The Utilization of Corn Stalk, Pine Bark, Pine Leaves, Wheat and Wood Flour as an Extender for Plywood Bonding (옥촉서간(玉蜀黍幹), 송수피(松樹皮), 소맥(小麥), 리기다송엽(松葉), 잣나무엽(葉) 및 목분말(木粉末)을 이용(利用)한 합판(合板)의 접착증량(接着增量)에 관(關)한 연구(硏究))

  • The purpose of this study was to evaluate the substitutional possibility of new extender instead of wheat flour, which is extending for plywood adhesives in Korea. As the extending materials corn stalk, pine bark, Pitch and Korean pine leaves, wheat, or wood flour were selected and prepared for the extending powders, dried at $103{\pm}2^{\circ}C$ during 24 hours in the drying oven, followed by being pulverized into 60-100 mesh powder. The extenders were mixed with urea formaldehyde resin in the ratio of 5, 10, 15 or 20%. After plywoods were manufactured by the above extended ratios, dry and wet shear strength and wood failures were analyzed and discussed. The results at the study may be summarized as follows ; 1. In urea formaldehyde resin dry shear strength in plywood extended by wheat flour showed the highest value. 2. Among the extenders in 10 and 20% extension of urea formaldehyde resin wet shear strength of wood flour was higher than that of wheat powder. They had no significant difference statistically. 3. Among the extenders of 5% extension of water soluble phenol formaldehyde resin dry shear strength of plywood extended by Korean pine leaf powder showed the highest value, while wheat powder showed the highest value among 10, 15 and 20% extentions. 4. In water soluble phenol formaldehyde resin the best results of wet shear strength showed in wheat powder. 5. Among the extenders in 15 and 20% extension of water soluble phenol formaldehyde resin, dry and wet shear strength in plywood of corn stalk powder were the highest value next to wheat powder.

  • The objectives of this study were to investigate 1) the effects of Selenium(Se), Vitamin E (Vit. E) or recombinant Bovine Somatotropin(rBST) administration on fresh and frozen/thawed semen characteristics and 2) the effect of taurine on frozen/thawed semen characteristics in Hanwoo sires Hanwoo sires were randomly assigned to five groups (1. control, 2. rBST, 0.09mg/kg body weight (BW), 3. Vito E 1,500IU/kg BW, 4. Se 0.l mg/kg BW, 5. Vit. E 1,500IU plus Se 0.1 mg/kg BW). The administration of Se, Vit. E and rBST for each experimental group were given 6 times at 15 days interval by intramuscular injection. The administration of Se, Vit. E or rBST in Hanwoo sires didn't affect semen volume and pH values, but sperm viability was significantly increased comparing to the control group. Also, frozen/thawed semen analysis showed that the sperm viability increased, but any other effects were not found in total sperm :lumber, motility and abnormality among treatments. The addition of taurine in semen freezing extender had a beneficial effects on frozen/thawecl semen characteristics in all groups. The administrations of rBST, Vit. E and Se did not affect the sperm capacitation and acrosome reaction, either the ratio of F pattern(uncapacitated and acrosome intact sperm) or AR pattern(capacitated and acrosome-reacted sperm), but the ratio of B patten(capacitated and acrosome intact sperm) of treatment groups was significantly higher than that of control group, These results indicated that the viability, motility and quality of semen in Hanwoo sires were slightly increased by the injection of rBST, Vit. E and Se, and the addition of taurine in semen freezing extender were also increased the semen characteristics after thawing.

