Differential Coupling of G$\alpha$q Family of G-protein to Muscarinic $M_1$ Receptor and Neurokinin-2-Receptor

  • The ligand binding signals to a wide variety of seven transmembrane cell surface receptors are transduced into intracellular signals through heterotrimeric G-proteins. Recently, there have been reports which show diverse coupling patterns of ligand-activated receptors to the members of Gq family $\alpha$ subunits. In order to shed some light on these complex signal processing networks, interactions between G$\alpha$q family of G protein and neurokinin-2 receptor as well as muscarinic M$_{1}$ receptor, which are considered to be new thearpeutic targets in asthma, were studied. Using washed membranes from Cos-7 cells co-transfected with different G.alpha.q and receptor cDNAs, the receptors were stimulated with various concentrations of carbachol and neurokinin A and the agonist-dependent release of [$^3H$]inositol phosphates through phospholipase C beta-1 activation was measured. Differential coupling of Gaq family of G-protein to muscarinic M$_{1}$ receptor and neurokinin-2 receptor was observed. The neurokinin-2 receptor shows a ligand-mediated response in membranes co-transfected with G$\alpha$q, G$\alpha$11 and G$\alpha$14 but not G$\alpha$16 and the ability of the muscarinic $M_1$ receptor to activate phospholipase C through G$\alpha$/11 but not G$\alpha$14 and G$\alpha$16 was demonstrated. Clearly G$\alpha$/11 can couple $\M_1$ and neurokinin-2 receptor to activate phospholipase C. But, there are differences in the relative coupling of the G$\alpha$14 and G$\alpha$16 subunits to these receptors.

  • Heterotrimeric G Proteins transduce ligand binding to a wide variety of seven transmembrane cell surface receptors into intracellular signals. The currently accepted model for the activation of G protein suggests that ligand-activated receptor accelerates GDP-GTP exchange reactions on the ${\alpha}$ subunit of the heterotrimeric G protein. At least seventeen distinct isoforms of the G${\alpha}$ subunit protein have been identified in mammalian organisms. Among them, the G${\alpha}$q family consists of five members whose ${\alpha}$ subunits show different expression patterns. G${\alpha}$q and G${\alpha}$11 seem to be almost ubiquitously expressed, whereas G${\alpha}$14 is predominantly expressed in spleen, lung, kidney and testis. G${\alpha}$16 and its murine counterpart G${\alpha}$15 are expressed in hematopoietic cells and has been shown to couple a wide variety of receptors to phosphoinositide-specific phospholipase C activity. Beta-isoforms of phospholipase C were shown to be activated by all members of G${\alpha}$q family, i.e., G${\alpha}$q, G${\alpha}$11, G${\alpha}$l4 and G${\alpha}$16 subunits either in reconstitution system. or in experiments using cDNA transfection with intact Cos-7 cells.

  • The xynA gene encoding an alikali-tolerant endo-1,4-${\beta}$-xylanase (XYN) was cloned from the alkalophilic Bacillus pumilus A-30. The nucleotide sequence of a 974-bp DNA fragment containing the xynA was determined. An ORF of 684 nucleotides that encoded a protein of 228 amino aicds was detected. Asparagine-71 of XYN from B. Pumilus A-30 showed to be highly conservative in alkaline xylanases of family G/11, upon comparing the amino acid sequences of 17 family G/11 xylanases. Site-directed mutation of N71D of the xynA gene resulted in a decrease of 12.4% in the specific acitivity and a significant decline in the enzyme activity in the alkaline pH range.

Introduction Community medicine with the concept of comprehensive medical care and an ideal medical care delivery system not only for an individual or family but for the whole community has emerged. In April 1970, the Presbyterian Medical Center started a hospital based community health service project in order to improve the health of the people in rural areas. Prior to commencing a comprehensive medical care system, a family survey was needed. The major objective of this survey was to obtain information concerning the people and their environment so as to be able to plan and implement a comprehensive medical care program in Soyang-Myun. 2. Survey Method An interview using a family record form was carried out for each household. This family record form was designed to get information about demography, family planning, environmental sanitation and vital statistics. Prior to beginning, the members of the survey team were trained in interviewing techniques for three days. The team consisted of a public health nurse, four nurse-aides, a sanitarian and four health extension workers who are working in our project, The survey was carried out during the period November 1971 to March 1972. 3. Project area 1) Population of Soyang-Myun was 11,668; male, 5,962 and female, 5,706. Effect of Several Species of the Family Rubiacea on Cytotoxicity and Apoptosis in HL-60 cells

  • Herbal medicines have been utilized to treat a variety of diseases, including cancer. Several species of the family rubiaceae have been reported to have antitumor activity. In this study, we report the cytotoxicity and antitumor activity exhibited dy the methanol extracts prepared from Rubia radix (RRME), Uncaria gambir (UGME) and Oldenlandia diffusa (ODME) (family: Rubiaceae) against human promyleloid leukemia cell line, HL-60. The cytotoxicity of RRME (2~20 ${\mu}g/ml$), UGME (20~200 ${\mu}g/ml$) and ODME (20~200 ${\mu}g/ml$) were assessed dy the MTT reduction assay. IC50 values for RRME, UGME and ODME were 11.0, 99.5 and 106.1 ${\mu}g/ml$, respectively. When the HL-60 cells were treated with RRME (10 ${\mu}g/ml$), UGME (120 ${\mu}g/ml$) and ODME (140 ${\mu}g/ml$) for 24 h, several apoptotic characteristics such as DNA fragmentation and morphologic changes were observed. Furthermore, flow cytometric analysis was peformed to determine the percent of apoptotic cells. The poupulation of sub-G1 hypodiploid cells was increased 37.49% in RRME treatment, 12.49% in UGME treatment and 7.21% in ODME treatment compared with untreated control cells (2.64%). To further confirm apoptotic cell death, we assayed caspase-3, -8 and -9 activities in RRME, UGME and ODME-treated cells. After treatment of RRME, UGME and ODME for 12 h, caspase-3, -8 and -9 activities significantly increased.compared to untreated control cells. These results show that RRME, UGME and ODME induced apoptotic cell death in HL-60 cells and may have a possibility of potential antitumor activities.

