지방분화가 유도된 3T3-L1 세포와 고지방식이로 유도된 마우스에서 보리순 물추출물의 항비만 효과 (Anti-obesity Effects of Barley Sprout Young Leaf on 3T3-L1 Cells and High-fat Diet-induced Obese Mice)

  • Background: An imbalance in energy intake and expenditure can cause obesity, which is a major risk factor for chronic diseases such as heart disease, type 2 diabetes, insulin resistance, cancers and hyperlipidemia. Methods and Results: In this study, we evaluated the anti-obesity effects of a water extract from the young leaves of barley sprout (BS) in 3T3-L1 cells and in high-fat diet (HFD)-induced obese mice (HF). Lipid accumulation measurement indicates that BS markedly inhibited adipogenesis by reducing lipid droplet production in a dose-dependent manner. Furthermore, the mRNA expression of adipogenic transcription factors peroxisome proliferator-activated receptor-${\gamma}$ and fatty acid synthetase, CCAAT/enhancer binding protein-${\alpha}$ and fatty acid binding protein 4 in 3T3-L1 cells was significantly inhibited by BS treatment. In an in vivo test, the BS-administered group of HFD-induced mice showed less body weight gain, and lower liver and epididymal white adipose tissue weights. The BS-treated mice showed decreased serum levels of leptin and lipids compared to untreated HFD mice and the levels of adiponectin and the HDL-cholesterol/total cholesterol ratio increased. These results indicate that BS inhibits body fat accumulation by reducing the mRNA expression of lipogenesis transcription factors and increasing serum adipokine concentration in in vitro and in vivo tests. Conclusions: BS reduced high fat diet-induced weight gain and had a positive effect on dyslipidemia.

High Density Lipoprotein from Egg Yolk (EYHDL) Improves Dyslipidemia by Mediating Fatty Acids Metabolism in High Fat Diet-induced Obese Mice

  • We investigated the effect of high density lipoprotein from egg yolk (EYHDL) on serum, hepatic and fecal lipid and fatty acids (FAs) levels and on gene expression involved in FAs metabolism. Male KM mice were fed either normal diet (ND; n=20), high fat diet (HFD; n=20), or high fat diet containing EYHDL (EYHDL; 0.6 mg/g, every day by oral gavage, n=20) for 100 days. At the end of the experiment, the effects of treatments on biochemical parameters, FAs profiles and involved gene expression were analyzed. Our results revealed that EYHDL markedly suppressed the body weight gain, accumulation of abdominal fat tissues, serum concentrations of LDL-cholesterol (LDL-C) and triglycerides, hepatic triglycerides and cholesterol accumulation, while increased serum concentration of HDL-cholesterol (HDL-C). EYHDL intake also increased total cholesterol (TC) excretions compared with HFD group. Moreover, it alleviated the severity of fatty liver and improved glucose and insulin tolerance compared with HFD. More importantly, EYHDL partially normalized FAs profiles in serum, liver and fecaces and neutralized the HFD-induced upregulation of SREBP-1c, Acaca, Fasn, GPAT and Scd1. In conclusion, our findings indicate that EYHDL may have the potential to improve metabolic disturbances that occur in HFD mice and can be considered as an appropriate dietary recommendation for the treatment of metabolic syndrome (MetS).

  • The present study investigated the anti-obesity effects of Mycoleptodonoides aitchisonii (MA) in mice fed a high-fat (HF) diet. Two groups were fed either a normal control diet or an HF (45% kcal fat) diet for 12 weeks and three groups were fed an HF diet supplemented with powdered MA (MAP, 1%, 3%, and 5%) for 12 weeks. The anti-obesity effects of MAP supplementation on body weight, fat mass development, and lipid-related markers were assessed. Consumption of an HF diet resulted in increased body weight, serum lipids, relative adipose tissues weight, and liver fat accumulation. However, administration of MAP significantly decreased body weight gain, food intake, food efficiency ratio, hepatic cholesterol level, and adipose tissue weight in a dose-dependent manner. In addition, treatment with MAP significantly reduced the occurrence of fatty liver deposits and steatosis, and inhibited an HF diet-induced increase in adipocyte size. These results suggest that dietary supplementation with MAP exerts anti-obesity effects and indicate that MAP could be used as a functional food to control obesity.

