A Rapid and Simple Method for Construction and Expression of a Synthetic Human Growth Hormone Gene in Escherichia coli

  • A cDNA, encoding the human growth hormone (hGH), was synthesized based on the known 191 amino acid sequence. Its codon usage was optimized for a high level expression in Escherichia coli. Unique restriction sites were incorporated throughout the gene to facilitate mutagenesis in further studies. To minimize an initiation translation problem, a 624-bp cassette that contained a ribosome binding site and a start codon were fused to the hGH-coding sequence that was flanked between the EcoRI and HindIII sites. The whole fragment was synthesized by an overlapped extension of eight long synthetic oligonucleotides. The four-short duplexes of DNA, which were first formed by annealing and filling-in with a Klenow fragment, were assembled to form a complete hGH gene. The hGH was cloned and expressed successfully using a pET17b plasmid that contained the T7 promoter. Recombinant hGH yielded as much as 20% of the total cellular proteins. However, the majority of the protein was in the form of insoluble inclusion bodies. N-terminal amino acid sequencing also showed that the hGH produced in E. coli contained formyl-methionine. This study provides a useful model for synthesis of the gene of interest and production of recombinant proteins in E. coli.

  • Such trend offers new fashion designs as a formative art with creative spontaneity. The purpose of this study is to seek to work out fashion design methods whereby fashions can be developed into an art form embracing handicraft premium textures, geometric formativeness and traditional beauty so as to satisfy the individualist expression desires of modern people who pursue practicality, originality, and beauty of simplicity. This art form also allows new images to be expressed. The corresponding methods studied include the space and technique of patchwork that can create artistry and aesthetic functionality into differentiated levels of images, and geometric ion from Mondrian' works. Fashion designs based on patchwork technique and the characteristics of Mondrian's works reveal that the patchwork technique using diverse materials is an artistic technique with high handicraft value. This technique provides new value to traditional aesthetic materials of clothing, and that Mondrian's unique designs are very effective in developing new fashion designs because they provide artistry and unique effects to modern fashion expression.

현대 패션에 표현된 몸의 파편화 (Fragmentation of the Body in Fashion)

  • Freed from its traditional confinement to the human body, postmodernism in fashion exposes the defectiveness of body and abstracts from the body under. As the 20th century art put premium on self-expression, the body itself became a powerful medium of expression in fashion. Using 'body' to analyze the clothing form, my study develops a framework by which to classify the fragmentation of the body in fashion. In order to inquire the formative style and aesthetic values expressed in fragmentation of the body in fashion, my study examines subjects from the discourse on the body to the fashion collections of the late 20th and 21st century, The results of the study are as follows. Fragmentation of the body in fashion means the break away from the idealized and standardized body for mass productions. It tends to experiment with extreme exaggeration in form, refusing to subscribe to the traditional values that build on the balance and symmetry of the body. The formative aspects of fragmentation are achieved through body casting, displacement, and deconstruction. The absence of physicality in fashion opposes the sartorial convention and symbolism that results in the discord between signifiant and $signifi{\tilde{e}}$ of clothing. Fashion continues to explore forms and images that transcend the traditional representations of the clothed body. As a type of intimate architecture, fashion always mediates the dialogue between clothes and body, or fashion and figure. My study suggests a framework to analyze fragmentation of the body in fashion, focusing on the relationship between the clothes and body.

  • 본 논문에서는 Alamouti[1], Tarokh[14], Xia[11]에 의해 도입된 직교 시공간 부호를 포함한 임의의 선형 직교 시공간 블록 부호에 대해, 느린 레일라이 페이딩 채널(slow Rayleigh fading channel)의 쌍간 오율(pairwise error probability)의 정확한 표현이 2개의 전송된 부호어 행렬(codeword matrices) 사이의 부호어 심볼 거리(codeword symbol distance)가 아니라, 2개의 메시지 벡터(messgae vector) 사이의 메시지 심볼 거리(message symbol distance)의 식으로 유도된다. 1차원 성분의 심볼 오율을 이용하여, 선형 직교 시공간 부호의 심볼 오율의 정확한 폐형의 표현이 QPSK, 16-QAM, 64-QAM과 256-QAM에 대해 유도된다.

Cloning, Expression in Escherichia coli, and Enzymatic Properties of a Lipase from Pseudomonas sp. SW-3

  • The lipase gene (lipA) and its activator gene (lipB) of Pseudomonas sp. SW-3 were cloned and sequenced. The lipB was found to be present immediately downstream of lipA. The deduced amino acid sequences of lipA and lipB showed a high level of homology to those of other lipases belonging to the family I.1 of bacterial lipases. When lipA was expressed in Escherichia coli using T7 promoter, an active lipase was produced in cells carrying both lipA and lipB, but not in cells harboring only lipA. Recombinant lipase (rPSL) overproduced in an insoluble form was solubilized in the presence of 8 M urea, purified in a urea-denatured form and refolded by removing urea in the presence of the Ca$\^$2+/ ion. rPLS had maximum activity at pH 8.0 and 50$^{\circ}C$, was stable at pHs from 7.0 to 9.0 and below 50$^{\circ}C$, and showed the highest activity toward the p-nitrophenyl ester of palmitate (Cl6).

