A review of forest trees micropropagation and its current status in Korea (국내 임목류 기내증식 연구현황 및 전망)

  • Plant micropropagation techniques include bud cultures using apical or axillary buds, organogenesis through callus culture or adventitious bud induction, and somatic embryogenesis. In Korea Forest Research Institute (KFRI), the first tissue culture trial in woody plant was initiated from the bud culture of hybrid poplars (Populus alba x P. glandulosa) in 1978. Since then several mass propagation techniques have developed from conifer and hardwood species, resulting in allowing practical application to Poplars, Birches and some oak species. In addition, useful micropropagation and genetic resources conservation techniques were established in some rare and endangered tree species including Abeliophyllum distichum. Among various in vitro propagation techniques, somatic embryogenesis is known to be the most efficient plant regeneration system. Since the first somatic embryo induction was reported in Tilia amurensis by KFRI in 1986, various protocols for direct or indirect somatic embryogenesis systems have developed in conifer and hardwood species including Larix leptolepis, Pinus rigida x P. taeda F1, Kalopanax septemlobus and Liliodendron tulipifera, etc. However, most of these technologies have been developed using juvenile tissues, i.e. immature zygotic embryos or mature embryos. Therefore it has been difficult to directly application to tree breeding program due to their unproven genetic background. Recently remarkable progresses and new approaches have been achieved in mature tree somatic embryogenesis. In this article we reviewed several micropropagation techniques, which have been mainly developed by KFRI and recent international progresses.

Comparison of Multi-Label U-Net and Mask R-CNN for panoramic radiograph segmentation to detect periodontitis

  • Purpose: Periodontitis, the most prevalent chronic inflammatory condition affecting teeth-supporting tissues, is diagnosed and classified through clinical and radiographic examinations. The staging of periodontitis using panoramic radiographs provides information for designing computer-assisted diagnostic systems. Performing image segmentation in periodontitis is required for image processing in diagnostic applications. This study evaluated image segmentation for periodontitis staging based on deep learning approaches. Materials and Methods: Multi-Label U-Net and Mask R-CNN models were compared for image segmentation to detect periodontitis using 100 digital panoramic radiographs. Normal conditions and 4 stages of periodontitis were annotated on these panoramic radiographs. A total of 1100 original and augmented images were then randomly divided into a training (75%) dataset to produce segmentation models and a testing (25%) dataset to determine the evaluation metrics of the segmentation models. Results: The performance of the segmentation models against the radiographic diagnosis of periodontitis conducted by a dentist was described by evaluation metrics(i.e., dice coefficient and intersection-over-union [IoU] score). MultiLabel U-Net achieved a dice coefficient of 0.96 and an IoU score of 0.97. Meanwhile, Mask R-CNN attained a dice coefficient of 0.87 and an IoU score of 0.74. U-Net showed the characteristic of semantic segmentation, and Mask R-CNN performed instance segmentation with accuracy, precision, recall, and F1-score values of 95%, 85.6%, 88.2%, and 86.6%, respectively. Conclusion: Multi-Label U-Net produced superior image segmentation to that of Mask R-CNN. The authors recommend integrating it with other techniques to develop hybrid models for automatic periodontitis detection.

Genetic Analysis of Complementary Gene Interactions of Pb and Pp Genes for the Purple Pericarp Trait in Rice (흑미의 자색종자과피(Purple pericarp) 형질을 결정하는 상보적 유전자 Pb와 Pp 유전자들의 상호관계 분석)

  • The Purple pericarp (Prp) trait is a trait often bred for in black rice. Generally, the Prp trait is displayed in the color variations of seeds following the 9:3:4 purple, brown, and white ratio, respectively. The Prp trait is a recessive epistasis of two gene interactions; however, it is caused by the two complementation genes Pb and Pp. Here we present a study of the genetic characteristics of the Prp trait using an $F_1$ hybrid with a Pbpb Pppp genotype. This hybrid generated four seed colors with the following numbers: 3 dark purple, 6 medium purple, 3 brown, and 4 white (or 9 purple, 3 brown, and 4 white). However, further biochemical analysis of the all progenies divided them into two groups. One group had the Pb_ Pp_ allelic constitutions and contained cyanidin 3-O-glucoside (C3G) in both the dark purple or medium purple seeds. The other group, however, was absent of C3G in both the brown and white seeds, resulting in a ratio of 9:7, respectively. This segregation revealed the extended Mendelian 9:7 ratios of the complementary gene interactions with a good fitness in ${\chi}^2$ analysis. Further analysis revealed that brown seeds with the Pb_ pppp genotype corresponded with a null C3G, indicating that the Brown pericarp trait in rice is caused by a dominant allele of the Pb gene. Therefore, we conclude that the production of C3G is a main phenotype of the black and purple colored rice in the Prp trait, and it is governed by the complementary gene interactions between Pb and Pp genes.

