Effects of Environmental Factors on the Eye Direction in Juvenile Starry Flounder Platichthys stellatus (강도다리(Platichthys stellatus) 변태 과정에서 안구 이동의 방향에 관여하는 환경적 요인)

  • The direction of eye movement in Pleuronectiformes is species-specific. In farmed flounder, deviations in eye direction can reduce marketability. Therefore, we explored the effects of genetic and environmental factors on the direction of eye movements in the starry flounder Platichthys stellatus to address this economic challenge. Four pairs of female and male starry flounder broodstock, with eyes positioned to the right or left, were used in a mating experiment. The experimental groups were established with breeding seawater temperatures set at 10, 14, and 18℃ for the water temperature experiment. Whole-body samples of juveniles were collected to analyze the expression of genes related to eye movement and direction. The mating experiment results showed no significant differences in abnormal eye movement. In the water temperature experiment, the rate of abnormal eye movement was significantly higher at 32.19±1.33% in the 18℃ group than in the 10 and 14℃ groups. Genetic analysis of eye movement related to water temperature revealed that the expression of eye migration genes was significantly higher at 10℃. Moreover, no significant differences were observed in mating experiments. In conclusion, water temperature and not mating affected the eye movement of starry flounder.

A Case of Ocular Sparganosis in China: Episode of Migration from Muscle Cone to Subconjunctiva

  • To improve our understanding of the migration of sparganum in humans, we report a case of ocular sparganosis having the migratory episode from the muscle cone to the subconjunctiva. A 34-year-old woman was admitted to the Hospital of Anhui Medical University (Hefei, China), in December 2019. She presented with conjunctival hemorrhage and recurrent pain in the left eye. A foreign body was found in the muscle cone of the eye. Two months later, a ribbon-like white material was found under the conjunctiva on slit-lamp examination. A long and slender, actively moving parasite was extracted by surgery. The extracted worm was approximately 8 cm long and 2 mm wide. The worm was whitish, wrinkled, ribbon shaped, and had a slightly enlarged scolex. The worm sample was morphologically identified as a plerocercoid larva (sparganum) of the Spirometra tapeworm. Her conjunctival blood suffusion and eye pain ceased within 1 week after operation. She has been in good health without any symptoms during the 2-year follow-up. A case of ocular sparganosis, in which larval worm migrated from the muscle cone to the subconjunctiva is reported from China.

Morphological Specificity in Cultured Starry Flounder Platichthys stellatus Reared in Artificial Facility

  • The starry flounder Platichthys stellatus, like all flatfish, exhibits conspicuous lateral asymmetry in numerous traits, most obvious of which is the migration of one eye to the other side of the head during metamorphosis. Additional changes related to eye migration include asymmetrical pigmentation, and a behavioral shift from larvae that exhibit upright, open-water swimming to juveniles and adults that lie on the ocean floor, eye side up. However, the morphology of these juveniles has been quite plastic in recent years, a phenomenon which is thought to be related to a diverse suite of semi-intensive and intensive larviculture methods. The cause of morphological abnormalities in the farmed flatfish is poorly understood. In the present study, we observe the features of morphological specificity and abnormality of immature fish (mean total length 23 cm) and survey the occurrence frequency of the specificity and abnormality of juvenile (mean total length 6.70 cm) in artificial culture facility. We find 2 types of abnormality (e.g., albino in ocular side and hypermelanosis in blind side) and 1 type of specificity (e.g., lateral polymorphism). These considerably differ from normal individuals (has sinistral eye and pigmented on only one side) by several characteristics (dextral eye, ocular side albinism, blind side hypermelanosis). The incidences of albinism, hypermelanosis, and body reversal are $10.1{\pm}2.56%$, $91.7{\pm}1.7%$, and $13.1{\pm}1.1%$, respectively. These suggest that these morphometric and morphological differences occur more in artificial environment during and just after metamorphosis.

