Cow Faeces in In vitro Digestibility Assays of Forages

  • The present study investigated the use of cow faeces as a source of micro-organisms instead of sheep rumen liquor in in vitro digestibility assays of forages. Initially 40 forage samples comprising ryegrass, wheat, maize and barley were screened to select a wide range of digestibility (327-794 g/kg) of forages. Finally 8 forage samples were assessed using rumen liquor as well as faecal liquor. The absolute organic matter digestibility (OMD) values using faecal liquor were lower than those with rumen liquor. However, the relationship between OMD using rumen liquor and faecal liquor was highly significant (p < 0.001). The $R^2$ value exceed 0.90. The results suggest that micro-organisms from faecal liquor are capable of digesting forage samples.

Isolation and Identification of Acid- and Bile-Tolerant Lactobacillus salivarius subsp. salivarius from Human Faeces

  • This study was conducted to isolate lactobacilli having characteristics to be used as health adjuncts with fermented milk products. Acid tolerant strains were selected in Lactobacilli MRS broth adjusted to pH 4.0 from human faeces. Bile tolerant strains were examined in Lactobacilli MRS broth in which 1.0% bile salt was added. Microhemagglutination tests using swine erythrocytes were performed to select lactobacilli having adherence properties to survive in the intestinal tract. By examination of these characteristics the strain Nam 27, which was isolated from adult faeces, was selected and identified as Lactobacillus salivarius subsp. salivarius based on carbohydrate fermentation and 16S rDNA sequencing.

Comparison of Rabbit Caecal Content and Rabbit Hard Faeces as Source of Inoculum for the In vitro Gas Production Technique

  • In order to find an alternative source of inoculum to caecal content for studying the fermentation activity of rabbit hindgut, caecal content and faeces of 25 hybrid Hyla rabbits were used as inocula for an in vitro gas production trial. About 1 g of three substrates (dehydrated alfalfa meal, dehydrated beet pulp, barley) was weighed, in quadruplicate per inoculum, in 120 ml bottles; 75 ml of anaerobic medium and 4 ml of reducing solution were added and bottles were placed at $39^{\circ}C$. Caecal content and faeces were diluted respectively 1:2 (CI) and 1:8 (FI) with anaerobic medium and were introduced in the respective bottles (10 ml). Gas production was recorded 20 times at 2-24 h intervals throughout fermentation (96 h). The fermentation characteristics (i.e. degraded organic matter, OMd; potential gas production, A; fermentation rate, Rmax; time at which it is reached, Tmax; pH, volatile fatty acid, VFA) were studied by inoculum and feedstuffs. The feedstuffs, according to their chemical composition, showed very different fermentation characteristics. In particular, OMd, A and Rmax allowed feedstuff classification as follows: barley>beet pulp>alfalfa. The inocula differ (p<0.05) in Tmax, were higher for CI (15.53 vs. 11.96 h) and in VFA production. In particular, CI produced higher levels of acetate (38.9 vs. 33.4 mM/g OM incubated, p<0.01) and isobutyrate (0.72 vs. 0.42, p<0.01) but less propionate (7.1 vs. 10.3, p<0.01) and butyrate (11.3 vs. 14.0, p<0.01). However, the trend of gas production, similar for the inocula according to the fermented substrate, and the good regression equation to estimate some caecal fermentation parameters from faeces suggest that, after standardisation, the faeces could be used as an alternative inoculum for gas tests in rabbit.

돼지에서 살모넬라균(菌)을 분리(分離)하는 방법(方法)에 관한 연구(硏究) (Studies on the Methods for the Isolation of salmonella from Pigs)

