Possible Abnormalities of Chimeric Chicken Caused by the Introduction of Exogenous Genes Into Chicken Embryos via Primordial Germ Cells (PGCs)

  • In chicken, exogenous genes introduced into germinal crescent region (GCR) of the early developmental stage, where primordial germ cells (PGCs) were concentrated, were successfully transferred to the gonads via PGCs. The foreign genes were also confirmed to be successfully incorporated into F1 and F2 generations. We tried to incorporate the exogenous genes into PGCs by lipofection, then the DNA mixture was injected into GCR at stage 3-5 or 9-11 of embryonic development (Hamburger and Hamilton, 1951). The manipulated eggs were incubated, and hatched chicks were reared until sexual maturation. F1 generation was obtained from the DNA-treated chicken (DNA-chicken) mated with normal birds. Furthermore, F2 generation was also obtained from the F1 chicken mated with normal birds. The transfer of introduced foreign genes were confirmed by marker gene detection methods and PCR analysis in the hatched chicks, F1 and F2 generations. However, in our experiments, DNA-chickens showed abnormal characteristics such as low egg production rate, abnormal appearance and decreased number of spermatozoa. In the case of F1 chicken, low egg production and the deterioration of sperm capacity for insemination in male chicken were observed.

  • This paper proposes an dffcient method for test pattern generation.Current test pattern genration systems generate a test vester for fault $F_{i+l}$ independently of the computation previously done for faults F1,F2...,Fi The proposed algorithm generates a test vector for fault $F_{i+l}$ by inheriting the test vector for fault Fi. A new test vector is grnerated from inherited values by gradually changing the inhderited values .The inherited values may partially activate a fauog and propagate the fault signal,Normally,this reduses the number of decision steps and backtracks in the second search.Experimental results for well-Known benchmark circuts show that the proposed algorithm is very efficient with small backtrack kimit;in combination eith other algorithms,it is very efficient for arbitrary backtrack limits.

Stable Transmission and Expression of TPO Transgene up to 10 Generation in the Transgenic Mice (형질전환 생쥐에서 제10세대까지 TPO 유전자의 안정적 전이와 지속적인 발현)

  • The pBT-L transgenic mice carrying human TPO gene in conjunction with bovine $\beta$-casein promoter express human TPO in milk during lactation. In this study, stability of germ line transmission and expression of pBT-L transgene integrated into host chromosome were monitored up to generation F10 of transgenic pBT-L/15 line. When male mouse of generation F8 was crossbred with normal females, approximately half of offsprings (51.3$\pm$18.98%) were identified as transgenic mice. Generation F9 and F 10 mice also showed similar transmission rates (43.8$\pm$18.98% and 71.4$\pm$26.98%, respectively), implying that pBT-L transgene can be transmitted stably up to long term generation in the transgenic mice. Expression levels of human TPO from milk of generation F9 and F10 mice were 1.1$\pm$0.33 mg/ml and 1.1 $\pm$0.45 mg/ml, respectively, which are similar to expression level of generation F2 mice. In conclusion, our results suggest that transgenic animals once established will continuously pass their transgenes to the progeny through the breeding program with the same productivity of human protein in their milk.

Saucerneol F, a New Lignan Isolated from Saururus chinensis, Attenuates Degranulation via Phospholipase Cγ1 Inhibition and Eicosanoid Generation by Suppressing MAP Kinases in Mast Cells