Breeding of (춘잠기 강건 다수성 새로운 누에품종 )

  • A new silkworm variety "DaeBakJam" for spring rearing season is F1 hybrid between Jam155, a japanese race bred from introduction breeding and Jam156, a chinese race from introduction breeding. In the local adaptability test performed at 8 local areas in spring of 2010, the hatchability rate of DaeBakJam was recorded 96% similar to DaePoongJam. The larval period was 3 hour shorter than DaePoongJam. The pupation percentage was recorded 0.2% higher than DaePoongJam. Single cocoon weight (2.72 g) and Cocoon yield (25.4 kg) was higher than DaePoongJam. The concentration of DNJ was measured 3.28 mg in DaePoongJam, lower than DaePoongJam (3.48 mg). Test of the artificial diet of DaeBakJam showed bad adaptability of the artificial diet for larval period from 1st to 3rd, but KumOkJam showed excellent adaptability of the artificial diet. In the Paecilomyces tenuipes production ability test, the pupal weight of DaeBakJam was 1.69 g/individual, higher than DaePoongJam (1.59 g/individual).

Breeding of WhangBoJama Sex-Limited Yellow Silk Silkworm Variety Suitable for Spring Rearing Season (춘잠기 강건 다수성 한성황견 누에품종 "황보잠" 육성)

  • A new silkworm variety "WhangBoJam" for spring rearing season is F1 hybrid between Jam317, a japanese strain bred from introduction breeding and Jam318, a chinese strain from introduction breeding. In the local adaptability test performed at 8 local areas in spring of 2012, the hatchability rate of WhangBoJam was recorded 95% similar to Kum HwangJam. The larval period was 8hours more long than KumHwangJam. The pupation percentage was recorded 6.6% higher than KumHwangJam. Single cocoon weight was recorded 2.29 g similar to KumHwangJam and cocoon yield(21.3 kg) was higher than KumHwangJam. The concentration of DNJ was measured to 0.22% in WhangBoJam, lower than DaePoongJam(0.26%). In the Paecilomyces tenuipes production ability test, the pupal weight of Whang BoJam was 1.33 g/individual, higher than KumHwangJam (1.19 g/individual).

Effects of Permeable Cryoprotectants on Viability of Mammalian Embryo Model (침투성 동결보호제가 포유류 초기배자의 생존성에 미치는 영향)

  • The objective of this study was to evaluate the toxicities of permeable cryoprotectants and finally to establish the cryopreservation method of surplus embryos obtained during assisted reproductive technology (ART). Toxicities of permeable cryoprotectants, dimethyl sulfoxide (DMSO), ethylene glycol (EG), Glycerol, and 1,2-PROH were investigated using a murine embryo model. Female $F-{_1}$ mice were stimulated with gonadotropin, induced ovulation with hCG and mated. Two cell embryos were collected and cultured after exposure to among DMSO, EG, Glycerol, and 1,2-PROH. Embryo development was evaluated up to the blastocyst stage. The total cell count of blastocysts that were treated with DMSO and Glycerol at the 2-cell stage was significantly lower than that were treated with EG ($81.1{\pm}15.1$), 1,2-PROH ($88.0{\pm}21.1$) or the control ($99.9{\pm}21.3$) (p<0.001). On comparison of four cryoprotectant treated groups, the DMSO and Glycerol treated group showed a decreased cell count compared with the EG and 1,2-PROH treated group (p<0.05). Both DMSO ($14.7{\pm}1.3$), EG ($12.1{\pm}1.1$), Glycerol ($15.2{\pm}1.8$), and 1,2-PROH ($11.5{\pm}1.3$) treated groups showed higher apoptosis rates of cells in the blastocyst compared with the control ($6.5{\pm}0.7$, p<0.0001). In addition, the DMSO or Glycerol treated group showed more apoptotic cells than the EG or 1,2-PROH treated group (p<0.001). The potential toxicity of cryoprotectants was uncovered by prolonged exposure of murine embryos to among DMSO, EG, Glycerol, and 1,2-PROH at room temperature. When comparing four permeable cryoprotective agents, EG and 1,2-PROH appeared to be less toxic than DMSO and Glycerol at least in a murine embryo model.

