An optimized microwave-assisted extraction method for increasing yields of rare ginsenosides from Panax quinquefolius L.

  • Background: Rare ginsenosides in Panax quinquefolius L. have strong bioactivities. The fact that it is hard to obtain large amounts of rare ginsenosides seriously restricts further research on these compounds. An easy, fast, and efficient method to obtain different kinds of rare ginsenosides simultaneously and to quantify each one precisely is urgently needed. Methods: Microwave-assisted extraction (MAE) was used to extract nine kinds of rare ginsenosides from P. quinquefolius L. In this article, rare ginsenosides [20(S)-Rh1, 20(R)-Rh1, Rg6, F4, Rk3, 20(S)-Rg3, 20(R)-Rg3, Rk1, and Rg5] were identified by high performance liquid chromatography (HPLC)-electrospray ionization-mass spectrometry. The quantity information of rare ginsenosides was analyzed by HPLC-UV at 203 nm. Results: The optimal conditions for MAE were using water as solvent with the material ratio of 1:40 (w/v) at a temperature of $145^{\circ}C$, and extracting for 15 min under microwave power of 1,600 W. Seven kinds of rare ginsenosides [20(S)-Rh1, 20(R)-Rh1, Rg6, F4, Rk3, Rk1, and Rg5] had high extraction yields, but those of 20(S)-Rg3 and 20(R)-Rg3 were lower. Compared with the conventional method, the extraction yields of the nine rare ginsenosides were significantly increased. Conclusion: The results indicate that rare ginsenosides can be extracted effectively by MAE from P. quinquefolius L. in a short time. Microwave radiation plays an important role in MAE. The probable generation process of rare ginsenosides is also discussed in the article. It will be meaningful for further investigation or application of rare ginsenosides.

Analyzing of the Essential Oil Chemical Constituents in Artemisia lavandulaefolia and its Pharmacological Property on Antibacterial Activity

  • Objective: The aim of this work is to investigate the antibacterial activity of the essential oil obtained from Artemisia lavandulaefolia (A. lavandulaefolia), as the development of microbial resistance to antibiotics make it essential to constantly look for new and active compounds effective against pathogenic bacteria. Method: The aerial parts of A. lavandulaefolia (1 kg) were subjected to steam distillation for 3 h, using a modified Clevenger type apparatus in order to obtain essential oil. Diethyl ether was the extracting solvent kept at 25?. The essential oil were analyzed by gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS). The essential oil and the composition were tested for antimicrobial activities against 15 different genera of oral bacteria. Ninety-nine compounds accounting for 94.74$\%$</TEX> of the oil were identified. The main compounds in the oil were 1,8-cineole (5.63$\%$), yomogi alcohol (4.49$\%$), camphor (4.92$\%$), a-caryophyllene (16.10$\%$), trans-a-famesene (5.09$\%$), a-terpineol (3.91$\%$), borneol (5.27$\%$), cis-chrysanthenol (6.98$\%$), and a-humulene oxide (3.33$\%$). The essential oil and its compounds were tested for antimicrobial activity against 10 different genera of oral bacteria. Conclusion: The essential oil of A. lavandulaefolia exhibited considerable inhibitory effects against all obligate anaerobic bacteria (MICs, 0.025 - 0.05 ㎎/ml) tested, while their major compounds demonstrated various degrees of growth inhibition

미세캡슐내의 캡사이시노이드의 새로운 분석법 (A New Method for Analysis of Capsaicinoids Content in Microcapsule.)