  • During our on-going studies to identify bioactive compounds in medicinal herbs, we found that saucerneol F (SF), a naturally occurring sesquilignan isolated from Saururus chinensis (S. chinensis), showed in vitro anti-inflammatory activity. In this study, we examined the effects of SF on the generation of 5-lipoxygenase (5-LO) dependent leukotriene $C_4$ ($LTC_4$), cyclooxygenase-2 (COX-2) dependent prostaglandin $D_2$ ($PGD_2$), and on phospholipase $C{\gamma}1$ ($PLC{\gamma}1$)-mediated degranulation in SCF-induced mouse bone marrow-derived mast cells (BMMCs). SF inhibited eicosanoid ($PGD_2$ and $LTC_4$) generation and degranulation dose-dependently. To identify the molecular mechanisms underlying the inhibition of eicosanoid generation and degranulation by SF, we examined the effects of SF on the phosphorylation of $PLC{\gamma}1$, intracellular $Ca^{2+}$ influx, the translocation of cytosolic phospholipase $A_2$ ($cPLA_2$) and 5-LO, and on the phosphorylation of MAP kinases (MAPKs). SF was found to reduce intracellular $Ca^{2+}$ influx by inhibiting $PLC{\gamma}1$ phosphorylation and suppressing the nuclear translocations of $cPLA_2$ and 5-LO via the phosphorylations of MAPKs, including extracellular signal-regulated protein kinase-1/2 (ERK1/2), c-Jun N-terminal kinase (JNK), and p38. Taken together, these results suggest that SF may be useful for regulating mast cell-mediated inflammatory responses by inhibiting degranulation and eicosanoid generation.

Transmission and Death Rates in Transgenic Mice Containing Growth Hormone Receptor Gene (성장호르몬수용체 유전자를 지닌 형질전환생쥐의 세대전달율 및 치사율)

  • To study the signaling effect of growth hormone (GH) in vivo on animal physiology, transgenic mice containing GH Receptor (GHR) gene fused to metallothionein promoter were produced by DNA microinjection into one-cell stage embryos. Three founder mice were produced with transgenic mice with approximately 4~6 copies of GHR genes and transgene was transmitted into the progeny. The founder mice were mated with normal mice to produce F$_1$ mice, and intergation and transmission of transgene were checked by polymerase chain reaction and Southern blot methods. Transmission rate of GHR transgenic mice were 20~50% in F$_1$ generation and 50% in F$_2$ generation which means that some founder mice were mosaic and transgene in F$_1$ mice was transmitted to F$_2$ progeny with Mendelian ratio. Death rate of GHR transgenic mice after birth was about 10~30% in F$_1$ and F$_2$ progenies indicating that GHR gene may affect death of transgnenic progeny.

  • More than 90 percent of domestic consumption of the lettuce occupied leaf lettuce, and the farmer preferred high yield, late bolting and stable expression of anthocyanins. So, this research focused on making resistant to disease, late bolting, and good expression of anthocyanin for lettuce. Based on these characteristic's parents, 11 crosses were created. So we advanced the generation and investigated each generation from F1 to F7. In this year, we selected 90 combination 221 lines which was F2 42 combination 137 lines, F3 4 combination 17 lines, F4 11 combination 27 lines, F5 7 combination 30 lines, F6 1 combination 2 lines, F7 3 combination 8 lines. Among the selected 17 lines, 5007, 5014, 6004, 6005, 6021, 6023, 7001, 7002, 7006, 7007, 7010, 7011, 7015, 7018, 7019, and 7021 which were late bolting and good expression of anthocyanins were investigated their yield. Finally, we selected 5 lines 7001, 7002, 7011, 7018 and 7022 in this year.

  • The purpose of this study was to evaluate the histological toxicity of chitosanoligosaccharide on the rat. A healthy male of Wistar rat that weighted $250{\pm}350g$ was used for experiment The experimental group was divided into five groups. Group 1 was control group which treated with general food Group 2 was $F_1$ generation which was born by mating of 0.1% (1 mg/ml) chitosanoligosaccharide was supplied by feeding ad libitum for 30 days. Group 3 was $F_2$ generation which was born by mating $F_1$ generation. Group 4 was treated with 90 days of 0.1% (1 mg/ml) chitosan oligosaccharide. Group 5 was treated with 365 days of 0.1% (1 mg/ml) chitosanoligo saccharide. All experimental groups were used to 10 rat. The results were as follow: The RER dilation was observed Group 4. However, there were no significantly changes of ultrastructures in the other groups compared to the control. It was concluded that chitosanoligosaccharide can be used for nontoxic natural material.

