Objectives : This experimental study was designed to investigate the effect of SBY-III extract on the weight, cell size of epididymal fat-pad, fat accumulation area in liver, serum lipid level and UCP1 mRNA in brown adipose tissue of high fat diet-fed obese rats continued by high fat diet and regulated by normal Diet. Methods : The body weight gain, weight of the internal organs(epididymis, liver, brown adipose tissue), insulin, triglyceride, total cholesterol, total lopod, free fatty acid, expression of UCP1 mRNA were measured in high fat diet-fed obese rats continued by high fat diet and regulated by normal diet. The experimental study are divided into exp-I and exp-II. Each study was administered normal diet, high fat diet and SBY-III according to each situation. Normal group is normal diet for 8 weeks. Exp-I are divided into control group(high fat diet for 8 weeks) and sample group(high fat diet for 8 weeks and SBY-III for last 2 weeks). Exp-II are divided into control group(high fat diet for 6 weeks and normal diet for 2 weeks) and sample group(high fat diet for 6 weeks and normal diet with SBY-III for 2 weeks). These were then compared mutually. Results : 1. Irrespective of diet control, sample group taken SBY-III showed the more effective decrease of weight gain than control group and diet control-fed sample group with SBY-III showed the more effective decrease of weight loss including weight gain than control group. 2. Irrespective of diet control, sample group taken SBY-III showed the more effective decrease cell size of epididymal fat-pad, fat accumulation area in liver than control group. 3. Non diet control-fed sample group taken SBY-III showed the more effective decrease of serum triglyceride, total lipid, free fatty acid than control group and diet control-fed sample group taken SBY-III showed the decrease of serum triglyceride, free fatty acid than control group. 4. Only diet control-fed sample group taken SBY-III showed the decrease of UCP1 volume. Conclusions : These results shows that SBY-III has effects on anti-obesity, especially keeping pace with diet control.

We investigated the effects of gambigyeongsinhwan(GGH)(4) on body weight and non-alcoholic fatty liver disease(NAFLD) examined whether blood total cholesterol, LDL-cholesterol, free fatty acid and triglyceride levels and hepatic lipid accumulation are inhibited by it in high fat diet-fed obese male mice. 8 weeks old, high fat diet-fed obese male mice were divided into 5 groups: C57BL/6N normal, control, GGH(4)-1, GGH(4)-2 and GGH(4)-3. After mice were treated with GGH(4) for 8 weeks, we measured body weight gain, food intake, feeding efficiency ratio, fat weight, plasma ALT, leptin and lipid levels. We also did histological analysis for liver and fat on the mice. Compared with controls, GGH(4)-treated mice had lower body weight gain and adipose tissue weight, the magnitudes of which were prominent in GGH(4)-2. Compared with controls, GGH(4)-treated mice had lower feeding efficiency ratio and blood leptin level, the magnitudes of which was prominent in GGH(4)-2. Compared with controls, GGH(4)-treated mice had lower blood plasma total cholesterol, LDL-cholesterol, free fatty acid and triglyceride levels. Compared with controls, GGH(4)-2 treated mice had lower blood plasma ALT concentration. Consistent with their effects on body weight gain, the size of adipocytes were significantly decreased by GGH(4), whereas the adipocyte number per unit area was significantly increased, suggesting that GGH(4) decreased the number of large adipocytes. Hepatic lipid accumulation was decreased by GGH(4). In conclusion, these results suggest that GGH(4) exhibits anti-obesity effects through the modulation of feeding efficiency ratio and plasma obesity parameters. Moreover, it seems that GGH(4) also contributes to improve NAFLD through the regulation of plasma ALT and hepatic triglyceride accumulation.
Objectives : To identify the effect of Taeyeumjowee-tang on the treatment of obesity Methods : After having been fed a high-fat diet for 6 weeks, obese white rats were classified into three groups. Control group 1 was fed a high-fat diet for 2 more weeks. Control group 2 was fed a normal diet for 2 weeks. The Sample group was fed Taeyeumjowee-tang extract with a normal diet for 2 weeks. The result of the comparison among the three groups was as follows. Results : The Sample group decreased significantly as compared with the Control 1 and 2 groups in changes of the body weight, fat cell size, fat accumulation, insulin, triglyceride, total cholesterol, total lipid, free fatty acid and UCP1. Conclusions : Taeyeumjowee-tang has significant effect on the treatment of obesity resulting from a high-fat diet. Especially, it was a sure remedy for Control group 2 that was fed normal1y after having a high-fat diet.
