Genetic Variation of Wild and Hatchery Populations of the Korean Ark Shell, Scapharca broughtonii Assessed by Microsatellite Markers (Microsatellite 마커를 이용한 한국산 피조개, Scapharca broughtonii Schrenck 집단의 유전적 다양성)

  • The genetic variation of Ark Shell, Scapharca broughtonii black was estimated using six polymorphic microsatellite (MS) loci in 443 individuals collected from five populations in Korea. The mean numbers of alleles per locus in five populations were 10-28. The mean number of alleles per locus in Jinhae Hatchery (JHH) population showed the least value as 15.5, but that in Gangjin (GJ) population showed the most value as 20.3. The mean expected heterozygosity in Saryangdo (SR) population showed the least value as 0.817, but that in Gangjin (GJ) population showed the most value as 0.831. In Jinhae hatchery(JHH) population, the mean expected heterozygosity was 0.822, there was no significant difference from those of wild population. The $F_{ST}$ values in Gangjin (GJ) population showed significant difference from those of the other populations, which revealed Gangjin (GJ) population is genetically different from the other populations. The $F_{ST}$ values among Jinhae Hatchery (JHH) population, Jinhae (JH) population and Saryangdo (SR) population showed lower values than the others, which implies there was a gene flow among these three populations. The $F_{ST}$ value and genetic distance between Jinhae (JH) population and Saryangdo (SR) population showed the least value as 0.0001 and 0.0386, indicating that these two populations were genetically the same.

A Study of the Family Caregiver's Burden for the Elderly with Chronic disease in a Rural Area (일부 농촌 지역 노인 만성질환자 가족의 부담감에 관한 연구)

  • The purpose of this study was to analysis level on family caregiver's burden for the elderly with chronic disease in a rural area and to choose priority care group, thereby facilitating the development of interventions to reduce the caregiver's burden. For this purpose, data were collected by questionaire from June 10 to October 8, 1994. The instruments for data collection were Caregiver Burden Inventory by Novak(1989) and Zarit et al(1982), severity of dementia by Hughes Scales(1982), ADL by Lawton(1971), patients' family caregiving activity by pre-survey and reference review(Lee, 1993 ; Jang, 1990 ; Yoo, 1982). The subjects were 213 family caregiver of elderly with chronic disease in a rural area. The data was analysed by the use of t-test, ANOVA, correlation and multiple regression. The results were as follows ; 1. Total burden was evaluated below average, the mean of family burden was 46.98. By the diagnostic classification, Hypertension was 27.37, DM 32.46, CVA 62.96, Dementia 61.24. 2. Significant variables which were correlated to the family caregiver's burden were the patient's disease diagnosis (F=33.82, p<0.001), severity of dementia(F=30.52, p<0.001), the status of disease management(F=11.53, p<0.001), ADL(F=10.54, p<0.001), PADL(F=7.50, p<0.001), income(F=7.17, p<0.001), caregiver's health status(F=24.53, p<0.001), a view of patient's prognosis (F=22.17, p<0.001), relationship with the patient(F=33.82, p<0.001), the number of hours per day spent on caregiving(F=77.52, p<0.001), level of intimacy of caregiver and patients(F=8.75, p<0.001), level of helping(F=4.90, p<0.01), the frequency of caregiving activity(F=3.80, p<0.01), the number of admission(F=5.54, p<0.01), the length of caregiving(F=4.43, p<0.01), other chronic patient in family(t=2.81, p<0.01), caregiver's job(F=3.11, p<0.01), the duration of illness(F=2.98, p<0.05), caregiver's religion(F=2.93, p<0.05), medical security(F=3.89, p<0.05), caregiving's helper(t=2.42, p<0.05). 3. PADL was the most important predictor to family caregiver burden(R2=0.6611). In addition to this, IADL, caregiver's health status, the length of caregiving. level of intimacy of caregiver and patients, patient's age, the patient's disease diagnosis and patient's job accounted for 76% of family caregiver burden. 4. The criteria of priority care group were as follows ; the mean of family caregiver burden was above 58, above of moderate ADL, the number of hours per day spent on caregiving above of 8 hours, above of moderate dementia. By the diagnostic classification, number of priority care group, Hypertension was 4 (8.0%), DM 4(8.0%), CVA 34(64.1%), Dementia 45(75.0%).