  • 건강도쇄돈(健康屠殺豚)의 장간막임파선(腸間膜淋巴膳)과 분(糞)(대장내용물(大腸內容物))으로부타 살모넬라균(菌)의 분리방법(分離方法)을 비교연구하여 다음과 같은 결론을 얻었다. 1. 비교적 다른 세균에 의한 오염도(汚染度)가 낮은 장간막임파선(腸間膜淋巴膳)을 검사하였을 경우, 증균배지(增菌培地)로서는 tetrathionate broth(TB)가 selenite broth(SB) 보다 살모넬라균(菌) 분리율(分離率)이 월등히 높았으며, 선택배지(選擇培地)로서는 brilliant green agar (BG)가 SS agar(SS) 보다 역시 월등히 높았다. 2. 여러가지 장내세균(腸內細菌)에 의한 오염도(汚染度)가 높은 분(糞)을 검사하였을 경우에는, 살모넬라균(菌)의 분리율(分離率)에 있어서 증균배지(增菌培地)로 사용한 TB와 SB의 우열(優劣)을 인정할만한 차이를 보지 못하였으며, 선택배지(選擇培地)로 사용한 BG와 SS의 성적도 비슷하였다. 증균배지(增菌培地)나 선택배지(選擇培地)를 어느 한가지 배지(培地)만 사용한 경우보다는 두가지 배지(培地)를 동시에 같이 사용함으로써 살모넬라균(菌)의 분리율(分離率)이 크게 높았다. 3. Brilliant green McConkey broth 를 증균배지(增菌培地)로 사용하였을 경우에는 장간막임파선(腸間膜淋巴膳)과 분(糞)의 두 가지 재료중 어느것이나 TB 보다 살모넬라균(菌)의 분리율(分離率)이 훨씬 낮았다. 분(糞)을 brilliant green McConeky agar 에 직접 배양(培養)하는 방법은, TB나 SB에 증균배양(增菌培養)을 한 뒤에 BG나 SS의 증균배지(增菌培地)를 사용하는 방법보다 또한 훨씬 분리율(分離率)이 낮았다. 4. TB 및 SB의 증균배양시간(增菌培養時間)은 $37^{\circ}C$에서 24, 48 및 72 시간(時間)등 세가지를 비교해본 결과 장간막임파선(腸間膜淋巴膳)은 48 시간(時間) 배양(培養)했을 때에 살모넬라균(菌)의 분리율(分離率)이 가장 높았으나 분(糞)은 24 시간(時間)배양했을 때에 가장 분리율(分離率)이 높았다.

Effect of Nonstarch Polysaccharide-Rich By-Product Diets on Nitrogen Excretion and Nitrogen Losses from Slurry of Growing-Finishing Pigs

  • An experiment was conducted to investigate the effect of diet for growing-finishing pigs with high level of non-starch polysaccharides (NSP) from by-products on nitrogen excretion and nitrogen losses from slurry during storage. Sixteen commercial crossbred barrows of about 68 kg BW were randomly allotted to one of four diets. The control diet was formulated using tapioca and rice as basal energy sources. In the other diets, tapioca was replaced by either coconut expellar, rice bran or beer by-product. The diets differed mainly in the amount and compostition of NSP. After a 12-day adaptation period, urine and faeces were collected separately in metabolism cages for 9 days. Urine and faeces from the first four days were used to analyse the nitrogen partitioning. Urine and faeces from the last 5 days were mixed as slurry. The slurry was sampled at the end of the collection period and again after 30 days storage, to analyse for nitrogen to calculate the losses. Increasing dietary NSP reduced urinary nitrogen and nitrogen losses from the slurry during storage. The pigs fed the diet based on beer by-product excreted the most nitrogen via faeces and the least nitrogen via urine. Nitrogen losses from slurry of pigs fed the beer by-product were from 34 to 65% lower than from the other three diets. It is concluded that including NSP-rich by-products in the diet of growing-finishing pigs reduces urinary nitrogen excretion and nitrogen losses from slurry during storage.

Effect of Fermented Fruits on the Growth Performance, Shedding of Enterobacteriaceae and Lactobacilli in Post-weaning Pigs

  • The aim of this study was to investigate the effect of fermented fruits (FF) on the growth performance, Enterobacteriaceae and Lactobacillus counts in faeces of the post-weaning piglets. A total of twenty-four 4 weeks old Landrace${\times}$ Large White${\times}$Duroc with initial body weight of 6 kg were used in this study. The piglets were housed individually in metabolic cage and randomly assigned to four groups with six piglets per group. The piglets were fed on basal diet without antibiotic (AF), basal diets with antibiotic (Ab), basal diet with 10% (w/w) fermented fruit (10% FF) and basal diet with 20% (w/w) fermented fruit (20% FF). Faecal samples were taken directly from the rectum of each piglet and cultured for Enterobacteriaceae and Lactobacillus counts. In the growth performance, the piglets of Ab and 10%FF had significantly higher (p<0.05) average daily gain than those of 20%FF. However, no differences (p>0.05) were observed between AF, Ab and 10%FF. Studies showed that the use of fermented fruits (FF) could significantly (p<0.05) reduce Enterobacteriaceae population in piglets' faeces compared to the use of normal feed (AF) and antibiotic (Ab). However, the Lactobacillus population in the faeces was increased in those piglets fed with diets added with FF.

