Control and prevention of foot and mouth disease (FMD) by vaccination remains unsatisfactory in endemic countries. Indeed, consistent and new FMD epidemics in previously disease-free countries have precipitated the need for a worldwide control strategy. Outbreaks in vaccinated animals require that a new and safe vaccine be developed against foot and mouth virus (FMDV). FMDV can be eradicated worldwide based on previous scientific information about its spread using existing and modern control strategies.
Fig mosaic disease (FMD) is a viral disease that spreads in all Tunisian fig (Ficus carica L.) orchards. RT-PCR technique was applied to leaf samples of 29 fig accessions of 15 fig varieties from the fig germplasm collection of High Agronomic Institute (I.S.A) of ChattMariem, to detect viruses associated to FMD. Analysis results show that 65.5% of the accessions (19/29) and 80.0% (12/15) of the fig varieties are infected by FMD-associated viruses. From all fig accessions, 41.4% of them are with single infection (one virus) and 24.1% are with multi-infections (2 virus and more). Viruses infecting fig leaf samples are Fig mosaic virus (FMV) (20.7%), Fig milde-mottle-associated virus (FMMaV) (17.25%), Fig fleck associated virus (FFkaV) (3.45%), and Fig cryptic virus (FCV) (55.17%). A reliable protocol for FCV and FMMaV elimination from 4 local fig varieties Zidi (ZDI), Soltani (SNI), Bither Abiadh (BA), and Assafri (ASF) via in vitro culture of 3 meristem sizes was established and optimized. With this protocol, global sanitation rates of 79.46%, 65.55%, 68.75%, and 70.83% respectively for ZDI, SNI, BA, and ASF are achieved. For all sanitized varieties, the effectiveness of meristem culture for the elimination of FCV and FMMaV viruses was related to meristem size. Meristem size 0.5 mm provides the highest sanitation rates ranging from 70% to 90%.
Objective: Foot and mouth disease (FMD) and porcine reproductive and respiratory syndrome (PRRS) are major diseases that interrupt porcine production. Because they are viral diseases, vaccinations are of only limited effectiveness in preventing outbreaks. To establish an alternative multi-resistant strategy against FMD virus (FMDV) and PRRS virus (PRRSV), the present study introduced two genetic modification techniques to porcine cells. Methods: First, cluster of differentiation 163 (CD163), the PRRSV viral receptor, was edited with the clustered regularly interspaced short palindromic repeats-CRISPR-associated protein 9 technique. The CD163 gene sequences of edited cells and control cells differed. Second, short hairpin RNA (shRNAs) were integrated into the cells. The shRNAs, targeting the 3D gene of FMDV and the open reading frame 7 (ORF7) gene of PRRSV, were transferred into fibroblasts. We also developed an in vitro shRNA verification method with a target gene expression vector. Results: shRNA activity was confirmed in vitro with vectors that expressed the 3D and ORF7 genes in the cells. Cells containing shRNAs showed lower transcript levels than cells with only the expression vectors. The shRNAs were integrated into CD163-edited cells to combine the two techniques, and the viral genes were suppressed in these cells. Conclusion: We established a multi-resistant strategy against viral diseases and an in vitro shRNA verification method.
Foot-and-mouth disease (FMD) is one of the most infectious diseases affecting cloven-hoofed animals including cattle, sheep, goats, and pigs. Seven serotypes of foot-and-mouth disease virus with multiple subtypes within each serotype have been identified until now. In particular, it has been demonstrated that the outbreak of the serotype Asia1 reported from China, Mongolia and North Korea since 2005 is mostly classified into genetic group V. Though it has been recommended that Asia1 Shamir strain can be used as a high priority vaccine by World References Laboratory for FMD, the complete nucleotide sequences of the strain has not yet been determined. In this study, to be prepared for Asia1 type viruses that may be brought into Korea, the complete genome sequence of this vaccine strain Asia1 Shamir including its 5' and 3' non-coding region was identified.
Foot-and-mouth disease (FMD) and avian influenza (AI) are highly pathogenic viral disease which affects the livestock industry worldwide. Outbreak of these viruses causes great impact in the livestock industry; thus, disease infected animals were immediately disposed. Burial is the commonly used disposal method for deceased animals. However, there is potential for secondary environmental contamination, as well as the risk that infectious agents persisting in the environment due to the limited environmental controls in livestock burial sites during the decomposition of the carcasses. Therefore, this study aimed to investigate the detection of FMD and AI viruses from animal carcass disposal sites using real-time reverse transcription PCR. Soil samples of more than three years post-burial from livestock carcass disposal sites were collected and processed RNA isolation using a commercial extraction kit. The isolated RNA of the samples was used for the detection of FMDV and AIV using qRT-PCR. Based on the qPCR assay result, no viral particle was detected in the soil samples collected from the animal disposal sites. This indicates that 3 years of burial and their carcass disposal method is efficient for the control or at least reduction of spread infections in the surrounding environment.
