Dendrobium moniliforme (DM) is a valuable and versatile herbal medicine with the anecdotal claims of antioxidant and anti-inflammation. In the present study, we investigated the whitening and anti-wrinkling effects of DM under various conditions with B16F10 melanoma cells and human dermal fibroblasts. The DM extract inhibited melanin contents and tyrosinase activity in a dose-dependent manner, compared with untreated group. Treatment of the DM extract effectively suppressed the ${\alpha}$-MSH-stimulated melanin formation, tyrosinase activity and dendrite outgrowth. Moreover, the ${\alpha}$-MSH-induced mRNA expressions of tyrosinase-related protein-1 (TRP-1) and tyrosinase-related protein-2 (TRP-2) and protein expression of tyrosinase were significantly attenuated by DM treatment. We also investigated the DM increased the production of type I procollagen and inhibited TNF-${\alpha}$-induced mRNA expressions of MMP-1, -3 in the human dermal fibroblast. These results indicate that DM may be a great cosmeceutical ingredient for its whitening and anti-wrinkle effects.
Seventy-three fungal species belonging to forty-three genera were isolated from 40 samples of Saccharrum officinarum (collected from Naage-Hamadi canal in Qena Governorate, Egypt). Aspergillus, Trichoderma, Mucor and Pythium were the most common genera on the two isolation media. The dominant species of Aspergillus were A. niger, A. flavus, A. ustus, A. terreus and A. wentii. Some species were dominant on 40 g/l sucrose such as Aspergillus niger, A. flavus, Emericella nidulans, Trichoderma viride, Torula herbarum and Mamaria echinoeotryoides, while the dominant species on 10 g/l glucose were Mucor circinelloides, Aspergillus niger, Torula herbarum and Trichoderma viride. Mycotoxins including aflatoxins $B_1,\;B_2,\;G_1\;and\;G_2$, zearalenone and diacetoxyscirpenol were detected in the examined samples of Saccharrum officinarum. The mycelial growth of A. flavus, A. niger, Fusarium moniliforme and Torula herbarum decreased with the increase in Dimethoate concentrations, although 25 ppm was less effective than the higher levels of the insecticide ($75{\sim}200\;ppm$). Dimethoate stimulated the activity of Go-Tin A. niger, F. moniliforme and T. harbarum, while the Go-T activity was inhibited in A. flavus with the Dimethoate treatments.
The incidence of total molds, Fusarium species, and the estrogenic mycotoxin,zearalenone, in Korean grain samples were investigated . The majority of molds infecting grain were identified as belonging to the genus Alternaria , with an average infection rate of kernels of 43% and 32% in rice and baley, respectively. Fusarium speciens were less common, with average infection rates of 13% and 19% in reice and barley, respectively. A number of field fungi including Curvularia and Dactylaria were also observed. Among the Fusarium speices, 71 of 94 Fusarium isolates were identified as F.semitectum. A few F. moniliforme and F. equiseti were observed linked immunosorbent assay (ELISA) or high-performance liquid chromatography(HPLC). In addition, deoxynivalenol was not deteted by ELISA . However, thepresence of molds, including Fusarium species, may pose possbile health hazards to persons consuming those grains.