Soil Organic Carbon of Soil Series from 2003 to 2010 in Korea

  • Soil organic carbon (SOC) of soil series is necessary to calculate soil C sequestration due to IPCC default categorized by climate regions and by soil types. The 3,400 thousand data were downloaded from agricultural soil information system and analyzed to get averages of soil order, soil series, and textual family for the three different soil management practices in Korea. The SOC content was $13.3{\pm}5.38g\;kg^{-1}$ in paddy field, $13.7{\pm}7.19g\;kg^{-1}$ in upland field, and $15.2{\pm}8.22g\;kg^{-1}$ in orchard soil, respectively. As SOC in orchard was 10% greater than that in upland, orchard must be managed with applying compost. The SOCs of inceptisols, which was largely distributed in Korea, were $13.6{\pm}5.48g\;kg^{-1}$ in paddy field, $14.1{\pm}7.38g\;kg^{-1}$ in upland field, and $15.3{\pm}8.20g\;kg^{-1}$ in orchard soil, respectively. The SOCs of alfisols were $13.6{\pm}4.96g\;kg^{-1}$ in paddy field, $13.7{\pm}6.99g\;kg^{-1}$ in upland field, and $15.6{\pm}8.59g\;kg^{-1}$ in orchard soil, respectively. The SOCs of entisols were $11.7{\pm}5.16g\;kg^{-1}$ in paddy field, $12.8{\pm}7.05g\;kg^{-1}$ in upland field, and $13.7{\pm}7.81g\;kg^{-1}$ in orchard soil, respectively. The SOCs of ultisols were $12.7{\pm}4.79g\;kg^{-1}$ in paddy field, $12.7{\pm}6.22g\;kg^{-1}$ in upland field, and $16.3{\pm}8.49g\;kg^{-1}$ in orchard soil, respectively. The fact that soils containing greater clay content in textual family had also more SOC content revealed that SOC could be also dependent on some soil properties as well as soil order. Because SOC differences among soil series representing same textual family were greater than those among textual family, SOC differences should be mainly affected by management practices such as compost application.

A Study on the Flora in Tanyang Limestone Area (단양 석회암지역의 식물상에 관한 연구)

  • Biomass and soil properties of plant communites was investigated in the limestone ares, Tanyang, Ch'ungbuk Province in Korea. Plants was classified in calcicoles and calcifuges within plant communities occurring in the limestone area. Biomass of plant populatins in the limestone area was measured that Stellaria aquatica was the lowest(0.58g/plant), Chrysanthemum boreale was the highest(8.87g/plant) and that Pennisetum alopecuroides, Miscanthus sinensis was 6.67g/plant, 5.76g/plant, respectively. Diameter of breast height among the tree layer communities was investigated that Pinus densiflora was the widest(20cm), Juniperus rigida was the narrowest(6cm) and that Quercus aliena, Populus ${\times}$ tomentiglandulosa was 10cm, 12cm, respectively. The high of density in the tree layer was found out Populus ${\times}$ tomentiglandulosa, Juniperus rigida, Quercus aliena, Pinus densiflora in this order. Dominant species in the study area were Pinus densiflora, Quercus aliena, Juniperus rigida, Quercus dentata in the tree layer, Lespedeza bicolor, Buxus microphylla, Rhus chinensis in the shrub layer, Humulus Japonicus, Erigeron canadensis, Xanthium strumarium, Oenothera odorata Persicaria hypropiper on the riverside, Pennisetum alopecuroides, Vicia amoena, Chrysanthemum boreale on the roadside. Soil properties of the limestone area was investigated around the protrusion of rock in the upper region, around the cave and underlayer accumulated by corrosion in the study area. Lime-chlorosis was observed in the upper region, underlayer and around the cave. It was observed that biota of kum-gul and dungbong-gul was Rhinolophus ferrumequinum korai, Epanerchodus kimi, Antrokoreana gracilipes, Diestrammena japonical. Dominant species were Humulus japonicus, Stellaria aquatica, Hydrocotyle maritima, Phragmites communis, Miscanthus sinensis around the inlet of a kum-gul. It was classified that plants in the study area was 34 order, 53 family, 135 species in all. Calcicoles were 11 order, 16 family, 18 species and calcufuges were 6 order, 8 family, 11 species of them all.

An Ethnographic approach to the Study of The Psychological Welfare of Teenage Heads of Family (아동가장의 심리적 복지에 관한 문화기술적 접근)

  • Employing an ethnographic approach, this case study analyzed the psychological welfare of 2 teenage and one early-twenties heads of family. The subjects were Sun-Ju (female, age 22, seamstress), Ji-Hyun (female, age 14, junior high school student), and Seung-Hwan (male, age 16, junior high school student). This study was processed between 9 March 1996 and 11 May 1996. The results show that teenage and early-twenties heads of family have ambivalent feelings, meaning two extreme psychological conditions that are not consistent. Three types of teenage-early twenties heads of family ambivalence were caused by diverse role dimensions (e.g., nursing, supporter, beneficiary, and independent subject roles). Revision of the present welfare system, particularly the protection system For teenage-early twenties heads of family was recommended. Further research is also needed to determine various factors harmful to their psychological welfare.
