부산해운대 동백섬 연안천해에 분포하는 노래미 개체군의 식성을 파악하기 위해 노래미의 위내용물을 분석하고 환경생물을 조사했다. 노래미의 주된 먹이 생물은 amphipoda 및 isopoda에 속하는 갑각류였다. 특히, Gammaridae는 모든 계절에서 어느 연령군의 노래미의 위내용물로부터 우점적으로 출현했다. Caprella sp.는 산란직전의 $8{\sim}9$월에 많이 나타났다. Synidotea laevidorsalis는 위내용물에서 개체수로서는 조성비가 낮았지만, 용적으로서는 Gammaridae 다음으로 조성비가 높았다. 저연령어는 크기가 작은 Gammaridae, Caprella sp., Dynamene sp.를 많이 먹었으며, 고연령어는 크기가 큰 S. laevidorsalis, Polychaeta, Reptantia를 많이 먹었다. 노래미 암수 간의 먹이 생물 차는 없었다. 연중 적극적(positive)으로 선택섭식된 먹이 생물은 Caprella sp.였다. Gammaridae는 봄과 가을(10,11월)에는 적극적으로 섭식되었으나, 여름과 9월에는 소극적(negative)으로 섭식되었다. 먹이 생물의 섭식량은 고연령군일수록 많았다. 노래미가 성장할수록 섭식하는 먹이 생물의 종류는 다양했다. 소형의 먹이 생물은 노래미의 어느 연령군의 위내용물에서도 고루 출현하였다. 한편 대형의 먹이 생물의 경우에는 노래미의 나이가 많을수록 위내용물에서 출현하는 크기가 증가했다.
Objectives: To circumvent the limitations of the current golden standard method, colony-forming unit (CFU) assay, for viability of Bacille Calmette-$Gu{\acute{e}}rin$ (BCG) vaccines, we developed a new method to rapidly and accurately determine the potency of BCG vaccines. Methods: Based on flow cytometry (FACS) and fluorescein diacetate (FDA) as the most appropriate fluorescent staining reagent, 17 lots of BCG vaccines for percutaneous administration and 5 lots of BCG vaccines for intradermal administration were analyzed in this study. The percentage of viable cells measured by flow cytometry along with the total number of organisms in BCG vaccines, as determined on a cell counter, was used to quantify the number of viable cells. Results: Pearson correlation coefficients of FACS and CFU assays for percutaneous and intradermal BCG vaccines were 0.6962 and 0.7428, respectively, indicating a high correlation. The coefficient of variation value of the FACS assay was less than 7%, which was 11 times lower than that of the CFU assay. Conclusion: This study contributes to the evaluation of new potency test method for FACS-based determination of viable cells in BCG vaccines. Accordingly, quality control of BCG vaccines can be significantly improved.
시장에서 수집한 갈변병에 감염된 느타리버섯으로부터 125 균주를 분리하였고 그 중 45 균주는 병원성 균주로 조사되었다. WLFO 6 strains, WLFO 6 strains으로 나타났고, WLRO의 몇 균주는 병원성 검정 실험에서 약한 병원성을 나타내기도 했다. WLFO 6균주는 모두 느타리버섯의 갈변병 주원인균으로 알려진 P. tolaasii로 미생물 신속 동성을 (MIDI, gas chromatograph-microbial identification system)에 의해 동정되었고 WLRO는 P. gingeri, P. fluorescens biotype A and type C. 로 동정되었다. 그 외의 선발된 Pseudomonas spp.는 P. gingeri, P. agarici, P. fluorescens biotype B, P. chloroaphis, non-pathogenic P. tolaasii, P. putida biotype A, B 등으로 동정이 되었다. 분리된 병원성 세균의 세포배양 여과액으로 버섯에서의 갈변과 조직 함몰을 실험하였다. 병원성 검정에서 약한 병원성을 보인 분리균들은 갈변 또는 조직함몰이 나타나지 않았으나 강한 병원성을 나타냈던 균들은 두 현상이 모두 나타났다. P. tolaasii에 의해서 생성되는 세포외독소인 tolaasin의 활성을 조사하기 위하여 600 nm에서 용혈 활성을 측정하였다. P. tolaasii로 동정된 6 균주는 0.8∼0.9, 약한 병원성의 WLROs는 0.9∼1.0 그리고 Pseudomonas spp.는 10∼1.2로 나타났다. 공초점 현미경 기술을 이용하여 신선한 버섯에서의 조직을 optical sectioning image와 vertical sectioning image로 관찰하였고 또한 P. toiaasii에 의하여 오염된 조직부위의 영상을 회득하였다.

유전자 변형 작물은 생산성 측면에서 많은 장점이 있지만 이를 섭취할 경우 잠재적인 위험 요소들에 의해 많은 문제가 대두대고 있다. 본 연구는 저항성유전자를 이입한 배추에서 Profillin, Tubulin-${\alpha}$ (Tub-${\alpha}1$), Heat-shock protein (Bchsp 17.6) and Ubiquitin conjugating enzyme (UBE)의 발현과 이를 30일간 섭취한 마우스에서 ${\beta}$-actin(${\beta}$-act), ${\beta}$-2-microglobulin (B2m), Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and ${\beta}$-glucuronidase (Gus)의 발현 정도를 RT-PCR을 통해 알아보았다. 실험 결과 저항성유전자를 이입한 배추와 그렇지 않은 배추의 유전자 발현 패턴은 큰 차이를 보이지 앓았으며, 이를 섭취한 마우스 장기에서도 발현에 따른 큰 차이는 나타나지 않았다.

