Isolation and Genetic Characterization of Protease-Producing Halophilic Bacteria from Fermenting Anchovy (발효중인 멸치액젓에서 분리한 단백질분해효소 생산 호염성 세균의 유전적 특성)

  • Three protease-producing halophilic bacteria were isolated from fermenting anchovy. Isolated FAM 10, FAM 114, and FAM 115 were found to grow optimally at salt concentrations of 2-4%, 10%, and 6%, respectively, and could grow in salinity of up to 18-22%. The salinity conditions for optimum protease production were 6% in FAM 10 and 10% in FAM 114 and FAM 115. The protease activity of FAM 10 was gradually inhibited by the addition of NaCl up to 10%, and was not evident at 14%, whereas FAM 114 and FAM 115 displayed protease activity at 14% NaCl and could not be measured at 18%. These results demonstrated that the three isolated strains belong to protease-producing, moderately halophilic bacteria. Strain FAM 10, FAM 114, and FAM 115 were identified as Salinivibrio sp., Halobacillus sp., and Halobacillus sp. respectively, based on comparative analyses of the 16S rRNA gene and the 16S-23S intergenic space sequence (IGS), biochemical testing, and Gram staining. Salinivibrio sp. FAM 10 had two 16S rDNAs containing different sequences at position 191 and four IGSs that harbored no tRNA gene and tRNA genes for isoleucine, alanine, glutamate, lysine, and/or valine. Halobacillus sp. FAM 114 and FAM 115 had completely identical 16S rRNA gene sequences and showed 99% identity to the sequences of various Halobacillus strains. The three IGSs found in the genome of both strains displayed 99% sequence identity with Halobacillus aidingensis and Halobacillus sp. JM-Hb, and had $IGS^0$ with no tRNA gene and $IGS^{IA}$ with tRNA genes for isoleucine and alanine.

  • FAM92A1 (named FAM92A1-271) belongs to the family of proteins with conserved DUF1208 domains. Its function remains elusive. We identified two novel transcript variants (FAM92A1-251, FAM92A1-289) of FAM92A1. The presence of these transcripts in cancerous and normal cells, as well as their influence on cell prolifera-tion and apoptosis, were investigated. The subcellular location of FAM92A1 was determined by fluorescence microscopy. We found that FAM92A1-271 and FAM92A1-289 were highly expressed in both normal and cancerous cells, but FAM92A1-251 was only expressed at a mo-derate level in both types of cell. Overexpression of FAM92A1-271, FAM92A1-251 and FAM92A1-289 inhibited cell proliferation, caused S-phase arrest and induced apoptosis. Subcellular localization showed that FAM92A1 localizes to the nucleus. Our results show that FAM92A1 has different splicing variants, and that it may take part in regulating cell proliferation and apoptosis.

FAM46B inhibits cell proliferation and cell cycle progression in prostate cancer through ubiquitination of β-catenin

  • FAM46B is a member of the family with sequence similarity 46. Little is known about the expression and functional role (s) of FAM46B in prostate cancer (PC). In this study, the expression of FAM46B expression in The Cancer Genome Atlas, GSE55945, and an independent hospital database was measured by bioinformatics and real-time PCR analysis. After PC cells were transfected with siRNA or a recombinant vector in the absence or presence of a ${\beta}$-catenin signaling inhibitor (XAV-939), the expression levels of FAM46B, C-myc, Cyclin D1, and ${\beta}$-catenin were measured by western blot and realtime PCR. Cell cycle progression and cell proliferation were measured by flow cytometry and the CCK-8 assay. The effects of FAM46B on tumor growth and protein expression in nude mice with PC tumor xenografts were also measured. Our results showed that FAM46B was downregulated but that ${\beta}$-catenin was upregulated in patients with PC. FAM46B silencing promoted cell proliferation and cell cycle progression in PC, which were abrogated by XAV-939. Moreover, FAM46B overexpression inhibited PC cell cycle progression and cell proliferation in vitro and tumor growth in vivo. FAM46B silencing promoted ${\beta}$-catenin protein expression through the inhibition of ${\beta}$-catenin ubiquitination. Our data clearly show that FAM46B inhibits cell proliferation and cell cycle progression in PC through ubiquitination of ${\beta}$-catenin.

Noise-tolerant Image Restoration with Similarity-learned Fuzzy Association Memory

  • In this paper, an improved FAM is proposed by adopting similarity learning in the existing FAM (Fuzzy Associative Memory) used in image restoration. Image restoration refers to the recovery of the latent clean image from its noise-corrupted version. In serious application like face recognition, this process should be noise-tolerant, robust, fast, and scalable. The existing FAM is a simple single layered neural network that can be applied to this domain with its robust fuzzy control but has low capacity problem in real world applications. That similarity measure is implied to the connection strength of the FAM structure to minimize the root mean square error between the recovered and the original image. The efficacy of the proposed algorithm is verified with significant low error magnitude from random noise in our experiment.

