Effects of a Butanol Fraction of Alisma canaliculatum and of Selenium on Blood Glucose Levels and Lipid Metabolism in Streptozotocin-Induced Diabetic Rats

  • The purpose of this study was to investigate the effects of a butanol fraction of fraction of Alisma canaliculatum All. Braun et Bouche (Ac), and of selenium (Se), on plasma gllucose and lipid levee in streptozotocin (STD-induced diabetic rats. Male Sprague-Dawley rats, fed the AIN-93 recommended diet, were divided into five groups: a non-diabetic control group (no STZ treatment), and four 572-induced diabetic groups which consisted of a diabetic-control group, an Ac-treated group, an Ac-Se treated group, and a Se-treated group. Diabetes was induced in the rats by an injection of STZ into the tail vein at a dose of 45 mg/kg body weight. The butanol (BuOH) fraction of Ac was orally administered at a rate of 400 mg/kg body weight for 21 days to both the Ac and Ac-Se groups. The supplementation of selenium in the Se and Ac-Se groups was achieved by adding (freshly, every day) 2 mg of Se as Na$_2$SeO$_3$ per kg of feed. The rats'body weights and hematocrit (Hct) levels were measured, along with plasma levels of glucose, insulin, cholesterol, HDL-cholesterol, triglyceride (TG), and free fatty acids (FFA). Aminotransferase activities were also analyzed. The non-diabetic rats gained weight, while the diabetic rats lost weight - except in the Ac-Se group, which maintained their initial weight. The blood glucose levels of the Ac group and the Se group were significantly lower than for the diabetic-control group. The plasma triglyceride levels were lowered when both Ac and Se were administered to diabetic rats. The concentrations of plasma FFA in the Ac-Se group were significantly lower compared with the diabetic-control group. Plasma cholesterol levels and alanine aminotransferase activity in the Ac, Ac-Se, and Se groups were significantly lower when compared with the diabetic-control group. Aspartate aminotransferase activity was significantly lower in the Se group compared to the other diabetic groups. These data show that treatment with a butanol fraction of Ac in combination with Se has no synergistic effect. Plasma glucose levels tended to be low when Se was administered to diabetic rats. Supplementation of Se in diabetic rats did not elicit a significant increase in plasma insulin levels or result in hypolipemic effects.

Formulation and Quality Evaluation of Chicken Nuggets Supplemented with Beef and Chicken Livers

  • This study explores the potential of utilizing meat byproducts, specifically chicken and beef liver, to enhance the nutritional value of processed foods like chicken nuggets. Proximate analysis was conducted on the livers, including moisture, ash, fat, and protein content, and degradation potential was observed. Antioxidant potential was analyzed through 2,2-diphenyl-1-picrylhydrazyl (DPPH). The total phenolic content (TPC), oxidative stability through peroxide value (POV), and free fatty acid (FFA) were performed to evaluate quality changes during seven-day storage. The radical scavenging activity showed that beef liver has excellent antioxidant capacity (61.55%- and 195.89- mM gallic acid equivalent for DPPH and TPC, respectively) compared to chicken liver and significantly increased the antioxidant potential of nuggets by 5%-10%. POV and FFA values increased with increased storage days for the liver and its incorporation in nuggets. However, the values remained under the 10 meq/kg threshold. Incorporating the livers into chicken nuggets led to a significant (p=0.000) improvement in nutritional content, particularly a 1.5%-2% increase in protein, with a similar increase in mineral content. Texture and sensory evaluations indicated favorable consumer acceptability for liver-enriched nuggets. Overall, this research shows the value of adding liver as a functional ingredient to enhance the nutritional profile of processed foods.

Effects of Allium victorials Extract on Lowing Lipid, Anti-oxidation and Concentration of Inflammatory Mediators in Rats Fed High Oxidized Fat (산마늘추출물이 과산화지질급여 비만쥐의 지질강하, 항산화효과 및 염증매개물질의 생산에 미치는 영향)

  • This study was carried out to effects of Allium victorials extract on lowering lipid, anti-oxidation and concentration of inflammatory mediators in rats fed high oxidized fat. Concentration of free fatty acid(FFA), triglyceride(TG), total cholesterol and LDL-cholesterol in plasma decreased in the Allium victorials extract groups and plasma HDL-cholesterol concentration revealed a tendency to increase in Allium victorials extract groups. Concentration of total cholesterol and TG in liver showed a tendency to decrease in Allium victorials extract groups. Concentration of thiobarbituric acid(TBARS) in plasma and liver showed a lower values in Allium victorials extract groups than that of control group. Activities of glutathione peroxidase(GSH-Px), superoxide dismutase(SOD) and catalase(CAT) in liver showed a tendency to increase in Allium victorials extract groups. Concentration of nitrogen oxide(NO), ceruloplasmin and ${\alpha}1$-acid glycoprotein in plasma showed a lower values in Allium victorials extract groups than that of control group. These results indicate that the Allium victorials extract have an functional material for lowering lipid, anti-oxidation and anti-inflammatory effect.