Mon1 is a guanine nucleotide exchange factor subunit that activates the Ypt7 Rab GTPase and is essential for vacuole trafficking and autophagy in eukaryotic organisms. Here, we identified and characterized the function of Mon1, an ortholog of Saccharomyces cerevisiae Mon1, in a human fungal pathogen, Cryptococcus neoformans. Mutation in mon1 resulted in hypersensitivity to thermal stress. The mon1 deletion mutant exhibited increased sensitivity to cell wall and endoplasmic reticulum stress. However, the mon1 deletion mutant showed more resistance to the antifungal agent fluconazole. In vivo studies demonstrated that compared to the wild-type strain, the mon1 deletion mutant attenuated virulence in the Galleria mellonella insect model. Moreover, the mon1 deletion mutant was avirulent in the murine inhalation model. These results demonstrate that Mon1 plays a crucial role in stress survival and pathogenicity in C. neoformans.
The second article of 'Effects of heat treatment on the nutritional quality of milk,' titled 'Destruction of microorganisms in milk by heat treatment' and authored by Dr. Seong Kwan Cha, who worked at the Korea Food Research Institute, covers the heat-stable microorganisms that exist in milk after pasteurization. The article focusses on the microbiological quality of raw milk and market milk following heat treatment, and is divided into four sub-topics: microbiological quality of raw milk, survey and measurement of microorganisms killed in raw milk, effect on psychrophilic and mesophilic microorganisms, and effect of heat treatment methods on thermoduric microorganisms. Bacillus spp. and Clostridium spp. are sporeforming gram-positive organisms commonly found in soil, vegetables, grains, and raw and pasteurized milk that can survive most food processing methods. Since spores cannot be inactivated by LTLT (low temperature long time) or HTST (high temperature short time) milk pasteurization methods, they are often responsible for food poisoning. However, UHT (ultra high temperature) processing completely kills the spores in raw milk by heating it to temperatures above $130^{\circ}C$ for a few seconds, and thus, the UHT method is popularly used for milk processing worldwide.
Incidents of seafood and massive fish kills have been rapidly increasing in both frequency and geographical distribution and the socioeconomic impacts brought by those incidents. However, the biological origins of those marine toxins have not been well clarified. Most of the marine organisms investigated are filter-feeder, which accumulate toxins from their food and/or their symbiotic microalgae. We have examined the action on rabbit platelets of marine toxins isolated from cultured dinoflagellates and sponge collected at Okinawa. Maitotoxin (MTX) is a water-soluble toxin isolated from the cultured dinoflagellate Gambierdiscus toxicus which causes a seafood poisoning in tropical regions. Zooxanthellatoxin A (ZT-A) was isolated from exteracts of cultured symbiotic dinoflagellate Symbiodinium sp. (socalled zooxanthella) from flatworms of the genus Amphiscolops collected at Okinawan marine sponge Theonella sp. MTX caused a disaggregation and a dissolution of large aggregates. ZT-A caused a dissolution of small aggregates followed by a increment of light tranmission. TZ-A caused an initial and transient shape change followed by a sustained aggregation and a increment of large aggregates. The objective of this study was to investigate the potential of the application of lactic acid bacteria (LAB) from kimchi as a starter culture in the production of fermented sausages. To achieve this, a submerged model medium that contained LAB as part of a complex system of kimchi (0.5, 1.0, 1.0, 3.0, and 5.0%) and lyophilized kimchi powder (0.2 and 0.5%) was fermented for 120 h. During the fermentation period, the growth of total viable organisms and LAB, and the changes in the pH and the titratable acidity, were investigated. The initial LAB counts ranged from 6.4 to 7.7 Log CFU/mL for the kimchi media, and from 6.9 to 6.9 Log CFU/mL for the kimchi powder media. In all the kimchi batches, the LAB increased logarithmically, and the highest LAB counts (around 9 Log CFU/mL) were reached in 24 h. An evident lag phase of the LAB was observed in the kimchi powder samples and reached 8.8 Log CFU/mL in 8 h. The decrease in the pH and the formation of lactic acid were rapid in the kimchi batches, and reached pH values of 3.4-3.5 in 12 h. With these results, the LAB that was integrated with the addition of kimchi or kimchi powder demonstrated its potential utility as a substitute for starter culture.
The present study was designed to characterize the bioaccumulation of organochlorine pesticides (OCPs) in marine organisms (zooplankton, oyster, crab, and goby) on different trophic level. In the present study, sedentary bivalve (oyster) showed strong correlations in OCPs levels with surface sediment in the study area. This indicates the two compartments can be used as alternative for pollution monitoring of OCPs even in narrow scale in space. Bioaccumulation and trophic transfer of OCPs was strongly associated with their hydrophobicity (i.e., KOW). HCHs with log KOW < 5 did not show any enrichment through food-chain. However, log BAF values of OCPs with log KOW > 5 positioned over the 1:1 lines of log BAF and log KOW of the top predator, indicating the greater fugacities in the higher trophic level and thus the occurrence of biomagnification via ingestion. Based on trophic transfer factors (TTF), more hydrophobic OCPs with log KOW > 5 were enriched by several to several ten times in the highest trophic level relative to the lowest trophic level. This finding can be used in the establishment of marine environmental water quality criteria by considering biomagnification factors (TTF in this study) of OCPs.