병원성 Fusarium에 의한 아스파라거스 감염은 비병원성 Fusarium을 5일과 7일 전에 접종하였을 때 방제효과가 있었다. 비병원성 F. oxysporum은 F. moniliforme에 대하여 방제효과가 있었고, F. solani는 F. oxysproum에 대하여 방제효과가 있음이 밝혀졌다. 실험에 사용된 Fusarium 균들은 모두 주근과 측근의 말단 부위, 상처부위, 그리고 표피의 세포벽 사이를 통하여 감염하였다. 경우에 따라 감염하는 동안 appressorium과 유사한 구조를 형성하기도 하였고, 직접 감염하는 경우도 있었다. 병원성 그리고 비병원성 Fusarium 균 모두 공통적으로 생장점 부위를 통하여 감염하였다. 병원성이 강한 Fusarium 균의 경우 비병원성 균들보다 감염의 속도가 빨랐고 더욱 생장이 왕성하였다. F. solani는 생장속도나 기주 조직 침입속도가 매우 느렸다. 기주 감염의 결과 처음에는 cortical rot을 유발시켰고 나중에는 tracheary elements를 감염하고 결국은 조직의 괴사를 유발하는 것이 관찰되었다. 비병원성 F. oxysporum은 표피조직에 두터운 균사층을 형성하였고, 이는 병원성 Fusarium 균에 대한 방제효과를 나타내는 원인을 제공한 것으로 여겨진다. F. solani는 측근의 생성을 촉진시켜 표면적을 증대시킨 것으로 여겨진다. 결론적으로 AVFO와 F. solani를 이용하여 아스파라거스에 발생하는 병원성 Fusarium균의 침입을 저지할 수 있는 생물적 방제가 가능함이 밝혀졌다.
Antifungal activity of seven natural antimicrobials, such as $\varepsilon$-polylysine, extract of Yucca shidigera, TLS (vitamin B1 derivative), BMB-FS, chitooligosaccharide, KDSP 001, and KDSP 002 were investigated for their applications in functional beverage. Five fungi including Aspergillus niger, Penicillium citrinum, Rhizopus oryzae, Fusarium moniliforme, and Mucor rouxii were applied as test fungi and mininum inhibitory concentrations (MICs) of antimicrobials were examined. TLS exhibited the strongst antifungal activity among tested antimicrobials and the growth of all fungi was inhibited at 100 ppm. The antifungal activity of BMB-FS appeared different. The growth of Fusarium moniliforme was inhibited by BMB-FS at 100 ppm, but it exhibit antifungal activity on P. citrinum, and M. rouxii at 1000ppm. MICs of TLS appeared to be 60 ppm for four test fungi except A. niger (100 ppm). These results indicate the possible usage of TLS and BMB-FS as natural antimicrobials in functional beverage.
From the sample seeds of rice cultivar Palkoeng with the brown spot symptom, Helminthosporium oryzae was detected in $86\~92\%$ of hulls including empty glumes, lemma, palea, and rachilla in five to seven days of incubation. When the seeds were dehulled, the percentage detection decreased to $74\~83\%$ of pericarps. The fungus sporulated on whole surface of the seed and was detected within about ten cell layers deep of endosperm but not detected in embryo. Fusarium moniliforme was found in embryo as well as pericarp and endosperm. The frequency of F. roseum, Trichoconis padwickii, Curvularia spp., Atternaria tenuis, Cladosporium cladosporoides and Phoma sp. was less than three percent of hulls and pericarps. In seven days of incubation, $60\~87\%$ of the germinating seeds showed the symptom of H. oryzae while $13\~40\%$ were apparently healthy. This fungus in hilum of infected rice transmitted through pericarp to plumule shoot and radicle of the germinating seed.
A total of 34 species belonging to 21 genera of fungi were recorded on passion fruits of both pure and hybrid origin in Uganda, however, the pure type exhibited wider spectrum (28 species and 16 genera) than the hybrid type (21 & 15). Also, yeasts (unidentified and Rhodotorula mucilaginosa) were also encountered in high numbers. Moreover, the mean count of all mycobiota obtained from the pure type was higher than that of hybrid, despite the bigger size of the later. Members of yeasts and Cladosporium followed by Phoma, Penicillium, Fusarium and Alternaria species dominated on passion fruits of pure origin, while only C. cladosporioides, F. solani and yeasts dominated on the hybrid type. Treatment with Na-hypochlorite exhibited inhibitory effects on the total mycobiotic propagules as well as the dominant species from fruits of both types. The current results, therefore, suggest the use of Na-hypochlorite to control the post-harvest mycobiota associated with passion fruits. Regarding the mycobiota contaminating passion juice, yeasts were found to be the major contaminants with Candida parapsilosis being the most common. Moulds constituted only a minor proportion with Acremonium strictum followed by Fusarium chlamydosporum, F. moniliforme, F. acuminatum and F. solani as the most dominant species. In the heat-treated juice samples, the counts of the most commonly encountered mycobiota (both yeasts and molds) were significantly inhibited or completely eliminated. Some unidentified Bacillus species were also recovered from the juice, however, their counts in the heated samples were increased but insignificantly.
