  • 미쇄(微碎)한 치자분말(粉末)에서 Orange-Yellow 색소(色素)를 추출(抽出)하면 짧은 시간(時間)에서 $log\;t^{1{\cdot}15}$에 비례(比例)하였고 최적(最適) 추출시간(抽出時間)은 30분(分)이 선정(選定)되었으며 온도(溫度)가 상승(上昇)함에 따라 $log\;T^{3{\cdot}73}$에 비례(比例)하였고 최적(最適) 추출온도(抽出溫度)는 $40^{\circ}C$가 선정(選定) 되었으며 시료(試料) 0.1 g당(當) 용매량(溶媒量)의 증가(增加)에는 $log\;S^{3{\cdot}7}$의 색소(色素) 추출량(抽出量)을 나타냈고, 지방(脂肪)을 완전(完全)히 제거(除去)한 시료(試料)는 전혀 제거(除去)하지 않은 시료(試料)보다 $0.6(O.D\;448\;m{\mu})$만큼의 좋은 추출효과(抽出效果)를 보였다. 이들을 綜合하여, $P_{total}{\simeq}1.15\;log\;t+3.73\;log\;T+3.7\;log\;S-4X-6.4$ $P_{total}$: total amounts of extracted orange-yellow pigment from Gardenia. t: extraction time $(0.5{\sim}60\;min)$ T: extraction temperature $(5{\sim}50^{\circ}C)$ S: volume of solvent $(5{\sim}50\;ml)$ X: fat contents $(0{\sim}15%)$ 와 같은 수정(修正)된 실험식(實驗式)을 유도했다.

미역 추출액의 품질 특성에 미치는 온도의 영향 (Effect of Temperature on Some Quality Characteristics of Aqueous Extracts of Sea Mustard)

  • 열수출시 미역 추출액의 품질 특성에 미치는 온도의 영향을 조사하였다. 추출액의 고형분, 단백질의 농도와 수율은 초기추출시간 동안 추출 온도 $50^{\circ}C$에서 $100^{\circ}C$로 증가할수록 높아졌다. 그러나 상징액율은 $50^{\circ}C$$100^{\circ}C$에서 가장 낮은 값을 보였으며 $100^{\circ}C$에서 1시간 추출할 때 고형분과 단백질 수율은 각각 21.56%와 4.76%였다. 점도는 $100^{\circ}C$에서 2시간 추출할 때 가장 높았으며 다른 온도의 추출액 점도에 비해 $2.5{\sim}4.0$배 높았다. 탁도는 추출 1시간 이후 차츰 감소하였으며 고형분과 단백질의 초기 추출속도에서 계산된 활성화에너지는 1.18과 3.9 Cal/mole이었다.

  • 고품질의 아가콩 음료개발을 위해 반응표면분석법에 의한 추출조건을 최적화 하였다. 아가콩의 최적 볶음조건은 250 ℃, 30분으로 설정하였다. 추출시간, 추출온도에 따른 아가콩의 품질지표인 pH, 색도 및 이소플라본 함량은 1% 이내에서의 유의적인 영향을 미치는 것으로 나타났다. 추출온도와 추출시간이 길어질수록 이소플라본 함량은 높은 것으로 나타났다. 반응표면분석의 이소플라본 추출 공정 최적화 결과 추출온도는 99.5 ℃, 추출시간은 1.7 h으로 나타났으며, 이 조건에서 이소플라본의 최적 수율은 10.63 ㎍/mL로 예측되었다.