The Effects of Prostaglandin $F_{2\alpha}(PGF_{2\alpha})$ Treatment on Hormone Concentrations and Follicular Development in Early Postpartum Korean Native Goats (한국 재래 산양에 있어서 Prostaglandin $F_{2\alpha}(PGF_{2\alpha})$의 투여가 호르몬 함량 및 난포의 발육에 미치는 영향)

  • These experiments were conducted to evaluate the effects of the administration of exogenous PG $F_{2{\alpha}}$ on hormone concentrations and follicular development in early postpartum(pp) Korean native goats. 1. Plasma PG $F_{2{\alpha}}$ concentrations in PG $F_{2{\alpha}}$ treated goals showed a gradual increase from a low on day 2 (GR-1 : 10 day-treatment group: 6.35$\pm$0.5 and GR-2; 4 day-treatment group: 0.22$\pm$0.2 pg/$m\ell$, respectively) to reach a peak of 21.18$\pm$1.6 or 4.21 $\pm$0.4 pg/$m\ell$ on day 4 after treatment of PG $F_{2{\alpha}}$. 2. Plasma PG $F_{2{\alpha}}$ concentrations in GR-1 goats averaged 9.08 $\pm$1.2 pg/$m\ell$ compared with 5.44$\pm$ 1.8 pg/$m\ell$ in GR-2 goats the day before treatment of PG $F_{2{\alpha}}$. Mean PG $F_{2{\alpha}}$ concentrations thereafter remained low during the treatment period but PG $F_{2{\alpha}}$ concentrations did not differ between the two group. 3. Plasma concentrations of estradiol-17 $\beta$ (E,) in PG $F_{2{\alpha}}$ - treated group were decreased gradually until 2 days after PG $F_{2{\alpha}}$ treatment but mean $E_2$ concentrations began to increase on 3 days and were Inaximal on the 4 days after treatment. 4. Plasma lulenizing hormone (LH) concentrations in PG $F_{2{\alpha}}$ - treated goats were slightly higher than in controls but mean LH concentrations did not differ between the two treatment groups. 5. Plasma prolactin (PRL) concentrations were suppressed in both GR-1(10 day-treatment group) and GR-2(4 day-treatment group) goats compared to saline controls but mean PRL concentrations remained lower in PG $F_{2{\alpha}}$ treated animals during post-treatment period. 6. The mean number of small and medium follicles present when PG $F_{2{\alpha}}$ was administrated was similar in all does but the increase in number of large follicles($\geq$4mm) tended to be higher in PG $F_{2{\alpha}}$ treated group than controls. These results suggest that concentrations of PG $F_{2{\alpha}}$ and estradiol-17$\beta$ were positively correlared with follicular diameter. We conclude that PG $F_{2{\alpha}}$ treatment stimulates follicular development similarly in both GR-1 and GR-2 group.

  • To investigate sliding friction and wear behaviors of WC-Co/WC-Co pairs containing different WC grain sizes, the ball-on-disc test in air were carried out, where WC grain sizes a $0.5\;{\mu}m$ (F.G.) and $1.5\;{\mu}m$ (C.G.). The wear volume of F.G. pin for F.G. pin/C.G. disc is larger than that of F.G. pin for F.G. pin/F.G. disc due to higher friction coefficient, and the surfaces after wear test are richer in oxygen compared to those before test. Furthermore, the wear debris, which is composed of nona-scale grain, after the wear test are remarkably richer in oxygen than to those before test.

