  • The immunosuppressive and cell cycle arrest agent rapamycin works by binding together two proteins: the FK506 binding protein (FKBP12) and the FKBP-rapamycin associated protein (FRAP). A 2.7 $\AA$ resolution crystal structure of the triple complex of human FK506 binding protein (FKBP12), rapamycin, and FKBP12-rapamycin binding domain (FRB) of FRAP, reveals two proteins bound together through rapamycin' s ability to simultaneously occupy two different hydrophobic binding pockets. • This study compared the antioxidant effects of two kinds of black raspberry extract, obtained from fruits of Rubus coreanus and Rubus occidentalis, which can be found in Korea. The fruits of R. coreanus and R. occidentalis were each extracted with 0%, 25%, 50%, 75%, and 100% ethanol (EtOH). Among the extracts of these two varieties, 50% EtOH extract of R. occidentalis showed the highest contents of total polyphenols ($46.96{\pm}2.78mg/g$) and flavonoid compounds ($11.77{\pm}0.81mg/g$). The 50% EtOH extract of R. occidentalis showed the highest antioxidant activity ($84.77{\pm}0.97%$) in terms of DPPH radical scavenging activity. On the contrary, 25% EtOH extract of R. occidentalis showed the best antioxidant activity ($29.65{\pm}2.41%$) in terms of ABTS radical scavenging activity. In the results of ferric reducing antioxidant power (FRAP) assay, 50% EtOH extract of R. occidentalis showed the highest antioxidant activity ($0.49{\pm}0.02%$). In the cytotoxicity test stimulated with $H_2O_2$, the extracts of 75% and 100% EtOH from R. occidentalis showed the highest cell viability ($93.54{\pm}3.37%$ and $97.19{\pm}0.74%$, respectively). According to our results, extracts of R. occidentalis showed higher antioxidant activities than extracts of R. coreanus. Especially, total polyphenol and flavonoid contents of R. occidentalis showed the highest significant correlation with FRAP by Pearson's correlation (P=0.005 and P=0.013, respectively).

  • The aim of this study was to determine the chemical and intracellular antioxidant activities of ${\beta}$-carotene and lycopene and to compare their quantitative structure-activity relationship (QSAR). In our previous study, the second ionization energy of lycopene was higher than that of ${\beta}$-carotene, as calculated by QSAR. Chemical antioxidant activities of ${\beta}$-carotene, lycopene, and Trolox were examined by measuring ferric reducing antioxidant power (FRAP) and 2,2'-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity. Intracellular antioxidant activities were evaluated by intracellular reactive oxygen species (ROS) and DNA fragmentation. The FRAP of lycopene was higher than that of ${\beta}$-carotene (P<0.05), and the two carotenoids had similar antioxidant activities in DPPH radical scavenging activity assay. Trolox had the greatest chemical antioxidant activities (P<0.05). When RAW264.7 cells were treated with lipopolysaccharide (LPS) (100 ng/mL) for 20 h, intracellular ROS and DNA fragmentation significantly increased (P<0.05). RAW 264.7 cells pretreated with ${\beta}$-carotene ($4{\mu}M$) and lycopene ($0.4{\sim}2{\mu}M$) for 4 h formed significantly less intracellular ROS than LPS-treated control cells (P<0.05), whereas cells with Trolox did not reduce production of intracellular ROS. In addition, cells pretreated with $2{\mu}M$ lycopene produced less intracellular ROS than those treated with ${\beta}$-carotene (P<0.05). DNA fragmentation of cells with ${\beta}$-carotene and lycopene was similar to that of LPS-treated control cells as measured by Hoechst staining. The antioxidant ability of lycopene was greater than that of ${\beta}$-carotene in the QSAR, FRAP, and intracellular ROS assays (P<0.05). ${\beta}$-Carotene and lycopene had lower antioxidant activities as measured by FRAP (P<0.05) but higher intracellular protective effects against LPS-induced oxidative stress in comparison with Trolox.

  • This study was carried out to investigate antioxidant compounds (total polyphenol and total flavonoid) and antioxidant activities (DPPH radical scavenging activity, ABTS radical scavenging activity, FRAP activity, and reducing power] of the mycelium and fruit body of Sparassis crispa depending on extraction temperature ($60^{\circ}C$ and $95^{\circ}C$). For total polyphenols, total flavonoids, FRAP activity, and reducing power, the mycelium of S. crispa extracted at $95^{\circ}C$ showed the highest contents and activities. The mycelium of S. crispa extracted at $60^{\circ}C$ and fruit body of S. crispa extracted at $95^{\circ}C$ showed the highest DPPH radical scavenging activity and ABTS radical scavenging activity, respectively. This study suggests that the antioxidant activities of S. crispa extracted at $95^{\circ}C$ are better than those of S. crispa extracted at $60^{\circ}C$, and the mycelium contained more antioxidant compounds than the fruit body.