Tolerance of Corn, Sorghum, Sorghum-Sudangrass Hybrid, and Pearl Millet to Simazine and Alachlor (옥수수, 수수, 수수-수단그라스 교잡종 및 진주조의 Simazine과 Alachlor에 대한 저항성)

  • In 1988 the tolerance of corn, sorghum, sorghum-sudangrass. and pearl millet hybrids to simazine and alachlor was tested in field during the growing season and pots during the summer and fall. In field and summer pot experiments(Exp.) the above mentioned four crops were tested at the ratios of simazine WP (50% ai, g/10a) ; alachlor EC (43.7% ai. ml/10a) of 130: 0, 100: 0, 70: 200, 0: 300 and 0: 400 and a sorghum hybrid was tested at 0, 50, 100, 200, 300, and 400ml/10a of alachlor and 70g/10a of simazine+ 200ml/10a of alachlor in fall pot Exp. In corn emergence rate, percent stand, plant height of seedlings, and dry matter(DM) yield were not affected by simazine and alachlor in all Exps. In sorghum and sorghum-sudangrass early growth and DM yield were not affected by simazine and alachlor in field Exp. In contrast, simazine reduced height and dry weight of seedlings slightly without any deterimental effects on emergence and survival rates. but alachlor reduced survival rate, plant height, and dry weight of seedlings significantly in summer pot Exp. In fall Exp. alachlor did not affect emergence rate of a sorghum hybrid, but survival rate, plant height, and dry weight of seedlings reduced with increased levels of alachlor when applied higher than 100ml/10a. In pearl millet simazine did not affect emergence rate, plant height, and DM yield in field, but reduced survival rate, plant height, and dry weight of seedlings in summer pot Exp. However, alachlor reduced DM yield significantly due to a lower percent stand even in the field. In summer pot Exp. although emeregence rate was slightly reduced, all seedlings were dead after emergence. Simazine did not control grasses such as Digitaria sanguinalis, Setaria viridis, Echinochloa crusgalli effectively, but controlled broadleaf weeds. Alachlor controlled all grasses, Porluraca oleracea, and Amaranthus mangostanus, but did not control Acalypha australis and Chenopodium album. A combination of simazine and alachlor controlled weeds more effectively than either simazine or alachlor alone.

  • A new silkworm variety "Daepoongjam" for spring rearing season is $F_1$ hybrid between Jam151, a Japanese race bred from introduction breeding and Jam152, a Chinese race from introduction breeding. In the local adaptatability test performed at 8local areas in spring of 2005, the heavy cocoon yield of "Daepoongjam" was recorded 6% higher than "Kumokjam". The concentration of Deokynojirimycin (DNJ) was measured 4.22 mg in "Daepoongjam", similar to "Kumokjam" (4.26 mg). In the Peacilomyces tenuipes production ability test, the pupal weight of "Daepoongjam" was 1.61 g/individual 13% heavier than "Kumokjam" (1.41 g/individual)

Genetic Relationships among the Parental Bombyx mori Strains of the Current F$_1$ Hybrid Silkworm based on RAPD (RAPD를 이용한 장려누에품종의 원종간 유전적 유연관계)

  • The genetic relationships among the twenty parental silkworm, Bombyx mori strains authorized in Korea were evaluated using RAPDs-PCR(Random Amplified Polymorphic DNAs-Polymerase Chain Reaction). Twenty-six different 10-mer oligonucleotide primers were used to screen genetic characteristics of parental twenty silkworm strains by RAPD-PCR analysis. 24 primers showed different banding patterns among the strains. Based on these RAPD patterns, the genetic relationships among the silkworm strains were analyzed by UPGMA(Unweighed Pair-Group Method with Arithmetic average) method. The phylogenetic relationships in the twenty silkworm strains were classified into two major sub-groups at the genetic similarity coefficient of 0.60. The first sub-group included Jaml13, Jaml 19, Jaml20, Jam123, Jam1 25 and Jam 127. Jamll4, Jam1 2 I, Jam 122, Jam 124, Jam1 26, Jam 128, Jam129, Jam 130, Jam 131, Jam1 32, Jam133, Jam134, Jam301 and Jam302 were included in the 2nd group. The genetic distance values among Jam1 14, Jam120 and Jam127 were lower than those among the other strains, while Jam129 is very closely related to Jam131 as showing coefficient value of 1 .O.

Breeding of SooOkJam an Artificial Diet Adaptable Silkworm Variety, for Spring Rearing Season (애누에 인공사료 적합성 봄누에 품종 )

  • A new silkworm variety SooOkJam for spring rearing season is F1 hybrid between Jam157, a japanese race bred from introduction breeding and Jam158, a chinese race from introduction breeding. In the local adaptability test performed at 8 local areas in spring of 2011, the hatchability rate of SooOkJam was recorded 2% higher than the authorized silkworm variety, DaePoongJam. The larval period was recorded 12 hour shorter than DaePoongJam. The pupation percentage was recorded 0.8% higher than DaePoongJam. Single cocoon weight (2.40 g), Cocoon yield (22.6 kg) and best cocoon rate (95.5%) was similar to DaePoongJam. The concentration of DNJ was measured 4.12 mg in SooOkJam, lower than DaePoongJam (4.45 mg). Test of the artificial diet of SooOkJam showed excellent adaptability of the artificial diet for larval period from 1st to 3rd, similar to KumOkJam. In the Paecilomyces tenuipes production ability test, the pupal weight of SooOkJam was 1.61 g/individual, similar to DaePoongJam (1.65 g/individual).