오령지 물추출물이 혈관내피세포의 chemokine 생성에 미치는 영향 (Effects of Faeces Trogopterori on the Production of Chemokine in HUVECs)

  • In order to validate the use of Faeces Trogopterori as an anti-inflammatory drug in the traditional Korean medicine, I have investigated the effect of water-soluble extract of F. Trogopterori (EFT) on the production of monocyte chemoattractant protein-1 (MCP-1), of which chemokine stimulates the migration of mononuclear cells, in human umbilical vein endothelial cells (HUVECs) stimulated with tumor necrosis factor-alpha. The extract inhibited dose-dependently MCP-1 production without its cytotoxic effect on HUVECs, as measured by enzyme-linked immunosorbent assay, and significantly decreased mRNA levels of MCP-1, as determined using reverse transcription polymerase chain reaction. These results suggest that F. Trogopterori may have therapeutic potential in the control of endothelial disorders caused by inflammation.

Effect of Helminthiasis on Zinc Metabolism

  • The effect of helminthiasis on zinc metabolism was monitored using endogenous $^{65}Zn$ after intraperitoneal injection of 1 g of $^{65}Zn$ as zinc chloride. In the first experiment zinc turnover was investigated in 18 male weanling rats, which were randomly divided into 3 groups. One group was infected with 73 third stage larvae of Nippostrongylus brasiliensis per gram body weight ; the other groups were the pair-fed and ad lib-fed controls. The route of loss of zinc was investigated in the second experiment with the same design using 18 animals with a lower dose of infection (33 larvae per gram body weight). The biological half life of endogenous $^{65}Zn$ was lower (p<0.05) in the infected group as compared to the controls. In the later phase of infection (9th to 16th day) there was reduced retention of $^{65}Zn$ and increased loss (p<0.05) of $^{65}Zn$ from the body though urine and faeces. It was concluded that infection of N. brasiliensis was accompanied by increased loss of endogenous Zn through faeces and urine.

수념산 전탕액이 배양 심근세포에 미치는 영향 (Effects of Sujeom-san Water Extract in Cultured Rat Myocardial Cells)

  • To test the protective effect of herbal medicine against oxygen free radical-induced myocardiotoxicity, cytotoxicity of xanthine oxidase/hypoxanthine (XO/HX) was examined using MTT, TBARS, and beating rate assay in the presence of water extract of Sujeom-san(SJS) or single consituents of its prescription. Myocardial toxicity was evaluated in neonatal rat myocardiocytes in cultures. In the present paper, XO/HX resulted in a decrease in viability and beating rate and increases in lipid peroxidation in cultured myocardial cells. In the effect of SJS water extract, it showed effects from the cardiocytotoxicity induced by XO/HX treatment such as increases in beating rate and decreases in lipid peroxidation. In the effect of Rhizoma Corydalis (RC), Faeces Trogopterori (FT), Fructus Amomi Tsaoko (FAT) and Myrrha on the cardiocytotoxicity, they were significantly effective in blocking the XO/HX-induced cardiocytotoxicity by increase of beating rate in FAT and FT group as well as decrease of lipid peroxidation in FT and RC group. These results show that oxygen free radical elicits toxic effects in cultured myocardial cells derived from neonatal rat, and suggest that water extract of Sujeomsan, Rhizoma Corydalis, Faeces Trogopterori, Fructus Amomi Tsaoko or Myrrha is very effective in the prevention of xanthine oxidase/hypoxanthine- induced cardiotoxicity.

MEASUREMENTS OF METABOLIC PARAMETERS IN LACTATING DAIRY COWS WITH NEAR INFRARED SPECTROSCOPY ANALYSIS USING CATTLE FAECAL SAMPLES

  • The knowledge of the nutrition in animal production is difficult to assess due of difficulty to determine the amount and quality of feeds intake, mainly if forages are the most important compound in the diet. It must be able to know responses to the metabolic process in lactating dairy cows earring out feeds evaluation trials. These metabolism studies with cows, requires measurements of: I) The amount of all feeds consumed. II) Excretion of faeces. III) Excretion of urine. Taking as a basis these trials, forage and total intake, dietary digestibility and balances of nitrogen and energy can be calculated. However, these feeds evaluation experiments with animals are very time consuming and expensives. The faeces excreted by animals containt undigested residues of the diet consumed. For this reason, their analysis can be an successful tool to determine the amount and quality of feed intake and other important biological parameters. The aim of this work was to know if faeces analysis by NIR could be used to determine with enough accuracy some attributes of different lactating dairy cows diets, using a global equation, developed on an heterogeneus population. For this purpose a total of 79 faecal samples from eight grass, three grass silages and two maize silages metabolic trials, on six cows each one, were used to constitute the initial population. The calibration equations were developed to predict forage and total intake, organic matter digestibility, digestibility coefficient of crude energy and digestibility energy. The combination of several trials with different diets and animal conditions gave promising results.