Chitosan, hydroxycitrate and L-carnitine have been known to be antiobesity components. The purpose of this study was to evaluate the combined effects of chitosan, hydroxycitrate and L-carnitine mixture as a potential antiobesity supplement in overweight women. Pre-menopausal healthy females who were overweight (percent ideal body weight > 110) were included in this study. Forty-nine subjects randomly received a placebo (n = 25) or antiobesity-supplement (n = 24), which was a mixture of chitosan, hydroxycitrate, and L-carnitine. Before and after the eight-week experimental period, anthropometric parameters, blood components and computerized tomography were measured. At baseline, the two groups were well matched in terms of age, body mass index and lipid profile. After the eight weeks of potential antiobesity supplementation, the subjects' body fat percent had decreased significantly (p < 0.001) by 5.6% (39.1 $\pm$ 1 vs 36.9 $\pm$ 1%) while lean body mass increased (p < 0.01). Vsceral fat area at the L4 vertebra decreased significantly (p < 0.01) by 8.6% in the supplemented group and the total fat area at the L4 vertebra showed a tendency to decrease (p = 0.051) by 2.4%. Also, in the group given the antiobesity-supplement rather than the placebo, the fasting triglyceride level decreased significantly (p < 0.05) by 10.0%. In addition, serum total cholesterol levels in the antiobesity-supplement group showed a tendency to decrease (p=0.159) by 2.7% (194 $\pm$ 6 vs 189 $\pm$ 6 mg/dl). No side effects were found in either group during the intervention. In conclusion, the present study demonstrated that taking a mixture of chitosan, hydroxycitrate, and L-carnitine as a potential antiobesity supplement for eight weeks produced advantageous changes in the weight and visceral fat accumulation of overweight women without any side effects. This study was performed to investigate the effects of ethanolic extract of Ulmus davidiana root (UE) on lipid metabolism in mice fed a high-fat diet (HF) for 7 weeks. Forty male ICR mice were randomly divided into four groups; normal diet group (N), high-fat diet group (HF), HF with 0.5% UE (HF-L) and 1% UE (HF-H) group. Body weight, body weight gain, and liver weight in the HF group was significantly higher than in the N group, while those of the HF-L and HF-H group were unchanged. UE improved HF-induced dyslipidemia by reducing serum triglyceride, total cholesterol, and the atherogenic index. There was no difference in serum HDL-cholesterol among experimental groups. However, the HDL-cholesterol/total cholesterol ratio was significantly increased in the HF-L and HF-H group. Histological analysis showed that HF-fed mice developed hepatocellular microvesicular vacuolation as a result of fat accumulation. These changes were attenuated by 1% UE supplementation. In addition, hepatic triglyceride and cholesterol levels in the HF-H group significantly reduced. Taken together, these results demonstrated that lipid levels in the blood and liver were reduced by UE, suggesting that it might be beneficial for the prevention and treatment of hyperlipidemia and fatty liver.

Whole grain cereal (WGC)-rich diets provide macronutrients that are important for the regulation of energy metabolism. The current study evaluated whether WGCs had a preventive effect on sarcopenic obesity in high-fat diet (HFD)-induced obese mice. C57BL/6N mice were fed a normal diet (ND), ND+WGC, HFD, and HFD+WGC for 12 weeks. WGCs significantly reduced body weight gain, food efficiency ratio, fat mass, and adipocyte size in HFD-induced obese mice. WGCs attenuated HFD-induced nonalcoholic fatty liver disease by decreasing liver weight and hepatic fat accumulation. In addition, WGCs increased muscle strength and muscle mass in HFD-induced obese mice as well as in ND mice. Taken together, WGCs can be employed as functional food materials for the prevention of sarcopenic obesity by inhibiting fat accumulation and increasing muscle mass.