High-level Expression and Characterization of the Human Interleukin-10 in the Milk of Transgenic Mice

  • Interleukin-10 (IL-10) is a homodimeric protein with a wide spectrum of anti-inflammatory and immune activities. It inhibits cytokine production and expression of immune surface molecules in various cell types. The transgenic mice carrying the human IL-10 gene in conjunction with the bovine $\beta$-casein promoter produced the human IL-10 in milk during lactation. Transgenic mice were generated using a standard method as described previously. To screen transgenic mice, PCR was carried out using chromosomal DNA extracted from tail or toe tissues with a primer set. In this study, stability of germ line transmission and expression of IL-10 gene integrated into host chromosome were monitored up to generation F15 of a transgenic line. When female mouse of generation F9 was crossbred with normal male, generation F9 to F15 mice showed similar transmission rates (66.0$\pm$20.13%, 61.5$\pm$16.66%, 41.1$\pm$8.40%, 40.7$\pm$20.34%, 61.3$\pm$10.75%, 49.2$\pm$18.82%, and 43.8$\pm$25.91%, respectively), implying that the IL-10 gene can be transmitted stably up to long term generation in the transgenic mice. For ELISA analysis, IL-10 expression levels were determined with an hIL-10 ELISA and a mIL-10 ELISA kit in accordance with the supplier's protocol. Expression levels of human IL-10 from milk of generation F9 to F13 mice were 3.6$\pm$1.20 mg/ml, 4.2$\pm$0.93 mg/ml, 5.7$\pm$1.46 mg/ml, 6.3$\pm$3.46 mg/ml, and 6.8$\pm$4.52 mg/ml, respectively. These expression levels are higher than in generation F1 (1.6 mg/ml) mice. We concluded that transgenic mice faithfully passed the transgene on their progeny and successively secreted target proteins into their milk through several generations, although there was a little fluctuation in the transmission frequency and expression level between the generations.

Yield Reductions in the $F_2$ and $F_3$ Generations of Highly Promising Varietal Crosses of Corn (유망시되는 옥수수 품종간교잡의 $F_2$$F_3$에 있어서의 수량감소)

In order to evaluate the performance of the $F_2$ and $F_3$ generations of $F_1$ varietal crosses which in past trials yielded better than or as high as the recommended double-cross hybrids, eleven yellow and five white-endosperm $F_1$ varietal crosses, their $F_2$ and $F_3$ generations, and the parental open-pollinated varieties together with the standard double-cross hybrids were tested in performance trials in the 1963 wet and 1963-64 dry seasons. The former trials, however, failed due to a strong typhoon that damaged the crop. 2. Net hybrid vigor was highly manifested in grain yield in the $F_1$ varietal crosses, which on the average was 39.9 per cent of the mid-parent yields. Reduction in heterosis was 27.2 per cent in the $F_2$and 30.3 per cent in the $F_3$ generation, respectively. 3. A fairly high degree of reduction in yield occurred in the $F_2$ and $F_3$ generation. On the average, the $F_2$ generation decreased by 17.6 per cent and the $F_3$ by 20.7 per cent. The losses in two generations were exhibited in two levels; four crosses decreased by 10.9 per cent and twelve combinations by 22.0 per cent. The advanced generations of one yellow cross, Cuban Yellow Flint ${\times}$ Hawaiian Flinty Dent, and three white flint crosses, Bicol White Flint ${\times}$ Eto Blanco, College White Flint ${\times}$ Eto Blanco and Bicol White Flint ${\times}$ Nariono 330b, yielded as high as Philippine Hybrid No.1 and Philippine Hybrid No.4, respectively. 4. No significant differences in yield were obtained between the $F_2$ and $F_3$ populations, showing that genetic equilibrium was reached in $F_2$, as may be expected in view of the Hardy-Weinberg law on panmixis. 5. As in past trials, Hawaiian Flinty Dent, a variety, yielded as high as Philippine Hybrid No. 1.

  • It is generally known that the automotive road wheel involves the non-proportional multiaxial loading condition, therefore the measuring dynamic stress and strain in driving state is very important to predict an endurance characteristic of the automotive road wheel. In this study, the ultimate driving test using F1 circuit with respect to 2 kinds of velocity conditions have been carried out in order to measure dynamic stress, strain of the wheel and acceleration of a vehicle. Based on the measured results, the characteristics of dynamic stress generation have been analyzed, and factors which have effect on the dynamic stress generation have been studied.