구제역은 발생 시 전염이 쉽게 일어나며 심각한 경제적 피해를 일으키는 질병이다. 구제역의 방역정책은 발견 직후 빠른 살처분이 최선책이나, 전파 속도나 상황 등에 따라 타지역 백신 접종 등의 방법을 시행할 수도 있다. 이러한 방법을 적용하기 위해서는 구제역을 빠르고 정확하게 진단할 필요성이 있다. 개발된 진단법들은 구제역의 확진, 혈청형의 동정, 백신 접종 후 항체의 생성 확인 등에 사용된다. 많은 진단법들이 개발되었지만 아직은 빠른 시간 내에 검출이 가능하며 동시에 정확성도 가진 방법이 드물다. 그렇기에 기존의 방법들을 개선시킨 새로운 진단법이 필요하다. 현재는 대부분 혈청학적 진단법인 ELISA에 의존하거나 분자 유전학적 기술인 PCR을 사용한다. 가장 최근 기술은 그 둘을 합치는 방법으로, 어떻게 하면 더 신속하고 저비용이면서, 민감하고 정확한 방법이 될 수 있을지 연구가 진행되고 있다.
PURPOSE: To examine the cumulative (7 days) effect of breaking up prolonged sitting on systemic endothelial function in sedentary men. METHODS: Thirty sedentary men ($33.93{\pm}5.72years$) participated in two randomized 7 days sitting trial (Sit group (control) vs. Breaks group). The protocol of Breaks group is as follows: 4-minute of moderate-intensity marching in place (walking) every 1 hour during business hour (total: 8 breaks/day). Assessment of brachial artery endothelial function using flow-mediated dilation (FMD) and arterial stiffness indices (augmentation index, arterial pressure and pulse wave velocity) were measured before and after 7 days treatment. RESULTS: Brachial artery FMD significantly increased after 7 days breaking up prolonged sitting treatment (Breaks groups, $9.65{\pm}2.61$ to $9.62{\pm}2.6%$) compared with 7 days prolonged sitting (Sit group, $8.37{\pm}3.41$ to $10.11{\pm}3.75%$) (interaction effect, p=.004). Arterial pressure (AP) significantly increased after treatment (Breaks group, $2.75{\pm}2.19$ to $2.38{\pm}1.63mmHg$, p=.002) in Sit group but there was no change (Sit group, $1.00{\pm}3.18$ to $2.50{\pm}9.23mmHg$) in Breaks groups (interaction effect, p=.008). CONCLUSIONS: These finding show that 7 days regular breaking up prolonged sitting improve in FMD, compared with prolonged sitting. Therefore, regular breaking up prolonged sitting may improve systemic endothelial function in sedentary men.
Background: Foot-and-mouth disease (FMD) is a highly contagious viral disease in livestock that has tremendous economic impact nationally. After multiple FMD outbreaks, the South Korean government implemented a vaccination policy for efficient disease control. However, during active surveillance by quarantine authorities, pig farms have reported an insufficient antibody positivity rate to FMD. Objective: In this study, the spatial and temporal trends of insufficiency among pig farms were analyzed, and the effect of the number of government veterinary officers was explored as a potential preventive factor. Methods: Various data were acquired, including national-level surveillance data for antibody insufficiency from the Korea Animal Health Integrated System, the number of veterinary officers, and the number of local pig farms. Temporal and geographical descriptive analyses were conducted to overview spatial and temporal trends. Additionally, logistic regression models were employed to investigate the association between the number of officers per pig farm with antibody insufficiency. Spatial cluster analysis was conducted to detect spatial clusters. Results: The results showed that the incidence of insufficiency tended to decrease in recent years (odds ratio [OR], 0.803; 95% confidence interval [95% CIs], 0.721-0.893), and regions with a higher density of governmental veterinary officers (OR, 0.942; 95% CIs, 0.918-0.965) were associated with a lower incidence. Conclusions: This study implies that previously conducted national interventions would be effective, and the quality of government-provided veterinary care could play an important role in addressing the insufficient positivity rate of antibodies.