Effect of Glycerol and Ethylene Glycol on Post-Thawed Sperm Function in Jeju Horse (제주마의 동결정액 제조에 있어 Glycerol과 EthyleneGlycol이 동결 융해 후 정자의 기능에 미치는 영향)

  • Cryopreservation induces sublethal damage to the spermatozoa, which leads to their reduced fertile life. This study was designed to determine effect of glycerol and ethylene glycol as cryoprotectant in extender on improve the freezability of Jeju horse semen. The semen was cryopreserved with glucose-EDTA extender containing each 5% glycerol, 5% ethylene glycol, 8% glycerol or 8% ethylene glycol, respectively. Post-thawed sperm were evaluated motility, viability, Membrane integrity and acrosome integrity. Post-thawed sperm motility were not significantly differences among treatments. However, sperm viability were significantly higher (p<0.05) in 8% glycerol ($39.85%{\pm}11.41$) than in 5% glycerol treatment ($18.08%{\pm}1.61$). In membrane integrity, swelling sperm ratio was significantly higher (p<0.05) in 8% glycerol ($34.12%{\pm}11.02$) than other treatments. In the percentage of capacitated sperm assessed by CTC staining, F pattern was significantly higher in 8% ethylene glycol than 5% glycerol and 5% ethylene glycol (p<0.05). B pattern ratio was significantly increased in 5% ethylene glycol compared with 8% glcerol and 8% ethylene glycol (p<0.05). Moreover, 8% ethylene glycol treatment was significantly decreased AR pattern ratio compared with other treatments (p<0.05). It is concluded that treatment of 8% glycerol was improved the sperm viability and 8% ethylene glycol was improved the sperm ascrosome integrity after thawing. However, they were not significantly difference between 8% glycerol and 8% ethylene glycol on post-thawed sperm viability. Therefore, 8% ethylene glycol was more effective sperm cryoprotectant than 8% glycerol in Jeju Horse.

Effects of Zardaverine in Freezing Extender on Kinetic Characteristics of Post-Thawed Boar Sperm (동결보존액에 Zardaverine의 첨가가 동결-융해 후 돼지 정자의 운동학적 특성에 미치는 영향)

  • This study investigated the effect of Zardaverine supplementation in freezing extender, on kinetic characteristics of post-thawed boar sperm. Cryopreservation of boar sperm is an important technique of assisted reproductive technology and genetic resource banking. Although this technique is particularly useful, freeze-thaw cycles associated with sperm cryopreservation significantly reduce sperm quality. Semen from mature Duroc boars were collected and cryopreserved in freezing extenders (LEY) treated with varying concentrations of Zardaverine (0, 20, 50, 75, 100 𝜇M). The time-dependent kinetic characteristics of post-thawed spermatozoa were determined after thawing by applying computer-assisted sperm analysis (CASA). We observed that the motility immediately after thawing was significantly higher in 20 𝜇M stocks than in control (0 𝜇M) and the other treatments (p<0.05). Curvilinear velocity (VCL) in 0 𝜇M and 20 𝜇M stocks were significantly higher than the other treatment groups, except 75 𝜇M (p<0.05). Higher average path velocity (VAP) was obtained at 20 𝜇M as compared to 100 𝜇M, whereas amplitude of head lateral displacement (ALH) was significantly higher at 20 𝜇M than 50 𝜇M and 100 𝜇M (p<0.05). No differences were obtained for Straight-line velocity (VSL) and Linearity (LIN). In conclusion, our results indicate that Zardaverine improves the motility, VCL, VAP, and ALH of post-thawed boar sperm.

Effect of Sugar Kind Added in Tris-buffer on Acrosome Damage of Post-thaw Spermatozoa in Canine (Tris-buffer에 첨가되는 당의 종류가 동결.융해정자의 첨체 손상에 미치는 영향)