  • 플라보노이드성분 함량이 높은 밀싹을 이용하여 유효성분을 추출하고, 중심합성계획모델을 이용하여 추출공정을 최적화하였다. 중심합성계획모델의 반응치로는 추출수율과 플라보노이드성분 함량을 설정하고, 독립변수인 추출시간, 주정/초순수 부피비, 추출온도에 따른 주효과도와 교호효과도를 해석하였다. 추출수율의 경우 추출시간과 추출온도가 상대적으로 큰 영향을 미쳤으며, 플라보노이드성분 함량의 경우에는 추출시간의 영향이 가장 크게 나타났다. 추출수율과 플라보노이드성분 함량을 모두 고려한 결과 최적추출조건은 추출시간(2.44 h), 주정/초순수의 부피비(50.00 vol%), 추출온도($54.41^{\circ}C$)이었으며, 이때 추출수율은 30.14 wt%, 플라보노이드성분 함량은 $35.37{\mu}g\;QE/mL\;dw$이었다. 이 조건의 실제 실험결과 추출수율(29.92 wt%), 플라보노이드성분 함량($35.32{\mu}g\;QE/mL\;dw$)으로 오차율은 각각 0.39%, 0.74%이었다. 이는 두 개의 반응치를 동시에 분석하는 다중분석 종합분석임에도 높은 정확도를 나타낸 것으로 본 연구에서의 최적화과정 신뢰도가 우수한 것으로 사료된다.

Optimization of Extraction Conditions for the Antibacterial Activity by Clove against Streptococcus mutans Using Evolutionary Operation-Factorial Design Technique

  • This study was conducted in order to elucidate the optimum conditions for the extraction of clove that can be used to elicit antibacterial activity against Streptococcus mutans using the evolutionary operation (EVOP)-factorial design technique. Higher antibacterial activity was achieved in a higher extraction temperature of $80^{\circ}C$ ($r=0.7983^{**}$) and in a longer extraction time of 26 hr ($r=0.6867^*$). Antibacterial activity was not effected by differentiation of ethanol concentration in the extraction solvent (r=-0.0683). The maximum antibacterial activity of clove against S. mutans as determined by the EVOP-factorial design technique was obtained at an extraction temperature of $80^{\circ}C$, an extraction time of 26 hr and a 50% ethanol concentration. Furthermore, the population of S. mutans decreased from an initial concentration of 6.850 to 4.195 log CFU/mL in the third set that is more than 2.6 log cycles by EVOP-factorial design technique.

Optimum Conditions for the Extraction of Effective Substances from the Stem of Opuntia fiscus-indica

  • The conditions for the extraction of effective substances from the stem of Opuntia ficus-indica were optimized by a response surface methodology. The total extract yield was maximized under the temperature of $97.77^{\circ}C$, at a time duration of 145.82 min and a water to sample ratio 16.59 mL/g. Moreover, the optimum conditions for the extraction of effective substances were as follows: $84.95^{\circ}C$, 156.50 min and a water/sample ratio of 7.46 mL/g for the phenolics content; and $97.11^{\circ}C$, 139.03 min and a water/sample ratio of 10.91 mL/g for the pectin content. The range of optimum extraction conditions in consideration of the physicochemical properties of the extracts were shown to be as $95-100^{\circ}C$ as the extraction temperature, 120-180 min as extraction time and a water to sample ratio of 5-18 mL/g.

  • II부에서는 I부에서 3가지 기계제연방식 중에서 가장제연성능이 우수한 것으로 나타난 배기방식의 풍량 0.6∼2.4 ㎥/s와 두 화재크기 200 ㎾와 2 ㎿에 대한 공간내 온도와 연기입자의 분포를 조사하였다. I 부와 동일한 수치법을 동일한 계산영역에 적용한 결과, 배기풍량은 제연성능에 큰 영향을 미치는 것으로 나타났고, 두 화재크기에서 풍량의 증가가 제연성능을 개선하는 비슷한 경향을 보였다. 주어진 조건에서 대피로의 온도를 $60^{\circ}C$ 이하로 유지하기 위해서는 2.4 ㎥/s 이상의 배기풍량이 필요함을 알 수 있었다.

