Objectives : The present study was designed to investigate the anti-diabetic effects of Mori Folium (Morus alba L. of Moraceae) extract (MFE) on high fat diet (HFD) and streptozotocin (STZ)-induced type II diabetes mellitus in mice. Methods : The mice (C57BL/6J) were fed HFD for 8 weeks and then was induced with a single injection of STZ (75 mg/kg). The diabetic mice were divided into four groups [(STD, HFD, HFD + MFE and HFD + quercetin (QUR)] and administered with MFE or OUR for 4 weeks. Fasting blood glucose, lipid profile (triglycerides and cholesterol etc.), glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), insulin and leptin were measured every 2 weeks. Results : Body weight gain was lower in the MFE and QUR groups than HFD group. The fasting blood glucose was lower in the MFE and QUR groups. Oral glucose and insulin tolerance were decreased in the MFE and QUR groups. The levels of serum total cholesterol, triglycerides, and LDL cholesterol were reduced in the MFE and QUR groups. The HDL cholesterol was much higher in the MFE and QUR groups than HFD group. The levels of GOT, GPT and atherogenic index were decreased in the MFE and QUR groups. The serum insulin and leptin concentrations were reduced in the MFE and QUR groups. Conclusions : These results showed that MFE could decrease blood glucose level and lead to an amelioration in dyslipidemia states on HFD/STZ-induced type II diabetes mellitus in mice.

BACKGROUND/OBJECTIVES: Jerusalem artichoke has inhibitory activity against ${\alpha}$-glucosidase and decreases fasting serum glucose levels, which may be related to its fructan content. The biological activity of fructan can be influenced by the degree of polymerization. Thus, in this study, the inhibitory effects of original and fermented purple Jerusalem artichoke (PJA) on ${\alpha}$-glucosidase were compared in vitro. Additionally, the anti-diabetes effect of Lactobacillus plantarum-fermented PJA (LJA) was studied in a non-insulin-dependent diabetes mellitus animal model (C57BIKsJ db/db). MATERIALS/METHODS: The water extract of PJA was fermented by L. plantarum, and two strains of Bacillus subtilis to compare their anti-${\alpha}$-glucosidase activities in vitro by ${\alpha}$-glucosidase assays. The anti-diabetes effect of LJA was studied in a non-insulin-dependent diabetes mellitus animal model (C57BIKsJ db/db) for seven weeks. During the experiment, food intake, body weight, and fasting blood glucose were measured every week. At the end of the treatment period, several diabetic parameters and the intestinal ${\alpha}$-glucosidase activity were measured. RESULTS: The LJA showed the highest ${\alpha}$-glucosidase inhibitory activity in vitro. In the in vivo study, it resulted in a significantly lower blood glucose concentration than the control. Serum insulin and HDL cholesterol levels were significantly higher and the concentrations of triglycerides, non-esterified fatty acids, and total cholesterol were significant lower in mice treated with LJA after seven weeks. In addition, the intestinal ${\alpha}$-glucosidase activity was partially inhibited. CONCLUSIONS: These results suggested that LJA regulates blood glucose and has potential use as a dietary supplement.