  • 미세캡슐화된 고추올레오레진 capsicum을 사용하여 core material인 올레오레진을 파괴하지 않고, wall material를 제거함으로써 올레오레진 중의 capsaicinoids 함량을 측정하는 방법을 확립하였다. 미세캡슐을 용해할 수 있는 용매로서 dimethyl sulfoxide(DMSO)를 선발하였으며, DMSO에 혼합되는 용매로서 탄수화물을 침전시킬 수 있으면서 올레오레진에 대하여 높은 용해성을 가진 용매인 에탄올을 wall material 침전에 이용하였다. Wall material은 DMSO사용량에 대한 미세캡슐의 사용량의 비율이 $7.5{\sim}10%(w/v)$범위에서 최적이었으며 DMSO 사용량의 8배 이상의 에탄올을 첨가하였을 때 완벽한 추출이 되었다. 또한 동량의 에탄올을 사용하여 추출하였을 때 1회 추출 보다는 2회 반복추출이 완벽하게 capsaicinoids를 용해 할 수 있었다. 가열처리조건은 $55^{\circ}C$, 30분에서 DMSO가 wall materials를 용해시키기에 용이하였고 에탄올 첨가후 capsaicinoids를 추출하기 위한 vortexing time은 40㎐에서 3분 이상의 혼합에서 일정한 값을 보여주었고, 원심분리조건은 21000rpm의 회전속도에서 15분이상이 capsaicinoids함량에 최고치를 나타내었다. 확립된 방법은 고추올레오레진 미세캡슐의 제조 및 저장등의 capsaicinoids의 함량변화측정에 응용이 기대된다.

자초 뿌리를 이용한 직물의 염색에 관한 연구 (The Fabrics Dyeing using Gromwell Roots)

  • For the efficiency in extracting gromwell colorant, methanol, ethanol and aceton were used as solvents. Also, to compare the results when dyed in various conditions and on various fabrics, the ratio of water-dye mixture, pH, temperature, and varieties of fabrics -wool, silk, cotton, nylon, ramie- were selected. L, a, b, ΔE, munsell and K/S value of each sample was measured and compared for the practical use. Water-dye mixture of 5:5 ratio showed the optimum dyeability. Not to mention wool and silk, but also cotton showed a satisfying dying result at the acidity of pH4. As the temperature increased, the dyeability of all three fabrics improved greatly. Especially, wool showed the highest improvement in terms of dyeability as the temperature increased. K/S values for wool and nylon showed superior results to the others in the gromwell colorant. The resulting color turned out somewhat different depending on the solvents used and the sort of fabric that was tested on. When the gromwell colorant extracted with methanol or ethanol was applied, the color of fabrics came out as purple or purpleblue. When the colorant extracted with acetone was used, the color of fabrics came out to be redpurple. The colorfastness to light showed low glades regardless to whichever solvents were used. The grades of colorfastness to laundering were recorded low numerical values, and its record became even lower when the gromwell colorant extracted with aceton was applied. The staining grade of the colorfastness to laundering showed a good grade in the range of 4 to 5. All the dyed fabrics showed a excellent drycleaning fastness.

Analysis and Exposure Assessment of Perchlorate in Korean Dairy Products with LC-MS/MS

  • Objectives: Perchlorate is an emerging contaminant that is found everywhere, including various foods. Perchlorate is known to disturb the production of thyroid hormones and leads to mental disorders in fetuses and infants, as well as metabolic problems in adults. In this study, we attempted to establish an LC-MS/MS method for measuring perchlorate in dairy products and used this developed method to investigate perchlorate levels in Korean milk and yogurt samples. Methods: The developed method of perchlorate analysis requires a shaker and 1% acetic acid/acetonitrile as the extracting solvent. Briefly, the samples were extracted and then centrifuged (4000 rpm, 1hour), and the supernatant was then passed through a $Envi^{TM}$ Carb SPE cartridge that had been prewashed sequentially with 6 mL of acetonitrile and 6 mL of 1% acetic acid in water. The final volume of the sample extract was adjusted to 40 mL with reagent water and the final sample was filtered through a 0.20-${\mu}m$ pore size PTFE (Polytetrafluoroethylene) syringe filter prior to LC-MS/MS. Results: The average levels of perchlorate in milk and yogurt samples were $5.63{\pm}3.49\;{\mu}g/L$ and $3.65{\pm}2.42\;{\mu}g/L$, respectively. The perchlorate levels observed in milk samples in this study were similar to those reported from China, Japan, and the United States. Conclusions: The exposure of Koreans to perchlorate through the consumption of dairy products was calculated based on the results of this study. For all age groups, the calculated exposure to perchlorate was below the reference of dose (0.7 ${\mu}g/kg$-day) proposed by the National Academy of Science, USA, but the perchlorate exposure of children was higher than that of adults. Therefore, further investigation of perchlorate in other food samples is needed to enable a more exact assessment of exposure of children to perchlorate.

