Gene Expression study of human chromosomal aneuploid

  • Chromosomal copy number changes (aneuploidies) are common in human populations. The extra chromosome can affect gene expression by whole-genome level. By gene expression microarray analysis, we want to find aberrant gene expression due to aneuploidies in Klinefelter (+X) and Down syndrome (+21). We have analyzed the inactivation status of X-linked genes in Klinefelter Syndrome (KS) by using X-linked cDNA microarray and cSNP analysis. We analyzed the expression of 190 X-linked genes by cDNA microarray from the lymphocytes of five KS patients and five females (XX) with normal males (XY) controls. cDNA microarray experiments and cSNP analysis showed the differentially expressed genes were similar between KS and XX cases. To analyze the differential gene expressions in Down Syndrome (DS), Amniotic Fluid (AF)cells were collected from 12 pregnancies at $16{\sim}18$ weeks of gestation in DS (n=6) and normal (n=6) subjects. We also analysis AF cells for a DNA microarray system and compared the chip data with two dimensional protein gel analysis of amniotic fluid. Our data may provide the basis for a more systematic identification of biological markers of fetal DS, thus leading to an improved understanding of pathogenesis for fetal DS.

Sequence Variations in the Non-Coding Sequence of CTX Phages in Vibrio cholerae

  • This study focused on the variations in the non-coding sequences between ctxB and rstR of various CTX phages. The non-coding sequences of CTX-1 and CTX-cla are phage type-specific. The length of the non-coding region of CTX-1 and CTX-cla is 601 and 730 nucleotides, respectively. The non-coding sequence of CTX phage could be divided into three regions. There is a phage type-specific Variable region between two homologous Common regions (Common regions 1 and 2). The non-coding sequence of RS1 element is similar to CTX-1 except that Common region 1 is replaced by a short RS1-specific sequence. The non-coding sequences of CTX-2 and CTX-cla are homologous, indicating the non-coding sequence of CTX-2 is derived from CTX-cla. The non-coding region of CTX-O139 is similar to CTX-cla and CTX-2; however, it contains an extra phage type-specific sequence between Common region 2 and rstR. The variations in the non-coding sequences of CTX phages might be associated with the difference in the replication efficiency and the directionality in the integration into the V. cholerae chromosome.

다운증후군의 Dyrk1A 의존적 뇌기능저하의 치료: 인간 Dyrk1A 특이적 shRNA 발굴 (Treatment of Dyrk1A-dependent Mental Retardation of Down Syndrome: Isolation of Human Dyrk1A-specific shRNA)

  • 다운증후군은 추가적으로 존재하는 인간염색체 21번에 위치한 유전자의 과발현으로 발병한다. 다운증후군 환자에서 보이는 여러 증상들 중 학습과 기억능력 저하와 같은 뇌기능 저하는 다운증후군 환자가 독립적인 생활을 영위하는데 가장 큰 걸림돌이 된다. 인간염색체 21번에 위치하는 Dyrk1A는 신경발달에 중요한 역할을 하는 단백질로 Dyrk1A를 과발현 하는 형질전환 생쥐에서 심각한 해마 의존적 학습과 기억 장애가 보고되었다. 본 연구에서는 인간 Dyrk1A를 과발현 하는 형질전환 생쥐와 RNA interference (RNAi) 방법을 이용하여 endogenous mouse Dyrk1A의 발현은 정상적으로 유지하면서 exogenous human Dyrk1A 발현은 특이적으로 저해함으로써 인간 Dyrk1A 과발현에 의한 학습과 기억 능력저하를 회복시킬 수 있는지 동물모델에서 검증하기 위한 첫 단계로 인간 Dyrk1A 특이적 lentiviral short hairpin RNA (shRNA)를 발굴하였다. 발굴된 shRNA를 이용한 형질전환 모델생쥐에서의 증상의 회복 가능성 검증은 다운증후군의 뇌기능저하 치료제 개발에 중요한 정보를 제공할 것이다.

A Commensal Thermophile, Symbiobacterium toebii: Distribution, Characterization, and Genome Analysis

  • A commensal thermophile, Symbiobacterium toebii, isolated from hay compost (toebii) in Korea commensally interacted with a thermophilic Geobacillus toebii sp. nov., which was a new species within the genus Geobacillus on the basis of the phenotypic traits and molecular systematic data. S. toebii required the crude extracts and/or culture supernatant of the Geobacillus toebii for axenic growth and could grow on the temperature between 45 and $70^{\circ}C$ (optimum: $60^{\circ}C$; 2.4 h doubling time) and pH 6.0 and 9.0 (optimum: pH 7.5). The G+C content of the genomic DNA was $65 mol\%$, and the major quinones were MK-6 and MK-7. A phylogenetic analysis of its 16S rDNA sequence indicated that Symbiobacterium toebii was closely related with solely reported Symbiobacterium thermophilum. The presence of the commensal thermophile 16S rDNA and accumulation of indole in all the enriched cultures indicate that Symbiobacterium toebii is widely distributed in the various soils. The genome of S. toebii constituted a circular chromosome of 3,280,275 base pairs and there was not an extra-chromosomal element (ECE). It contained about 4,107 predicted coding sequences. Of these protein coding genes, about $45.6\%$ was encoded well-known proteins and annotated the functional assignment of 1,874 open reading frames (ORFs), and the rest predicted to have unknown functions. The genes encoding thermostable tyrosine phenol-lyase and tryptophan indole-lyase were cloned from the genomic DNA of S. toebii and the enzymatic production of L-tyrosine and L-tryptophan was carried out with two thermostable enzymes overexpressed in recombinant E. coli.