Purpose: This study was done to investigate the effect of Tai Chi Qigong exercise program on insulin resistance and blood glucose in patients with type 2 diabetes. Method: This was a quasi-experimental study used a non-equivalent control group pretest-posttest design. Participants included 39 patients with type 2 diabetes mellitus patients (ages between 40-70)(experimental group, 19, control group, 20). The Tai Chi Qigong exercise program consisted of 5 minute warming up, 30-minute main exercise, and 5 minute cooling down, and was conducted twice a week for 10 weeks. The outcome variables were insulin resistance (proinsulin, C-peptide, insulin, insulin sensitivity index) and blood glucose (fasting, post prandial 2hr, HbA1c). Data was analyzed using ANCOVA, with pre-test values as covariates, to examine difference between pre and post measures between the two groups. Results: The Tai Chi Qigong was effective in lowering postprandial 2hr blood glucose. Conclusion: Twice a week 10 week-Tai Chi Qigong exercise seems to be too short to improve insulin excretion or resistance, and fasting blood sugar and HbA1C in patients with type 2 DM. A Tai Chi Qigong program at least 3 times per week or longer is recommended.
Nuclear receptor coactivator 6 (NCOA6) is a transcriptional coactivator of nuclear receptors and other transcription factors. A general Ncoa6 knockout mouse was previously shown to be embryonic lethal, but we here generated liver-specific Ncoa6 knockout (Ncoa6 LKO) mice to investigate the metabolic function of NCOA6 in the liver. These Ncoa6 LKO mice exhibited similar blood glucose and insulin levels to wild type but showed improvements in glucose tolerance, insulin sensitivity, and pyruvate tolerance. The decrease in glucose production from pyruvate in these LKO mice was consistent with the abrogation of the fasting-stimulated induction of gluconeogenic genes, phosphoenolpyruvate carboxykinase 1 (Pck1) and glucose-6-phosphatase (G6pc). The forskolin-stimulated inductions of Pck1 and G6pc were also dramatically reduced in primary hepatocytes isolated from Ncoa6 LKO mice, whereas the expression levels of other gluconeogenic gene regulators, including cAMP response element binding protein (Creb), forkhead box protein O1 and peroxisome proliferator-activated receptor γ coactivator 1α, were unaltered in the LKO mouse livers. CREB phosphorylation via fasting or forskolin stimulation was normal in the livers and primary hepatocytes of the LKO mice. Notably, it was observed that CREB interacts with NCOA6. The transcriptional activity of CREB was found to be enhanced by NCOA6 in the context of Pck1 and G6pc promoters. NCOA6-dependent augmentation was abolished in cAMP response element (CRE) mutant promoters of the Pck1 and G6pc genes. Our present results suggest that NCOA6 regulates hepatic gluconeogenesis by modulating glucagon/cAMP-dependent gluconeogenic gene transcription through an interaction with CREB.

To test their roles in rumination behavior, metabolites (acetic acid, propionic acid, butyric acid and glucose) were intravenously infused into the jugular vein of goats during fasting. The heads of four female goats tested were locked in a stanchion of cages in an experimental room. Ruminating behaviors with regard to number of ruminations, ruminating time, number of remastications and remasticating time were significantly decreased by acetic acid infusion (p < 0.05), and tended to be depressed more on values in butyric acid infusion or glucose infusion than those in pre-infusions, but propionic acid infusion decreased a little. Those data suggest that rumination receptors sensitive to VFAs and glucose are more likely to be situated in the area, where they would respond too blood levels.

The expression of SGLT-1 mRNA has been reported in the small intestine of mammals and the rectum of chickens. However, the expression and functional significance of SGLT-1 in bovine rectum is not known. In this study, we studied the effects of fasting and grazing on SGLT-1 gene expression in biopsy epithelial tissue of bovine rectum. In Japanese Black beef cattle, i) SGLT-1 gene expression was measured by quantitative real-time PCR in the biopsy rectal epithelia samples obtained through an endoscope, ii) SGLT-1 gene expression in the rectal epithelial tissues increased at 48 and 72 h after fasting correlating with a decrease in body weight. iii) SGLT-1 gene expression decreased after one month from the start of grazing (May to June) and then stabilized until the end of the grazing period (June to October) in the rectal epithelial tissues of grazing cattle. In conclusion, it is clear that SGLT-1 gene expression in the rectal epithelial tissue is increased by a restricted dietary condition.