Extracts of Codonopsis lanceolata Decrease $CCl_4$-Induced Hepatotoxicity on High Fat Diet in Rats

  • This study was designed to investigate the effect of Codonopsis lanceolata extracts on carbon tetrachloride (CCl₄)­induced hepatotoxicity in rats fed high, medium or low fat diet. The liver weight was increased not by the level of fat in diet but by administration of CCl₄. Activities of serum aspartate aminotransferase (AST) and alkaline phosphatase (ALP) were significantly increased by CCl₄, the activities of AST and alanine aminotransferase (ALT) were significantly decreased in case of CCl₄ treatment with C. lanceolata extracts. In high fat diet with CCl₄ administration, the content of total lipid in liver was significantly increased, which was significantly decreased by the administration of C. lanceolata extracts. Administration of C. lanceolata water extracts remarkably prevented the tissue injury such as lipid degeneration, infiltration of inflanunatory cells and necrosis caused by CCl₄, but those methanol extracts did not affect histopathological changes of the liver as water extract. The data suggest that intake of C. lanceolata extracts would prevent lipid accumulation and hepatotoxicity induced by high fat diet and CCl₄ abministratim, and the water extract of C. lanceolata is supposed to be more effective than those methanol extract.

고지방식이 유도성 지방간 쥐 모델에서 간의 자기공명분광 분석을 이용한 지질 양성자 조성 변화 연구 (The Study of Lipid Proton Composition Change in a Rat Model of High Fat Diet Induced Fatty Liver by Magnetic Resonance Spectroscopy Analysis)

  • The purpose of this study is to investigate the changes in lipid proton (LP) composition according to the induced obese fatty liver and to use it as basic data for treatment and diagnosis of fatty liver in the future. The phantom study was conducted to identify differences between STEAM and PRESS Pulse sequences in LP concentration. A high-fat diet (60%) was administered to eight Sprague-Dawley rats to induce obesity and fatty liver disease. Baseline magnetic resonance imaging /spectroscopy data were obtained prior to the introduction of high-fat diet, and data acquisition experiments were performed after eight weeks using procedures identical to those used for baseline studies. The six lipid proton metabolites were calculated using LCModel software. The correlation between the fat percentage and each LP, revealed that the methylene protons at 1.3 ppm showed the highest positive correlation. The α-methylene protons to carboxyl and diallylic protons showed negative correlation with fat percentage. The methylene proton showed the highest increase in the LP; however, it constituted only 71.86% of the total LP concentration. The methylene proton plays a leading role in fat accumulation in liver parenchyma.

  • 누에(Bombyx mori)번데기는 창상치료용 드레싱, 간보호활성, 항유전독성, 포도당농도 조절, 항암활성 등에 다양하게 적용되고 있다. 본 연구에서는 붉은동충하초(Cordyceps militaris)를 이용하여 발효시킨 누에번데기분말의 에탄올 추출물(Ethanol extract of silkworm pupae fermented with Cordyceps militaris, ESfC)의 항비만효능을 분석하기 위하여, SD랫드로부터 분리된 일차지방세포(Primary adipocytes)에서 농도별로 ESfC를 처리하여 free glycerol release, cAMP농도를 측정하였고, 고지방사료(High fat diet, HFD)가 처리된 C57BL/6N 비만마우스에 ESfC를 투여하여 지방조직과 간조직에서 지방축적(Fat accumulation)을 관찰하였다. 먼저, 지방감소효과를 나타내는 지표물질로서 cordycepin의 존재는 HPLC분석을 통하여 ESfC에서 확인하였다. 또한, free glycerol수준과 cAMP농도는 고농도 ESfC가 처리된 일차지방 세포에서 유의적으로 증가하였지만 이들 수준은 저농도와 중농도의 ESfC로처리세포에서는 유의적인 변화가 없었다. HFD-유도 비만모델에서, 고지방사료의 투여에 의해 유의적으로 증가된 지방무게와 지방세포의 크기는 ESfC처리그룹에서 감소되었다. 간 무게와 지방방울(Lipid droplet)의 수는 고지방사료의 투여에 의해 증가하였지만 ESfC투여에 의해 유의적으로 감소하였다. 또한, 유사한 회복효과는 perilipin과 HSL의 인산화 수준뿐만 아니라 ATGL의 발현수준에서도 관찰되었다. 따라서 이상의 결과는 ESfC가 지방일차세포에서 지방분해를 촉진하고, HFS유도 비만모델에서 지방 축적을 억제하는 효과를 나타냄을 보여주고 있으며, 향후 비만환자에 처리하기 위한 약물로서 사용가능성을 제시하고 있다.