  • This study was carried out to compare anti-oxidant activities of chaff vinegar liquors. We examined the effects of several kinds of chaff vinegar liquor (CA; CA1, chaff vinegar liquor; CA2, wood vinegar liquor; CA3, chaff vinegar liquor with red ginseng; and CA4, chaff vinegar liquor with rosemary) by 1,1-diphenyl-2-picrylhydrazyl (DPPH), ferric reducing activity of plasma (FRAP) assay, and $Cu^{2+}$ reduction. The results showed that CA have potential in reducing DPPH, FRAP and $Cu^{2+}$ activity. CA was increased its anti-oxidant activity by the fermentation of rosemary extract. The present results suggest that the chaff vinegar liquor could be used for anti-oxidant agents and/or be developed for anti-oxidative potentiation of prototypes.

  • Objectives: FDY003 is a raw material for medicine consisting of a natural product that is expected to have the advantages of low side effects and high efficacy. In this study, we predict the efficacy and the standardization of the drug by method validation of anticipated index compounds and the measurement of antioxidant activity. Methods: FDY003 is prepared by extracting and purifying 70% of ethyl alcohol (EtOH). The method validation of cordycepin and chlorogenic acid was determined by high-performance liquid chromatography-photo diode array (HPLC-PDA) and the content of FDY003 was calculated. In order to monitor the biological activity of FDY003, antioxidant activity was measured by 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2-azino bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and ferric-reducing antioxidant power (FRAP). The equivalent values of antioxidants such as trolox, ascorbic acid, gallic acid, and caffeic acid were measured by ABTS and FRAP. Results: Chlorogenic acid and cordycepin were both found suitable for method validation in HPLC and FDY003 containing 9.92±0.50 and 17.97±0.27 ㎍/g, respectively. In DPPH, the electron donating ability (EDA) value of FDY003 was increased in a concentration dependent manner. FDY003 confirmed antioxidant activity by ABTS and FRAP. Conclusions: FDY003 contains certain components including cordycepin and chlorogenic acid and has antioxidant ability by various mechanisms. Therefore, it is expected that FDY003 is capable of various physiological activities including anti-cancer activity.

  • In this study, bioactive constituents and biological activities of aronia, blackcurrant, and maquiberry were investigated. The 60% ethanol extracts were prepared and evaluated for total polyphenol, total flavonoid, and total anthocyanin contents. Biological activities, including 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activities, ferric reducing antioxidant power (FRAP), reducing power, tyrosinase inhibitory activity, and nitrite scavenging activity were also examined. Maquiberry showed the highest bioactive constituents as shown based on total polyphenol (73.66 mg GAE/g), total flavonoid (28.08 mg/g), and total anthocyanin (12.82 mg/g) contents compared to aronia and blackcurrant. Maquiberry also exhibited the highest DPPH and ABTS radical scavenging activity, FRAP, and reducing power at a concentration of 5 mg/mL, which were superior to those of ascorbic acid (a positive control). Therefore, maquiberry could be used as a potential source of antioxidants and functional food substances.

  • Although several analytical methods for measuring total antioxidant capacity (TAC) have been applied to biological samples, there were often dissimilar results due to the different principles of methods applied. Thus, this study aimed to validate four conventional analytical methods for measuring plasma TAC, including the ABTS assay, DPPH assay, FRAP assay, and ORAC assay, by comparing with urinary 8-isoprostane concentration. In addition, TAC results were compared with antioxidant enzyme activities including superoxide dismutase (SOD) and glutathione peroxidase in erythrocyte, and catalase in plasma. Plasma TAC measure by ABTS assay was strongly correlated with the result by FRAP assay. Plasma TAC by FRAP and ORAC assays were negatively correlated with erythrocyte SOD activity. The agreement among the four TAC assay methods and 8-isoprostane was determined using 95% prediction limits of linear regression, expressed as the mean of 8-isoprostane ${\pm}95%$ prediction limits. The ABTS method better agreed with 8-isoprostane than the other methods, demonstrating narrow prediction of limits. Furthermore, only plasma TAC determined by the ABTS assay was inversely correlated with urinary 8-isoprostane (r = -0.35, p < 0.05). In summary, the ABTS assay would be an appropriate method to measure overall plasma antioxidant capacity and predict the body's antioxidant status.

  • This study was conducted to investigate the antioxidant activity and the bioactive compounds found in 70% ethanol extracts taken from Sanguisorbae officinalis L. leaves(SO) cultivated in Korea. The extracts were tested for their total phenolic contents (TPC), total flavonoid contents (TFC), phenolic compounds, and antioxidative activities using various in vitro assay such as DPPH, ABTS radical scavenging activity, FRAP activity, and reducing power. The TPC and TFC were found to be $119.3{\pm}1.54mg$ gallic acid and $59.6{\pm}1.43mg$ rutin at mg of 70% ethanol extracts, respectively. Catechin was the major material among the phenolic compounds in SO extracts. The DPPH radical scavenging activity, ABTS radical scavenging activity, FRAP activity, and reducing power of SO extracts were increased in a dose-dependent manner. These results suggest that SO extracts could be considered as a good source of natural antioxidants and functional food ingredient.