톳 추출액을 이용한 친환경 항균성 모발 염색 (Ecofriendly Antimicrobial Hair Coloration Using Sargassum fusiforme Extract)

  • To overcome the harmful effects caused by conventional oxidative hair dyes, natural colorants becomes more popular in the hair dyeing. By extracting Sagassum fusiforme powders with aqueous alkaline solution as a solvent at 130℃ for 60 minutes, a fucoxanthin concentration of up to 216㎍/ml can be obtained. UV/Vis analysis was used to prove the presence of fucoxanthin in the extract powder. A K/S value of 23.8 can be obtained when wool fabrics were dyed with the extract at 120℃ for 60 minutes under pH 2. The color fastness properties of the dyed wool fabrics were very good as indicated by rating 4 for laundering(color change), rating 3 or higher for rubbing, and rating 5 for light irradiation. The dyed wool fabric was found to have antimicrobial activity against Staphylococcus aureus. The antioxidant and antimicrobial activity of the Sagassum fusiforme extract can be an effective functional hair colorant. Hair dyeing with the extract formulation at 45℃ for 40 minutes under pH 5 accomplished a K/S value of 8.9. The color fastness of the dyed hair showed rating 3 against light irradiation, which increased to rating 5 with after-mordanting of tannin acid.

갈화(Puerariae flos)추출물이 Rat 혈중 Ethanol 농도에 미치는 영향 (Effect of Arrowroot Flower (Puerariae flos) Extract on Lowering of Ethanol Concentration in Rat Blood)

  • 갈화(Puerariae flos)추출물이 rat의 혈액, 뇌 및 간의 ethanol농도를 유의차있게 감소시켰다. 혈중 ethanol의 감소 효과는 ethanol을 투여한지 1시간 경과한 때에 대조군과 가장 많은 차이를 보였으며 열수 추출물이 80% ethanol 추출물보다 혈중 ethanol 농도를 더 크게 감소시킴이 관찰되었다. Ethanol 투여 10분 전에 갈화추출물 투여가 1시간 전 혹은 10분 후에 투여한 군에 비해서 ethanol 농도 저하에 더 효과적이었으며, 간과 뇌 조직에서도 ethanol의 농도가 저하되는 현상이 관찰되었다. 그러나 이 때 acetaldehyde는 검출되지 않았다. 혈중 ethanol 농도를 가장 효율적으로 감소시키는 갈화 추출물의 적정량은 55.7 mg/kg body weight이었다. 이번 실험을 위해 새로이 시도된 dichloromethane에 의한 시료 추출과 GC를 이용한 분석법은 간단, 신속 하면서도 좋은 분석결과를 보였다.

Optimization of Extraction Conditions for the 6-Shogaol-rich Extract from Ginger (Zingiber officinale Roscoe)

  • 6-Shogaol, a dehydrated form of 6-gingerol, is a minor component in ginger (Zingiber officinale Roscoe) and has recently been reported to have more potent bioactivity than 6-gingerol. Based on the thermal instability of gingerols (their dehydration to corresponding shogaols at high temperature), we aimed to develop an optimal process to maximize the 6-shogaol content during ginger extraction by modulating temperature and pH. Fresh gingers were dried under various conditions: freeze-, room temperature (RT)- or convection oven-drying at 60 or $80^{\circ}C$, and extracted by 95% ethanol at RT, 60 or $80^{\circ}C$. The content of 6-shogaol was augmented by increasing both drying and extraction temperatures. The highest production of 6-shogaol was achieved at $80^{\circ}C$ extraction after drying at the same temperature and the content of 6-shogaol was about 7-fold compared to the lowest producing process by freezing and extraction at RT. Adjustment of pH (pH 1, 4, 7 and 10) for the 6-shogaol-richest extract (dried and extracted both at $80^{\circ}C$) also affected the chemical composition of ginger and the yield of 6-shogaol was maximized at the most acidic condition of pH 1. Taken together, the current study shows for the first time that a maximized production of 6-shogaol can be achieved during practical drying and extraction process of ginger by increasing both drying and extracting temperatures. Adjustment of pH to extraction solvent with strong acid also helps increase the production of 6-shogaol. Our data could be usefully employed in the fields of food processing as well as nutraceutical industry.

