Untreated Copra Cake and Evaluation of Its Fuel Properties for Biodiesel

  • This study evaluated the microbial conversion of coconut oil waste, a major agro-residue in tropical countries, into single cell oil (SCO) feedstock for biodiesel production. Copra cake was used as a low-cost renewable substrate without any prior chemical or enzymatic pretreatment for submerged growth of an oleaginous tropical mangrove fungus, Aspergillus terreus IBB M1. The SCO extracted from fermented biomass was converted into fatty acid methyl esters (FAMEs) by transesterification and evaluated on the basis of fatty acid profiles and key fuel properties for biodiesel. The fungus produced a biomass (8.2 g/l) yielding 257 mg/g copra cake SCO with ~98% FAMEs. The FAMEs were mainly composed of saturated methyl esters (61.2%) of medium-chain fatty acids (C12-C18) with methyl oleate (C18:1; 16.57%) and methyl linoleate (C18:2; 19.97%) making up the unsaturated content. A higher content of both saturated FAMEs and methyl oleate along with the absence of polyunsaturated FAMEs with ≥4 double bonds is expected to impart good fuel quality. This was evident from the predicted and experimentally determined key fuel properties of FAMEs (density, kinematic viscosity, iodine value, acid number, cetane number), which were in accordance with the international (ASTM D6751, EN 14214) and national (IS 15607) biodiesel standards, suggesting their suitability as a biodiesel fuel. The low cost, renewable nature, and easy availability of copra cake, its conversion into SCO without any thermochemical pretreatment, and pelleted fungal growth facilitating easier downstream processing by simple filtration make this process cost effective and environmentally favorable.