The purpose of this study was to assess the effects of mulberry leaf extract supplement on blood glucose, glycated hemoglobin ($HbA_{1C}$) and serum lipids in type II diabetic patients, and also to assess safety in liver function after mulberry leaf extract supplement. The study was a randomized placebo-controlled trial and total 23 type II diabetic patients were divided into a MLE group taking 1,000 mg mulberry leaf extract supplement per day as experimental group and a placebo group taking 1,000 mg cellulose Powder supplement per day for 12 weeks. After 2 weeks of wash-out period, fasting blood glucose, $HbA_{1C}$, serum lipid levels and liver function test were analyzed before and after treatment of 12 weeks. The general baseline characteristics, nutrient intake and life style factors of study subjects were similar between two groups during intervention. The concentrations of fasting blood glucose and $HbA_{1C}$ (p<0.05) decreased significantly after mulberry leaf extract supplement in MLE group, while there were no changes found in placebo group. We also found it showed that mulberry leaf extract supplement for 12 weeks decreased significantly (p<0.05) the fasting blood glucose in poor fasting blood glucose group and $HbA_{1C}$ concentration in poor $HbA_{1C}$ group. The concentrations of LDL-cholesterol (p<0.05) and triglyceride (p<0.01) decreased significantly in MLE group after 12 weeks of taking the supplement, while there were no changes found in placebo group. The mulberry leaf extract supplement for 12 weeks didn't show hepatotoxicity. These results suggested that mulberry leaf extract supplement could be effective in improving fasting blood glucose and $HbA_{1C}$ levels in the diabetic patients, specially having high concentrations of fasting blood glucose and $HbA_{1C}$ among type II diabetic patients.

This study investigated the effect of Corni Fructus (Cornus officinalis Sieb. et Zucc.) extract on blood glucose and insulin resistance in db/db mice. Seven weeks old male mice were divided into normal control group (NC), diabetic control group (DC) and Corni Fructus treated diabetic group (DCF). Over an 8-week experimental period, Corni Fructus extract was administered orally at 500 mg/kg BW/day. Corni Fructus inhibited increase in blood glucose level during the OGTT (oral glucose tolerance test). At 8 weeks after beginning of the experiment, blood glucose level in the DCF group was significantly lower (p<0.01) than the DC group. Final fasting serum glucose and triglyceride in the DCF group were significantly lower (p<0.05) than the DC group by 32% and 41% respectively. Serum insulin did not differ among the NC, DC and DCF groups. The mRNA expression of adiponectin, GLUT 4 and PPAR-$\gamma$ in adipose tissue in the DC group were significantly lower than the NC group and they were higher in the DCF group than the DC group by 76%, 130% (p<0.05) and 43%, respectively. In conclusion, these results indicated that Corni Fructus would have antidiabetic effects via improving insulin resistance in favor of higher glucose utilization and reducing blood glucose level in db/db mice.
Objectives: This study investigated the effects of water extract of Aconitum carmichaeli Debx (ACD) on obesity and glucose tolerance in high fat diet induced obese mice. Methods: Five-week-old C5BL/6 mice were fed a high fat diet (HFD) containing or not containing ACD (100 or 300 mg/kg) for 16 weeks. Body weight, food intake, fasting blood glucose, and body temperature were checked every week and then organs, blood serums were collected after 16-week treatment. Furthermore oral glucose tolerance test (OGTT) was carried out after treatment. Results: ACD treated mice showed no significant decreases in body weight and adipose tissue weight as compared with HFD mice. Lipid accumulations in liver and serum lipid levels were not different between ACD treated and HFD mice. However, ACD extract administration significantly and dose-dependently reduced fasted plasma glucose and glucose tolerance as determined by OGTT. Conclusions: The present study demonstrates that ACD might prevent diet-induced glucose tolerance in mouse models of obesity.
Purpose: The purpose of this study was to investigate the computer input frequency of blood glucose self testing in type2 diabetic patients, for three months. Method: 39 participants were recruited from the endocrinology outpatient department of a tertiary care hospital in an urban city. The computer input frequencies were measured by patients' log in and input of http://www.biodang.com. Glycosylated haemoglobin was determined by a high performance liquid chromatography technique and fasting blood glucose was analyzed by the glucose oxidase method. Diabetes knowledge was measured by a 19 item diabetes knowledge test. Results: The computer input frequency of before breakfast blood glucose was 33.5 for three months. The total blood glucose input frequency tended to be lower in female, aged patients, middle school graduates, those who have no spouse and job, those who have no insulin treatment, obese subjects, and hyperglycemia patients, than in their counterparts. The diabetic knowledge was positively correlated with the computer input frequency of blood glucose self testing. Conclusion: An educational program should be developed to increase the computer input frequency of blood glucose self testing in type2 diabetic patients.