Wogonin attenuates vascular remodeling by inhibiting smooth muscle cell proliferation and migration in hypertensive rat

  • Wogonin, extracted from the roots of Scutellaria baicalensis Georgi, has been shown to suppress collagen deposition in spontaneously hypertensive rats (SHRs). This study was performed to investigate the role and mechanism of wogonin underlying vascular remodeling in SHRs. After injection of SHRs with 40 mg/kg of wogonin, blood pressure in rats was measured once a week. Masson's trichrome staining was conducted to observe the changes in aortas and mesenteric arteries. Vascular smooth muscle cells (VSMCs) isolated from rat thoracic aortas were treated with Angiotensin II (Ang II; 100 nM) in the presence or absence of varying concentrations of wogonin. The viability and proliferation of VSMCs were examined using Cell Counting Kit-8 assay and 5-ethynyl-2'-deoxyuridine assay, respectively. The migration of VSMCs was examined using wound healing assay and transwell assay. We found that wogonin administration alleviated hypertension, increased lumen diameter, and reduced the thickness of the arterial media in SHRs. Ang II treatment enhanced the viability of VSMCs, which was inhibited by wogonin in a concentration-dependent manner. Wogonin reversed Ang II-induced increases in the viability, proliferation, and migration of VSMCs. Moreover, wogonin inhibited Ang II-induced activation of mitogen-activated protein kinase (MAPK) signaling in VSMCs. Overall, wogonin repressed the proliferative and migratory capacity of VSMCs by regulating the MAPK signaling pathway, thereby attenuating vascular remodeling in hypertensive rats, indicating that wogonin might be a therapeutic agent for the treatment of vascular diseases.

Organ-specific Toxocara canis larvae migration and host immune response in experimentally infected mice

  • We investigated organ specific Toxocara canis larval migration in mice infected with T. canis larvae. We observed the worm burden and systemic immune responses. Three groups of BALB/c mice (n=5 each) were orally administered 1,000 T. canis 2nd stage larvae to induce larva migrans. Mice were sacrificed at 1, 3, and 5 weeks post-infection. Liver, lung, brain, and eye tissues were collected. Tissue from 2 mice per group was digested for larval count, while the remaining 3 mice underwent histological analysis. Blood hematology and serology were evaluated and compared to that in a control uninfected group (n=5) to assess the immune response. Cytokine levels in bronchoalveolar lavage (BAL) fluid were also analyzed. We found that, 1 week post-infection, the mean parasite load in the liver (72±7.1), brain (31±4.2), lungs (20±5.7), and eyes (2±0) peaked and stayed constant until the 3 weeks. By 5-week post-infection, the worm burden in the liver and lungs significantly decreased to 10±4.2 and 9±5.7, respectively, while they remained relatively stable in the brain and eyes (18±4.2 and 1±0, respectively). Interestingly, ocular larvae resided in all retinal layers, without notable inflammation in outer retina. Mice infected with T. canis exhibited elevated levels of neutrophils, monocytes, eosinophils, and immunoglobulin E. At 5 weeks post-infection, interleukin (IL)-5 and IL-13 levels were elevated in BAL fluid. Whereas IL-4, IL-10, IL-17, and interferon-γ levels in BAL fluid were similar to that in controls. Our findings demonstrate that a small portion of T. canis larvae migrate to the eyes and brain within the first week of infection. Minimal tissue inflammation was observed, probably due to increase of anti-inflammatory cytokines. This study contributes to our understanding of the histological and immunological responses to T. canis infection in mice, which may have implications to further understand human toxocariasis.

The Role of nerve Growth Factor on Corneal Wound Healing in Dogs

  • To investigate the modulation of nerve growth factor (NGF) during corneal epithelial wound healing and the effect of topical NGF on corneal epithelial wound healing in dogs. An axial epithelial defect was created in the right eye using 6mm axial corneal mechanical debridement while the left served as an unwounded control. The tears were collected from both eyes during 1 week and the corneal epithelium was processed for the measurement of NGF at day 0 and 7. The NGF content of tears and corneal epithelium was determined by enzyme-linked immunosorbent assay. In another experiment, the animals were divided into 3 groups. The right eyes in each group were treated every six hours with 200 ug/ml of recombinant human (rh) NGF, murine NGF, or 600 ug/ml of anti-NGF blocking antibody. The left eye of each animal was treated with bovine serum albumin (BSA) to serve as controls. Wound healing was analyzed using NIH image software. Tear NGF was markedly increased in the wounded eyes, relative to tears from control eyes during the early healing period. The NGF content of the corneal epithelium was elevated in the wounded eye (p=0.024). Time to wound closure and rate of epithelial migration were not significantly different between the NGF treated or the NGF antibody treated, and the control BSA treated eyes. Corneal epithelial wounding increased NGF content only on the wounded side during the early healing period. Neither topical recombinant human or murine NGF affected corneal epithelial wound healing in the normal dog.