구제역은 발굽이 갈라진 동물에서 나타나는 바이러스성질병으로, 가축전염병 중에서 가장 전염력이 강한 질병의 하나이다. 2010년 12월 10일, 경북 안동시 와룡면 주계리 773-1번지에 조성된 구제역 가축 살처분 대량매몰지의 침출수와인근 지역(안동시 와룡면 주계리 777번지) 지하수의 물리화학적 특성을 1년간 분석하였다. 2011년 3월, 5월, 7월, 9월 및 11월에 각각 침출수 시료를 회수하여 수분함량, 고형분, pH, brix, 총당, 환원당, 조지질, 회분 함량 등을 분석한 결과, 매몰 이후 겨울동안 거의 변화가 없었으나, 3월~7월 사이, 특히 온도가 $18.4^{\circ}C{\sim}27.8^{\circ}C$를 유지한 5월~7월 사이 매립지내 침출수의 특성이 급격히 변화되며, 유기물질의 빠른 분해가 나타남을 확인하였다. 특히, pH 6.41(3월)에서 7.97(9월)로 지속적인 증가, brix 및 고형분 함량의 급격한 감소와 함께 마그네슘, 칼슘 및 철분과 같은 생체 미네랄도 3월 침출수에는 각각 1073, 4311 및 56.2 ppm에서 11월 침출수에는 151, 78, 및 0.1 ppm으로 감소함을 고려할 때, 침출수내 세균 증식이 침출수의 특성변화의 주요 원인으로 작용하리라 판단된다. 실제 침출수내 일반세균 분석 결과 5월-9월 사이 급격한 세균 증식이 나타남을 확인하였다. 한편 침출수 재처리의 기본 자료를 확보하기 위해, 침출수내 유기물질의 친수성산, 소수성산(fulvic acid 및 humic acid), 반친수성산의 상대적인 분포를 측정하여 유기물질 기원을 분석하였다. 그 결과 침출수는 전형적인 분뇨연계 시설 방류수와 유사한 유기물질 성상을 나타내었으며, 인근 지하수는 전형적인 논, 흙 기원의 유기물질 성상을 나타내었다. 본 연구결과는 구제역 매몰지 침출수의 거동, 침출수의 재처리 및 환경위해 및 식품보건학적 위해 분석의 기초자료로 이용될 것이다.
목 적 : 대혈관 합병증은 1형 당뇨병에서 주된 사망 원인으로 합병증을 예방하고 조기에 발견하는 것은 중요하므로, 저자들은 소아-청소년기에 1형 당뇨병으로 진단된 젊은 성인에서 대혈관 합병증의 발생 여부를 알아보고자 혈관초음파 검사를 통해 동맥경화의 초기 변화를 측정하였고, 이 변화와 관련된 위험인자를 분석하기 위해 본 연구를 시행하였다. 방 법 : 서울대학교병원에서 18세 이전에 1형 당뇨병으로 진단받고 추적 관찰 중인 20대의 72명의 환자들과 건강한 20명의 대조군을 대상으로 하였다. 환자들에서 고혈압과 고지혈증의 빈도 및 관련 인자를 알아보았으며, 환자군과 대조군 간에 혈관 초음파 검사를 통해 측정한 FMD, mean IMT를 비교하였고, 위험인자와의 상관성을 분석하였다. 결 과 : 72명의 대상 환자 중 32명(44.4%)에서 고혈압이 있었고, 고혈압군은 정상 혈압군 보다 남자의 빈도(P=0.03)와 평균 체질량지수(P=0.04)가 더 높았다. LDLc이 100 mg/dL 이상으로 측정된 환자는 31명(N=69, 44.9%)이었고, 전체 환자군의 LDLc은 평균 당화혈색소(r=0.32, P=0.008), 하루 총 인슐린 사용량(r=0.27, P=0.02)과 양의 상관관계를 보였다. Mean IMT는 환자군이 대조군 보다 유의하게 더 두꺼웠고($0.43{\pm}0.06$ mm vs $0.39{\pm}0.06$ mm, P=0.03), FMD는 두 군 사이의 통계적 유의성은 없었으나, 환자군 내에서 사춘기발현 이후의 유병기간이 길수록 FMD는 감소하는 경향을 보였다(r=-0.34, P=0.01). 결 론 : 20대의 1형 당뇨병 환자에서도 동맥경화가 진행되고 있으므로 대혈관 합병증의 위험인자에 대한 선별검사 및 이의 조절이 필요하며, 1형 당뇨병이 있는 젊은 성인뿐만 아니라 청소년기의 환자에서도 대혈관 합병증의 위험인자에 대한 주기적인 평가가 필요할 것으로 생각된다.