THE EFFECT OF Fam83h KNOCKDOWN ON THE AMELOGENIN GENE EXPRESSION IN THE AMELOBLAST CELL LINE (Fam83h 발현 억제에 의한 조법랑세포 Amelogenin 발현 변화)

  • Amelogenesis imperfecta, one of the dental genetic disease, is clinically and genetically complex disease. Amelogenesis imperfecta can be classified into three major categories according to clinical phenotype; hypoplastic, hypomaturation, and hypocalcification. Recently a novel gene, Fam83h, was identified to cause autosomal dominant hypocalcification amelogenesis imperfecta, however its functional role in the pathogenesis of enamel defect is not known yet. So this study was aimed to identify the knockdown effect of Fam83h gene on the amelogenin mRNA expression via shRNA transfection into immortalized ameloblast cell line. The result showed that the knockdown of Fam83h did not influence the amelogenin expression. Further study of the functional role of Fam83h gene should be performed to understand the complex nature of amelogenesis as well as molecular pathogenesis of amelogenesis imperfecta.

Adenovirus vector-mediated FAM176A overexpression induces cell death in human H1299 non-small cell lung cancer cells

  • FAM176A (family with sequence similarity 176 member A) is a novel molecule related to programmed cell death. A decreased expression of FAM176A has been found in several types of human tumors in including lung cancers. In the present study, we investigated the biological activities of FAM176A on the human non-small cell lung cancer cell line H1299 cells. We constructed a recombinant adenovirus 5-FAM176A vector (Ad5-FAM176A) and evaluated the expression and anti-tumor activities in vitro. Cell viability analysis revealed that the adenovirus-mediated increase of FAM176A inhibited the growth of the tumor cells in a dose- and time-dependent manner. This inhibitory effect was mediated by both autophagy and apoptosis that involved caspase activation. In addition, cell cycle analysis suggested that Ad5-FAM176A could induce cell cycle arrest at the G2/M phase, all of which suggested that adenovirus-mediated FAM176A gene transfer might present a new therapeutic approach for lung cancer treatment.

  • This paper is proposed a high Performance speed control of the synchronous reluctance motor through the SV-PWM(Space Vector Pulse Width Modulation) of FAM-PI(Fuzzy Adaptive Mechanism-PI). SV-PWM is controlled using FAM-PI control. SV-PWM can be maximum used maximum do link voltage and is excellent control method due to characteristic to reducing harmonic more than others. Fuzzy control has a advantage which can be robustly controlled. FAM-PI controller is changed fixed gain of PI controller using fuzzy adaptive mechanism(FAM) to match operating condition. The results on a speed controller of IPMSM are presented to show the effectiveness of the proposed gain tuner. And this controller is better than the fixed gains one in terms of robustness, even under great variations of operating conditions and load disturbance.

The Effect of Fam Tour Experience on Brand Equity and Revisit Intention in Rural Area (농촌지역의 팸투어 체험이 브랜드자산 및 재방문의도에 미치는 영향)

  • This study is about the effect of experience activities of fam tours in Korea on brand assets and revisit intention to revisit. In other words, the fam tour experience(feel, relate, sense, think, act) in research hypothesis 1 will have a positive (+) influence on brand assets(recognition, image, perceived quality, loyalty, and exclusive assets). In addition, it was set that the brand asset in Hypothesis 2 would have a positive (+) significant effect on the revisit intention. To verify this, 362 questionnaires were collected from MZ households in the metropolitan area and statistical analysis was conducted. The analysis results are as follows. First, in Hypothesis 1-1, the fam tour experience (feel, relate, act) has a positive (+) effect on awareness, In Hypothesis 1-2, the fam tour experience (relate, sense, think) has the effect of positive (+) on the image, In Hypothesis 1-3, the fam tour experience (feel, relate, sense, think, act) is the influence of perceived quality, In Hypothesis 1-4, the fam tour experience(sense, act) has a positive effect on loyalty, In Hypothesis 1-5, the fam tour experience (feel, relate, sense, think) had a positive (+) effect on exclusive assets. The brand assets(awareness, image, quality, loyalty, and exclusive assets) of the rural fam tour in Hypothesis 2 were significant for revisit intention.

Development of Flood Analysis Module for the Implementation of a Web-Based Flood Management System (웹기반 홍수관리시스템 구현을 위한 홍수분석모듈개발)

  • This study was to develop the flood analysis module (FAM) for implementation of a web-based real-time agricultural flood management system. The FAM was developed to apply for an individual watershed, including agricultural reservoir. This module calculates the flood inflow hydrograph to the reservoir using effective rainfall by NRCS-CN method and unit hydrograph calculated by Clark, SCS, and Nakayasu synthetic unit hydrograph methods, and then perform the reservoir routing by modified Puls method. It was programmed to consider the automatic reservoir operation method (AutoROM) based on flood control water level of reservoir. For a $15.7km^2$ Gyeryong watershed including $472{\times}10^4m^3$ agricultural reservoir, rainfall loss, rainfall excess, peak inflow, total inflow, maximum discharge, and maximum water level for each duration time were compared between the FAM and HEC-HMS (applied SCS and Clark unit hydrograph methods). The FAM results showed entirely consistent for all components with simulated results by HEC-HMS. It means that the applied methods to the FAM were implemented properly.

A Contact Stress Analysis in a FAM Process Using Variational Approximation Procedure (변분근사법을 이용한 FAM 과정의 접촉응력 해석)

  • A variational approximation procedure is introduced to study the contact stresses between a representative asperity and a feature generally happening in superfinishing processes such as FAM. After a description of the model under consideration is presented, a system of governing equation for the model is derived fullowed by the assumptions made in order to make progress in model development. Final computation is made to evaluate contact stresses on an elastic asperity tip in small scale in size and a computer simulation is performed for detailed surface profile variations on a representative feature. Numerical results are presented along with a discussion of the conclusions that can be drawn from this analysis.