Detection of Face Direction by Using Inter-Frame Difference

  • Applying image processing techniques to education, the face of the learner is photographed, and expression and movement are detected from video, and the system which estimates degree of concentration of the learner is developed. For one learner, the measuring system is designed in terms of estimating a degree of concentration from direction of line of learner's sight and condition of the eye. In case of multiple learners, it must need to measure each concentration level of all learners in the classroom. But it is inefficient because one camera per each learner is required. In this paper, position in the face region is estimated from video which photographs the learner in the class by the difference between frames within the motion direction. And the system which detects the face direction by the face part detection by template matching is proposed. From the result of the difference between frames in the first image of the video, frontal face detection by Viola-Jones method is performed. Also the direction of the motion which arose in the face region is estimated with the migration length and the face region is tracked. Then the face parts are detected to tracking. Finally, the direction of the face is estimated from the result of face tracking and face parts detection.

Late Complication of a Silicone Implant Thirty Years after Orbital Fracture Reconstruction

  • Alloplastic materials used for orbital fracture reconstruction can induce complications, such as infection, migration, extrusion, intraorbital hemorrhage, and residual diplopia. Silicone is one of the alloplastic materials that has been widely used for decades. The author reports a rare case of spontaneous extrusion of a silicone implant that was used for orbital fracture reconstruction 30 years earlier. A 50-year-old man was admitted to the emergency room for an exposed substance in the lower eyelid area of the left eye, which began as a palpable hard nodule a week earlier. The exposed material was considered to be implant used for previous surgery. Under general anesthesia, the implant and parts of the fibrous capsule tissue were removed. Several factors hinder the diagnosis of implant extrusions that occur a long period after the surgery. So, surgeons must be aware that complications with implants can still arise several decades following orbital fracture reconstruction, even without specific causes.

  • The research on Harmful Algal Blooms is generally in progress through field work, such as the naked eye and sampling. It was difficult to forecast exactly the course, from appearance of red tide to disappearance, with the established ways of investigation and analysis. Accordingly it is need to analyze environmental factors in time and space, the appearance of red tide and the path of its migration by more objective and scientific methods. In this study the remote sensing was used to diminish damage from the occurrence of red tide. Such as a temperature change of sea water and a change of tidal currents, the major cause for red tide. The probed data were utilized. The technique for forecast of red tide phenomenon on the south coast was researched by analyzing the cause of red tide, pollutant flowed from landand the possibility of application of the technique was showed.

  • The research on red tide is generally in progress through field work, such as the naked eye and sampling. It was difficult to forecast exactly the course, from appearance of red tide to disappearance. with the established ways of investigation and analysis. Accordingly it is need to analyze environmental factors in time and space, the appearance of red tide and the path of its migration by more objective and scientific methods. In this study, GIS is applied to analyse the space character of red tide and the interpolation of IDW(Inverse Distance Weight) is applied to assume the density distribution of red tide after gather data by using Arc/Info. After IDW interpolation, the sea area occurred over 1,000 cells/ml of red tide density is extracted with CON and SUM Function of Grid Module, and the density of the sea area is accumulated daily. As a result of this study, the distribution condition of red tide is found timely and spacially by applying GIS to the sea area of red tide, the results indicated that the spatial density and the cumulative frequency about the origin of red tide using GIS, the sea area demonstrated that the maximum density and the maximum frequency varied significantly over the Nammyun of Namhae-Is. with the maximum frequency being 49 times. accordingly if data about the areas of red tide will occur from the present are accumulated, the shifting route of red tide occurrence and extinction can be predicted.
