  • Partial dominance was exhibited by flowering(heading), panicle length, panicle numbers, $F_1$ag leaf length, 1000 kernel weight in $F_1$ and $F_2$ hybrids by 7 \times 7 and 5 \times 5 diallel crosses. Over dominance was exhibited by culm length, flag leaf width, appearance degree of panicle in $F_1$ generation of the crosses, and also over dominance was exhibited by kernel weight in $F_2$ generation of the crosses.

High-level Expression and Characterization of the Human Interleukin-10 in the Milk of Transgenic Mice

  • Interleukin-10 (IL-10) is a homodimeric protein with a wide spectrum of anti-inflammatory and immune activities. It inhibits cytokine production and expression of immune surface molecules in various cell types. The transgenic mice carrying the human IL-10 gene in conjunction with the bovine $\beta$-casein promoter produced the human IL-10 in milk during lactation. Transgenic mice were generated using a standard method as described previously. To screen transgenic mice, PCR was carried out using chromosomal DNA extracted from tail or toe tissues with a primer set. In this study, stability of germ line transmission and expression of IL-10 gene integrated into host chromosome were monitored up to generation F15 of a transgenic line. When female mouse of generation F9 was crossbred with normal male, generation F9 to F15 mice showed similar transmission rates (66.0$\pm$20.13%, 61.5$\pm$16.66%, 41.1$\pm$8.40%, 40.7$\pm$20.34%, 61.3$\pm$10.75%, 49.2$\pm$18.82%, and 43.8$\pm$25.91%, respectively), implying that the IL-10 gene can be transmitted stably up to long term generation in the transgenic mice. For ELISA analysis, IL-10 expression levels were determined with an hIL-10 ELISA and a mIL-10 ELISA kit in accordance with the supplier's protocol. Expression levels of human IL-10 from milk of generation F9 to F13 mice were 3.6$\pm$1.20 mg/ml, 4.2$\pm$0.93 mg/ml, 5.7$\pm$1.46 mg/ml, 6.3$\pm$3.46 mg/ml, and 6.8$\pm$4.52 mg/ml, respectively. These expression levels are higher than in generation F1 (1.6 mg/ml) mice. We concluded that transgenic mice faithfully passed the transgene on their progeny and successively secreted target proteins into their milk through several generations, although there was a little fluctuation in the transmission frequency and expression level between the generations.

  • We present a microscopic description for generation of 1/f noise in interfaces between cobalt thin film and silicon substrate. Along with surface resistance measurements and transmission electron diffraction observations. 1/f noise power spectral density has been measured for the interfacial structures at the liquid nitrogen temperature . The cobalt films have been deposited by the electron-beam evaporation technique onto p-type (100) silicon in the high vacuum condition. The measured noise power spectral density shows highest magnitude near the structural transition and metallization transition region. The noise magnitude rapidly decreased after the cobalt silicide nucleation. The noise parameter is concluded to be originated form the structural fluctuations.

  • Korean agriculture is currently experiencing difficulties as a cause of rural depopulation, aging of rural population, grain self-sufficiency rate decline, and deepening of climate change. It is necessary to ensure our country's agriculture industrial competitiveness in accordance with opening of FTA imports expanded. To ensure the underdeveloped competitive, Korean government defines the 3rd generation model from 1st generation model to extend the smart farms of Korean types. The agriculture smarting overcomes the growth limitations of agriculture, and efforts to develop 6th + ${\alpha}$ industry. In this paper, We define and verify the IoF(Internet of Farming)-Cloud based substantial services about 2rd generation model, and propose a greenhouse of IoF-Cloud testbed.

The increased GUS gene inactivation over generation in Arabidopsis transgenic lines (애기장대 형질전환 식물체의 세대경과에 따른 GUS유전자의 비활성화에 관한 연구)

  • The effect of transgene inactivation in T2, T3 and F2 generations was analyzed in progeny seedlings which had been generated by Agrobacterium (LBA4404/pBI121)-mediated transformation in Arabidopsis thaliana. In a system which investigated in the expression of $\beta$-glucuronidase(GUS)gene in kanamycin-resistant (ke $n^{R}$)seedlings, GUS inactivated seedlings were observed in 5 of 12 tested lines of T2 generation and the frequency of GUS inactivation was approximately 2.3%. Lines with multi-copies of T-DNA exhibited severe GUS gene inactivation with the frequency of 5.8% in T2 generation. In T3 generation lines exhibited GUS gene inactivation with the frequency of 1.3%. In contrast, inactivation increased dramatically up to 12.6% in multi-copy T-DNA line. A similar phenomenon was also found in F2 progeny from a transgenic line which had been crossed with wild-type Arabidopsis plant, WS-O (GUS gene inactivation frequency 9.9%). These results indicate that the foreign gene introduced into the plant was inactivated progressively in its transmission during subsequent generations and the transgenic line with multi-copies of T-DNA tended to show more increased inactivation.

Transmission and Death Rates in Transgenic Mice Containing Growth Hormone Receptor Gene (성장호르몬수용체 유전자를 지닌 형질전환생쥐의 세대전달율 및 치사율)

  • To study the signaling effect of growth hormone (GH) in vivo on animal physiology, transgenic mice containing GH Receptor (GHR) gene fused to metallothionein promoter were produced by DNA microinjection into one-cell stage embryos. Three founder mice were produced with transgenic mice with approximately 4~6 copies of GHR genes and transgene was transmitted into the progeny. The founder mice were mated with normal mice to produce F$_1$ mice, and intergation and transmission of transgene were checked by polymerase chain reaction and Southern blot methods. Transmission rate of GHR transgenic mice were 20~50% in F$_1$ generation and 50% in F$_2$ generation which means that some founder mice were mosaic and transgene in F$_1$ mice was transmitted to F$_2$ progeny with Mendelian ratio. Death rate of GHR transgenic mice after birth was about 10~30% in F$_1$ and F$_2$ progenies indicating that GHR gene may affect death of transgnenic progeny.