Objectives : Ojeok-san (OJS), an oriental herbal formula, has been used in Asian countries including Korea, China and Japan to treat the common cold and illnesses including fatigue and gastrointestinal disorders. The purpose of this study was to examine the anti-obesity effect and molecular mechanism of OJS, on adipogenesis in 3T3-L1 cells. Also, the effects of OJS in obese mice fed a high-fat diet on adiposity were examined.Methods : Preferentially, we analyzed the component of OJS and measured the stability of its component in OJS according to study periods using high performance liquid chromatography (HPLC). In vitro, 3T3-L1 cells were treated with OJS (50 to 200 μg/mL) during differentiation for 8 days. The accumulation of lipid droplets was determined by Oil Red O staining. The expressions of genes related to adipogenesis were measured by RT-PCR and Western blot analyses. For anti-obesty effect in vivo, we experimented for 8 weeks with four group (normal diet (CON), high-fat diet (HF), high-fat diet with OJS (HF+OJS) and high-fat diet with Bang-pung-tong-sung-san (HF+BTS) in comparison group HF+OJS).Results : OJS showed inhibitory activity on adipocyte differentiation at 3T3-L1 preadipocytes without affect cell toxicity as assessed by measuring fat accumulation and adipogenesis. In addition, OJS significantly reduced the expression levels of several adipocyte marker genes including proliferator activated receptor-γ(PPAR-γ) and CCAAT/enhancer-binding protein-α(C/EBP-α). Also OJS-administered mice showed significant inhibitory of body weights and abdominal adipose tissue weights.Conclusions : This study showed that traditional medicine OJS has an anti-obesity effect in vitro and in vivo. Thus, OJS could be developed as a supplement for reduction of body weight gain induced by an obesity.
The effects of feeding soybean oil (SBO) or rumen protected conjugated linoleic acid (RP-CLA) on CLA accumulation in milk, and performance of lactating dairy cows were studied. Twenty four Holstein Friesian crossbred lactating dairy cows, averaging $126{\pm}45days$ in milk, $15.6{\pm}2.43kg$ of milk and $452{\pm}51kg$ body weight were stratified randomly and assigned in a randomized complete block design (RCBD) to three treatments of 8 cows each. The treatments were control, 150 g of SBO and 150 g of RP-CLA supplementation. Performance parameters showed that DM intake, NELP intake and body weight change were similar across treatments, while CP intake was decreased by SBO and RP-CLA supplementation. Milk yield and milk composition were not significantly different among treatments, except for milk fat percentage and fat yield which were significantly decreased by 27% (p<0.05) and by 28% (p<0.01), respectively, by RP-CLA supplements compared with control treatment. Feeding RP-CLA reduced 3.5% FCM compared with the other treatments (p<0.003). Both SBO and RP-CLA supplementation reduced ${\geq}C18:0$ and CLA concentration in milk fat.
The anti-obesity potential of an ethanolic extract of the edible red alga Callophyllis japonica extract (CJE) was investigated in mice fed a high-fat diet (HFD). CJE administration into HFD mice revealed suppression of body weight, adipose tissue weight, serum total cholesterol, triglyceride, and glucose levels in a dose-dependent manner. Also, it reduced serum levels of glutamic pyruvic transaminase, glutamic oxaloacetic transaminase, and lactate dehydrogenase, as well as the accumulation of fatty droplets in liver tissue. CJE and its ethyl acetate fraction inhibited adipogenesis in 3T3-L1 adipocytes by down-regulating the adipocyte-specific transcriptional regulators. Taken together, these results suggest that CJE reduces obesity in mice fed an HFD by inhibiting lipid accumulation and adipogenesis in the adipose tissues.

Effect of pine pollen and defatted pine pollen on lipid accumulation and metabolism in serum and liver were investigated with rats fed high fat-supplemented diets for 6 weeks. Concentrations of total lipid and total cholesterol in serum significantly decreased in the experimental groups (treated with pine pollen and defatted pine pollen) compared with the control group. Concentration of triglyceride in serum was remarkably lower than that of control group. This was similar to that of normal group. Concentration of glucose in serum was significantly lower in the experimental group. It was observed from photographs of hepatic tissue in rats that pine pollen inhibits the lipid accumulation induced by high fat diets.