  • The aim of this study was conducted to evaluate the effect of sugar kinds and combination of various sugars on acrosome damage of post-thaw spermatozoa in canine. The extender used in this study was Tris-citric acid extender (Tris-buffer) supplemented with 20% Egg-yolk, 8% glycerol, 1% Equex STM paste, and 70 mM sugars such as monosaccharide (fructose and xylose) and disaccharide(trehalose). To evaluate of sugar combination, the sugars supplemented in Tris-buffer were combined such as control (fructose, xylose, trehalose), two combinations (Fru+Tre, Fru+Xyl, Tre+Xyl) and three combinations (Fru+Tre+Xyl). The acrosome damage rate of post-thaw spermatozoa in Eosin B & Fast Green stain in Fruc+Tre was higher than those in fructose, trehalose, xylose, Fruc+Xyl, Tre+Xyl, Fruc+Tre+Xyl (83.0$\pm$5.6 vs. 82.3$\pm$3.1%, 81.7$\pm$2.1%, 72.0$\pm$2.0%; 80.3$\pm$4.5%, 76.7$\pm$3.8%, 81.0$\pm$5.6). The motility after CASA analysis in Fru+Tre was higher than those in Fru+Tre+Xyl, Tre+Xyl, Fru+Xyl, Xylose, Trehalose, Fructose(79$\pm$6 vs 75$\pm$3, 74$\pm$8, 71$\pm$11, 70$\pm$4, 66$\pm$15, 63$\pm$ 12%). However, the progressive motility after CASA analysis in Fru+Tre group was higher than those in Fru+Tre+Xyl, Tre+Xyl, Fru+Xyl, Xylose, Trehalose, Fructose (67$\pm$7, 64$\pm$3, 62$\pm$6, 61$\pm$8, 60$\pm$2, 57$\pm$13, 53$\pm$10%). The results indicated that the acrosome damage & progressive motility of post-thaw spermatozoa in 70 mM Fruc+Tre (two combination) following Eosin B & Fast Green stain and CASA analysis.

Influence of Glycerol Concentration, Freezing Rate and Thawing Rate on Survival of Canine Spermatozoa Frozen (개 정자의 동결보존에 있어서 Glycerol 농도, 동결 및 융해 속도가 정자의 생존율 및 운동성에 미치는 영향)

  • This study was carried out to establish most suitable freezing condition, to evaluate the different condition of freezing and thawing rates on the viability and motility of frozen canine spermatozoa. The ejaculated semen was added to obtained 200∼400 ${\times}$ 10$^{6}$ /$m\ell$ with extender I and was cooled to 4$^{\circ}C$ over 30, 60 and 120 minutes. And then, semen was diluted with extender II containing 4, 6 and 8%(v/v) glycerol for 60 min, respectively and packaged in 0.5$m\ell$ straw, equilibrated far 30, 60 and 120 min at 4$^{\circ}C$ and cryopreserved in liquid nitrogen vapor at different distance(3, 5, 7 and 9 cm, respectively), plunged into nitrogen tank. Samples were thawed by placing straws into 27, 37, 47, 57$^{\circ}C$ water bath for 120, 20 and 12 sec, respectively. The results were as follows; 1. The survival and motility rate immediately post-thawing was significantly higher in samples frozen in 4% glycerol than 6 or 8% glycerol(P< 0.05). 2. According to equilibration time at 4$^{\circ}C$, the survival and motility rate immediately post-thawing was significantly higher in samples frozen after 60 min equilibration than 30 or 120 min equilibration(P<0.05). 3. Freezing in distance of 5 cm from liquid nitrogen yield better survival and motility rate than the others(3, 7 or 9 cm)(P<0.05). 4. The effect of thawing rate on sperm survival were higher when the thawing was done at 37$^{\circ}C$ for 120 sec(P<0.05).

The Effect of Seminal Plasma on Chilling and Freezing of Canine Spermatozoa (개 정액의 정장이 개정자의 냉각과 동결에 미치는 영향)