Effects of Ovarian Morphology and Culture Vessel on In vitro Development and Cell Number in Embryos of Korean Native Cows

  • The main purpose of this study was to improve the efficiency and quality of in vitro embryo production in Korean Native Cows (KNC). We examined the effects of ovarian morphologies (Experiment 1) and the culture vessel (Experiment 2) on in vitro maturation (IVM). We measured the subsequent development rates and cell numbers of blastocysts. In Experiment 1, the ovaries of KNC were divided into six groups, based on follicle and corpus luteum (CL) morphology. The development rates to the 2- and 8-cell stages were similar among the six groups. The development rates to blastocyst stages were significantly higher in the group without a CL or follicle (WOCL/F) than in the groups with follicular cysts (FCs), regressive CLs (RCLs) or cystic CLs (CCLs) (p<0.05). The cell number of the inner cell mass (ICM) of blastocysts in the FCs and RCLs groups, and the number of cells in the trophectoderm (TE) in the WOCL/F group, FCs, growing CLs (GCLs) and RCLs were significantly higher than in other groups (p<0.05). The total cell number (TCN) in the WOCL/F, FC and RCL groups was also significantly higher than in other groups (p<0.05). The ICM cell number/TCN ratio was significantly higher in the FC and RCL groups than in the GCL and DF groups (p<0.05). In Experiment 2, oocyte IVM was carried out in culture dishes, in 0.25- or 0.5-ml straws used for freezing sperm. The development rate to the 2-cell stage was significantly higher in the 0.5-ml straw group than in the 0.25-ml straw group. The development rates to the blastocyst stage were similar in the dish and the two straw groups. There were no differences in the cell numbers of ICM, TE or TCN or ICM cell number/TCN ratios between groups.

Natural Occurrence of Fusarium Mycotoxins in Field-collected Maize and Rice in Korea in 2009 (2009년산 옥수수와 벼에서의 Fusarium 곰팡이독소 자연발생량 조사)

  • To detect Fusarium mycotoxins, grain samples were collected from 32 rice fields all over the country and from 19 maize fields in eastern and midland provinces in Korea in 2009. Maize contamination with Fusarium species (54.9%) was higher than in rice (8.2%). Using Fusarium species specific PCR primer sets (Fg16 and VERT), 58 and 354 of total 506 isolates from maize samples were putatively identified as F. graminearum (11.5%) and F. verticillioides (70.0%), respectively. From rice samples, 276 of 315 isolates (87.8%) were putatively identified as F. graminearum but F. verticillioides was not identified. LC or LC-MS analysis of the samples revealed that fumonisin was the most commonly detected mycotoxin in maize samples but its level was below the regulation limit. Only two maize samples were contaminated with deoxynivalenol and zearalenone at the levels above the regulation limit. In rice samples, contamination with zearalenone was common but the levels were below the regulation limit. This study showed that most of the Korean maize and rice samples collected in 2009 were contaminated with Fusarium mycotoxins but the levels were below the Korean regulations for deoxynivalenol, fumonisin and zearalenone.

Association of Novel Polymorphisms in Lymphoid Enhancer Binding Factor 1 (LEF-1) Gene with Number of Teats in Different Breeds of Pig

  • Lymphoid enhancer binding factor 1 (LEF-1) is a member of the T-cell specific factor (TCF) family, which plays a key role in the development of breast endothelial cells. Moreover, LEF-1 gene has been identified as a candidate gene for teat number trait. In the present study, we detected two novel mutations (NC_010450.3:g. 99514A>G, 119846C>T) by DNA sequencing and polymerase chain reaction-restriction fragment length polymorphism in exon 4 and intron 9 of LEF-1 in Guanzhong Black, Hanjiang Black, Bamei and Large White pigs. Furthermore, we analyzed the association between the genetic variations with teat number trait in these breeds. The 99514A>G mutation showed an extremely significant statistical relevance between different genotypes and teat number trait in Guanzhong (p<0.001) and Large White (p = 0.002), and significant relevance in Hanjiang (p = 0.017); the 119846C>T mutation suggested significant association in Guanzhong Black pigs (p = 0.042) and Large White pigs (p = 0.003). The individuals with "AG" or "GG" genotype displayed more teat numbers than those with "AA"; the individuals with "TC" or "CC" genotype showed more teat numbers than those with "TT". Our findings suggested that the 99514A>G and 119846C>T mutations of LEF-1 affected porcine teat number trait and could be used in breeding strategies to accelerate porcine teat number trait improvement of indigenous pigs breeds through molecular marker assisted selection.