  • The purpose of this paper is to explore the syntactic criteria for determining a secondary predicates as a predicate modifier or a conjunction, and to formalize the semantic aspects of the [-ke] structure as a predicate in Korean. Syntactically, the [-ke] structure is considered to be a secondary predicate when the shared arguments appear in both the [-ke] structure and the main verb structure. On the other hand, if they do not appear in both structures, the [-ke] structure is considered to be a connective element. Semantically the [-ke] structure has numerous aspects such as depictives, resultatives, objectivity, and emphasis. The depictives of the secondary predicate can be formalize as $p{\wedge}q$ where p represents a propositional expression of the secondary predicate and q is a propositional expression of the main verb. Resultatives have the logical form $q{\rightarrow}{\Box}p$, because the consequence has to always be true. However, objectivity has the logical form $q{\rightarrow}{\diamondsuit}p$, because the consequence can be either true or false. Emphasis is represented as $q{\rightarrow}p{\uparrow}$ because the secondary predicate represents the polarity of the event.

Anxiety before dental surgery under local anesthesia: reducing the items on state anxiety in the State-Trait Anxiety Inventory-form X

  • Background: It is important to evaluate preoperative anxiety and prepare sedation when performing dental surgery under local anesthesia. Spielberger's State-Trait Anxiety Inventory (STAI) is useful for predicting preoperative anxiety. State anxiety is defined as a subjective feeling of nervousness. Reduction in the number of the state anxiety items (questions) will be clinically important in allowing us to predict anxiety more easily. Method: We analyzed the STAI responses from 1,252 patients who visited our institution to undergo dental surgery under local anesthesia. Multiple linear regression analysis was conducted for 9 groups comprising anxiety level determinations using the STAI; we then developed a coefficient of determination and a regression formula. We searched for a group satisfying the largest number of requirements for regression expression while setting any necessary conditions for accurately predicting anxiety before dental surgery under local anesthesia. Results: The regression expression from the group determined as normal for preoperative state anxiety was deemed the most suitable for predicting preoperative anxiety. Conclusion: It was possible to reduce the number of items in the STAI by focusing on "Preoperative anxiety before dental surgery."

Genetic association tests when a nuisance parameter is not identifiable under no association

  • Some genetic association tests include an unidentifiable nuisance parameter under the null hypothesis of no association. When the mode of inheritance (MOI) is not specified in a case-control design, the Cochran-Armitage (CA) trend test contains an unidentifiable nuisance parameter. The transmission disequilibrium test (TDT) in a family-based association study that includes the unaffected also contains an unidentifiable nuisance parameter. The hypothesis tests that include an unidentifiable nuisance parameter are typically performed by taking a supremum of the CA tests or TDT over reasonable values of the parameter. The p-values of the supremum test statistics cannot be obtained by a normal or chi-square distribution. A common method is to use a Davies's upper bound of the p-value instead of an exact asymptotic p-value. In this paper, we provide a unified sine-cosine process expression of the CA trend test that does not specify the MOI and the TDT that includes the unaffected. We also present a closed form expression of the exact asymptotic formulas to calculate the p-values of the supremum tests when the score function can be written as a linear form in an unidentifiable parameter. We illustrate how to use the derived formulas using a pharmacogenetics case-control dataset and an attention deficit hyperactivity disorder family-based example.

원통형 이상체에 의한 자력 변화율 텐서 반응식 (The Closed-form Expressions of Magnetic Gradient Tensor due to a Circular Cylinder)

  • 이 논문에서는 축 방향 대칭성을 가지는 원통형 이상체에 대한 자력 변화율 텐서의 해석해를 유도하였다. 일정한 방향으로 자화된 원통형 이상체에 대한 3성분 벡터 자력 반응식을 기존 연구에서 이미 유도하였으므로, 이를 직교좌표계에서 각 축 방향으로 미분하여 자력 변화율 텐서 반응식을 유도하였다. 원통형 이상체가 가지는 축 방향 대칭성 때문에 벡터 자력 반응식은 원통 좌표계의 변수를 포함한 식으로 표현되어 있으므로 직교 좌표계와 원통 좌표계 사이의 변수 미분 관계를 적용하여 자력 변화율 텐서를 유도하였다.

An Interference Analysis Method with Site-Specific Path Loss Model for Wireless Personal Area Network

  • In this paper, an interference analysis method with a site-specific path loss model for a wireless personal area network (WPAN) is proposed. The site-specific path loss model is based on geometrical optics and geometric probability to consider both site-specific radio propagation characteristics and a closed-form expression to obtain the mean interference from which the uniformly distributed multiple interferers are derived. Therefore, the proposed interference analysis method can achieve more computational simplicity than the Monte-Carlo (MC) simulation, which uses the ray-tracing (RT) technique. In addition, better accuracy than the conventional interference analysis model that uses stochastic method can also be achieved. To evaluate the proposed method, a signal to the interference-noise ratio with a mean interference concept for uniformly distributed interferers is calculated and compared in two simulation scenarios. As a result, the proposed method produces not only better matched results with the MC simulation using the RT technique than the conventional interference analysis model, but also simpler and faster calculation, which is due to the site-specific path loss model and closed-form expression for interference calculation.