  • Seminal plasma(SP) is usually removed from semen that is to be cryopreserved. However, some reports indicate that SP has beneficial effects on spermatozoa during chilling and freezing. The purpose of this study was to determine the effect of SP on sperm survival by adding SP to the extender before cooling and freezing canine spermatozoa. In replicate experiments, ejaculates obtained from four healthy dogs(1-4 years old) of various breeds were pooled, centrifuged at $300{\times}g$ for 10 min at $25^{\circ}C$, and the supernatant of seminal plasma was decanted. Spermatozoa were suspended in egg yolk-Tris(EYT) buffer. The study comprised two experiments: [Exp 1] Sperm were suspended in EYT extender containing either 0, 20, 40, 80 or 100% SP and were slowly cooled to $4^{\circ}C$ for 2h or held at $25^{\circ}C$ as controls. Sperm concentration was adjusted to $2{\times}10^8/ml$. [Exp II] Sperm samples, each of which contained $1{\times}10^8/ml$, were assigned to nine groups to be frozen. In the first four groups, sperm in EYT containing either 20, 40, 80 or 100% SP were cooled to $4^{\circ}C$, then diluted to contain final concentrations of EYT+0.6M glycerol and then were frozen. The final concentrations of SP were 10, 20, 40 or 50%. In the other four groups, sperm in EYT alone were first cooled slowly to $4^{\circ}C$, then diluted to contain final concentrations of EYT+0.6M glycerol plus 10, 20, 40 or 50% SP and then were frozen. Spermatozoa, which chilled in EYT alone and diluted to contain final concentrations of EYT+0.6M glycerol without seminal plasma, and then frozen, was regarded as control. Spermatozoa were frozen at $25^{\circ}C/min$ of cooling rate in plastic straws that were suspended above liquid nitrogen and thawed in water at $38^{\circ}C$ for 1 min. Sperm survival was assayed by determining progressive motility and integrity of plasma and acrosome membranes. Progressive motility was determined by microscopic examination at $200{\times}$ magnification. Membrane integrity was assessed by use of a double fluorescent dye, and acrosome integrity by staining sperm with Pisum sativum agglutinin. The results of the first experiment showed that adding SP did not improve motility of spermatozoa compared to those incubated without SP regardless of temperature. The results of the second experiment showed that spermatozoa suspended in EYT+0.6M glycerol containing SP exhibited the higher progressive motility before being frozen(P<0.05). However, frozen-thawed spermatozoa that had suspended in EYT+0.6M glycerol containing SP showed the similar or lower viability(P<0.05). In summary, although seminal plasma did not affect spermatozoa that were chilled in EYT without cryoprotectant(CPA), addition of seminal plasma to EYT containing CPA did significantly improved progressive motility of canine spermatozoa that were chilled.

In Vitro Fertilization of Pig Oocytes Matured In­Vitro by liquid Boar Spermatozoa (체외성숙 돼지 난포란의 액상정액을 이용한 체외수정)

  • The present study was carried out to investigate the effects of the maturation media such as a modified TCM-199 (mTCM-199) medium, modified Waymouth MB 752/1 (mWaymouth MB 752/1) medium or NCSU-23 medium on penetrability of pig oocytes by liquid boar sperm. Oocytes (30~40) were transferred into each well of a Nunc 4-well multidish containing 0.5 $m\ell$ maturation medium. When immature pig oocytes were cultured in mTCM-199, mWaymouth MB 752/1 and NCSU-23 maturation media for 44 h in 5% $CO_2$, in air at 38.5$^{\circ}C$, the germinal vesicle breakdown (CVBD) rates of the oocytes were 95.6, 94.1 and 94.9%, respectively, and the maturation rates (metaphase II) of oocytes were 92.5, 90.1 and 91.1%, respectively. No differences were observed among the maturation media. The spermrich portion of ejaculates with greater than 90% motile sperm were used in the experiment. The semen was cooled 22 to 24$^{\circ}C$ over 2 h period. The semen was diluted with Beltsville Thawing Solution (BTS) extender at room temperature to give 2$\times$10$^{8}$ sperm/$m\ell$ in 100 $m\ell$ plastic bottle. Liquid boar semen of 30 $m\ell$ in 100 $m\ell$ plastic bottle was kept at 17$^{\circ}C$ for 5 days. The sperm with greater than 70% motility after day 5 of storage were used for in-vitro fertilization (IVF). After 44 h maturation of immature oocytes, cumulus cells were removed and oocytes (30~40) coincubated far 6 h in 0.5 $m\ell$ mTCM-199 and mTBM fertilization media with 2$\times$1061$m\ell$ sperm concentration. At 6 h after IVF, oocytes were transferred into 0.5 $m\ell$ mTCM-199 and NCSU-23 culture media for further culture 6 or 42 h. Sperm penetration, polyspermy and male pronuclear formation of oocytes at 12 h after IVF, and developmental ability of oocytes at 48 h after IVF were evaluated. The oocytes in combination with NCSU-23 medium for maturation and mTBM medium for IVF increased male pronuclear formation (48.0%) compared to those in combination with mTCM-199 media for maturation and IVF, and mWaymouth MB 752il medium for maturation and mTCM-199 medium far IVF. The rates of cleaved embryos (2~4 cell stage) at 48 h after IVF were 24.1% in combination with mTCM-199 media for maturation, IVF and culture, 43.6% in combination with mWaymouth MB 75211 medium fur maturation and mTCM-199 media for IVF and culture, and 71.2% in combination with NCSU-23 medium for maturation, mTBM medium for IVF and NCSU-23 medium for culture. In conclusion, we found out the oocytes matured in vitro were fertilized by liquid boar sperm stored in BTS extender at 17$^{\circ}C$ for 5 days. We recommend the simple defined NCSU-23 medium for nuclear maturation, mTBM medium and liquid boar sperm for IVF, and NCSU-23 medium for embryo culture.