Some interactions in various soil conditions, numbers of microbial populations, root rot disease development and rates of spore germiation in three different location of soils were investigated. The calcium and magnesium contents were higher in replanted fields of ginseng (Panax ginseng) at Goesan. Potassium contents were high in replanted field at Poonggi and textural class of the soils was silt loam except for silt clay loam in first cultured field of ginseng at Goesan. For the germination process of Fusarium solani, F. moniliforme, F. oxysporum, and Alternaria panax, the percentage germination of fungal spores was high in double distilled water and Pfeffer's solution as media, whereas the lower rate of germination of spores was observed in soil extracts. Numbers of bacteria were high in replanted field soil at Gumsan, and propagules of fungi in replanted fields at Gumsan and Poonggi were higher than other soils, but higher numbers of actinomycetes were found in the first cultured field of ginseng at Goesan and Poonggi. Fungistasis was induced by higher microbial populations present in soil that was initiated when amended with garlic stalk, crushed bean and ginseng leaves. On the other hand, there was no fungistasis in soil amended with wheat and barley straw, and this tendency was a little difference on the soil sample.
Fusarium spp. were isolated from the postharvest fruit rot of oriental melon fruits at commercial fruit markets in Korea during 2001 to 2003. The decayed fruits were covered with the fungal mycelia and eventually soft rotted. The disease started at the fruit stalk area, the calyx end of the fruit and skin of fruit. As the disease advanced, white to pinkish mycelia covered with the surface of decayed fruit. The cultural and morphological characteristic of Fusarium spp. were compared with descriptions of those reported previously, and identified as Fusarium equiseti, F. graminearum, F. moniliforme, F. proliferatum, F. sambucinum, and F. semitectum. Pathogenicity of the isolates was proved by artificial wound and unwound inoculation onto the healthy fruits. Two days after inoculation, aerial mycelia were noticed on the wound inocultion region of the fruit and developed soft rot symptoms. Although Fusarium spp. causing fruit rot disease in oriental melon have been reported in Korea, identification of the those species was not described. Therefore, this is the first report of Fusarium spp. causing postharvest fruit rot on oriental melon in Korea.
In this study, five endophytic fungi belonging to the Aspergillus and Alternaria genera were isolated from Lagopsis supina. The antimicrobial activity of all fungal fermented extracts against Staphylococcus and Fusarium graminearum was tested using the cup-plate method. Among them, Aspergillus ochraceus XZC-1 showed the best activity and was subsequently selected for large-scale fermentation and bioactivity-directed separation of the secondary metabolites. Four compounds, including 2-methoxy-6-methyl-1,4-benzoquinone (1), 3,5-dihydroxytoluene (2), oleic acid (3), and penicillic acid (4) were discovered. Here, compounds 1 and 4 displayed anti-fungal activity against F. graminearum, F. oxysporum, F. moniliforme, F. stratum, Botrytis cinerea, Magnaporthe oryzae, and Verticillium dahlia with diverse MIC values (128-512 ㎍/ml), which were close to that of the positive control antifungal, actidione (64-128 ㎍/ml). Additionally, compounds 1 and 4 also exhibited moderate antibacterial activity against S. aureus, Listeria monocytogenes, Escherichia coli, and Salmonella enterica, with low MIC values (8-64 ㎍/ml). Moreover, compounds 1 and 4 displayed selective cytotoxicity against cancer cell lines as compared with the normal fibroblast cells. Therefore, this study proposes that the endophytic fungi from L. supina can potentially produce bioactive molecules to be used as lead compounds in drugs or agricultural antibiotics.