Influence of Total Abdominal Fat Accumulation on Serum Lipids and Lipoproteins in Korean middle-aged men (한국 중년 남성에서 복부지방 축적이 혈청 지질 및 지단백 농도에 미치는 영향)

  • Anthropometry, computed tomography(CT) at the umbilical level, nutrient intake, blood pressure, serum levels of lipids and lipoproteins and response of glucose, c-peptide, insulin, and free fatty acid(FFA) during oral glucose tolerance test(OGTT) were estimated on 11 normal-weight controls and 35 overweight and obese middle-agd men. The areas of total abdominal, subcutaneous and visceral were determined by CT scanning technique. Total abdominal fat area correlated the most significantly with the levels of serum lipids, lipoproteins and insulin among several obese indices. Compared with normal-weight controls, overweight and obese men with abdominal fat lower than 29000$\textrm{mm}^2$ showed an increase in waist-hip ratio, areas of total abdominal(35%), visceral and subcutaneous fat and C-peptide response area during OGTT, though age, percent ideal body weight, body mass index, % body fat, and all biochemical indices except C-peptide response area were not different between two groups. Overweight and obese men with abdominal fat greater than 29000$\textrm{mm}^2$ showed a higher values in total abdominal fat(85%), serum levels of triglyceride, total-and LDL-cholesterol, the ratio of LDL-to HDL-cholesterol, and response areas of FFA, insulin and C-peptide during OGTT than normal-weight controls. Overweight and obese men with great abdominal fat showed an increase in alcohol ingestion and percent calorie intake per total energy expenditure, compared with normal-weight controls. Our results indicate that obesity and a certain level of total abdominal fat accumulation is required to observe abnormal levels of serum lipids, lipoproteins and insulin in Korean middle-aged men. In addition, increased alcohol and calorie intake and decreased physical activity could partly explain total abdominal fat accumulation in men.

Cordyceps militaris alleviates non-alcoholic fatty liver disease in ob/ob mice

  • BACKGROUND/OBJECTIVES: Non-alcoholic fatty liver disease (NAFLD) is becoming an important public health problem as metabolic syndrome and type 2 diabetes have become epidemic. In this study we investigated the protective effect of Cordyceps militaris (C. militaris) against NAFLD in an obese mouse model. MATERIALS/METHODS: Four-week-old male ob/ob mice were fed an AIN-93G diet or a diet containing 1% C. militaris water extract for 10 weeks after 1 week of adaptation. Serum glucose, insulin, free fatty acid (FFA), alanine transaminase (ALT), and proinflammatory cytokines were measured. Hepatic levels of lipids, glutathione (GSH), and lipid peroxide were determined. RESULTS: Consumption of C. militaris significantly decreased serum glucose, as well as homeostasis model assessment for insulin resistance (HOMA-IR), in ob/ob mice. In addition to lowering serum FFA levels, C. militaris also significantly decreased hepatic total lipids and triglyceride contents. Serum ALT activities and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interleukin-6 (IL-6) levels were reduced by C. militaris. Consumption of C. militaris increased hepatic GSH and reduced lipid peroxide levels. CONCLUSIONS: These results indicate that C. militaris can exert protective effects against development of NAFLD, partly by reducing inflammatory cytokines and improving hepatic antioxidant status in ob/ob mice.

Flavor development in cheddar cheese (체다 치즈의 맛의 개발)

  • This study was carried out to find a cholesterol removal rate, flavor development, and bitter amino acid productions in Cheddar cheese treated with -cyclodextrin ($\beta$-CD): l) Control (no homogenization, no $\beta$-CD), and 2) Milk treatment (1000 psi milk homogenization, 1 % $\beta$-CD). The cholesterol removal of the cheese were 79.3%. The production of short-chain free fatty acids (FF A) increased with a ripening time in both control and milk treated cheese. The releasing quantity of short-chain FFA was higher din milk treated cheese than control at 5 and 7 mo ripening. Not much difference was found in neutral volatile compounds production between samples. In bitter-tasted amino acids, milk treatment group produced much higher than control. In sensory analysis, texture score of control Cheddar cheese significantly increased, however, that in cholesterol-reduced cheese decreased dramatically with ripening time.