Antioxidant and Antimicrobial Activity of Rosa multiflora Thunberg Fruits Extracts

  • In this study, we selected some material to have potential bioactivity from natural plants, confirmed as basic data for industrializing and tried to develope the food materials using them. DPPH, ABTS, antioxidant protection factor, TBARs and antimicrobial activity of extracts from Rosa multiflora Thunberg fruits were determined. The total phenolics extracted from Rosa multiflora were 12.08, 11.82, 11.1 and 12.6 mg/g when using water, 70% ethanol, 70% methanol and 70% acetone as the solvent, respectively. The optimum conditions for extracting the phenolic compounds were 70% ethanol over for 12 hrs(11.82 mg/g). The electron donating ability and inhibition rate on ABTS of the 70% ethanol extracts were 97% and 92.2%, respectively while the antioxidant protection factor(PF) of the water extracts and 70% ethanol extracts were 1.79 and 1.34 PF, respectively. The TBAR (thiobarbituric acid reactive substance) value were $1.3{\mu}M$ for the control and $0.15{\mu}M$ for the 70% ethanol extracts. The inhibitory activity against ${\alpha}$-amylase was 26% for the 70% ethanol extracts. The 70% ethanol extracts from Rosa multiflora Thunberg fruits exhibited antimicrobial activity against H. pylori, S. epidermidis, S. aureusand and E. coli with clear zone diameters of 14, 25, 14 and 13 mm, respectively when using $200{\mu}g/mL$ of the phenolic compounds. An HPLC analysis identified 6 major phenolic metabolites in the Rosa multiflora Thunberg fruits extracts: rosmarinic acid, caffeic acid, chlorogenic acid, courmaric acid, protocatechuic acid and quercetin. In particular, the content of rosmarinic acid was the highest in the 70% ethanol extracts. Therefore these results indicate that 70% ethanol extracts from Rosa multiflora Thunberg fruits can be useful as a natural antioxidant and in functional foods.

Quantification and comparison of functional phytochemicals in steamed and freeze-dried mature silkworm powders and freeze-dried mulberry leaves

  • Various health promoting effects of steamed and freeze-dried mature silkworm powder (SMSP) have been reported. However, it is not still clear which substances in SMSP are responsible for those health promoting effects yet. In this study, we examined and compared the quantities of phytochemicals in SMSP and freeze-dried mulberry leave powder (FMLP). To investigate the optimal solvent for extracting phytochemicals from SMSP and FMLP, we used four different solvents. Among them, 80% ethanol extracts from SMSP and FMLP contained the highest amount of five flavonoids examined. In addition, FMLP had high contents of flavonoids compared with those of SMSP. The amounts of rutin, isoquercetin, astragalin, quercetin, and kaempferol in FMLP were $5.078{\pm}0.187mg/g$, $5.162{\pm}0.083mg/g$, $2.989{\pm}0.061mg/g$, $3.317{\pm}0.236mg/g$, and $2.243{\pm}0.237mg/g$, respectively, while the amounts of rutin, isoquercetin, astragalin, quercetin, and kaempferol in SMSP were $0.171{\pm}0.024mg/g$, $0.252{\pm}0.032mg/g$, $0.374{\pm}0.031mg/g$, $0.645{\pm}0.063mg/g$, and $0.0512{\pm}0.047mg/g$, respectively. Taken together, SMSP could be a source for providing various and readily absorbable flavonoids.