프로폴리스 농도가 항균활성에 미치는 영향 (The effect of propolis concentration on the antibacterial activity)

  • The objetives of this study are to set up optimum extraction temperature, time and organic solvent for propolis extraction, to investigate chemical properties, and to develop health foods from propolis preparation. In this study, ethanol and ultrasonic extracts method performed to optimum extraction temperature was at 60, $20^{\circ}C$, optimum extraction time was at 12, 4 hours and optimum extraction amount of solvent was at 20, 15 times of propolis weight. When various ethanol solutions were used, whereas flavonoid content was highest in 70, 80% aqueous ethanol, respectively. So the ultrasonic extracts method used gave better results than the ethanol extracts method in this work. Extraction of propolis with etanol and ultrasonic extracts method was performed by using the water and various concentrations of aqueous ethanol as solvent. Sensitivity of propolis samples to Staphylococcus aureus was investigated and the results were shown. Samples of water extract did not inhibit microbial growth, where as 50% aqueous ethanol extract the largest inhibitory zone for Staphylococcus aureus, then decreased inhibition with increasing ethanol concentrations.

유출조건에 따른 인삼중의 Ginseng Extract와 Saponin 합량변화에 관한 연구 (Studies on the Extracting Methods of Ginseng Extract and Saponins in Panax Ginseng)

  • In order to investigate the optimal conditions which affects to extraction of ginseng extract and saponin in ginseng extract, experiment was carried out varing with ethanol percentage, extraction time, temperature, sol$.$vent and Plant Parts. The results art as follows: 1. The amounts of ginseng saponin was increased according to increanation of ethanol Percentage while the amounts of ginseng extract was decreased. 2. The amounts of ginseng extract was increased as the prolongation of extraction time, on the ether hand, ginseng saponin contents increased lentil 40hr. and decreased after that. 3. By the raise of extract temperature, both of the amounts of ginseng saponin and ginseng extract was increased two times and four times. respectively. 4. The total amounts ginseng extract was obtained 22.86u when the water used as the extraction solvent, 11.28% on ethanol and 11.04U on methanol, in the order. and the saponin contents gained when the extraction solvents of water, methanol and ethanol 7.47%, 12.36% and 12.77%, respectively. 5. It showed 9.23% of ginseng extract in epidermis and 8.4% of ginseng saponin in tail Part of raw ginseng and in the case of dried ginseng, ginseng extract and saponin showed the most amounts in epidermis of 18.28% and 19.35%, respectively. 6. The ratio of panaxadiol and panaxatriol contents of ginseng saponin was almost same when it was extracted varing with ethanol percentage and extraction time (duration), and the more alcohol percentage and the longer extraction time increased, the more fractional content of ginseng saponin was extracted.

Analysis of Anti-adipogenic Constituents of Cordyceps militaris Using High Performance Liquid Chromatography-Diode Array Detection in Different Samples: Comparison with Anti-adipogenic Activity

  • We previously isolated cordycepin, guanosine and tryptophan from Cordyceps militaris as antiadipogenic constituents. For the quality control of C. militaris for anti-adipogenic activity, simultaneous analytical method using high-performance liquid chromatography (HPLC)-diode array detection (DAD) was developed and validated. Quantitation of these compounds in various Cordyceps samples from different sources and various extraction methods were conducted using developed method. Our study shows that natural Cordyceps and host insect possess higher content than cultured ones and fruiting bodies, respectively. The content of cordycepin showed great difference in different C. militaris samples whereas trytophan content was similar in tested samples. Addition of water to extraction solvent greatly increased the yield of guanosine and tryptophan. High temperature and longer extraction time increased yield of guanosine, whereas the content of trytophan was decreased in high temperature during extraction with water. Extraction using ultrasonic apparatus slightly increased extraction efficiency. Cordycepin, however, has little variation in different extraction method tested. Strong anti-adipogenic activity was observed in the samples that contain all the three constituents. Taken together, quantitation of these compounds using developed analytical method might provide basic requirement for the anti-adipogenic activity of C. militaris.