Relative Analysis between Fertility and Protein Changes in Semen of Different Species in Pigs (돼지 품종간 정액 내에서 수정 능력과 단백질 변화와의 관계 분석)

  • The objective of this study was to investigate the relationship between fertility and protein pattern change using in vitro fertilization, analysis of sperm characteristics and two-dimensional gel electrophoresis in different pig types. In results, the viability and mitochondria integrity of sperm were higher significantly (p<0.05) but the portions of acrosome reaction was lower significantly (p<0.05) in Duroc and $F_1$ (potbellied ${\times}$ PWG miniature pig) than PWG miniature. On in vitro fertilization to investigate fertility, the fertility of $F_1$ semen war higher significantly (p<0.05) than in Duroc and PWG miniature pig. On the other hand, protein patterns showed similar function among the different boar semen. Especially, the heat shock 70 kDa 1-like and G patch domain-containing protein 4 were significantly (p<0.05) higher expressed in $F_1$ than in Duroc and PWG miniature pig. The proteins associated with mitochondria in Duroc were significantly (p<0.05) higher expressed than in $F_1$ and PWG miniature pig. The developmental rates to blastocyst stage of oocytes fertilized with sperm of $F_1$ pig were significantly (p<0.05) higher than in PWG miniature pig. However, phosphoglycerate kinase 2 and zinc finger protein 431 were significantly (p<0.05) higher expressed in PWG miniature pig than in $F_1$ and Duroc pigs. In conclusion, the results of the present study indicate that different proteins were expressed in different pig types, and were associated with a sperm functions and embryo development.

  • To evaluate the modifying effect of Kwao Kreu, Pueraria mirifica (PM) well-known as a rejuvenating folk medicine from Thailand, peri- and post-natal studies were carried out in rats. PM extract was administered to pregnant Sprague Dawley (SD) rats by oral gavage from gestation 6 (GD 6) to postnatal day 21 (PND 21). The amount of administered in this study was 0.042, 0.42 and 4.2 mg/kg/day, respectively. There were no treatment related changes of dams in deaths, clinical signs, and parturition. Treatment related changes in body weight, food consumption and lactation of dams were not observed. F1 fetuses in external abnormality, physical development, reflex/sensory functions and behavioral development were not found. No adults and F1 fetuses in organ weight was found with the exception of vagina and uterus of F1 fetuses. The results showed that PM extract, up to 4.2 mg, had no adverse effects on the peri- and post-natal development of rats. Therefore, PM extract has no adverse effects on peri- and post-natal development of rats.

Growth Performance of Offspring from Selected Korean, Cultured Japanese and Their Reciprocal Intraspecific Hybrids of Red Sea Bream, Pagrus major

  • Intraspecific hybrids have been proven to be a useful tool for increase growth rate in some freshwater fish species. However the effects of intraspecific hybrids in seawater fish species is little known. To assess whether intraspecific hybrids in red sea bream was associated with growth enhancement, growth performances of offspring from two inbred lines, selected Korean line (KORDI-F4) and cultured Japanese line (IPN) and two intraspecific hybrids lines (KORDI-F4♀×JPN♂ and JPN♀×KORDI-F4♂) of red sea bream were compared. There's no significant difference in body weight between offspring groups during seed production period (until 4 months old). At seven months old (after three months rearing trials on sea cages communally), the offspring from KORDI-F4 (41.0±11.1g) and JPN♀×KORDI-F4♂ (39.3±8.1g) showed significantly better performance in body weight than that of the offspring from KORDI-F4♀×JPN♂ (37.0±8.1g) and IPN (35.7±9.7g). At 15 months old, the offspring from JPN♀×KORDI-F4♂ showed best growth performances in body weight, weight gain, specific growth rate among four offspring groups. As the results, the intraspecific hybrids between cultured Japanese line and selected Korean line showed superior growth performances than their paternal and maternal inbred lines.