  • More than 90 percent of domestic consumption of the lettuce occupied leaf lettuce, and the farmer preferred high yield, late bolting and stable expression of anthocyanins. So, this research focused on making resistant to disease, late bolting, and good expression of anthocyanin for lettuce. Based on these characteristic's parents, 11 crosses were created. So we advanced the generation and investigated each generation from F1 to F7. In this year, we selected 90 combination 221 lines which was F2 42 combination 137 lines, F3 4 combination 17 lines, F4 11 combination 27 lines, F5 7 combination 30 lines, F6 1 combination 2 lines, F7 3 combination 8 lines. Among the selected 17 lines, 5007, 5014, 6004, 6005, 6021, 6023, 7001, 7002, 7006, 7007, 7010, 7011, 7015, 7018, 7019, and 7021 which were late bolting and good expression of anthocyanins were investigated their yield. Finally, we selected 5 lines 7001, 7002, 7011, 7018 and 7022 in this year.

Genetic diversity evolution in the Mexican Charolais cattle population

  • Objective: The aim was to characterize the genetic diversity evolution of the registered Mexican Charolais cattle population by pedigree analysis. Methods: Data consisted of 331,390 pedigree records of animals born from 1934 to 2018. Average complete generation equivalent, generation interval, effective population size (Ne), and effective numbers of founders (fe), ancestors (fa), and founder genomes (Ng) were calculated for seven five-year periods. The inbreeding coefficient was calculated per year of birth, from 1984 to 2018, whereas the gene contribution of the most influential ancestors was calculated for the latter period. Results: Average complete generation equivalent consistently increased across periods, from 4.76, for the first period (1984 through 1988), to 7.86, for the last period (2014 through 2018). The inbreeding coefficient showed a relative steadiness across the last seventeen years, oscillating from 0.0110 to 0.0145. During the last period, the average generation interval for the father-offspring pathways was nearly 1 yr. longer than that of the mother-offspring pathways. The effective population size increased steadily since 1984 (105.0) and until 2013 (237.1), but showed a minor decline from 2013 to 2018 (233.2). The population displayed an increase in the fa since 1984 and until 2008; however, showed a small decrease during the last decade. The effective number of founder genomes increased from 1984 to 2003, but revealed loss of genetic variability during the last fifteen years (from 136.4 to 127.7). The fa:fe ratio suggests that the genetic diversity loss was partially caused by formation of genetic bottlenecks in the pedigree; in addition, the Ng:fa ratio indicates loss of founder alleles due to genetic drift. The most influential ancestor explained 1.8% of the total genetic variability in the progeny born from 2014 to 2018. Conclusion: Inbreeding, Ne, fa, and Ng are rather beyond critical levels; therefore, the current genetic status of the population is not at risk.

  • This paper proposes an dffcient method for test pattern generation.Current test pattern genration systems generate a test vester for fault $F_{i+l}$ independently of the computation previously done for faults F1,F2...,Fi The proposed algorithm generates a test vector for fault $F_{i+l}$ by inheriting the test vector for fault Fi. A new test vector is grnerated from inherited values by gradually changing the inhderited values .The inherited values may partially activate a fauog and propagate the fault signal,Normally,this reduses the number of decision steps and backtracks in the second search.Experimental results for well-Known benchmark circuts show that the proposed algorithm is very efficient with small backtrack kimit;in combination eith other algorithms,it is very efficient for arbitrary backtrack limits.

Inheritance of Resistance to Phytophthora capsici by Inoculums in Korean Hot Pepper (고추 역병균의 접종원에 따른 역병 저항성의 유전 양식)

  • The study aims to identify the pathogenicity of Phytophthora. capsici isolates in major pepper-producing areas in Korea and the inherit genetic pattern of phytophthora blight resistance by inocula. With five kinds of testing materials including 'Kataguma (Sakata Korea)' peppers, a disease-susceptible material, '#308', a phytophthora blight resistance material, 'CM334', and their $F_1$ and $F_2$, respective isolates of P. capsici obtained from Icheon, Eumseong, Buan, Imsil and Yeongyang regions together with six kinds of peppers' inoculum including PA-159 (KACC No.40482) received from Korean Agricultural Culture Collection were used for inoculation. The disease-susceptible material '#308', the resistant material 'CM334' and the non-segregating generation of $F_1$ represented 4.94-5.00, 1.00-1.07, and 1.01-1.08 phytophthora blight incidence respectively in the group comparison by isolate. This result means that the phytophthora blight resistance was clearly distinguished among testing materials in the group comparison by P. capsici isolate. Moreover, $F_2$ segregating generation showed 1.79-2.31 phytophthora blight incidence which turned out to be identical in the group comparison by the six isolates of P. capsici isolate and with similarity between both the resistant and susceptible materials. Thus, the result proved that using the six isolates of P. capsici tested as inocula was suitable to investigate the phytophthora blight resistance. When it comes to group comparison of $F_2$ segregation generation, however, isolates were divided with PA-159 isolate being the center: a group consisting of isolates from Icheon, Buan, and Imsil and a group consisting of Yeongyang and Eumseong isolates with higher pathogenicity. The expected segregation ratio of the phytophthora blight resistance in $F_2$ generation by isolate was analyzed. PA-159 isolate showed 3:1 or 9:3:3:1, indicating that one to two genes were involved. On the other hand, results also proved that there is an interaction of genes since both Eumseong and Yeongyang isolates showed a segregation ratio of 11:5 while the Icheon isolate represented 12:3:1.