  • Mahwangsan is used in the treatment of asthma due to wind and cold(風寒喘). This is study was carried out to investigate the effect of Mahwangsan extract and In order to investigate the effects of Chungyeuldodamtang experimental studies were performed in hyperlipidemia Rats induced by cholesterol and Spontaneusly hypertension rats. The contents of serum total cholesterol, triglyceride, phospholipid and FFA and the changes of blood pressure and heart beat were measured. The results were summarized as follows ; 1. The content of Total-Cholesterol in the serum compared with control group tended to be decreased in Chungyeuldodamtang group, but did not show a significance. 2. The content of Triglyceride in the serum was significantly decreased in Chungyeuldodamtang group. 3. The content of Phospholipid in the serum was significantly decreased in Chungyeuldodamtang group. 4. The content of FFA in the serum tended to decreased in Chungyeuldodamtang group, but did not show a significance. 5. The S.H.R's blood pressure was significantly decreased in Chungyeuldodamtang group. 6. The S.H.R.'s Heart beat compared with control group tended to be decreased in Chungyeuldodamtang group, but did not show a significance. According to the above results, it is assumed that Chungyeuldodamtang has a valid effect on Hypertension and Hyperlipidemia.

Effect of CHCl$_3$fraction of Alisma canaliculatum with selenium on the plasma glucose and lipids levels in streptozotocin-induced diabetic rats (택사 CHCl$_3$분획물과 Selenium 보충이 당뇨 흰쥐의 혈당과 지질함량에 미치는 영향)

  • The purpose of this study was to investigate the effect of chloroform(CHC1$_3$) fraction of Alisma canaliculatum All. Braun et Bouche(Ac) with selenium(Se) on the plasma glucose and lipid levels in streptozotocin(STZ)-induced diabetic rats. Male Sprague-Dawley rats were divided into five groups. The normal and diabetic rats were separated into four groups: the STZ-control group, the Ac group, the Ac-Se group and the Se group. Diabetes was induced in the male rats by an injection of STZ into the tail vein at a dose of 45 mg/kg. The CHCl$_3$fraction of Ac(250 mg/kg) was administered orally for 14 days. The supplementation was achieved with the AIN-93 recommended diet by adding 2 mg/kg diet of selenium as Na$_2$SeO$_3$. which was prepared freshly everyday. The body weight, hematocrit(Hct), glucose, insulin, cholesterol, HDL-cholesterol, triglyceride(TG) and free fatty acids(FFA) concentrations in plasma were measured. The aminotransferase activities were also analyzed. The changes of body weight in the experimental groups were not significantly different from that of the STZ-control group, but diabetes hyperphagia accompanied changes with body weight loss in Ac-Se group. The levels of Plasma cholesterol were not significantly different among the experimental groups. The concentrations of FFA in the Ac-Se group increased significantly compared with the STZ-control group. The effect of Se alone significantly increased aspartate aminotransferase activity and alanine aminotransferase activity. The results showed that the treatment of CHCl$_3$fraction of Ac in combination with Se has no synergistic effect. There was a tendency for the plasma glucose levels to decrease when Se was administered into diabetic rats. Supplementation of Se in diabetic rats did not elicit a significant increase in plasma insulin levels and exhibited hypotriglyceridemic effect.

Effects of Pumpkin (Cucurbita moschata Duch.) Leaf Ethanolic Extracts on Lipid Oxidation and Microbial Activity in Refrigerated Raw Ground Pork

  • This study was conducted to evaluate the antioxidant effects of pumpkin leaf extracted using a 50% ethanol on ground pork during storage. The pumpkin leaf extracts were added at concentrations of 0.05 (PE-0.05), 0.1 (PE-0.1), and 0.2% (PE-0.2) to ground pork, and 0.05% of ascorbic acid (As-0.05) was added as a control. Each sample was collected after 1, 4, 7, and 10 d of storage and the pH, total viable counts (TVC), conjugated dienes (CD), free fatty acids (FFA), and thiobarbituric reaction substance (TBARS) values were measured. The pH of the pork samples decreased until day 7, and then increased thereafter, except for the control and PE-0.05 sample. Lower CIE $a^*$ values were observed for pork samples containing PE relative to As-0.05 at increasing storage time (p<0.05). The addition of PE decreased the TVC, CD, FFA and TBARS values levels in the ground pork when compared to the control during 10 d of storage. These results indicate that PE can produce notable effects on meat products, such as inhibiting lipid oxidation and discoloration.

Estimation of Flood Quantile in Ungauged Watersheds for Flood Damage Analysis Based on Flood Index of Natural Flow (미계측 유역의 홍수피해분석을 위한 자연유량의 홍수지표 기반 확률홍수량 산정)

  • In this study, flood quantiles were estimated at ungauged watersheds by adjusting the flood quantiles from the design rainfall - runoff analysis (DRRA) method based on regional frequency analysis. Comparing the flood frequency analysis (FFA) and DRRA, it was found that the flood quantiles estimated by the DRRA method were overestimated by 52%. In addition, a practical method was suggested to make an flood index using natural flows to apply the regional frequency analysis (RFA) to ungauged watersheds. Considering the relationships among DRRA, FFA, and RFA, we derived an adjusting formula that can be applied to estimate flood quantiles at ungauged watersheds. We also employed Leave-One-Out Cross-Validation scheme and skill score to verify the method proposed in this study. As a result, the proposed model increased the accuracy by 23.2% compared to the existing DRRA method.