청심연자탕(淸心蓮子湯)이 비만 유도 생쥐의 체중 및 혈청 지질에 미치는 영향 (Effects of Cheongshimyeonja-tang on Body Weight and Serum Lipid Levels in High Fat Diet Induced Obese Mice)

  • Objectives : This study was performed to investigate the inhibitory effects of Cheongshimyeonja-tang water extracts(CSYJ) on high fat diet-induced obesity and hyperlipidemia in C57BL/6J mice. Methods : The experimental animals were divided into four groups; normal diet-fed control(ND), high fat diet-fed control(HFD), HFD+CSYJ 150 mg/kg(CSYJ 150), and HFD+CSYJ 300 mg/kg(CSYJ 300). Obesity with hyperlipidemia was induced by feeding high fat diet(40%), and CSYJ was administrated orally into mice every day for 5 weeks. The effect of CSYJ on the serological parameters for Obesity with hyperlipidemia was evaluated. Results : CSYJ-treated groups revealed significantly reduced body weight and feed intake, as well as feed efficiency ratio, compared to HFD-fed group in dose-dependent manner. CSYJ reduced significantly the serum levels of triglyceride, total cholesterol and LDL-cholesterol elevated by intake of high fat diet feed, while the increased serum levels of HDL-cholesterol attenuated levels of atherogenic index and cardiac risk factor. It also reduced the blood levels of insulin and leptin in HFD group, and inhibited lipid accumulation in organs such as liver and abdomal adipose tissue. Moreover oral administration of CSYJ decreased significantly the blood level of alanine aminotransferase(ALT), aspartate aminotransferase(AST) and lactate dehydrogenase(LDH), and lipid peroxide(LPO), compared to HFD-fed group in dose-dependent manner. Conclusions : These results indicate that CSYJ could reduce high fat diet-induced obesity and hyperlipidemia, suggesting its clinical usefulness for declining body fat and hyperlipidemia.

Effects of Low Calorie Diet and Platycodon Grandiflorum Extract on Fatty Acid Binding Protein Expression in Rats with Diet-induced Obesity

  • Obesity can be defined as a metabolic disease due to an increased fat accumulation in the body caused by an imbalance of calorie intake and output The prevalence of obesity has increased substantially over the past 2-3 decades in developed and developing countries. The health impact of weight gain is so marked that obesity has now been classified as a major global public health problem In order to investigate the effect of diet conversion and oral administration of Platycodon grandiflorum extracts on the treatment of obesity, male Spraque-Dawley rats were divided into four groups: a group converted to normal diet (Control group), a group maintained high fat (30%) diet (H), and two groups with Platycodon grandiflorum extract added to the previously mentioned two groups. All animals were fed high fat diet for 7 weeks to induce the obesity. Then they were divided as mentioned above. Animals were fed experimental diet and Platycodon grandiflorum extract (150 mg/ml/rat/day) for 7 weeks. Body weight, adipose tissue weight (subcutaneous, epididymal, peritoneal fat pads) and serum lipids (total cholesterol and triglyceride) showed some differences among groups. The Platycodon grandiflorum feeding markedly decreased both body weight and adipose tissue weight in control group compared to H, high fat diet maintaining, group. Platycodon grandiflorum extracts significantly decreased the concentrations of serum lipids compared to H group. Fat cell numbers and sizes were significantly reduced in the oriental medicinal herb extract administrated group. Increased fatty acid binding protein (FABP) expression in high fat diet group was decreased by the dietary conversion to normal diet and the oral administration of Platycodon glandiflorum extracts. In contrast, there was no significant effect on FABP expression in the high fat maintenance group. In this study, the conversion from high fat diet to low fat or normal diet had a beneficial effect on body weight loss and serum lipid profiles. Dietary Platycodon glandiflorum extracts had an additive beneficial effect on the prevention and treatment of obesity.

자색고구마로부터 분리한 안토시아닌 분획물의 고지방식이로 유도된 인슐린 저항성과 간 지질 축적 개선 효과 (Inhibitory Effects of Anthocyanin-rich Fraction from Purple Sweet Potato on High Fat Diet-induced Insulin Resistance and Hepatic Steatosis)

  • Anthocyanins, a class of flavonoids, are natural water-soluble pigments, mainly found in vegetables and fruits. Anthocyanins have attractive pharmacological activities, such as anti-oxidant, anti-inflammatory, anti-cancer, and anti-diabetic effects. The purpose of this study was to investigate the inhibitory effects of the anthocyanin-rich fraction (ANF) from purple sweet potato on high fat diet-induced insulin resistance and hepatic steatosis. C57BL/6J mice were assigned to the following groups (n=8 per group): normal fat diet (NF); high fat diet (HF); high fat diet with ANF 50mg/kg (ANF50). Normal fat or high fat diets were fed for a total of 17 weeks, and ANF was orally administrated for 8 weeks (from 10 to 17 weeks, five times/week). In our results, there were no significant differences in body weight, food intake, and tissue weight upon ANF supplementation. The levels of serum triacylglycerol, total-cholesterol, and glucose were also not affected by ANF supplementation. However, ANF supplementation significantly decreased serum insulin and HOMA-IR levels as well as prevented hepatic fat accumulation in high fat-fed mice. These results show that ANF may be beneficial for improving high fat-induced insulin resistance and protecting against development of hepatic steatosis.