Preparation and Properties of Polyurethane Dispersions with Aromatic/Aliphatic Mixed Diisocyanate (방향족/지방족 혼합 Diisocyanate를 포함하는 Polyurethane 분산체의 제조와 성질)

  • An anionic polyurethane dispersions (PUDs) were synthesized from the poly (tetramethylene glycol) (PTMG, Mw = 2000 g/mol), mixed isocyanate of dicyclohexylmethane-4,4'-diisocyanate $(H_{12}-MDI)$ and 4,4'-diphenylmethane diisocyanate (MDI), and dimethylol propionic acid (DMPA) as anionic site, following a prepolymer mixing process. Triethylamine (TEA) was used as a neutralization agent and the ethylenediamine (EDA) as the chain extender of the prepolymer. The effects of the DMPA molar ratio and aromatic diisocyanate content in the mixed isocyanate on the particle size and viscosity of PUD were studied. Also, the mechanical and thermal properties of the PUD cast films were discussed according to the molar ratio of DMPA and aromatic isocyanate content. It was found that the particle size and the viscosity of an anionic PUD decreased with increasing DMPA molar ratio but increased with increasing aromatic isocyanate (MDI) content in the mixed isocyanate at the constant DMPA content. Tensile strength of the PUD cast films increased and elongation at break decreased with increasing DMPA content at the constant mixed isocyanate molar ratios. In thermal degradation temperature of PUD cast films, the effect of DMPA contents was great but the effect of aromatic isocyanate contents at the low DMPA content was very slight respectively.

Effect of Glycerol Concentration, Freezing Rate and Thawing Rate on Semen Characteristics in PoongSan-dog (풍산개 정자의 동결보존에 있어서 Glycerol 농도, 동결 및 융해속도가 정자성상에 미치는 영향)

  • This research was carried out in order to establish the production technique for Poong-san dog’s frozen semen, by examining the semen characteristic and the volume of glycerol added to the dilution solution, thawing temperature and sperm motility and viability as well as the motility using CASA according to time variation. Average semen volume was 5.9ml, sperm concentration 116.3×106 sperm/ml, total sperm number 789.3×106 sperm, motility 88.7±1.7% and viability 87.6±7.8%. When it was cryopreservation and thawed at different glycerol concentrated extender, it showed 52.7% motility and 57.7±10.3% viability at 7% glycerol, compared to other treatments. For semen cryogeny, at conditions of 5, 7cm and a height of 10cm for pre-cryogeny and maintaining the semen at 7cm from the surface of liquid nitrogen resulted in profitable motility and viability.
